The influence of thymus extracts on the chemotaxis of polymorphonuclear neutrophils (PMN) from patients with insulin-dependent diabetes mellitus (IDD).

The main goal of this study was to evaluate the possible influence of thymus extracts on the chemotaxis of PMN isolated from diabetic patients. The analyzed group consisted of fifty patients with insulin-dependent diabetes mellitus (IDD), and twenty healthy adults were taken as controls. The results showed, that PMN isolated from patients with IDD manifested the impaired chemotaxis toward zymosan-activated plasma (ZAP), plasma incubated with cellophane and supernate from E. coli culture. Both thymus extracts did not change the directed migration toward control plasma and toward ZAP. THYMEX-L did improve the migration of diabetic PMN toward plasma incubated with cellophane, whereas TFX-THYMOMODULIN ameliorated the impaired chemotaxis toward supernate from E. coli culture. Both thymus extracts can attract PMN obtained from healthy donors, but not the cells isolated from diabetic patients.

Polymorphonuclear neutrophils function in splenectomized patients.

Some essential functions of polymorphonuclear neutrophils (PMN) were evaluated in 30 patients splenectomized because of rupture of the spleen. These cells revealed normal random migration, adherence, unstimulated O2- and H2O2 production. Phagocytosis of viable staphylococci was higher than in controls, whereas chemotaxis, bactericidal capacity, aggregation and stimulated O2- and H2O2 production were significantly impaired. PMN from splenectomized patients manifested also the decreased intracellular myeloperoxidase activity. The percentage of cells with receptor for Fc IgG in peripheral blood was markedly decreased. Plasma of these patients induced increased adherence of autologous as well as control neutrophils. The possible mechanisms leading to the observed events are discussed.

Possible prognostic value of plasma activity augmenting polymorphonuclear neutrophils (PMN) adherence estimations in untreated patients with chronic myeloid leukemia.

Plasma activity augmenting polymorphonuclear neutrophils adherence was evaluated in 30 untreated patients with chronic myeloid leukemia. The estimations were done by the use of cells isolated from healthy persons. The plasma of some patients with CML induced the intensive adherence of PMN to plastic surface. Further observations suggested, that these patients were characterized by earlier blast crisis occurrence and poorer prognosis.

Polymorphonuclear neutrophils function in untreated patients with chronic myeloid leukemia.

Morphologically mature polymorphonuclear neutrophils (PMN) isolated from the peripheral blood of 30 patients were examined. These cells manifested the diminution of phagocytosis, random migration, chemotaxis, thermotaxis and bactericidal capacity. The surface charge of circulating PMN and the percent representation of cells with receptor for Fc IgG were significantly decreased. Stimulated as well as unstimulated oxygen consumption, production of O2- and H2O2 were significantly decreased, whereas the hexose monophosphate shunt activity was similar to that noticed in controls. The aggregation of leukemic PMN was practically normal, however their adherence was significantly higher. The possible mechanisms responsible for the observed PMN defects are discussed.

Evidence of polymorphonuclear neutrophils (PMN) activation in patients with insulin-dependent diabetes mellitus.

Polymorphonuclear neutrophils' chemotaxis, surface charge, superoxide anions generation, NBT (nitro blue tetrazolium) reduction and intracellular lysozyme, and beta-glucuronidase content were estimated in patients with type I diabetes mellitus in a similar state of metabolic control. The chemotaxis of diabetic cells toward bacterial chemotactic factors was similar to controls, whereas migration toward complement-derived chemoattractants was significantly reduced. Polymorphonuclear neutrophils isolated from diabetic patients, when unstimulated, produced significantly greater amounts of superoxide anions and reduced NBT more efficiently. They also revealed reduced surface charge and lower intracellular content of lysozyme, whereas beta-glucuronidase content was similar to controls. The results obtained seem to indicate that neutrophils in patients with insulin-dependent diabetes manifest signs of being in the activated state. The possible mechanisms of such stimulation are discussed.

Phagocytosis, bactericidal capacity, and superoxide anion (O2-) production by polymorphonuclear neutrophils from patients with diabetes mellitus.

Phagocytosis, bactericidal capacity and superoxide anion production of polymorphonuclear neutrophils (PMN) were estimated in 30 patients with well-controlled insulin-dependent diabetes (IDD) and in 50 patients with non insulin-dependent diabetes (NIDD). The estimations were additionally done in 20 elderly patients without glucose intolerance. The estimations of bactericidal capacity were performed in autologous-, zymosan activated-, inactivated- and control plasma. The phagocytosis of viable staphylococci was unchanged in all evaluated groups. The bactericidal capacity in all diabetic patients was significantly reduced. It was fully correctable in patients with IDD by suspension of cells in control or zymosan activated plasma. The improvement of PMN bactericidal capacity in patients with NIDD in similar conditions was less distinct. The superoxide anion production in patients with IDD was similar to values noticed in healthy persons. Whereas, the O2- production in patients with NIDD as well as in elderly patients were significantly reduced and correlated significantly with bactericidal capacity impairment. The possible mechanism of noticed disturbances were discussed.

