An efficient rhythmic component expression and weighting synthesis strategy for classifying motor imagery EEG in a brain-computer interface.

The recognition of mental states during motor imagery tasks is crucial for EEG-based brain-computer interface research. We have developed a new algorithm by means of frequency decomposition and weighting synthesis strategy for recognizing imagined right- and left-hand movements. A frequency range from 5 to 25 Hz was divided into 20 band bins for each trial, and the corresponding envelopes of filtered EEG signals for each trial were extracted as a measure of instantaneous power at each frequency band. The dimensionality of the feature space was reduced from 200 (corresponding to 2 s) to 3 by down-sampling of envelopes of the feature signals, and subsequently applying principal component analysis. The linear discriminate analysis algorithm was then used to classify the features, due to its generalization capability. Each frequency band bin was weighted by a function determined according to the classification accuracy during the training process. The present classification algorithm was applied to a dataset of nine human subjects, and achieved a success rate of classification of 90% in training and 77% in testing. The present promising results suggest that the present classification algorithm can be used in initiating a general-purpose mental state recognition based on motor imagery tasks.

GADD34-PP1c recruited by Smad7 dephosphorylates TGFbeta type I receptor.

The cascade of phosphorylation is a pivotal event in transforming growth factor beta (TGFbeta) signaling. Reversible phosphorylation regulates fundamental aspects of cell activity. TGFbeta-induced Smad7 binds to type I receptor (TGFbeta type I receptor; TbetaRI) functioning as a receptor kinase antagonist. We found Smad7 interacts with growth arrest and DNA damage protein, GADD34, a regulatory subunit of the protein phosphatase 1 (PP1) holoenzyme, which subsequently recruits catalytic subunit of PP1 (PP1c) to dephosphorylate TbetaRI. Blocking Smad7 expression by RNA interference inhibits association of GADD34-PP1c complex with TbetaRI, indicating Smad7 acts as an adaptor protein in the formation of the PP1 holoenzyme that targets TbetaRI for dephosphorylation. SARA (Smad anchor for receptor activation) enhances the recruitment PP1c to the Smad7-GADD34 complex by controlling the specific subcellular localization of PP1c. Importantly, GADD34-PP1c recruited by Smad7 inhibits TGFbeta-induced cell cycle arrest and mediates TGFbeta resistance in responding to UV light irradiation. The dephosphorylation of TbetaRI mediated by Smad7 is an effective mechanism for governing negative feedback in TGFbeta signaling.

Synthesis and cell affinity of functionalized poly(L-lactide-co-beta-malic acid) with high molecular weight.

A novel functionalized biodegradable poly(L-lactide-co-beta-benzyl malolactonate) (p-PLMA) with high molecular weight was synthesized through ring-opening copolymerization. Three p-PLMA copolymers with different beta-benzyl malolactonate content were synthesized. The molecular weight (M(w)) and tensile strength of the copolymer with 4 mol% beta-benzyl malolactonate content were 179,800 and 19.0MPa respectively, the molecular weight (M(w)) and tensile strength of p-PLMA decreased with beta-benzyl malolactonate content increasing. The hydrophilicity of the de-protected product: poly(L-lactide-co-beta-malic acid) (d-PLMA) increased with malic acid content increasing. The results of 3T3 mice fibroblasts cultivated on d-PLMA films showed that the cell adhesion on d-PLMA was better than that of PLLA and the cell attached efficiency of d-PLMA with 8 mol% malic acid content was the highest. The cells grew well both on the surface and inside of d-PLMA scaffolds. The cell affinity of d-PLMA was better than that of PLLA.

Classifying EEG-based motor imagery tasks by means of time-frequency synthesized spatial patterns.

OBJECTIVE: To develop a single trial motor imagery (MI) classification strategy for the brain-computer interface (BCI) applications by using time-frequency synthesis approach to accommodate the individual difference, and using the spatial patterns derived from electroencephalogram (EEG) rhythmic components as the feature description. METHODS: The EEGs are decomposed into a series of frequency bands, and the instantaneous power is represented by the envelop of oscillatory activity, which forms the spatial patterns for a given electrode montage at a time-frequency grid. Time-frequency weights determined by training process are used to synthesize the contributions from the time-frequency domains. RESULTS: The present method was tested in nine human subjects performing left or right hand movement imagery tasks. The overall classification accuracies for nine human subjects were about 80% in the 10-fold cross-validation, without rejecting any trials from the dataset. The loci of MI activity were shown in the spatial topography of differential-mode patterns over the sensorimotor area. CONCLUSIONS: The present method does not contain a priori subject-dependent parameters, and is computationally efficient. The testing results are promising considering the fact that no trials are excluded due to noise or artifact. SIGNIFICANCE: The present method promises to provide a useful alternative as a general purpose classification procedure for MI classification.

