[Pridiction of the Sex-effects of the Cocoon Quality Characters in Silkworm.].

In this paper , the sex-effects of the cocoon quality characters in silkworm was predicted with Mixed linear model úThe fact that the probability of effect variance and predictability of random gender of whole cocoon weight, cocoon shell weight, ratio of cocoon shell and pupa weight reached a level of extreme significance showed that the gender effect of the four traits was extremely significant, which matched with the reality completely. The predictive values of gender effect of the four traits of female(male) were 0.248g (-0.247g), 2.423cg(-2.394)cg, -1.976%(1.992%) and 0.224g(-0.223g) respectively. Each trait showed single peak distribution after adjusting by sex-effects, which fitted for the request that quantitative traits should show continuously normal standard distribution if QTL analysis was taken.

[Study on location of QTLs controlling cocoon traits in silkworm].

On the basis of the molecular linkage map, mapmaker software QTLMapper 2.0 was used to analyze the QTLs effect of the whole cocoon weight, cocoon shell weight, ratio of cocoon shell and pupa weight of domestic silkworm. For these four cocoon quantitative traits, 7, 6, 2 and 8 effective QTLs were detected and mapped to 7, 5, 2 and 7 linkage groups, respectively. Complicated epistatic effects were found involved in the genetic variation of the whole cocoon weight and cocoon shell weight. For the whole cocoon weight, there were three pairs of QTLs with significant additive by additive interactions, in which, one pair had significant additive by dominance and dominance by dominance interactions. Whereas significant dominance were detected for three QTLs and significant additive effects one QTL had. For the cocoon shell weight, significant genetic effects, including epistatic effects were found for one pair of QTLs, significant dominance by dominance interaction for another pair of QTLs; one QTL had significant dominance and another QTL had additive by additive interaction. The ratio of cocoon shell and the pupa weight were controlled mainly by additive or dominance effects. No interaction between QTL was found for the ratio of cocoon. Most QTLs, associated with the pupa weight, had negative dominance effects. Only significant additive by additive interaction was found between one pair of QTLs. The 2nd, 3rd, 4th, 11th, 13th, 24th, 34th, 37th, and 40th linkage groups are the common chromosomal regions harboring QTLs of two or more cocoon quantitative traits. There are identical QTL or chromosomal region for the whole cocoon weight and cocoon shell weight, indicating they can be simultaneously improved by utilizing epistatic effects in breeding.

[The significance of Th1/Th2 function imbalance in patients with post-infarction cardiac insufficiency].

OBJECTIVE: To study the significance of Th1/Th2 function imbalance in patients with post-infarction cardiac insufficiency. METHODS: Forty-three MI (myocardial infarction) patients were divided into 2 groups one month after the onset according to the New York Heart Association (NYHA) classification system: group MI 1 (I, II) 25 patients and group MI 2 (III, IV) 18 patients. At the same time, the heart function was evaluated by two-dimensional echocardiography. Peripheral blood mononuclear cells (PBMCs) were collected from these patients. Cytokine-producing CD4 + T cells were quantified by 3-color flow cytometry after being stimulated with phorbol myristate acetate (PMA) and ionomycin. After being stimulated with PHA, the levels of IFN-gamma and IL-4 in culture supernatants were measured by ELISA. RESULTS: The frequencies of IFN-gamma-producing T cells were found to be significantly higher in group MI 2 (16.8%) than that in group MI 1 (13.1%). There was no significant difference on the frequencies of IL-4-producing peripheral T cells between the two groups. The IFN-gamma level and the ratios of IFN-gamma/IL-4 in group MI 2 were significantly higher than those in group MI 1, while there was no significant difference in IL-4 levels between the two groups. CONCLUSIONS: The Th-cell function was associated with heart function in post MI patients. The up-regulation of Th1 cell function was consistent with poor heart function, suggesting that Th1/Th2 cell function imbalance may participate in ventricular remodelling after MI.

[The changes and their clinical significance of D-dimer and platelet glycoprotein in patients with coronary heart disease].

