Measuring the parity of an N-qubit state.

We present a scheme for a projective measurement of the parity operator Pz = Product(N)i=1 sigma(i)z of N qubits. Our protocol uses a single ancillary qubit, or a probe qubit, and involves manipulations of the total spin of the N qubits without requiring individual addressing. We illustrate our protocol in terms of an experimental implementation with atomic ions in a two-zone linear Paul trap, and further discuss its extensions to several more general cases.

Globoid cell leukodystrophy (Krabbe disease): normal umbilical cord blood galactocerebrosidase activity and polymorphic mutations.

Globoid cell leukodystrophy is an inherited metabolic disorder of the central nervous system caused by deficiency of the lysosomal enzyme galactocerebrosidase. Haematopoietic stem cell transplantation is the only available effective treatment. The engraftment from normal donors provides competent cells able to correct the metabolic defect. Umbilical cord blood cells have proved to significantly decrease complications and improve engraftment rate compared to adult marrow cells in haematopoietic stem cell transplantation. Umbilical cord blood cells must be of sufficient activity to provide central nervous system recovery after engraftment is obtained. Galactocerebrosidase activity is known to be affected by two polymorphic alleles found at nucleotides 502 and 1637 of the cDNA for this gene. This enzyme activity and the polymorphic alleles noted above were analysed in 83 random samples of umbilical cord blood. The activity, assayed with the fluorogenic substrate 6-hexadecanoylamino-4-methylumbelliferyl-beta-galactopyranoside, in those with neither polymorphic allele was 4.6 +/- 1.7 units (nmol/h per mg protein). This optimal choice of cord blood was found in only 24% of specimens. Homozygotes for 1637T > C with activity of only 1.5 +/- 0.4 units represented 16% of the samples. Those heterozygous for 1637T > C with slightly better activity (2.3 +/- 0.7 units) represented 52% of the samples. Choice of umbilical cord blood for haematopoietic stem cell transplantation, therefore, requires consideration not only of cell quantity and HLA compatibility but also selection for normal alleles to obtain maximal enzymatic activity for central nervous system correction.

Elevated circulating levels of matrix metalloproteinase-9 and -2 in patients with symptomatic coronary artery disease.

BACKGROUND: Matrix metalloproteinases (MMP-9 and MMP-2) have been implicated in development of atherosclerosis and plaque rupture in acute coronary syndromes (ACS). AIM: To determine the relationship between circulating MMPs and symptomatic coronary artery disease. METHODS: Plasma levels of MMP-9 and MMP-2 were measured in patients with ACS, stable angina (SA) and in controls, using a quantitative gelatin zymography. These measurements were correlated with markers of systemic inflammation (hs-CRP) in all subjects and myocardial injury (troponin T) in patients with ACS. RESULTS: Plasma MMP-9 in ACS was greater than in SA, and was greater in SA than in controls (P < 0.01 ACS vs SA and controls, P < 0.01 SA vs control). Plasma MMP-2 was significantly greater in ACS than SA or controls (P < 0.01 vs SA and controls). There was strong overall relationship between hs-CRP and MMP-9 (r = 0.65, P < 0.0001) driven by a significant relationship in ACS patients (r = 0.58, P = 0.02), as there was no significant association in SA or controls. A weaker overall correlation was found between hs-CRP and MMP-2 (r = 0.39, P = 0.02), but no significant relationship was present for either of the two patient subgroups or controls. There was no correlation between levels of troponin T and MMP-9, MMP-2 or hs-CRP in ACS patients. CONCLUSION: Quantitative gelatin zymography identifies increased circulating levels of MMP-9 and MMP-2 in patients with symptomatic coronary disease. MMP-9 and MMP-2 are higher in ACS than SA patients and might have use as markers of plaque rupture or instability. The strong relationship between MMP-9 and hs-CRP in ACS patients suggests MMP-9 might be an additional marker and/or consequence of the inflammatory component in ACS.

