Foscarnet-induced changes in plasma concentrations of total and ionized calcium and magnesium in HIV-positive patients.

The time course and magnitude of foscarnet-induced changes in plasma concentrations of total and ionized calcium and magnesium were investigated in 13 male HIV-positive patients who had no active cytomegalovirus-associated disease. The patients had a mean age of 36 years (range 25-49 years) and a mean CD4 cell count of 550 cells/mm3 (range 130-1280 cells/mm3). Peak (mean +/- SD) plasma concentrations of foscarnet (0.89+/-0.10 mmol/l) were seen at the end of the period of drug infusion (90 mg/kg of foscarnet was infused over 2 hours) and declined with a terminal half-life of 5.7+/-0.7 hours. Plasma concentrations of total calcium declined over an 8-hour period, with the lowest concentration occurring after 4 hours (baseline: 2.29+/-0.09 mmol/l; lowest: 2.18+/-0.07 mmol/l; P < 0.001). By contrast, the lowest plasma concentration of ionized calcium occurred after 2 hours (baseline: 1.25+/-0.04 mmol/l; lowest: 0.99+/-0.05 mmol/l; P < 0.001), before gradually recovering to baseline levels over the next 10 hours. The mean maximal decrease in total calcium was 0.11+/-0.06 mmol/l, compared with 0.26+/-0.04 mmol/l for ionized calcium (P < 0.001). Plasma concentrations of total magnesium declined from 0.79+/-0.06 mmol/l (baseline) to 0.74+/-0.04 mmol/l (P < 0.05) after 4 hours and remained at this level after 8 hours. However, plasma concentrations of ionized magnesium fell steeply from 0.56+/-0.03 mmol/l to 0.39+/-0.03 mmol/l at 2 hours (P < 0.001), followed by a gradual recovery over the next 10 hours. The mean maximal decrease in total magnesium was 0.05+/-0.08 mmol/l, compared with 0.18+/-0.03 mmol/l (P < 0.001) for ionized magnesium. In summary, we found that foscarnet-induced changes in the plasma concentrations of total calcium and magnesium were dissociated from the corresponding changes in ionized calcium and magnesium. The maximal decreases in the plasma concentrations of total calcium and magnesium were smaller in magnitude and occurred much later than did the changes in ionized calcium and magnesium. The relative changes in the plasma concentration of ionized magnesium were greater than those of ionized calcium, indicating that foscarnet binds preferentially to the magnesium ion.

[Regulatory effect of lingqi anshen liquor on erythrocyte immune function and antioxidation in immunosuppressed mice].

The regulatory effect of Lingqi Anshen Liquor (LQASL) on erythrocyte immune function and antioxidation in cyclophosphamide induced immunosuppressed model mice was observed. After given LQASL for 7 days, the results showed that it could antagonize the inhibitory action of cyclophosphamide significantly, demonstrated by raising the level of superoxide dismutase activity, erythrocyte immune adhesive enhance factor and erythrocyte C3b receptor rosette forming rate, lowering the serum malondialdehyde, erythrocyte adhesive inhibitory factor and erythrocyte immunocomplex rosette forming rate. It suggested that LQASL can regulate the erythrocyte immune function and antagonize the injury of oxygen free radical in immunosuppressed mice.

Mitogenic response to TGF-beta in 3T3-F442A cells.

In 3T3-F442A cells, TGF-beta caused cellular proliferation in a time and dose-dependent manner. TGF-beta induced cyclin D1 and cdk2 proteins in 3T3-F442A cells. The mitogenic effect of TGF-beta was specific in nature. The antimitogenic agent, hGH, inhibited the mitogenic effect of TGF-beta and was associated with inhibition of cyclin D1 expression. The protein kinase c inhibitor, staurosporine, inhibited the mitogenic effect of TGF-beta. Taken together, these results suggest that TGF-beta affects expression levels of cell cycle-regulated proteins and its mitogenic effect is mediated through protein kinase C in 3T3-F442A cells.

Induction of tumor suppressor p21 protein by kinase inhibitors in MCF-7 cells.

The expression level of tumor suppressor p21 protein in response to protein kinase inhibitors was examined in MCF-7 cells. Both H7 (serine/threonine kinase inhibitor) and staurosporine (protein kinase C inhibitor) were able to induce p21 protein in a time- and dose-dependent manner. Induction of p21 by H7 but not staurosporine required the induction of p53 protein. Induction of p21 was preceded by the induction of p53 protein. Based on FACS analysis, both H7 and staurosporine act as antimitogenic agents.