OBJECTIVE: To explore the changes and their clinical significance of D-dimer and platelet glycoprotein (GP) in patients with coronary heart disease. METHODS: D-dimer and GP in 20 patients with stable angina (SA group), 48 patients with unstable angina (UA group), and 20 control cases were measured. The changes of D-dimer and GP in patients with and without coronary events were compared. The sensitivity of those changes in the diagnosis of coronary events was evaluated. RESULTS: There were significant differences of D-dimer and GP between UA group and SA group or control group (P < 0.01), while there was no significant difference between SA group and control group (P > 0.05). There were also significant differences of D-dimer and GP between patients with coronary events and patients without coronary events (P < 0.05). In the sensitivity test for detecting coronary events, D-dimer and GPIIb, GPIIIa were much more sensitive than other parameters. CONCLUSIONS: D-dimer and GPIIb, GPIIIa may be regarded as the indexes of coronary thrombosis and used for predicting the severity of coronary events.

Effect of femoral nutrient artery ligature on radionuclide uptake in rabbits.

To elucidate the effect of damaging the nutrient artery to a long bone, we used radionuclide imaging to survey the change of femoral blood supply after ligating the nutrient artery in rabbits. The radionuclide uptake of the femoral shaft, upper and lower metaphysis, and epiphysis decreased significantly at 1 hr postoperation (0.66, 0.74, 0.81, respectively, p <0.05), were close to normal on day 4 (0.96, 0.98, 1.02, respectively, p >0.05), slightly exceeded the contralateral level at 9 and 12 days, and returned to normal on day 16. This study shows that ligation of the nutrient artery of a long bone leads to an immediate decrease in the bone blood flow. However, so long as anastomoses among other bone vessels are intact, the recovery rate is speedy, and long-term, serious disorders of the bone blood supply do not ensue.

[Clinical and pathological features of 112 cases with ocular adnexal lymphoproliferative lesions].

OBJECTIVE: To investigate the histopathological classification and clinical features of ocular adnexal lymphoproliferative lesions. METHODS: The clinical, histomorphological and immuno-histochemical features of 112 cases of ocular adnexal lymphoproliferative lesions (116 paraffin specimens) were studied retrospectively. The lesions were classified according to the World Health Organization classification of tumors of haematopoietic and lymphoid tissues (2001). RESULTS: This group of patients' ages averaged at 49. The mean duration between the onset of the symptoms and the time of presentation was 22 months. Sixteen patients (14.3%) had bilateral lesions. Proptosis or local orbital mass was presented in 69 cases (61.6%). Reactive lymphoid hyperplasia was diagnosed in 11 cases (9.8%) and atypical lymphoid hyperplasia in 10 cases (8.9%). The rest 91 cases (81.3%) were diagnosed as lymphoma, among which 74 cases (81.3%) were extranodal marginal zone B-cell lymphoma of mucosa-associated lymphoid tissue (MALT-EMZL). CONCLUSIONS: MALT-EMZL is the most frequent type of ocular lymphoproliferative lesions in ocular adnexa. The ocular adnexal lymphoproliferative lesions show an indolent course clinically, tending to affect the old. Bilateral case is not uncommon.

[In vitro study on inhibition of human choroidal melanoma OCM-1 cell line by arsenic trioxide].

OBJECTIVE: To investigate the effects and mechanisms of arsenic trioxide (ATO) on human choroidal melanoma cell line OCM-1. METHODS: OCM-1 cells were cultured with 0.75 to 24.00 micromol/L arsenic trioxide for various durations, then cell viability was measured by MTT assay. The cell necrosis and apoptosis rates were observed by flow cytometry. The morphological changes of the cells were examined by electron microscopy. Glutathione peroxidase (GSH-Px) activities were tested. Mitochondrial membrane potential (MMP) was detected by confocal microscopy. RESULTS: Growth of OCM-1 cells was inhibited by ATO at concentrations of (1.5 to 24.0) micromol/L. However, there was no effect of 0.75 micromol/L ATO on the growth of OCM-1 cells. The inhibition showed both dose and time dependent effects (P < 0.05). The IC(50) was 16.8 micromol/L at 24 h. Flow cytometry analysis showed a positive correlation between the rate of cell necrosis and apoptosis and the concentration of ATO. The cell necrosis rates were higher than the cell apoptosis rates at various concentrations of ATO. OCM-1 cells cultured with ATO showed the classic morphologic characteristics of necrosis and apoptosis. GSH-Px activities and MMP decreased in a dose dependent manner. CONCLUSION: ATO inhibits the growth of OCM-1 cells. The mechanism of this effect is that ATO inhibits the GSH-Px activities, decreases the MMP and impairs mitochondrial energy synthesis, which induces necrosis and apoptosis of human choroidal melanoma COM-1 cells eventually.

