Relative selectivity of some conformationally constrained tryptamine analogs at 5-HT1, 5-HT1A and 5-HT2 recognition sites.

In an attempt to define pharmacophoric differences between 5-HT1, 5-HT1A and 5-HT2 recognition sites, a number of rigid analogs were studied and compared to analogous free chain tryptamines. Racemic partial ergolines RU 27849 and RU 28306 showed reduced potency at all 5-HT1 sites, but were at least equipotent to analogous tryptamines at the 5-HT2 site. A nonergoline-like constrained analog of tryptamine was similar in potency to RU 27849 at all 5-HT1 sites, but showed a 4-fold enhancement in potency over RU 27849 and tryptamine at the 5-HT2 site. At all 3 sites, 3-(tetrahydropyridyl) indoles (unless substituted at the indole 2-position) were the most potent rigid analogs studied and represent the most promising class for the development of selective compounds.

3H-imipramine binding in platelets: influence of varying proportions of intact platelets in membrane preparations on binding.

Human platelets possess high-affinity 3H-imipramine binding sites. A study in ten healthy volunteers showed that platelet preparations produced by mechanical disruption contained varying proportions of intact platelets, as measured by the cytoplasmic marker enzyme LDH. This may invalidate the protein reference. Higher proportions of membranes in the preparations lead to higher Bmax values of imipramine binding (fmol/mg protein). The use of intact platelets in imipramine binding studies is therefore preferable to membrane preparations, particularly in studies where the interindividual variation of binding parameters is of interest.

Nonspecific X-linked mental retardation--a review.

Mental retardation, in particular the "X-linked" type, has interested geneticists for many years. An increasing number of affected families have been to genetic counselling centres, and an effort is being made to find clinical and cytogenetic methods so a reliable diagnosis can be made. This would enable the detection of carriers and the opportunity to offer prenatal diagnosis. Many questions remain regarding X-linked mental retardation, its causes, diagnosis, and prevention. In this article we try to give an overview about the status of our present knowledge and the questions to be answered in the future.

[Objective determination of prostate size with ultrasound].

The rectal digital examination of the prostate is, despite the high diagnostic value, for a large part subjective in its evidence. By means of the ultrasound examination the size of the prostate may be objectified, which otherwise is difficult to be described, and thus may be used as a firm and reproducible relation size in the diagnosis and control of the course of disease of the prostate which are accompanied by an increase of the organ.

[Changes in the system of kidney pelvis and calices in bladder neck adenoma].

The development of a renal lesion following an adenoma of the vesical neck is one of the most important complications of this disease. For the determination of the frequency of changes of the renal calyces in the adenoma of the vesical neck the findings of 253 patients of the years 1976 and 1977, in whom for the first time such an excrescence was established were evaluated. 135 excretion urogrammes showed changes of the renal pelvis and the renal calyces. The main findings were a narrowing of the renal calyces with spindle-shaped necks of the calyces. The calyces themselves were deformed only in one part of the patients. Urinary stasis and cirrhoses of the kidney were not found. As cause of the narrowed renal calyces in correlation with the clinical and laboratory-clinical findings the chronic pyelonephritis, the latent dehydration of the patient and disturbances of the motility of the urodynamics are discussed. As conclusion of these results the opinion is uttered that all male persons from 45 years, who have clinically healthy kidneys and in whom for any reasons an excretion urography has been performed, and in which evaluation narrowed renal calyces were found, by all means must undergo the examination by an urologist for the exclusion of a disease at the vesical neck.

[The septic kidney].

After the definition of the notion of septic kidney with the help of a 10-year-survey cause, therapy and results of our clinical material are analysed. The obstructive urolithiasis is found to be the main cause of the development of septic picture of the disease. The nephrostomy is in suitable cases the operative therapy of choice. The relatively good results of the treatment are to be traced back to a close interdisciplinary cooperation of operative urology, intensive therapy and use of haemodialysis.

[Our first results with cryotherapy on the prostate].

It is reported on our first experiences with the cryotherapy in 15 patients with adenomas of the neck of the bladder and carcinoma of the prostate who all on account of high quantities of residual urine underwent a permannt catheterisation and in whom an open operation of Tur were contraindicated due to severe concurring secondary diseases. 2/3 of the patients we could free from the permanent catheter, in 1/3 an undisturbed miction free from residual urine was possible In 1/3, however, the catheter had to be kept also after the end of the treatment.

[Significance of prostatic biopsy in the diagnosis of chronic prostatitis].

It is reported on the importance of the prostate biopsy for the limitation of the prostate neurosis and the real chronic prostatitis. In own patients the tentative diagnosis "chronic prostatitis" could be proved histologically in only 8 out of 30 cases. We think the biopsy necessary in cases of chronic recurrent prostatitis, because the therapy depends extremely on the exact diagnosis and the complicating quota is relatively small in the prostate biopsy.
