Integration of genome mining and HiTES reveals secondary metabolic potential in marine-derived Aspergillus sp. WHUF0304.

AIMS: Marine-derived Aspergillus species are prolific producers of bioactive secondary metabolites, yet the majority of their biosynthetic gene clusters (BGCs) remain silent. This study aimed to integrate genome mining with high-throughput elicitor screening (HiTES) to unlock the metabolic potential of Aspergillus sp. WHUF0304 and identify elicitors that promote the accumulation of previously undetected metabolites. METHODS AND RESULTS: A high-quality genome of Aspergillus sp. WHUF0304 was assembled and annotated using multiple functional databases, revealing substantial secondary metabolic potential. antiSMASH analysis identified diverse BGCs, including NRPS/indole-related clusters potentially associated with indole diketopiperazine biosynthesis. A HiTES-inspired elicitor screening strategy was then applied to evaluate 42 small molecules for their ability to alter the metabolite profile of this strain. Among the tested elicitors, fluconazole was identified as the optimal inducer, triggering the production of several indole diketopiperazine-related differential metabolites. Subsequent activity-guided isolation led to the identification of a bioactive indole diketopiperazine dimer, cristatumin E, which exhibited antibacterial activity against Escherichia coli and Bacillus subtilis with minimum inhibitory concentrations (MICs) of 32 µg mL-1 and 256 µg mL-1, respectively. CONCLUSIONS: These findings demonstrate that integrating genomic and functional approaches effectively activates silent BGCs in marine fungi. The fluconazole-associated accumulation and subsequent isolation of cristatumin E, a bioactive indole diketopiperazine dimer, highlight the potential of elicitor-mediated activation to expand the detectable metabolite profile of Aspergillus sp. WHUF0304.

Systematic Genome Mining of Peptide Metallophore Pathways Uncovers Novel Dibenzo-α-Pyrone Siderophores in Streptomyces sp. HB-R818.

Metallophores are metal-chelating natural products that enable microorganisms to acquire essential metal ions and mediate processes such as iron uptake, quorum sensing, and interspecies competition. Metallophores also display potent antimicrobial and anticancer activities, highlighting their biomedical and biotechnological potential. Despite Streptomyces being prolific producers of bioactive metabolites, their metallophore pathways remain largely unexplored. Here, we systematically mined 519 reference Streptomyces genomes to elucidate the distribution, diversity, and structural features of metallophores and identified a new metallophore biosynthetic gene cluster (BGC) (ser) from sponge-derived Streptomyces sp. HB-R818. Using a metabologenomics-based strategy, five new siderophore analogs serobactins A-E (1-5) and known enterobactin (6) were isolated. These compounds show potential to inhibit tumor invasion and feature a unique dibenzo-α-pyrone scaffold in structure, formed through the cyclization of an extra 2,3-dihydroxybenzoic acid with 2,3-dihydroxybenzoyl serine. The BGC (ser) was validated by the nonribosomal peptide synthetase gene knockout; the biosynthesis of 1-6 was proposed.

Tunable instability mechanisms of polymer thin films by molecular self-assembly.

Incorporation of a block copolymer into a thin polymer film is observed to alter both the rate and mechanism by which the film dewets from an immiscible polymer substrate. Films with little or no copolymer dewet by classical nucleation and growth of circular holes, and the dewetting rate decreases with increasing copolymer concentration. Increasing the copolymer content at constant film thickness generates copolymer micelles that adsorb/aggregate along the polymer/polymer interface and promote nonclassical dewetting fluctuations similar in appearance to spinodal dewetting. At higher copolymer concentrations, dewetting proceeds after a lengthy induction period by the nucleation and growth of flower-shaped holes suggestive of film pinning or viscous fingering. Atomic force microscopy of the polymer/polymer interface after removal of the top film by selective dissolution reveals substantial structural development due to copolymer self-assembly.

Reversal of the TCR stop signal by CTLA-4.

The coreceptor cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) is pivotal in regulating the threshold of signals during T cell activation, although the underlying mechanism is still not fully understood. Using in vitro migration assays and in vivo two-photon laser scanning microscopy, we showed that CTLA-4 increases T cell motility and overrides the T cell receptor (TCR)-induced stop signal required for stable conjugate formation between T cells and antigen-presenting cells. This event led to reduced contact periods between T cells and antigen-presenting cells that in turn decreased cytokine production and proliferation. These results suggest a fundamentally different model of reverse stop signaling, by which CTLA-4 modulates the threshold for T cell activation and protects against autoimmunity.

[Stress analyses and comparison of the Kennedy III dentition defect restored with removable partial denture].

PURPOSE: To investigate the stress distribution of the abutment and oral mucosa in Kennedy III, restored with removable partial dentures, either unilateral or bilateral designed. METHODS: Five FEA models were established for the partial edentulous and RPDs, which were divided into group A and group B. According to the denture design, bilateral (B) or unilateral (A) designed. The 3-D finite element analyses were proceeded and calculated with the help of the software ANSYS. Paired t test was used for statistical analysis using SAS 6.2 software package. RESULTS: The maximum stress appeared near the cervical mesial area of the first bicuspid and the second molar. There was no significant difference between the two groups in the stress distribution of the abutment and oral mucosa (P>0.05). CONCLUSION: In regard to the stress of the abutments, unilateral designs have no excess stress formed when RPD was used to restore Kennedy III dentition defect.

[Characters of the surface of the cast-bonded keeper: an experimental study].