Therapeutic effects of astrocytes expressing both tyrosine hydroxylase and brain-derived neurotrophic factor on a rat model of Parkinson's disease.

Tyrosine hydroxylase (TH) and brain-derived neurotrophic factor (BDNF), expressed in normal astrocytes, were used in combination for the treatment of Parkinson's disease (PD) symptoms in a rat model. Normal neonatal rat astrocytes were co-transfected with a vector expressing BDNF (AAVBDNF) and a retroviral vector expressing TH (termed TH-BDNF-DA(+) cells), and then implanted into the striatum of PD rats induced by 6-hydroxydopamine. TH-BDNF-DA(+) cells compensated for a severe insufficiency of endogenous dopaminergic neurons in the PD rats, resulting in a significant improvement of PD symptoms. The decrease in the rotational rate of PD rats implanted with TH-BDNF-DA(+) cells was more marked than that in PD rats implanted with normal astrocytes expressing either TH or BDNF alone (termed TH(+) and BDNF(+) cells, P<0.01 and 0.001, respectively), and suggested a synergistic effect between TH and BDNF. In contrast, the rotational rate was not altered from the baseline in PD rats without treatment or implanted with parental rat astrocytes alone (P>0.05). BDNF protected the dopaminergic neurons from apoptosis induced by 6-hydroxydopamine, and significantly increased the long-term survival of TH-positive cells in the striatum. Our data indicate that the combined use of TH and BDNF has a synergistic therapeutic effect, and is more efficient for the treatment of PD than a single gene therapy using either TH or BDNF alone.

Voltammetric behavior of L-cysteine in the presence of CPB at a silver electrode.

The voltammetric behavior of L-cysteine at a silver electrode is described. L-Cysteine can be anodically accumulated at a silver electrode surface as a sparingly soluble silver salt; at more negative potentials, the insoluble compound is stripped cathodically yielding a small current peak at about -1.10 V (vs. SCE). In the presence of cetyl pyridine bromide (CPB), the stripping peak shifts slightly to a more negative potential, and the peak height increases significantly. Thus, the peak becomes more useful for the determination of L-cysteine. In contrast to other surfactants, CPB can improve the accumulation and stripping of L-cysteine obviously. The voltammetric behavior of cysteamine, 3-mercaptopropionic acid and homocysteine is discussed as well.

Rapid detection of the two common mutations in Ashkenazi Jewish patients with mucolipidosis type IV.

Among Ashkenazi Jewish individuals with mucolipidosis IV (ML IV), two mutations in the ML IV gene, IVS3-1A --> G and delEX1-EX7, account for more than 95% of disease alleles. The reported method of genotyping for the delEX1-EX7 mutation involves a cumbersome multistep procedure. In the present study, a new simplified one-step procedure is described that detects this mutation in both patients and carriers. An improved procedure is also described for detection of the IVS3-1A --> G mutation. Using these improved procedures, we have characterized the ML IV mutant alleles in 27 patients and 95 of their relatives from 22 families, and in 123 unrelated and unaffected Ashkenazi Jewish controls. Of the 27 ML IV patients, 16 patients (59.3%) were found to be homozygous for the IVS3-1A --> G mutation and 1 patient (3.7%) homozygous for the delEX1-EX7 mutation. Additionally, 9 patients (33.3%) were compound heterozygotes for IVS3-1A --> G/delEX1-EX7. Among the 123 Ashkenazi Jewish controls, two individuals were identified as heteroallelic with one IVS3-1A --> G mutation (carrier frequency: approximately 1 in 61); none showed the delEX1-EX7 mutation. The modifications described here provide a more facile means of genotyping patients and carriers and expand the possibilities for screening at-risk populations.

Electrochemical characteristics of thin nickel hexacyanoferrate films formed on gold and thiol self-assembled monolayers modified gold electrodes.

