Heat capacity microcalorimetry of the in vitro reconstitution of calf brain microtubules.

The self-assembly of calf brain tubulin, purified by the modified Weisenberg procedure, was examined in an adiabatic differential heat capacity microcalorimeter. Tubulin solutions at concentrations between 6 and 17 mg/mL were heated from 8 to 40 degrees C at heating rates between 0.1 and 1.0 deg/min in a pH 7.0 phosphate buffer containing 1 X 10(-3) M GTP, 1.6 X 10(-2) M MgCl2, and 3.4 M glycerol. The heat capacity change, deltaCp of the microtubule growth reaction was found to be -1600 +/- 500 cal/(deg mol) per 110 000 molecular weight tubulin dimer incorporated into microtubules, in agreement with the reported van't Hoff deltaCp value of -1500 cal/(deg mol) [Lee, J.C., & Timasheff, S.N. (1977) Biochemistry 16, 1754-1765]. The assembly reaction is characterized by a complex heat uptake pattern comprising both endothermic and exothermic processes.

Prediction of treatment effectiveness in a drug-free therapeutic community.

Treatment effectiveness for drug dependance is evaluated and compared with success rates reported in the literature. Program differences and varying follow-up procedures make assessment difficult. Identical pre- and posttreatment quantitative adjustment indices are recommended in order to improve the measurement of improvement. Education and length of stay in treatment are among the most important predictors of posttreatment drug usage and work akjustment. Education and improved treatment success with lower class youth are discussed.

Effects of retinoic acid on prostaglandin biosynthesis in guinea-pig skin.

Topical application of retinoic acid on guinea-pig skin resulted within 70 hours in erythema with a concomitant elevation of endogenous prostaglandin E2 (PGE2) in the treated areas of the skin. Prolonged daily treatment resulted in the development of severe scaly dermatoses and a corresponding decrease in the level of PGE2 in the skin. Examination of retinoic acid effects on the in vitro biosynthesis of PGE2 from arachidonic acid by extracts from guinea-pig skin and sheep vesicular gland demonstrated that retinoic acid inhibits prostaglandin synthesis in a concentration and time-dependent manner. These results indicate that retinoic acid may exert a regulatory role on prostaglandin biosynthesis in the skin.

Living with altered body image: the cancer experience.

A wide range of cancer problems (e.g. tumour, treatment and treatment delivery systems) cause alterations in body image for patients. Individuals' sense of self-worth is bound up with appearance, function and control of their bodies. This value system is developed through the process of socialization and the advertising in the fashion industries. Nurses must consider the value system that patients employ and the way in which they value different aspects of their bodies. Nurses cannot make effective care contributions to the problems of altered body image during the immediate acute phase of illness. It is at a later stage, normally at discharge, that patients experience a reaction to their altered body image.

Airborne asbestos levels in buildings: maintenance worker and occupant exposures.

A substantial number of air samples have been collected during the past few years to measure airborne asbestos levels in buildings with asbestos-containing materials (ACM). These samples fall into two categories: (i) samples collected to measure the exposure of workers while they were engaged in routine maintenance and repair activities; and (ii) samples collected during normal building activity to measure prevalent levels in buildings. The measurements derived from these samples have been compiled and summarized to provide estimates of the airborne asbestos exposure of workers engaged in routine maintenance and repair work and by other building occupants. These data indicate that maintenance and repair workers in buildings with ACM, after accounting for the frequency and duration of these types of activities, have annual exposure levels ranging from a median value of 0.002 fibers per cubic centimeter (f/cc) per year to 0.02 f/cc per year at the 90th percentile. Building occupants not involved in maintenance and repair work experience average exposure levels ranging from 0.00003 f/cc to 0.0005 f/cc.