Speciation of heavy metals in marine sediments from the East China Sea by ICP-MS with sequential extraction.

Twelve elements (V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Mo, Sn, Cd and Pb) in 24 sediment samples at eight sites (S1-S8) from the East China Sea were analyzed with the BCR sequential extraction (SE) protocol to obtain the metal distribution patterns in this region. The results showed that the heavy metal pollutions in S4 and S8 were more severe than in other sampling sites, especially Cd and Pb pollution. In the top sediments at S4 and S8, both the total contents and the most dangerous non-residual fractions of Cd and Pb were extremely high. More than 90% of the total concentrations of V, Cr, Mo and Sn existed in the residual fraction. Fe, Co, Ni, Cu and Zn mainly (more than 60%) occurred in the residual fraction. While Mn, Pb and Cd dominantly presented in the non-residual fractions in the top sediments. The metal distribution patterns with depth and the correlations between total organic carbon (TOC) and the total Fe-Mn content were also investigated. The results showed that, for most of the elements except Fe, the concentration of elements in fraction A in the top sediments was higher than that in other depth. The similar rule was also found in fraction B but not in fraction C. Besides, the distributions of V, Cd in fraction B and Pb, Cd, Cu in fraction C might be affected by TOC.

Estimation of the effective and functional human cortical connectivity with structural equation modeling and directed transfer function applied to high-resolution EEG.

Different brain imaging devices are presently available to provide images of the human functional cortical activity, based on hemodynamic, metabolic or electromagnetic measurements. However, static images of brain regions activated during particular tasks do not convey the information of how these regions are interconnected. The concept of brain connectivity plays a central role in the neuroscience, and different definitions of connectivity, functional and effective, have been adopted in literature. While the functional connectivity is defined as the temporal coherence among the activities of different brain areas, the effective connectivity is defined as the simplest brain circuit that would produce the same temporal relationship as observed experimentally among cortical sites. The structural equation modeling (SEM) is the most used method to estimate effective connectivity in neuroscience, and its typical application is on data related to brain hemodynamic behavior tested by functional magnetic resonance imaging (fMRI), whereas the directed transfer function (DTF) method is a frequency-domain approach based on both a multivariate autoregressive (MVAR) modeling of time series and on the concept of Granger causality. This study presents advanced methods for the estimation of cortical connectivity by applying SEM and DTF on the cortical signals estimated from high-resolution electroencephalography (EEG) recordings, since these signals exhibit a higher spatial resolution than conventional cerebral electromagnetic measures. To estimate correctly the cortical signals, we used a subject's multicompartment head model (scalp, skull, dura mater, cortex) constructed from individual MRI, a distributed source model and a regularized linear inverse source estimates of cortical current density. Before the application of SEM and DTF methodology to the cortical waveforms estimated from high-resolution EEG data, we performed a simulation study, in which different main factors (signal-to-noise ratio, SNR, and simulated cortical activity duration, LENGTH) were systematically manipulated in the generation of test signals, and the errors in the estimated connectivity were evaluated by the analysis of variance (ANOVA). The statistical analysis returned that during simulations, both SEM and DTF estimators were able to correctly estimate the imposed connectivity patterns under reasonable operative conditions, that is, when data exhibit an SNR of at least 3 and a LENGTH of at least 75 s of nonconsecutive EEG recordings at 64 Hz of sampling rate. Hence, effective and functional connectivity patterns of cortical activity can be effectively estimated under general conditions met in any practical EEG recordings, by combining high-resolution EEG techniques and linear inverse estimation with SEM or DTF methods. We conclude that the estimation of cortical connectivity can be performed not only with hemodynamic measurements, but also with EEG signals treated with advanced computational techniques.

Spatio-temporal cortical source imaging of brain electrical activity by means of time-varying parametric projection filter.

In the present study, we explore suitable spatio-temporal filters for inverse estimation of an equivalent dipole-layer distribution from the scalp electroencephalogram (EEG) for imaging of brain electric sources. We propose a time-varying parametric projection filter (tPPF) for the spatio-temporal EEG analysis. The performance of this tPPF algorithm was evaluated by computer simulation studies. An inhomogeneous three-concentric-spheres model was used in the present simulation study to represent the head volume conductor. An equivalent dipole layer was used to represent equivalently brain electric sources and estimated from the scalp potentials. The tPPF filter was tested to remove time-varying noise such as instantaneous artifacts caused by eyes-blink. The present simulation results indicate that the proposed time-variant tPPF method provides enhanced performance in rejecting time-varying noise, as compared with the time-invariant parametric projection filter.

