We have isolated and sequenced four overlapping cDNA clones to identify the full-length cDNA for topoisomerase II (PsTopII) from pea. Using degenerate primers, based on the conserved amino acid sequences of other eukaryotic type II topoisomerases, a 680 bp fragment was PCR-amplified with pea cDNA as template. This fragment was used as a probe to screen an oligo-dT-primed pea cDNA library. A partial cDNA clone was isolated that was truncated at the 3' end. RACE-PCR was employed to isolate the remaining portion of the gene. The total size of PsTopII is 4639 bp with an open reading frame of 4392 bp. The deduced amino acid sequence shows a strong homology to other eukaryotic topoisomerase II (topo II) at the N-terminus end. The topo II transcript was abundant in proliferative tissues. We also show that the level of topo II transcripts could be stimulated by exogenous application of growth factors that induced proliferation in vitro cultures. Light irradiation to etiolated tissue strongly stimulated the expression of topo II. These results suggest that topo II gene expression is up-regulated in response to light and hormones and correlates with cell proliferation. Besides, we have also isolated and analysed the 5'-flanking region of the pea TopII gene. This is first report on the isolation of a putative promoter for topoisomerase II from plants.

A fermentation system for the continuous ethanol production from sucrose and molasses using calcium alginate immobilized Saccharomyces cerevisiae strain HAU-1 has been optimised. Immobilization of active yeast cells (30%, w/v) was accomplished in 1.5% calcium alginate and these yeast-beads were employed for ethanol production in a vertical column reactor. High ethanol productivity was achieved with a medium containing 10%, w/v sucrose at a dilution rate of 0.20 h-1. Alginate beads containing 30-50% yeast cell--mass resulted in high productivity 60.4 g l-1 h-1). Sugarcane molasses containing 15% total sugars was fermented in column reactor to produce 6.0%, v/v ethanol in continuous system. These yields could be improved to 7.0% with overall 11% increase in fermentation--efficiency with the use of acid-clarified sugar cane molasses medium.

Continuous ethanol production by immobilized cells of Saccharomyces cerevisiae HAU-1 has been studied using synthetic and molasses based medium in column reactors. Immobilization of 30% yeast cells biomass (wet) in 1.5% calcium alginate gel resulted in the production of 20.8 g. l-1.h-1 alcohol at a dilution rate of 0.36 h-1 with approximately 1/3rd volume of the column reactor packed with gel beads. Optimum diameter of the beads was found to be 3.5 mm for efficient fermentation. The size of the column reactor (length to diameter ratio) also affected the productivity and fermentation efficiency due to gas hold-up and mass transfer effects. Molasses could also be fermented by this system but at a lower fermentation efficiency which could be improved, to some extent, by supplementation with nutrients.

A community based prospective study on measles was undertaken during 1986 to 1988 in Ramgarh village of Alwar district (Rajasthan) to elucidate epidemiological features of measles. The initial population of the village was 5258 with 2018 children (0-14 years) which rose to total population of 5923 with 2200 children in 1988. During the entire period of study, all the children (0-14 years) were covered regularly through monthly domicilliary visits by trained paramedical personnel under direct supervision of Medical Officers. A total of 208 measles cases were detected which gave an overall incidence rate of 31.5 per 1000 children (0-14 years) per year. Incidence rate was highest in children of 2-5 years of age group and lowest (4.6 per 1000) in 10-14 years of age group. The maximum number (86.5 per cent) of cases occurred during the first six months of year. The Kolmogrov-Smirnov statistical method validated the seasonal character of the disease (Vn = 5.36, p less than 0.01). A significant (P less than 0.005) rise in seropositivity with increase in age was observed in children (6-36 months) who had no previous history of measles and measles immunization during their life time. A higher rate of sero-conversion was observed in children vaccinated after 10-months of age than those before. No significant relationship of seroconversion following vaccination could be seen with age of vaccination (p greater than 0.50), sex and nutritional status (p greater than 0.10).

Four patients with aortitis and stenotic lesions of the aorta were successfully treated by percutaneous transluminal balloon angioplasty. The peak systolic gradient across the constrictions decreased from 82.5 +/- 35.7 to 37.5 +/- 18.5 mm Hg immediately after dilatation. Repeat angiography in the first three patients performed 2 months later showed a further decrease in the gradient to 13.8 +/- 9.5 mm Hg. There were no complications during or after the procedure, and all the patients have shown remarkable symptomatic relief during the follow-up.