Nickel hexacyanoferrate (NiHCF) film was prepared and characterized on gold and thiol self-assembled monolayers (SAMs)-modified gold electrodes. It was found that the film exhibited some different electrochemical characteristics compared with that found on a carbon electrode. In the presence of K+, the film exhibited a redox peak at about 0.5 V. The peak potential shifted linearly with the K+ concentration over the range of about 0.1 mM - 0.1 M with slopes of 54 - 60 mV per log[K+]. However, in solutions containing Na+, Li+ or NH4+ ion the film did not generate well-defined peaks, or even a visible redox peak. Therefore, the film showed a selective potential response to K+. The voltammetric behavior of NiHCF film varied with thiols, the preparation procedure and the solution pH. Under certain conditions, the characteristics of the film could be improved to some extent.

[Characteristic analysis of modulation transfer function curve of retina-brain system].

This study was aimed to find some parameters for characterizing the modulation transfer function (MTF) specific to diagnosing eye diseases. Least-square regression method was used for MTF curve (at logarithmic axis) fitting with quadratic equation so as or cubic equation to obtain the desired parameters. The data of normal and some eye diseases were fitted by quadratic and cubic equations. Three or four parameters were obtained from the curve fitting. The fitting curves were also expressed with figures along with the original data. For symmetric MTF, parameters k1, k2 and k3 were used to characterize the eye diseases. For asymmetric MTF, an additional parameter k4 was used to show the different injures of frequency channels.

Isolation and characterization of the normal canine beta-galactosidase gene and its mutation in a dog model of GM1-gangliosidosis.

The acid beta-galactosidase cDNA of Portuguese Water dogs was isolated and sequenced. The entire coding region of the gene consists of 2004 nucleotides encoding a protein of 668 amino acids. Its encoding sequence indicates approximately 86.5% identity at the nucleotide level and about 81% identity at the amino acid level with the encoding region of the human acid beta-galactosidase gene. The deduced amino acid sequence contains a 24-amino-acid putative signal sequence, six possible glycosylation sites, and seven cysteine residues. A homozygous recessive mutation, causing canine GM1-gangliosidosis, was identified at nucleotide G200-->A in exon 2 resulting in an Arg60-->His (mutation R60H) amino acid substitution. The mutation creates a new restriction enzyme site for Pml1. Genotyping 115 dog samples for this acid beta-galactosidase gene alteration readily distinguished affected homozygous recessives (n=5), heterozygous carriers (n=50) and normal homozygotes (n=60). DNA mutation analysis provided a method more specific than enzyme assay of beta-galactosidase for determination of carriers.

Quality control of postoperative acute pain service.

To establish an effective method of continuous quality control of acute pain service, a retrospective study on incident reporting during postoperative analgesia period was conducted. Incidents were reported and analyzed in 1507 patients who received epidural postoperative analgesia, and the results of satisfaction of pain relief was compared with those of incident analysis. In this study, an incident was defined as any factor that might or had affected patient's safety during analgesia period. Our results showed that 1203 incidents were reported in 641 of 1507 patients, of which 122 incidents were critical. 78.3% of all incidents were detected by acute pain service stuff. The most common incidents included complications, insufficient analgesia and problems with delivery circuits. Human factors were involved in 28.9% of the incidents, most being associated with technical failure due to unskillfulness, poor communications between APS stuff and patients and lack of cooperation with surgeons and nurses. The general satisfaction rate of the patients was 90.8%. There was a very significant difference between the satisfaction of the patients who suffered from incidents and who did not (P < 0.001). It is concluded that incidents affect the satisfaction of the patients who received postoperative pain relief. Incident reporting is a more effective method for quality control of acute pain service.

In vivo and in vitro glioma cell killing induced by an adenovirus expressing both cytosine deaminase and thymidine kinase and its association with interferon-alpha.

