[Apoptosis of K562 cells induced by nociceptin/orphanin FQ].

The study was to investigate the proliferation and apoptosis effect of nociceptin/orphanin FQ (OFQ) on 562 cells in vitro. Inhibition of K562 cells proliferation was measured by MTT assay. Morphological assessment of apoptosis was performed with Wright staining and transmission electron microscope. The apoptosis peak was measured by flow cytometry. DNA fragmentation was visualized by agarose gel electrophoresis. The results showed that OFQ time-dependently and no-dose-dependently inhibited the proliferation of K562 cells at concentrations of 10(-6) - 10(-13) mol/L. Discrete maximum of cytolytic activity was detected at concentrations of 10(-6) - 10(-7), 10(-9), 10(-12) mol/L. Compared with the control group K562 cells, the cells treated with OFQ at concentration of 10(-9) mol/L for 72 hours showed typical characteristics of apoptosis under transmission electron microscope. Apoptosis peak was found by FCM at concentration of 0, 10(-7), 10(-8), 10(-9) mol/L of OFQ for 72 hours, apoptosis rates were 0%, 22.8%, 23.8% and 26.5% respectively. DNA agarose gel electrophoresis revealed nuclear fragmentation (DNA ladder). It is concluded that OFQ can inhibit the proliferation of K562 cells and induce the apoptosis in K562 cells.

Ethanol reduces p38 kinase activation and cyclin D1 protein expression after partial hepatectomy in rats.

BACKGROUND/AIMS: Chronic ethanol consumption inhibits liver regeneration. We examined the effects of chronic ethanol consumption on two mitogen-activated protein kinases in relation to induction of cell cycle proteins after partial hepatectomy (PH). METHODS: Male Wistar rats were ethanol-fed (EF) or pair-fed (PF) for 16 weeks before PH. Hepatic activation of extracellular signal regulated kinase (ERK)1/2, p38 kinase and expression of cyclinD1, cyclin-dependent kinase-4 (cdk4) and proliferating cell nuclear antigen (PCNA) were studied. RESULTS: In PF rats, PH-induced p38 activation was evident at 2h and was maximal at 12h. There was a close temporal relationship between p38 activation, cyclin D1 and PCNA expression. Alcohol exposure reduced p38 activation, cyclin D1 and PCNA, each by approximately 50%. ERK1/2 activation occurred during the first 2h post-PH in both EF and PF rats, and there was no later increase in PF rats. In vivo inhibition of p38 suppressed PCNA expression whereas the effect of ERK1/2 inhibition was inconsistent. CONCLUSIONS: p38 kinase activation is linked temporally with cyclin D1 expression after PH and appears to exert cell cycle control in the adult liver. p38 signaling also appears to be a target for the inhibitory effect of chronic alcohol on liver regeneration.

Measuring gene flow in the cultivation of transgenic cotton (Gossypium hirsutum L.).

Transgenic Bt cotton NewCott 33B and transgenic tfd A cotton TFD were chosen to evaluate pollen dispersal frequency and distance of transgenic cotton (Gossypium hirsutum L.) in the Huanghe Valley Cotton-producing Zone, China. The objective was to evaluate the efficacy of biosafety procedures used to reduce pollen movement. A field test plot of transgenic cotton (6 x 6 m) was planted in the middle of a nontransgenic field measuring 210 x 210 m. The results indicated that the pollen of Bt cotton or tfd A cotton could be dispersed into the environment. Out-crossing was highest within the central test plot where progeny from nontransgenic plants, immediately adjacent to transgenic plants, had resistant plant progeny at frequencies up to 10.48%. Dispersal frequency decreased significantly and exponentially as dispersal distance increased. The flow frequency and distance of tfd A and Bt genes were similar, but the pollen-mediated gene flow of tfd A cotton was higher and further to the transgenic block than that of Bt cotton (chi2 = 11.712, 1 degree of freedom, p < 0.001). For the tfd A gene, out-crossing ranged from 10.13% at 1 m to 0.04% at 50 m from the transgenic plants. For the Bt gene, out-crossing ranged from 8.16% at 1 m to 0.08% at 20 m from the transgenic plants. These data were fit to a power curve model: y = 10.1321x-1.4133 with a correlation coefficient of 0.999, and y = 8.0031x-1.483 with a correlation coefficient of 0.998, respectively. In this experiment, the farthest distance of pollen dispersal from transgenic cotton was 50 m. These results indicate that a 60-m buffer zone would serve to limit dispersal of transgenic pollen from small-scale field tests.

A Konowledge-based Protein Modeling System and Its Evaluation III. A Sensitive Method for Evaluation Protein Model.

A new scoring table of compatibilities of residues with environments in a protein has been derived from environment-dependent amino acid substitution tables. The environments of the residues in the 3D structure are described by: (1) the secondary structure; (2) the solvent accessibility; (3) the hydrogen bonding. On the basis of the scoring table, an assessment method of protein models is proposed. Compared with the famous software Profile-3D, The method not only shows a more effective test of the accuracy of a 3D protein model, but also exhibits a higher sensitivity to the local misfolding. It is an important module in our protein modeling system PMODELING.