Phosphorylation and activation of the DNA binding activity of purified Stat1 by the Janus protein-tyrosine kinases and the epidermal growth factor receptor.

The activation of Janus protein-tyrosine kinases (Jaks) and the subsequent phosphorylation and activation of latent signal transducers and activators of transcription (Stats) are common elements in signal transduction through the cytokine receptor superfamily. To assess the role and specificity of Jaks in Stat activation, we have utilized baculovirus expression systems to produce Stat1 and the Jaks. Co-expression of Stat1 with Tyk2, Jak1, or Jak2 resulted in the specific tyrosine phosphorylation of Stat1 at Tyr701, the residue phosphorylated in mammalian cells stimulated with interferon gamma. Alternatively, Stat1, purified to apparent homogeneity from insect cell extracts, was phosphorylated at Tyr701 in Jak immune complex kinase reactions. Phosphorylation of purified Stat1 was necessary and sufficient for the acquisition of DNA binding activity. The specificity in both systems was indicated by the inability of a Jak2 catalytically inactive mutant (Jak2-Glu882) or the Tec protein-tyrosine kinase to phosphorylate Stat1. However, immune complex-purified epidermal growth factor receptor was capable of phosphorylating purified Stat1 at Tyr701 and activating its DNA binding activity in in vitro reactions.

Effects of tumor necrosis factor-alpha on antimitogenicity and cell cycle-related proteins in MCF-7 cells.

Tumor necrosis factor-alpha (TNF-alpha) demonstrated antimitogenic activity in MCF-7 cells (estrogen receptor-positive human breast cancer cells) in a dose- and time-dependent manner (EC-50 of 2.5 ng/ml). This antimitogenic effect of TNF-alpha was accompanied by a decreased number of cells in S phase in a dose- and time-dependent manner. Based on growth arrest experiments using aphidicolin, it is apparent that TNF-alpha acted in early G1 phase. It did not show antimitogenic effects once cells reentered the S phase based on [3H]thymidine incorporation into DNA and cell cycle analysis. Specificity of TNF-alpha was established by using monoclonal anti-human TNF-alpha antibody. On the basis of Western immunoblot analysis of Rb, p53 and cell cycle inhibitory protein (Cip1) (p21) proteins, TNF-alpha decreased Rb protein expression in a dose- and time-dependent manner whereas it increased the expression level of tumor suppressor p53 protein. TNF-alpha also increased the expression level of Cip1 (p21) protein in a dose-dependent manner. This induction of Cip1 (p21) protein was preceded by the induction of p53 protein in MCF-7 cells. Cip1 (p21) protein associated with cyclin D was also increased. Tumor suppressor Rb protein expression was increased during G1 to S phase progression. Cyclin D protein expression levels were not changed in response to TNF-alpha treatment, although serine/threonine kinase inhibitors such as H7 and the protein kinase C inhibitor staurosporine decreased cyclin D expression levels in MCF-7 cells. Based on experiments with staurosporine, it appears that TNF-alpha does not utilize a protein kinase C pathway in MCF-7 cells. Other cell cycle-related proteins such as Cdk2, Cdc2, and Cdk4 did not show any change in response to TNF-alpha. TNF-alpha did not affect complexes between cyclin D and Cdk2, Cdk4, and Rb proteins in MCF-7 cells. Taken together these results suggest that Rb, p53, and Cip1 (p21) proteins mediate TNF-alpha antimitogenic activity, and TNF-alpha induces growth arrest in the G1 phase in MCF-7 cells.

Effect of human growth hormone and insulin on [3H]thymidine incorporation, cell cycle progression, and cyclin D expression in 3T3-F442A preadipose cells.

Cellular growth- and cyclin D expression-regulating activities of GH and insulin were investigated in 3T3-F442A preadipose cells under serum-free culture conditions. The present report provides evidence that the proliferative potential of 3T3-F442A cells is reduced by GH in a time- and concentration-dependent manner based on [3H]thymidine incorporation assay and cell cycle analysis. In contrast, treatment of 3T3-F442A cells with insulin resulted in cellular proliferation. The insulin-induced proliferation of 3T3-F442A cells was diminished in the presence of GH. In an effort to define biochemical events relevant to the regulatory activities of GH and insulin on the proliferation of 3T3-F442A cells, the effects of these peptides on the expression of cyclin D were studied using Western blotting. Treatment of 3T3-F442A cells with insulin led to an increase in cyclin D expression relative to that in untreated cells. The insulin-elicited expression of cyclin D was time and dose dependent. In addition, the ability of insulin to induce cyclin D expression was reduced by GH. Our experimental results indicate that proliferation of 3T3-F442A cells was regulated by GH and insulin. The regulatory effects of GH and insulin are mediated at least in part by the alternating expression of cyclin D protein.