Replication of herpes simplex virus 1 depends on the gamma 134.5 functions that facilitate virus response to interferon and egress in the different stages of productive infection.

The ability of the gamma(1)34.5 protein to suppress the PKR response plays a crucial role in herpes simplex virus pathogenesis. In this process, the gamma(1)34.5 protein associates with protein phosphatase 1 to form a large complex that dephosphorylates eIF-2alpha and thereby prevents translation shutoff mediated by PKR. Accordingly, gamma(1)34.5 null mutants are virulent in PKR-knockout mice but not in wild-type mice. However, gamma(1)34.5 deletion mutants, with an extragenic compensatory mutation, inhibit PKR activity but remain avirulent, suggesting that the gamma(1)34.5 protein has additional functions. Here, we show that a substitution of the gamma(1)34.5 gene with the NS1 gene from influenza A virus renders viral resistance to interferon involving PKR. The virus replicates as efficiently as wild-type virus in SK-N-SH and CV-1 cells. However, in mouse 3T6 cells, the virus expressing the NS1 protein grows at an intermediate level between the wild-type virus and the gamma(1)34.5 deletion mutant. This decrease in growth, compared to that of the wild-type virus, is due not to an inhibition of viral protein synthesis but rather to a block in virus release or egress. Virus particles are predominantly present in the nucleus and cytoplasm. Notably, deletions in the amino terminus of the gamma(1)34.5 protein lead to a significant decrease in virus growth in mouse 3T6 cells, which is independent of eIF-2alpha dephosphorylation. In correlation, a series of deletions in the amino-terminal domain impair nuclear as well as cytoplasmic egress. These results indicate that efficient viral replication depends on the gamma(1)34.5 functions required to prevent the PKR response and to facilitate virus egress in the different stages during virus infection.

Motor imagery task classification for brain computer interface applications using spatiotemporal principle component analysis.

Classification of single-trial imagined left- and right-hand movements recorded through scalp EEG are explored in this study. Classical event-related desynchronization/synchronization (ERD/ERS) calculation approach was utilized to extract ERD features from the raw scalp EEG signal. Principle Component Analysis (PCA) was used for feature extraction and applied on spatial, as well as temporal dimensions in two consecutive steps. A Support Vector Machine (SVM) classifier using a linear decision function was used to classify each trial as either left or right. The present approach has yielded good classification results and promises to have potential for further refinement for increased accuracy as well as application in online brain computer interface (BCI).

[Effect of herceptin combined with Doxorubicin on rat cardiotoxicity].

BACKGROUND & OBJECTIVE: Herceptin is a humanized monoclonal antibody for treating the patients with metastatic breast cancers overexpressing human epidermal growth factor receptor (HER-2). Herceptin combined with doxorubicin markedly increased the incidence of cardiac dysfunction in clinical trials. The current study was designed to investigate the cardiotoxicity aggravated by Herceptin combined with doxorubicin in rats. METHODS: Thirty-six female rats were randomized into four groups (n=9): control group, Herceptin group, doxorubicin group, Herceptin combined with doxorubicin group. The value of malon dialdehyde (MDA) was measured by thiobarbituric acid (TBA) method and the activity of glutathione peroxidase (GSH-Px) was measured by 5,5'-dithiobis-2-nitrobenzoic acid (DTNB) method. The damage and apoptosis of myocardium were observed by electron microscopy and tdt-mediated dUTP nick end labeling (TUNEL). The ratio of myocardial apoptosis cells was quantified using flow cytometry. RESULTS: The value of MDA in control group and Herceptin group were (0.70+/-0.03) and (0.73+/-0.05) nmol x(mg x protein)(-1), respectively; the activity of GSH-PX in control group and Herceptin group were (81.31+/-0.13) and (78.43+/-0.15) U x(mg x protein)(-1),respectively;the two groups were similar. The value of MDA in doxorubicin group and Herceptin combined with doxorubicin group were (0.88+/-0.10) and (0.94+/-0.13) nmol x(mg x protein)(-1), respectively; the activity of GSH-PX in doxorubicin group and Herceptin combined with doxorubicin group were(67.88+/-0.24) and(63.37+/-0.28) U x(mg x protein)(-1),respectively. The most prominent ultrastructural damages were mitochondrial swelling with fragmentation of cristae and vacuoles formation in doxorubicin group and Herceptin combined with doxorubicin group. Herceptin markedly increased the ratio of apoptosis of myocardial cells from (5.35+/-0.27) % in doxorubicin group to (8.27+/-0.38) % in Herceptin combined with doxorubicin group as measured by flow cytometry (P< 0.05). CONCLUSION: Herceptin alone had little toxic effects on rat cardiomocytes, but Herceptin combined with doxorubicin can aggravate rat cardiotoxicity. Herceptin can also increase the number of the apoptosis cell in rat myocardium.