An adenovirus, AdCDTK, expressing both bacterial cytosine deaminase (CD) and herpes simplex virus thymidine kinase (HSVTK) was constructed and introduced into glioma cells. AdCDTK selectively rendered glioma cells sensitive to both 5-fluorocytosine (5-FCyt) and ganciclovir (GCV) (termed AdCDTK/5-FCyt-GCV). AdCDTK/5-FCyt-GCV not only potently mediated apoptosis and the arrest of glioma cell growth in vitro, but also significantly increased the survival time of glioma-bearing rats as compared with controls. The 90-day survival time was observed in 50% of rats. Interferon-alpha (IFN-alpha) further enhanced the tumor cell killing of AdCDTK/5-FCyt-GCV. In the group of AdCDTK/5-FCyt-GCV/IFN-alpha, the average survival time was significantly increased, and the average tumor size was smaller than that in the group of AdCDTK/5-FCyt-GCV. Ninety-day survival increased from 50% in the group of AdCDTK/5-FCyt-GCV to 75% in the group of AdCDTK/5-FCyt-GCV/IFN-alpha. Complete tumor regression was observed in 50% of rats in the group of AdCDTK/5-FCyt-GCV/IFN-alpha. The data indicate that AdCDTK/5-FCyt-GCV induces glioma cell killing greater than that induced by either CD/5-FCyt or HSVTK/GCV alone. IFN-alpha synergistically enhances this effect by increasing apoptosis.

Chicken FK506-binding protein, FKBP65, a member of the FKBP family of peptidylprolyl cis-trans isomerases, is only partially inhibited by FK506.

The chicken FK506-binding protein FKBP65, a peptidylprolyl cis-trans isomerase, is a rough endoplasmic reticulum protein that contains four domains homologous to FKBP13, another rough endoplasmic reticulum PPIase. Analytical ultracentrifugation suggests that in FKBP65 these four domains are arranged in a linear extended structure with a length of about 26 nm and a diameter of about 3 nm. All four domains are therefore expected to be accessible to substrates. The specificity of FKBP65 towards a number of peptide substrates was determined. The specific activity of FKBP65 is generally lower than that of FKBP12 when expressed as a per domain activity. The substrate specificity of FKBP65 also differs from that of FKBP12. Inhibition studies show that only one of the four domains can be inhibited by FK506, a powerful inhibitor of all other known FKBPs. Furthermore, the same domain seems to be susceptible to inhibition by cyclosporin A. No other FKBPs were shown to be inhibited by cyclosporin A. It is also shown that FKBP65 can catalyse the re-folding of type III collagen in vitro with a kcat/Km = 4.3 x 10(3) M-1.s-1.

Multiple respiratory gas monitoring causes changes of inspired oxygen concentration in closed anesthesia system.

Effect of multiple respiratory gas monitoring (MRGM) on inspired concentration of oxygen in circuit system during closed anesthesia was studied in 51 adult patients scheduled for abdominal surgery. Required flow rate of fresh oxygen (OFR), inspired oxygen concentration (FiO2) and oxygen saturation of pulse oximeter (SpO2) were measured continuously. Patients were equally divided into three groups at random, group C (no MRGN used), group M1 (using MRGM with its tail gas returned to circuit system), group M2 (using MRGM without tail gas returned). The results revealed that during 180 min of closed anesthesia, OFR required in group C and M1 were about 200-230 ml/min, and in group M2 it was about 400 ml/min. In group C FiO2 decreased by about 10% after 60 min of closed anesthesia (P < 0.01, 60 min vs 0 min) and then stayed stable at this level. In group M1, FiO2 decreased by 16% at 60 min and 34% at 180 min and the decrease was significantly greater than that in group C (P < 0.01). In group M2, FiO2 remained constant during closed anesthesia, which was much higher than those in group C and M1. The tail gas of Capnomac Ultima MRGM contained less oxygen than its sample gas drawn from circuit system simultaneously.

[Description of finger shape in the process of growing during infancy].

In this paper, a new method of estimating bone age is reported that is based on the use of spatial description parameters and fourier frequential descriptors, which presents the boundary shapes detected in finger bones X-ray films of 0-, 3-, 6-, 9-month baby according to their bone shape changes in different stages.