[A survey on HIV and HV infection of heroin addicts in Linchang and Kunming].

Serum samples were collected from 353 heroin addicts in Linchang Prefecture and Kunming Area, Yunnan Province for detecting anti-HIV with ELISA and Western Blot, and 319 of them also for detecting various infection markers of hepatitis A, B, C and D with ELISA. Results showed no anti-HIV was found among them, overall infection prevalence rate of hepatitis A, B, C and D was 78.11%, 72.73% for males and 90.91% for females, and 1.57%, 63.97%, 51.85% and 5.79% positive rates for anti-HAV-IgM, HBV markers, anti-HCV and HDAg and/or anti-HD-IgM, respectively. Prevalence of mixed infection with different types of hepatitis was 40.07%, and 35.35% for mixed HBV and HCV. Infection rate of hepatitis B and C in Kunming Area was significantly higher than that in Linchang Prefecture, and so did in Han nationality than in other minority nationalities. It suggests hepatitis viruses infection is caused by drug abuse, use of non-sterilizing syringes with others.

[Effects of intracerebroventricular administration of norepinephrine on paraventricular neurosecretory cells].

Extracellular electrical activities of paraventricular nucleus (PVN) cells were recorded in anaesthetized adult rats by using glass microelectrodes. After antidromical identification of the magnocellular neurosecretory cells, the effects of intracerebroventricular (i.c.v.) injection of norepinephrine (NE) on such cells were observed. Of 49 neurons of antidromic stimulation, phasic, fast continuous and slowly irregularly. firing units constituted 42.9%, 36.7%, and 20.4% of the total population respectively. With i.c.v. injection, NE inhibited the activity of most phasically firing units, but enhanced that of most fast continuous and slowly irregular firing units. The inhibition effect of NE could be partially antagonized by i.c.v. injection of phentolamine (2 micrograms/microliters), showing that the effect is possibly mediated by alpha-receptor. The above findings indicate that NE plays different roles in different PVN magnocellular neurons having different patterns of firing activity.

Four outbreaks of human trichinellosis in Henan Province.

Four outbreaks of human trichinellosis caused by eating pork together had occurred successively in two rural areas and two cities of Henan Province, where the disease is endemic. Of the 110 persons involved, 54 had the onset. All of them had the history of eating pork. In the outbreak of Dengzhou, in those persons who dined together, men were all uninfected and 13 women were infected. The difference found between sexes was suggested to be related with spirit drinking. In these 54 patients, the latent period ranged from 3 to 28 days. The relevant clinical symptoms and signs were fever, edema, myalgia, rash, headache, nausea, abdominal pain and diarrhea. Vomiting was uncommon. Most patients were hospitalized and all cases recovered in two weeks after proper treatment with albendazole.

[A research of acute hemodynamics and hormonal changes of intravenous captopril in patients with cor pulmonale and pulmonary hypertension].

In 20 patients of cor pulmonale with heart failure and respiratory failure and in 11 healthy men, after intravenous captopril injection, acute hemodynamic and hormonal change were observed. The results indicated that intravenous captopril injection rapidly reduced cardiac preload and afterload, reduced pulmonary arterial pressure, increased cardiac output, inhibited renin system activity, decreased plasma level of thyroxin and atrial natriuretic peptide. A rapid symptomatic improvement in patients with acute or severe cor pulmonale was shown and there was no significant effect on blood gas immediately.

Assisted refolding of recombinant prochymosin with the aid of protein disulphide isomerase.

Protein disulphide isomerase (PDI) was shown to be able to accelerate the refolding of unfolded recombinant prochymosin and to enhance the overall yield of active protein. Unlike previous reports in this study PDI was found to be active at pH values as high as 11. The coincidence of the similar apparent optimum pH values of uncatalysed and PDI-catalysed reactions suggests that conditions favourable to spontaneous refolding of proteins may help PDI to catalyse thiol/disulphide interchange. Under the conditions described here no exogenously added dithiothreitol was required for PDI-catalysed renaturation, implying that the disulphide form of PDI was reduced to its active form by the free thiol groups in prochymosin molecules.

