FK330, a novel inducible nitric oxide synthase inhibitor, prevents ischemia and reperfusion injury in rat liver transplantation.

Nitric oxide (NO), produced via inducible NO synthase (iNOS), is implicated in the pathophysiology of liver ischemia/reperfusion injury (IRI). We examined the effects of a novel iNOS inhibitor, FK330 (FR260330), in well-defined rat liver IRI models. In a model of liver cold ischemia followed by ex vivo reperfusion, treatment with FK330 improved portal venous flow, increased bile production and decreased hepatocellular damage. FK330 prevented IRI in rat model of 40-h cold ischemia followed by syngeneic orthotopic liver transplantation (OLT), as evidenced by: (1) increased OLT survival (from 20% to 80%); (2) decreased hepatocellular damage (serum glutamic oxaloacetic transaminase/glutamic pyruvic transaminase levels); (3) improved histological features of IRI; (4) reduced intrahepatic leukocyte infiltration, as evidenced by decreased expression of P-selectin/intracellular adhesion molecule 1, ED-1/CD3 cells and neutrophils; (5) depressed lymphocyte activation, as evidenced by expression of pro-inflammatory cytokine (TNF-alpha, IL-1beta, IL-6) and chemokine (IP-10, MCP-1, MIP-2) programs; (6) prevented hepatic apoptosis and down-regulated Bax/Bcl-2 ratio. Thus, by modulating leukocyte trafficking and cell activation patterns, treatment of rats with FK330, a specific iNOS inhibitor, prevented liver IRI. These results provide the rationale for novel therapeutic approaches to maximize organ donor pool through the safer use of liver grafts despite prolonged periods of cold ischemia.

Induction of systemic lupus erythematosus-like syndrome in syngeneic mice by immunization with activated lymphocyte-derived DNA.

OBJECTIVES: Systemic lupus erythematosus (SLE) is the prototype of autoimmune disease and the mechanisms underlying the disease have not yet been elucidated. Thus, animal models of SLE would facilitate investigation of pathogenetic mechanisms involved in the development of the disease. This study characterizes a murine model of SLE-like syndrome induced by syngeneic activated lymphocyte-derived DNA (referred to as ALD DNA). METHODS: Normal BALB/c mice were immunized subcutaneously with highly purified ALD DNA. Anti-double-stranded DNA (anti-dsDNA) antibodies were determined by enzyme-linked immunosorbent assay. Other SLE-associated autoantibodies were examined by indirect immunofluorescence and anti-ENA (extractable nuclear antigen) profile assay. Pathological changes were analysed by light microscopy and electron microscopy. Kidney cryostat sections were viewed by immunofluorescence for the presence of glomerular IgG and C3 deposits. Proteinuria was measured by Coomassie brilliant blue assay. RESULTS: High levels of anti-dsDNA antibodies and other autoantibodies frequently appearing in SLE were detectable in the sera of ALD DNA-immunized mice. Glomerulonephritis and glomerular deposition of IgG plus C3 were observed in the kidney sections. Moreover, proteinuria was seen in the immunized mice. CONCLUSIONS: SLE-like syndrome can be induced by ALD DNA in normal mice. This induced model may be useful for elucidating the mechanisms involved in autoimmunity to DNA and the development of SLE.

Enhanced late components of AEFs associated with the temporal lobe lesions.

We recorded auditory evoked magnetic fields (AEFs) by presenting pure tone bursts once every 4000 ms in 11 patients with a brain lesion in or in the vicinity of the auditory cortex. AEFs on the damaged side revealed several enhanced deflections in late-latency AEFs (slow AEFs), peaking at approximately 320 (DI), 1030 (D2) and 1600 (D3) ms post-stimulus in eight patients. All the dipoles of slow AEFs were concentrated in the superior temporal regions which were not involved by brain lesions. D1, D2 and D3 dipoles were uniformly upward, downward and upward, respectively. The dipole moment varied from 12 to 122 nAm and had no consistent relationship with latency. This is the first report describing slow AEFs in cases with temporal lobe lesions.

Oil spill model development and application for emergency response system.

The paper introduces systematically the developing principle of CWCM 1.0 oil spill model based on Lagrange system and oil spill fate processes in environment, reviews two oil spill incidents of "East Ambassador" in Jiaozhou Bay and "Min Fuel 2" in the mouth of Pearl River, and designs the predictable system simulating oil spill applied in contingency plans. It is indicated that CWCM 1.0 has met preliminarily the demands for functions of precision simulating and predicting oil spill, and can plan an important role to support oil spill response.

