Effect of monoclonal antibodies on the entry and development of Plasmodium vivax sporozoite in cultured cells.

We have observed the effect of monoclonal antibodies against sporozoites (2F2,NV3,2E10) and blood stages (4B2,8E3) of P. vivax on entry and subsequent development of P. vivax sporozoites in HepG2--A16 cells in vitro. The results demonstrated that inhibitory effects of monoclonal antibodies on the attachment and entry were found to be related to the antibody concentration. At 25 micrograms/ml, the percentages of inhibition of sporozoite invasion were 100% (2F2), 76%(NVS3) and 10.5% (2E10). Even if the invasion was not totally inhibited, the presence of abnormal exoerythrocytic schizonts suggested that continued effect of antibodies after sporozoite penetration still existed. No significant effects of 4B2 (4.5%) and 8E3 (3.4%) were recorded as compared with normal mouse serum. These findings indicate that inhibition of sporozoite invasion assay may be useful for determining the protective effect of anti-sporozoite antibodies in vitro.

Determination of L-glutamate using flow injection analysis with immobilized L-glutamate oxidase reactor.

L-Glutamate oxidase (GOD) and horseradish peroxidase (HRP) were covalently coupled on alkylamine pretreated controlled pore glass (CPG) by means of glutaraldehyde. The immobilized enzymes were packed into a teflon tube and used in flow injection analysis (FIA) system for L-glutamate determination. A good linearity range was obtained at 0.1-2.0 mM, and the coefficient of variation was 0.7% (n = 8). More than 80 samples were measured within an hour. The stability of the immobilized GOD reactor was good, retaining 50% of its initial activity after 4 months storage in buffer at 4 degrees C. When the concentration of L-glutamate remained lower than 2.5 mM, the determination of L-glutamate in this system was not affected by pH and temperature within the range of 6.0-8.0 and 20-35 degrees C, respectively. The system was applied to determine L-glutamate in broth samples during L-glutamate fermentation and good correlations were achieved between results obtained with the FIA system, L-glutamate oxidase kit and Warburg's method.

The glutamate biosensor and its application to flow injection analysis system.

A micro-enzyme electrode was fabricated by cross-linking L-glutamate oxidase with glutaraldehyde on aminopropyl-platinized platinum wire. A flow injection analysis system with glutamate sensor was used for L-glutamate determination. The peak current is linearly related to the L-glutamate concentration in the range of 0.02-2.0 mM, with good performance, accuracy (CV = 0.4%), fast response (< 60s), and stability (> 20 days). The system was applied to determine the concentration of L-glutamate in a fermentation broth. The recovery rate was in the range of 98.7-107.5%.

[Etiologic study on the relationship between HBV, HCV and HCC].

A total of 110 HCC patients were sampled randomly in 20 townships of Qidong county (a higher incidence area of HCC in China) from Mar. to Sept. 1989. Each patient was paired with two healthy persons as control. The results showed that the prevalences of HBV and HCV in 110 patients were 85.45% and 17.27% respectively, and were significantly higher that in control (50.45% and 4.09%). Using Conditional Logistic Regression Model. we found that the main risk factors of HCC in Qidong were HBsAg and Pre-S2 for HBV and anti-HCV for HCV. The OR value were 10.22, 2.96 and 2.63, respectively. Drinking tea was the only protective factor (OR = 0.36). Among 110 HCC patients. 13.64% were positive for both HBV and HCV. The OR value was the highest (6.48). 88.18% patients were either HBV or HCV positive. The OR values were similar (3.90 and 3.89). It suggests that the most important risk factor of HCC in Qidong was HBV infection because it had a higher prevalence among the population while the HCV infection having an equal OR value was also a risk factor.

[In vitro cultivation of the exoerythrocytic stage of Plasmodium vivax (southern China isolate)].

An in vitro culture system for the exoerythrocytic (EE) stage of Plasmodium vivax was first developed in our laboratory in China. Anopheles stephensi mosquitoes were infected by membrane feeding with heparinized blood from a volunteer. After 14-18 days, the mosquito salivary glands were aseptically dissected in culture medium and ground in a tissue grinder to form sporozoite suspension. Sporozoites were counted and added to the cultures of monolayer hepatoma cells at the number of 4.9 x 10(4)-1.3 x 10(5) per cover glass. Sometimes, 8-10 pairs of infected gland were added directly to the cultured cells. On day 7, EE schizonts of P. vivax were found in cultures. In addition, normal erythrocytes (type o) were added to the cultures on day 10 at a concentration of 10(8) per dish. Fourteen days later, erythrocytes in culture supernatant were collected and thin blood films were made. Numerous intra-erythrocytic P. vivax parasites were identified on the films after Giemsa staining. Most intra-erythrocytic forms were rings and early large trophozoites. Schuffer's dots were present in many of the infected cells which were pale and obviously enlarged. These results indicated that the in vitro hepatic cycle of P. vivax was established.

Beta-carotene enhances plaque detection by fluorescence attenuation in an atherosclerotic rabbit model.

Improved detection of plaque during cardiovascular procedures could enhance the outcome of diagnosis and therapy. We evaluated a new method to stain plaque using a special intravenous preparation of beta-carotene (beta-C) in an in vivo model. beta-C was used to enhance the absorption coefficient of plaque and decrease the laser-induced fluorescence emission. Using this approach the difference in fluorescence emission was accentuated between normal artery and atherosclerotic plaque. Twenty-nine NZW rabbits were divided into five groups, each receiving a different intervention. This included the administration of beta-C to rabbits on a normal or a high cholesterol diet, with or without endothelial debridement. Aortae were examined grossly, by histology, and relative total fluorescence was detected at 886 sites using 488 nm or 514 nm laser excitation. At 488 nm excitation, unstained plaque attenuated total fluorescence twice as much as normal controls (7.55 +/- 1.46 vs. 15.06 +/- 3.12; P < 0.0001); beta-C stained plaque attenuated total fluorescence 17 times more than normal controls (0.89 +/- 0.29 vs. 15.06 +/- 3.12; P < 0.0001). Total fluorescence from unstained plaque was eight times greater than plaque stained with beta-C (7.55 +/- 1.46 vs. 0.89 +/- 0.29; P < 0.0001). Results obtained using 514 nm excitation were similar. The attenuation effect persisted up to 8 weeks following beta-C administration. Thus, beta-C stained plaque displayed fluorescence attenuation, which suggests that pretreatment with beta-C may greatly enhance plaque detection. This may be useful as a guide during plaque removal procedures.

[Antimalarial activity of deoxoqinghaosu].

Compound 3 (deoxoqinghaosu) and compound 4 (deoxocarbaqinghaosu) were synthesized from arteannuic acid (2). Primary exploration of their antimalarial effect against K173 strain of Plasmodium berghei shows that compound 3 is somewhat more effective than the natural compound--qinghaosu. However, the carba-analogue compound 4 is almost ineffective. The enhanced activity of compound 3 is supposedly ascribed to its higher lipophilicity and, hence, higher affinity for plasmodial membrane. Further studies are in progress.