PURPOSE: This study is aimed to analyze the characters of the surface of the keeper after cast-bonded in varied temperatures. METHODS: Twelve keepers were divided into three groups: the gold alloy group, the Ni-Cr alloy group and the Ti group with four samples in each group. A sample that contained a Magdisc 500 keeper was fabricated with the cast-bonded technique. Then, the surface of the sample was observed under scanning electron microscope (SEM) and its elements were analyzed with the electron probe. RESULTS: The results showed that there was no oxide film on the surface of the keeper in the gold alloy group and the Ni-Cr alloy group, but there was an oxide film on the surface of the keeper in the Ti group. However, remains of the investment materials were found on the surface of the sample of three groups. CONCLUSIONS: The study suggested that the casting temperature affect the characters of the surface of the cast-bonded keeper. Gold alloy or Ni-Cr alloy is of better properties for clinical use.

Amnioinfusion for the prevention of the meconium aspiration syndrome.

BACKGROUND: It is uncertain whether amnioinfusion (infusion of saline into the amniotic cavity) in women who have thick meconium staining of the amniotic fluid reduces the risk of perinatal death, moderate or severe meconium aspiration syndrome, or both. METHODS: We performed a multicenter trial in which 1998 pregnant women in labor at 36 or more weeks of gestation who had thick meconium staining of the amniotic fluid were stratified according to the presence or absence of variable decelerations in fetal heart rate and then randomly assigned to amnioinfusion or to standard care. The composite primary outcome measure was perinatal death, moderate or severe meconium aspiration syndrome, or both. RESULTS: Perinatal death, moderate or severe meconium aspiration syndrome, or both occurred in 44 infants (4.5 percent) of women in the amnioinfusion group and 35 infants (3.5 percent) of women in the control group (relative risk, 1.26; 95 percent confidence interval, 0.82 to 1.95). Five perinatal deaths occurred in the amnioinfusion group and five in the control group. The rate of cesarean delivery was 31.8 percent in the amnioinfusion group and 29.0 percent in the control group (relative risk, 1.10; 95 percent confidence interval, 0.96 to 1.25). CONCLUSIONS: For women in labor who have thick meconium staining of the amniotic fluid, amnioinfusion did not reduce the risk of moderate or severe meconium aspiration syndrome, perinatal death, or other major maternal or neonatal disorders.

CTLA-4 up-regulation of lymphocyte function-associated antigen 1 adhesion and clustering as an alternate basis for coreceptor function.

Although cytotoxic T lymphocyte antigen-4 (CTLA-4) negatively regulates T cell activation, the full range of functions mediated by this coreceptor has yet to be established. In this study, we report the surprising finding that CTLA-4 engagement by soluble antibody or CD80 potently up-regulates lymphocyte function-associated antigen 1 (LFA-1) adhesion to intercellular adhesion molecule-1 (ICAM-1) and receptor clustering concurrent with IL-2 inhibition. This effect was also observed with CTLA-4 ligation and not with other coreceptors. T cell antigen receptor (TcR)-induced lymphocyte function-associated antigen 1 function was also dependent on CTLA-4 expression as observed with reduced adhesion/clustering on CTLA-4(-/-) primary T cells. CTLA-4 up-regulated adhesion was mediated by regulator for cell adhesion and polarization type 1 (Rap-1) as shown by anti-CTLA-4-induced Rap-1 activation as well as Rap-1-N17 blockade and Rap-1-V12 mimicry of adhesion/clustering. Our findings identify a potent role for CTLA-4 in directing integrin adhesion and provide an alternate mechanism to account for aspects of CTLA-4 function in T cell immunity.

[3-D FEA of the stress state of abutment and its periodontal tissue with three different retention styles in the absence of mandibular anterior teeth].

PURPOSE: To study the stress of the abutment and its periodontal tissue with different designs of retainer (type of retainer, place)in the same situation of dentural defect. METHODS: Based on the 3-D FEA database of RPD, the stress state of the abutment and its periodontal tissue was analyzed to choose the optimal retainer with the same situation of dentural defect. RESULTS: With three different retainer designs, the retainer on the second premolar and the first molar exerted the smallest stress on the abutment and its periodontal tissue, while the retainer on the first premolar and the first molar had the largest stress. CONCLUSION: Analysis for the health condition of abutment and its periodontal tissue is imperative for the selection of different designs of retainers.

Dewetting behavior of a block copolymer/homopolymer thin film on an immiscible homopolymer substrate.

Numerous previous studies have established that the addition of a microphase-ordered AB diblock copolymer to a thin homopolymer A (hA) film can slow, if not altogether prevent, film rupture and subsequent film dewetting on a hard substrate such as silica. However, only a few reports have examined comparable phenomena when the hA/AB blend resides on a soft B-selective surface, such as homopolymer B (hB). In this work, the dewetting kinetics of thin films composed of polystyrene (PS) and a symmetric poly(styrene-b-methyl methacrylate) (SM) diblock copolymer on a poly(methyl methacrylate) substrate is investigated by hot-stage light microscopy. Without the SM copolymer, the dewetting rate of the PS layer is constant under isothermal conditions and exhibits Arrhenius behavior with an apparent activation energy of approximately 180 kJ/mol. Addition of the copolymer promotes a crossover from early- to late-stage dewetting kinetics, as evidenced by measurably different dewetting rates. Transmission electron microscopy reveals the morphological characteristics of dewetted PS/SM films as functions of film thickness and SM concentration.