Mathematical models of the early embryonic cell cycle: the role of MPF activation and cyclin degradation.

Recent advances in cell biology indicate that the interactions between two proteins, cdc2 and cyclin, together with the activity of the cdc2/cyclin complex called MPF in the cytoplasm form the basis of a universal biochemical control mechanism for the cell division cycle in eukaryotes. Based on experimental facts that total cdc2 level is constant throughout the cell cycle and that onset of mitosis is subsequent to activation of MPF, we propose and analyze two different but related models--an ordinary differential equations model and a delay differential equations model--for the control of the early embryonic cell division cycle. Assuming very general reaction terms in the model equations, it is shown that MPF activation and rapid cyclin degradation triggered by active MPF drive cells to alternate between interphase and mitosis, the two phases of the cell cycle.

Quiescence and transient growth dynamics in chemostat models.

A chemostat model for microbial growth taking into account that cells can exist in two states, active and quiescent, is proposed and analyzed. Cells adapt to environmental (nutrient supply) fluctuations by changing from one state to another with asymmetric thresholds. Depending on the growth conditions, steady-state growth is attained with either a homogeneous population of active cells or a heterogeneous population of both active and quiescent cells. Transient oscillatory growth that has been observed in bacterial and algal chemostat cultures can be a result of such an adaptation mechanism.

Cytokine receptors and signal transduction.

The past few years have seen an explosion in the identification, cloning and characterization of cytokines and their receptors. The pleiotropic effects of many of the growth factors and the considerable redundancy in the actions of growth factors have contributed to a mass of descriptive literature that often seems to defy summary. Only recently have common concepts begun to emerge. First, cytokines mediate their effects through a large family of receptors that have evolved from a common progenitor and retain structural and functional similarities. Within the haematopoietic system, the cytokines are not usually instructive in differentiation, but rather supportive, and may contribute to some differentiation-specific responses. The patterns of expression of cytokine receptors are therefore a product of differentiation and provide for changes in physiological regulation. The second important concept that is emerging is that the cytokines mediate their mitogenic effects through a common signal-transducing pathway involving tyrosine phosphorylation. Thus, although the cytokine receptor superfamily members do not have intrinsic protein tyrosine kinase activity, by coupling to activation of tyrosine phosphorylation they may affect cell growth by pathways that are common with the large family of growth factor receptors that contain intrinsic protein tyrosine kinase activity. The coupling of cytokine binding to tyrosine phosphorylation and mitogenesis requires a relatively small membrane-proximal domain of the receptors. This region has limited sequence similarity which may be required for the association of individual receptors with an appropriate kinase. Activation of kinase activity results from the dimerization or oligomerization of receptor homodimers or heterodimers. Again this requirement is similar to that seen with the growth factor receptors which have intrinsic protein tyrosine kinase activity. The protein tyrosine kinases that couple cytokine binding to tyrosine phosphorylation are members of the Jak family of kinases. The ubiquitous expression of these kinases provides a common cellular background on which the cytokine receptors can function and on which unique functionally distinct receptors have evolved. In particular, tyk2 is required for the responses initiated by IFN-alpha while Jak2 has been implicated in the responses to G-CSF, IL-3, EPO, growth hormone, prolactin and IFN-gamma.(ABSTRACT TRUNCATED AT 400 WORDS)

Tec kinase associates with c-kit and is tyrosine phosphorylated and activated following stem cell factor binding.

Stem cell factor (SCF) plays a crucial role in hematopoiesis through its interaction with the receptor tyrosine kinase c-kit. However, the signaling events that are activated by this interaction and involved in the control of growth or differentiation are not completely understood. We demonstrate here that Tec, a cytoplasmic, src-related kinase, physically associates with c-kit through a region that contains a proline-rich motif, amino terminal of the SH3 domain. Following SCF binding, Tec is tyrosine phosphorylated and its in vitro kinase activity is increased. Tyrosine phosphorylation of Tec is not detected in the response to other cytokines controlling hematopoiesis, including colony-stimulating factor-1 (CSF-1), granulocyte-macrophage colony-stimulating factor (GM-CSF), and interleukin-3 (IL-3). Conversely, the cytoplasmic kinase JAK2 is activated by IL-3 but not by SCF stimulation. The activation of distinct cytoplasmic kinases may account for the synergy seen in the actions of SCF and IL-3 on hematopoietic stem cells.