[Studies on random amplified polymorphic DNA fingerprinting of Cortex Magnoliae Officinalis].

OBJECTIVE: To identify Chinese traditional medicine (CTM) "Hou-pu" (Cortex Magnoliae Officinalis), its counterfeits and substitues. METHODS: Total genomic DNA samples of ten plant species were amplified by randomly amplified polymorphic DNA (RAPD). RESULTS: Ten samples were able to be distinguished through their amplified DNA banding patterns on the agarose gels after electrophoresis. CONCLUSION: RAPD is able to identify "Hou-pu", its counterfeits and substitutes quickly and truly, which is also quite valuable for correctly introducing plant.

Skeletal abnormalities in Pth-null mice are influenced by dietary calcium.

We have examined the role of PTH in the postnatal state in a mouse model of PTH deficiency generated by targeting the Pth gene in embryonic stem cells. Mice homozygous for the ablated allele, when maintained on a normal calcium intake, developed hypocalcemia, hyperphosphatemia, and low circulating 1,25-dihydroxyvitamin D(3) [1,25(OH)(2)D(3)] levels consistent with primary hypoparathyroidism. Bone turnover was reduced, leading to increased trabecular and cortical bone volume in PTH-deficient mice. When mutant mice were placed on a low-calcium diet, renal 25-hydroxyvitamin D 1 alpha-hydroxylase expression increased despite the absence of PTH, leading to a rise in circulating 1,25(OH)(2)D(3) levels, marked osteoclastogenesis, and profound bone resorption. These studies demonstrate the dependence of the skeletal phenotype in animals with genetically depleted PTH on the external environment as well as on internal hormonal and ionic circulatory factors. They also show that, although PTH action is the first defense against hypocalcemia, 1,25(OH)(2)D(3) can be mobilized, even in the absence of PTH, to guard against extreme calcium deficiency.

Epidermal growth factor increases coactivation of the androgen receptor in recurrent prostate cancer.

Growth of normal and neoplastic prostate is mediated by the androgen receptor (AR), a ligand-dependent transcription factor activated by high affinity androgen binding. The AR is highly expressed in recurrent prostate cancer cells that proliferate despite reduced circulating androgen. In this report, we show that epidermal growth factor (EGF) increases androgen-dependent AR transactivation in the recurrent prostate cancer cell line CWR-R1 through a mechanism that involves a post-transcriptional increase in the p160 coactivator transcriptional intermediary factor 2/glucocorticoid receptor interacting protein 1 (TIF2/GRIP1). Site-specific mutagenesis and selective MAPK inhibitors linked the EGF-induced increase in AR transactivation to phosphorylation of TIF2/GRIP1. EGF signaling increased the coimmunoprecipitation of TIF2 and AR. AR transactivation and its stimulation by EGF were reduced by small interfering RNA inhibition of TIF2/GRIP1 expression. The data indicate that EGF signaling through MAPK increases TIF2/GRIP1 coactivation of AR transactivation in recurrent prostate cancer.

Investigation of methylmercury and total mercury contamination in mollusk samples collected from coastal sites along the Chinese Bohai Sea.

This paper presents the investigation results of methylmercury and total mercury in gastropod and bivalve species collected from eight coastal sites along the Chinese Bohai Sea. The total mercury was directly determined by atomic fluorescence spectrometry (AFS), while the methylmercury was measured by a laboratory established high performance liquid chromatography-atomic fluorescence spectrometry system (HPLC-AFS). Certified reference material DORM-2 (Dogfish muscle) was used to validate the two methods and the obtained results proved to be in good agreement with the certified values. It was demonstrated that the mercury contamination was commonly existed in all selected mollusks, with methylmercury and total mercury concentration in the range of 4.8-168.4 and 6.7-453.0 ng Hg g(-)(1), respectively. Mollusks from HuLuDao were the most mercury contaminated, and those from PengLai took the second place. The species-dependent bioaccumulation capacity was observed in this study. Gastropods showed more capacity to bioaccumulate mercury than bivalves, and mercury contents in two kinds of gastropods (Rapana venosa and Neverita didyma) presented some uplifted trends with the dimensions increasing of the gastropods. Rapana venosa was found to be a potential biomarker to monitor mercury pollution in oceans. Evaluations were also made concerning about the ratio of methylmercury to total mercury.

Equivalent physical models and formulation of equivalent source layer in high-resolution EEG imaging.