[Separation and identification of thellungianin G from the root of Pimpinella thellungiana Wolff].

OBJECTIVE: To study the effective components in the root of Pimpinella thellungiana METHOD: Column chromatography with silica gel and ODS was employed for the isolation and purification of ingredients. Their structures were elucidated by spectral analysis. RESULT: Three compounds were obtained and identified as 2-(1',2'-epoxy)-4-methoxy pheryl-2-methyl-butyrate(I),4-(1'-propenyl) phenol(II) and 2-methyl butyric acid(III). CONCLUSION: Compound I is a new compound and named thellungianin G. Compounds II and III were separated from P. Thellungiana for the first time.

[Chemical constituents of the root of Pimpinella thellungiana Wolff].

OBJECTIVE: To search for new effective hypotensive components from the Chinese folk drug, the root of Pimpinella thellungiana. METHOD: A new compound was isolated from etheral extract of the drug by chromatographic method and identified by physical, chemical and spectroscopic methods. RESULT: The compound has been identified as 2-(1',2'-dihydroxy)propyl-4-methoxy phenol. CONCLUSION: The compound was obtained from the root of P. thellungiana for the first time.

[Bilateral ureter within single nipple for cutaneous ureterostomy after radical cystectomy].

OBJECTIVE: To evaluate the effect of bilateral ureter within single nipple for cutaneous ureterostomy after radical cystectomy. METHOD: Bilateral ureter within single nipple for cutaneous ureterostomy was performed on 36 patients after radical cystectomy. RESULT: The nipple was formed well, and urinary drainage was patent. No death or severe complication occurred. The patients have been followed up for 3 months to 3.5 years. Urinary, electrolyte and renal functions were within normal levels. No pyelonephritis or hydronephrosis was observed in IVP, retrograde pyelography and/or sonographic examination of kidneys, no nipple constriction. A right ureter was pulled to the left retroperitoneum and formed a nipple with ureter left on the left abdominal wall. CONCLUSION: This is a simple and safe operation, especially suitable to the old and weak patients.

[Studies on the chemical constituents of the root of Pimpinella thellungiana].

Four compounds were isolated from the roots of Pimpinella thellungiana Wolff., their structures were identified as palmitic acid(I), 4-propenylphenol(II), pinoresinol(III), 2-methyl-2-hydroxy-5-methoxy berzo(d) hydrofuran-3-one(IV) by physicochemical constants and spectral analysis (UV, IR, MS, 1HNMR, 13CNMR, 13C-1HCOSY, DEPT), IV of them is a new compound, Pharmacological tests showed IV has a hypotensive effect.

[Expression of proliferating cell nuclear antigen (PCNA) in squamous cell carcinoma of the oral cavity].

Proliferating cell nuclear antigen (PCNA) was identified by immunohistochemistry in 52 cases. PCNA-positive cells had been detected in the basal cells of normal epithelium known to contain proliferating cells. PCNA scores of normal epithelium was 1 as "low", and carcinomas were 3 or 4 as "high". The PCNA scores of high-grade carcinoma was significantly higher than the low-grade carcinoma. The study shows that expression of PCNA correlated with carcinoma grade.

Identification of patterns of transmission of Salmonella within swine production systems using pulsed field gel electrophoresis (PFGE) and repetitive sequence polymerase chain reaction (REP-PCR): a quantitative analysis.

Pulsed field gel electrophoresis (PFGE) using 3 enzymes (Spe I, Xba I, Avr II) and repetitive sequence polymerase chain reaction (REP-PCR) with 3 primers (BOX, ERIC, REP) were compared with respect to their validity as a method for identifying transmission of Salmonella on swine farms. Sixty-eight isolates of Salmonella were obtained from feces of swine, cats, mice, and birds, insect body parts, water and floor samples, and boot scrapings collected on 9 swine farms in Illinois USA. Genetic distances between isolates were calculated using the Dice matching coefficient. Cluster analysis of distance matrices was conducted using the UPG-MA algorithm. There was no significant difference between PFGE and REP-PCR in the genetic diversity detected; however, REP-PCR differentiated between 14 pairs of isolates which PFGE identified as identical. There were no significant differences between PFGE and REP-PCR in identifying all or most close genetic links as isolates from the same farm, the same building, and from the same sampling visit, suggesting ecological validity for both methods. Thus, REP-PCR should be considered as an acceptable and perhaps preferable alternative to PFGE as a genotyping method for studies of Salmonella transmission.