[Effect of electroacupuncture on proliferation of astrocytes after spinal cord injury].

OBJECTIVE: To explore the mechanism of acupuncture in treatment of spinal cord injury (SCI). METHODS: Adult Wistar rats were used to make SCI model by Allen's method. The SCI rats were treated with electroacupuncture (EA) at acupoints of the Governor Vessel for 3 days, 1 week, 2 weeks or 4 weeks. Normal group and spinal cord injury group were used as controls. The number and morphology of astrocytes in each group were investigated by electron microscope, immunohistochemistry and in situs hybridization methods. The expression of glial fibroblast acid protein (GFAP) mRNA in the injured spinal cord was detected by reverse transcription polymerase (RP-PCR). RESULTS: The mitochondria and ribosomes of astrocytes in the EA group increased. The number of astrocytes increased after SCI, in gray matter being more than that in the white matter, in the caudal being more than that in the rostral. The expression of GFAP mRNA in the EA group was significantly lower than that in the control group. CONCLUSION: Electroacupuncture can inhibit the reactive proliferation of the astrocytes after spinal cord injury and prevent formation of the glial scar.

[Clinical study on laparoscopic technique for vaginoplasty using an ileum with long vascular pedicle].

OBJECTIVE: To study the operative methods and clinical outcome of laparoscopic ileal colpopoiesis with long vascular pedicle. METHODS: We reviewed 31 cases which had received ileal colpopoiesis, including 25 cases of congenital absence of vagina and 6 cases of male-to-female trans-sex. All cases were operated on by laparoscopic (8 cases) and laparoscopy assisted (23 cases) ileal colpopoiesis. The postoperative results were analyzed through physical examination and interview regarding the patient's functional status and satisfaction during sexual intercourse. RESULTS: Ileal colpopoiesis were preformed successfully on all 31 cases. Follow-ups of 3-13 months were done for all of them. Patients used vaginal mould for at least 8 months. All artificial vaginas had excellent cosmetic results, including adequate vaginal length, good lubrication, and appearance and physical functions similar to female natural vagina, and sexual function was reportedly satisfactory. One of the patients had an operative complication of an intestinal obstruction, which was cured by a second operation and other treatments. CONCLUSIONS: The laparoscopic and laparoscopy assisted ileal vaginoplasty is a feasible, safe and ideal method of current vaginoplasty due to its minor damage, excellent cosmetic results and similarity to normal vaginal features. However, vaginal mould should be used for a long period of time (at least 8 months) after an operation, to prevent vaginal orifice shrinkage.

[Border-line smooth-muscle tumor of the uterus: analysis of 131 patients].

OBJECTIVE: To investigate whether the border-line uterine smooth-muscle tumor is different from leiomyoma or leiomyosarcoma in history and clinical manifestations. METHODS: The medical records of 131 surgically treated patients suffering from the so-called cellular leiomyoma or mitotically active leiomyoma of the uterus treated from 1984 to 2002 were retrospectively reviewed. All pathological sections of these patients were reviewed by a senior pathologist. Chi-square test and Kaplan-Meier life table were used for statistical analysis. RESULTS: The overall 5-year survival rate of patients with the border-line uterine smooth-muscle tumors was 100%, however, 19.1% (24/131) of whom developed a late recurrence and 8 patients had repeated recurrences with a more shortened course and aggressive potential changes of mitosis and cellular atypia. The overall 5-year survival rate the patients with recurrence was 91.7%, but only 75.0% for those with repeated recurrences. CONCLUSION: Patients with border-line uterine smooth-muscle tumor do possess some difference in nature from the leiomyoma or leimyosarcoma. Long-term follow-up is very important for these patients after surgery.