Intravascular degranulation of polymorphonuclear neutrophils during hemodialysis. The effects of pretreatment with colchicine.

Polymorphonuclear neutrophil (PMN) counts, plasma activity inducing PMN aggregation, augmenting PMN adherence and chemotactic activity were estimated during hemodialysis (HD) with cuprophane membranes. All these plasma activities are accepted as related to the presence of activated complement (C). The same estimations were made for hemodialyzed patients pretreated with colchicine. The results, especially comparison of plasma activities in blood entering and leaving the dialyzer, indicate that significant amounts of activated C components are generated within the patients vascular bed. The PMN entrapped in the pulmonary microcirculation that undergo subsequent degranulation are most probably the source of these C components. This concept was confirmed by the results for patients pretreated with colchicine, who had significantly less of the activities generated in the patients own circulation.

The influence of thymus extract (TFX) on the phagocytosis and the bactericidal capacity of polymorphonuclear neutrophils from patients with insulin-dependent diabetes mellitus.

The influence of thymus extract (TFX) on phagocytosis and bactericidal capacity of polymorphonuclear neutrophils (PMN) from patient with insulin-dependent diabetes was studied. In previous observations PMN from these patients revealed the serum-related defect of bacterial killing. The preincubation of cells with standard dose of TFX significantly increased ingestion rate and recovered the bactericidal capacity to normal values. The augmentation of bactericidal capacity due to preincubation with TFX appeared to be dose-dependent and not related to other blood cells participation. The increase of bacterial killing was also evident, when TFX was applied after phagosome formation. The presented studies indicate that, among numerous peptides, thymus extracts contain those exerting their hormone-like activity on polymorphonuclear neutrophils. The observations may be of clinical importance.

Unsaturated vitamin B12 binding capacity and serum lysozyme activity during the marrow granulocytes reserve tests.

Unsaturated vitamin B12 binding capacity (UBBC) and serum lysozyme activity (LZM) were estimated durinng the endotoxin, prednisone and hydrocortisone marrow granulocyte reserve (MGR) pool tests. Our results showed, that no additional mechanism except the shift of MGR from marrow caused granulocytosis after typhoid vaccine administration. While the prednisone, when given orally diminished additionally the number of the physiologically destroyed neutrophils. The hydrocortisone, however, showed the results very similar to those obtained after typhoid vaccine adminstration. Thus the hydrocortisone test seems to be most useful. It gives as good information as typhoid vaccine test but does not show its side-effects.

[Tissue migration of neutrophil granulocytes in the course of antiproliferative treatment in Hodgkin's disease].

The ability of neutrophil granulocytes to mobilization in sterile inflammatory focus was studied in patients with Hodgkin's disease during treatment. Plastic chambers filled with patient's serum were used for this purpose. The migration ability of granulocytes in the group of patients was reduced already before beginning of treatment. It decreased even more 3 days after beginning of cytostatic treatment but on the 7th day the number of cells passing into the inflammatory exudate increased to values observed in healthy subjects. The observed changes were not correlated with fluctuations in the granulocyte count in the peripheral blood observed on the same days. It is suggested that the evaluation of migration ability of granulocytes to the inflammatory focus may provide better information about the defensive mechanisms of the organism than determination of the count of these cells in the peripheral blood.

[Effect of prednisone and typhoid vaccine on the size of systemic neutrophil granulocyte pool].

Changes of total granulocyte pool in the organism were evaluated after administration of prednisone or typhoid vaccine. For this purpose the unsaturated capacity of serum vitamin B12 binding was determined. After administration of prednisone the rise of granulocytes in peripheral blood was associated with a rise in the unsaturated capacity of vitamin B12 binding by the serum which was not observed after administration of typhoid vaccine. This observation confirmed the suggestions that corticosteroids not only mobilize the bone marrow reserve of granulocytes but prolong also their survival in the organism.

The influence of increasing glucose concentrations on selected functions of polymorphonuclear neutrophils.

The influence of increasing glucose concentrations on some essential functions in polymorphonuclear neutrophils (PMN) was evaluated in vitro in 20 healthy persons. PMN chemotaxis, adherence, phagocytosis and bactericidal capacity were estimated. Higher than physiologic glucose concentrations were found to diminish chemotaxis, phagocytosis and bactericidal capacity. The results obtained may indicate another possible mechanism that accounts for the increased susceptibility to infection observed in diabetic patients. However, PMN adherence rose parallel with increasing glucose concentrations reaching maximum values at 300 mg/dl (16.6 mmol/l). The changes of PMN adherence may suggest the participation of these cells in the development of microangiopathic injuries.