In high-resolution EEG imaging, both equivalent dipole layer (EDL) and equivalent charge layer (ECL) assumed to be located just above the cortical surface have been proposed as high-resolution imaging modalities or as intermediate steps to estimate the epicortical potential. Presented here are the equivalent physical models of these two equivalent source layers (ESL) which show that the strength of EDL is proportional to the surface potential of the layer when the outside of the layer is filled with an insulator, and that the strength of ECL is the normal current of the layer when the outside is filled with a perfect conductor. Based on these equivalent physical models, closed solutions of ECL and EDL corresponding to a dipole enclosed by a spherical layer are given. These results provide the theoretical basis of ESL applications in high-resolution EEG mapping.

Octamethylcyclotetrasiloxane exhibits estrogenic activity in mice via ERalpha.

Octamethylcyclotetrasiloxane (D4) is a low molecular weight cyclic silicone used in the synthesis of larger silicone polymers and in the formulation of a variety of personal care products. The effects of oral D4 exposure in mice on serum estradiol levels, uterine wet weight, and uterine peroxidase activity were investigated. Additionally, in vitro estrogen receptor binding activity was evaluated. Serum estradiol levels decreased in a dose-dependent manner after exposure to 100 mg/kg to 1000 mg/kg D4. Studies with adrenalectomized animals demonstrated that the decreased serum estradiol levels were not due to elevated serum corticosterone levels. Uterine wet weights in ovariectomized mice were significantly increased in a dose-dependent manner by exposure to 250-1000 mg of D4/kg, but not by exposure to other silicone compounds tested (hexamethylcyclotrisiloxane, decamethylcyclopentasiloxane, decamethyltetrasiloxane, and octaphenylcyclotetrasiloxane). Uterine peroxidase activity, a marker for estrogenic activity, was also significantly increased in D4-exposed mice, but not in mice exposed to the other siloxanes. Pretreating mice with the estrogen receptor antagonist ICI 182,780 completely blocked the D4-induced increase in uterine weight, and ovariectomized estrogen receptor-alpha knockout mice showed no increases in uterine weights when orally exposed to D4 or estradiol. In an in vitro estrogen receptor binding assay, D4 showed significant competition with (3)H-estradiol for binding to estrogen receptor-alpha, but not estrogen receptor-beta. The data presented here indicate that D4 has weak estrogenic activity, and that these effects are mediated through estrogen receptor-alpha.

Genomic expression discovery predicts pathways and opposing functions behind phenotypes.

Discovering states of genetic expression that are true to a high degree of certainty is likely to predict gene function behind biological phenotypes. The states of expression (up- or down-regulated) of 19200 cDNAs in 10 meningiomas are compared with normal brain by an algorithm that detects only 1 false measurement per 192000; 364 genes are discovered. The expression data accurately predict activation of signaling pathways and link gene function to specific phenotypes. Meningiomas appear to acquire aberrant phenotypes by disturbing the balanced expression of molecules that promote opposing functions. The findings expose interconnected genes and propose a role of genomic expression discovery in functional genomics of living systems.

Amino acid substitutions in the effector domain of the gamma(1)34.5 protein of herpes simplex virus 1 have differential effects on viral response to interferon-alpha.

The gamma(1)34.5 protein of herpes simplex virus 1 (HSV-1) is a virus-encoded protein phosphatase 1 (PP1) regulatory protein that contributes to viral resistance to interferon. It functions to block the shutoff of protein synthesis mediated by the double-stranded RNA-dependent protein kinase. This requires the carboxyl terminus of the gamma(1)34.5 protein to recruit PP1, forming a high-molecular-weight complex that dephosphorylates the alpha subunit of translation initiation factor eIF-2 (eIF-2alpha). In the present study, we introduced a series of point mutations into a region in the effector domain of the gamma(1)34.5 protein, which is adjacent to the PP1-binding domain. Analysis of these mutants in virus-infected cells shows that Ser209Ala, Ser209Asp, Ser218Ala, or Trp219Tyr substitution does not affect viral response to interferon-alpha. In contrast, Arg215Leu or Ser218Asp substitution rendered the virus hypersensitive to interferon-alpha, which correlates with the inability of these gamma(1)34.5 mutants to mediate dephosphorylation of eIF-2alpha. However, Arg215Leu or Ser218Asp substitution does not disrupt the formation of a high-molecular-weight complex required for eIF-2alpha dephosphorylation or binding of the gamma(1)34.5 protein to PP1. These results suggest that concerted action of the PP1-binding domain and the effector domain of the gamma(1)34.5 protein is required to confer HSV-1 interferon resistance.