Terahertz radiation from oscillating electrons in laser-induced wake fields.

Strong terahertz (1 THz= 10(12) Hz) radiation can be generated by the electron oscillation in fs-laser-induced wake fields. The interaction of a fs-laser pulse with a low-density plasma layer is studied in detail using numerical simulations. The spatial distribution and temporal evolution of terahertz electron current developed in a low-density plasma layer are presented, which enables us to calculate the intensity distribution of THz radiation. It is shown that laser and plasma parameters, such as laser intensity, pulse width, and background plasma density, are of key importance to the process. The optimum condition for wake-field excitation and terahertz emission is discussed upon the simulation results. Radiation peaked at 6.4 THz, with 900 fs duration and 9% bandwidth, can be generated in a plasma of density 5x 10(17) cm(-3) . It turns out that the maximum radiation intensity scales as n(3)(0) a(4)(0) when wake field is resonantly excited, where n(0) and a(0) are, respectively, the plasma density and the normalized field amplitude of the laser pulse.

Crystal structure of human ABAD/HSD10 with a bound inhibitor: implications for design of Alzheimer's disease therapeutics.

The enzyme 17beta-hydroxysteroid dehydrogenase type 10 (HSD10), also known as amyloid beta-peptide-binding alcohol dehydrogenase (ABAD), has been implicated in the development of Alzheimer's disease. This protein, a member of the short-chain dehydrogenase/reductase family of enzymes, has been shown to bind beta-amyloid and to participate in beta-amyloid neurotoxicity. We have determined the crystal structure of human ABAD/HSD10 complexed with NAD(+) and an inhibitory small molecule. The inhibitor occupies the substrate-binding site and forms a covalent adduct with the NAD(+) cofactor. The crystal structure provides a basis for the design of potent, highly specific ABAD/HSD10 inhibitors with potential application in the treatment of Alzheimer's disease.

[Identification of FIP1L1-PDGFRA fusion, and expression of signal transducer and activator of transcription 5 in hypereosinophilic syndrome].

OBJECTIVE: To determine whether FIP1L1-PDGFRA fusion exists in hypereosinophilic syndrome (HES) patients, explore the relationship between FIP1L1-PDGFRA fusion and clinical phenotypes, and observe and reveal the expression of signal transducer and activator of transcription 5 (STAT(5)) in granulocytes of HES and the biological significance thereof. METHODS: Specimens of peripheral blood were collected from 4 HES patients diagnosed based on the criteria of Chusid et al. Total RNA was extracted from granulocytes and cDNA was synthesized by reverse transcription. Nested-PCR was used to amplify the target fusion gene and the positive PCR fragments were sequenced directly. Total protein of the peripheral granulocytes was extracted. Western blotting was used to detect the expression of STAT(5) protein in the granulocyte lysates. RESULTS: FIP1L1-PDGFRA fusion genes were found in 3 of the 4 HES patients. The break points in PDGFRA were all located at exon 12, while in FIP1L1 the break points were highly variable, located at exon 8a, intron 8a, and exon 8 respectively. The patients with FIP1L1-PDGFRA fusion were susceptible to cardiac involvement. The expression of STAT(5) protein was upregulated in FIP1L1-PDGFRA positive HES patients, while STAT(5) protein expression was negative in HES patients without FIP1L1-PDGFRA fusion. CONCLUSION: FIP1L1-PDGFRA fusion has a universal significance for HES. The identification of FIP1L1-PDGFRA rearrangement is a useful molecular mark for HES diagnosis and works as the therapeutic target of imatinib. Furthermore, the activation of STAT(5), a downstream signal of the FIP1L1-PDGFRA fusion, indicates that HES is a malignant clonal disease of the hematopoietic tissue.
