Effect of Rumalon (glycosaminoglycan-peptide) on the articular tissue.

The effect of glycosaminoglycan-peptide complex (GPC) (Rumalon, made by Robapharm, Switzerland) on cells of the inflammatory periarticular infiltrate and on the articular chondrocytes was studied in experimentally induced papain arthropathy by means of image cytometry and biochemistry. The GPC therapy exhibited some antiinflammatory effect as documented by the reduction of DNA proliferative activity in the inflammatory infiltrate and lowered the activity of hydrolytic enzymes and enzyme inhibitors detected in chondrocytes.

Effect of selected antirheumatic drugs on the metabolism of cartilage and synovial tissue in experimental arthropathy.

The effects of several antirheumatic drugs on the activity of degradative enzymes in normal and pathologic knee joint cartilage and on the proliferative activity of synovial tissue cells were studied. Inflammatory arthropathy was induced in rabbits by intraarticular papain administration. Elevated contents of proteoglycanase and collagenase, together with an increase in serine and cysteine proteinase inhibitors, were found in animals with papain-induced arthropathy. Inflammation also accelerated the rate of proliferation of cells present in the synovial tissue. In the treated animals, the reduction in enzyme activity, decrease in inhibitor content and decreased DNA proliferation rate were registered to a different degree. The suppression of protein synthesis by nonsteroidal antiinflammatory drugs may explain our findings. The best therapeutic results were achieved with glycosaminoglycan polysulphate (Arteparon).

DNA microfluorometry of synovial effusion cells in rheumatic diseases.

A microfluorometric study of relative DNA content in lymphocytes and granulocytes from the synovial fluid of 26 patients with rheumatoid arthritis or seronegative reactive arthritis was carried out. Results obtained from individuals without treatment or treated with nonsteroid anti-inflammatory drugs were compared with measurements of 7 healthy persons. DNA content in granulocytes was more sensitive to effects of disease or therapy than in lymphocytes. Introduction of therapy resulted in decreased percentage of cells in synthetizing or mitotic phase of the cell division cycle (microfluorometric index showing non G0 (G1 population]. A possible local influence of regulatory substances produced in the arthritic joint on DNA metabolism of both cells under study was suggested.

[Study of crystals in synovial exudations with simultaneous determination of adequate enzymes].

The paper presents the results of study of 28 synovial exudations in which the uricase method was used for uric acid determination with simultaneous pyrophosphatase and cathepsin C and D activity determination. The presence of crystals in these exudations was investigated as well. From the study follows that a higher uric acid level and higher pyrophosphatase activity coincide with the finding of urate and pyrophosphate crystals. The reported method of examination applies also to other identifications of microcrystals or crystalline split of urates or calcium pyrophosphate dihydrate in the exudation. The activity of cathepsin D permits to conclude on potential participation of peptides and/or polypeptides in the formation of crystals in the exudation.
