Powdered diatrizoic acid for radiography of the respiratory tract. Part I. Experimental investigation.

Powdered diatrizoic acid as a contrast medium administered by inhalation and insufflation for visualization of the airways was tested in vitro and in 21 dogs. Good radiopacity of the contrast medium and its antibacterial activity was found in vitro. In the majority of animal experiments visualization of the bronchial tree down to segmental and partially to subsegmental bronchi with only minimal agglomeration of contrast medium and with good or very good demonstration of anatomic details was achieved. In the majority of dogs contrast medium was eliminated from the lungs within 18 hours. Arterial blood gases tested on 5 dogs showed only unimportant changes after contrast medium administration. No adverse reactions were observed. Histologic and ultrastructural examinations after contrast studies showed phagocytic reaction to diatrizoic acid, transient impairment of production of surfactant, reactive changes of bronchial mucosa, and no fibrotic changes in the long-term observation.

[Effects of D-penicillamine and encorton on the fibrotic processes in experimental liver cirrhosis in rats].

The aim of the study was to evaluate the effects of D-penicillamine (cuprenil), encorton and both drugs in combination on fibrotic processes in the rat liver damaged by chronic use of CCl4. The studies comprised 80 white Wistar rats divided into 8 experimental groups. Group I receiving every day oral methylocellulose for 12 weeks was a control group. In group II the rats were given only CCl4 for 12 weeks. In the remaining experimental groups beside CCl4 the animals received drugs in various doses and for various periods. At 12 weeks all animals were killed and post-mortem studies were performed. The liver sections for histopathological studies were fixed in 10% buffered formaline. Paraffin specimens were stained with hematoxylin and eosine. Colour reactions to collagen fibers were performed by using Heidenhein's method and to reticuline fibers by Gomori's method. In the assessment of the severity of fatty degeneration, inflammatory infiltrates and fibrosis the 3-point scale was used, ranging from + for minor changes, ++ for moderate changes, for severe changes, and 0 for no changes. Morphological analysis of the liver showed that chronic administration of CCl4 produced an experimental model of cirrhosis of the liver in rats. Concomitant use of CCl4 and cuprenil revealed its inhibiting action on the fibrotic process in the rats' liver. Inhibition of fibrosis varied and was related to the dose and time of its action. The most optimal was a low dose, 10 mg kg of body weight, whereas a double dose used during the experiment appeared less favourable. Similarly less effective action was exhibited after encorton. After combined use of both drugs the inhibitory effect was negligible. In addition hepatotoxic effects were found manifested by marked fatty degeneration of hepatocytes.

Occurrence and characteristics of sclerotic lesions in uterine arteries of sterile and multiparous pigs.

Sows and gilts of the Great White Polish (GWP) breed were classified into groups of 10 each: gilts 8 mo of age (A), sows over 1 yr old with an average litter size of five (BI) and eleven (BII), 2- to 3-yr-old sows with an average litter size of five (CI) and nine (CII), 4- to 6-yr-old sows with an average litter size of six (DI) and nine (DII). Gilts of group A were necropsied on d 10 of the estrous cycle while sows were necropsied 120 to 360 d postpartum. A blood sample obtained by vena cava puncture immediately before exsanguination was quantitated for total cholesterol, free cholesterol, cholesterol ester, total lipid, triglyceride, high and low density lipoprotein and chylomicrons. Postmortem angiograms of the uterine vasculature were evaluated for occurrence of arteriosclerotic lesions of the arteries and their branches. Sections of blood vessels from areas of restriction were examined histologically and quantitated for lipids and proteolytic and lipolytic enzyme activities. With the exception of high density lipoproteins and chylomicrons, serum concentrations of various lipids increased (P less than .01) with age. Restrictions of the lumen were found in uterine arteries and their branches of most gilts in group A and in sows in groups B, C and D, irrespective of litter size. Histology of uterine artery revealed preatherosclerotic lesions in groups B through D. No relationship between the incidence and degree of sclerotic lesions and litter size was evident. However, the incidence and degree of sclerotic lesions increased with age and parity. Results from histopathology were supported by results from measurement of lipids and enzyme activities of the uterine artery wall. Conception rates of sows in groups C and D were lower than those of group B. Many of these sows (70%) failed to conceive during 1 yr even though they displayed normal estrous cycles.

[Pathomorphological and histochemical changes in the liver of rabbits following fractional irradiation with Co-60].

Two groups of experimental animals, each consisting of 12 rabbits, were subjected to local fractional irradiation with cobalt 60. Group I received the total dose of 2580 R during 13 days, group II - 5100 R during 24 days. The effects of irradiation were estimated on the strength of histological examination of the liver immediately and after 7, 15, 30, 60 and 90 days after the last exposition. The histological sections were stained with haematoxylin and eosin, and colour reactions were performed for argentaffine and collagen fibres and for glycogen, neutral fats, alkaline and acid phosphatase, ATP-ase, glycose-6-phosphatase, non-specific esterase and succinic acid dehydrogenase. It was found that the dose of 2580 R is safe for the liver. The effects of irradiation were slight and limited to weak catabolic disturbances in the form of mild steatosis of the liver and of a transient and short-lived fall of glycogen and rise of hydrolytic enzymes. More pronounced and intense changes were observed in the other group of animals. During the early period, the changes were of a retrograde character and were typical of the acute post-irradiation effect. There was necrosis of the walls of the blood vessels, of the epithelium of the bile ducts and of the liver cells, accompanied by a rise in the hydrolytic enzymes and by a considerable fall of the level of glycogen and succinic acid dehydrogenase. During the late period (30-90 days), changes typical of the so-called post-irradiation hepatitis were found histologically.

Urogranoic acid as a radiopaque additive to the cyanoacrylic adhesive in transcatheter obliteration of renal arteries.

The purpose of the investigation was to study the application of radiopaque materials to the tissue adhesive base of the ester of n-butylic-alpha-cyanoacrylic acid for use in direct transcatheter renal artery obliteration. Lipiodol Ultra-fluid (Ethiodol) and urogranoic acid in quantities of 30--55% by weight of the adhesive were used as the radiopaque agents. The study was performed on seven dogs; obliterative material was administered through a telescopic, coaxial catheter system. Two dogs treated with the Lipiodol adhesive material died, which confirmed the inefficacy of Lipiodol for this purpose. In the other five dogs, treated by adding urogranoic acid to the adhesive, complete and permanent occlusion occurred and the adhesive remained radiopaque for 60--90 days. One of these dogs died of canine distemper infection. The complication of unpredictable embolization of the femoral artery appeared in a second dog. The same material has been used in two cases of nonoperative hypernephroma in humans. Permanent occlusion of the renal artery and elimination of hematuria were achieved. The obliterative material was sufficiently opaque for radiographic visualization.

Role of some biogenic amines in the pathomechanism of glomerulonephritis. I. Histological examinations of kidneys in acute and chronic serum sickness in rabbits.

Investigations were carried out on the role of biogenic amines in the development of glomerulonephritis in rabbits using the experimental model of acute and chronic serum sickness. The animals immunized with foreign-species albumin received additionally 5HT and some of them were given reserpine or PCPA for reducing the level of biogenic amines. The results of histological examinations demonstrated a clear cut inhibitory effect of PCPA and reserpine on the development of inflammatory changes in the glomeruli, and their intensification with simultaneous 5HT administration. The obtained results seem to suggest a significant role of biogenic amines in the pathogenesis of inflammatory lesions developing in renal glomeruli in acute or chronic serum sickness in rabbits.
