Cloning and expression of PDK4, FOXO1A and DYRK1A from the hibernating greater horseshoe bat (Rhinolophus ferrumequinum).

Pyruvate dehydrogenase kinase isoenzyme 4 (PDK4) cDNA was cloned from the brain of greater horseshoe bat (Rhinolophus ferrumequinum). The deduced amino acid sequence shares strong homology with these PDK4 of other mammals. Moreover, we partially cloned homologues of dual-specificity tyrosine-phosphorylated and regulated protein kinase 1A (DYRK1A), and forkhead box protein O1A (FOXO1A) from greater horseshoe bat. Among five different tissues tested, PDK4 mRNA was highly expressed in the heart, white adipose tissue and muscle, but weakly expressed in the brain and liver, while DYRK1A and FOXO1A were expressed in all five tissues. Moreover, the transcript levels of PDK4, DYRK1A, and FOXO1A were measured in the heart, white adipose tissue and muscle of hibernating and arousal greater horseshoe bats by Northern blot and real time PCR. The results showed that transcript level of PDK4 was significantly higher in white adipose tissue. Expression level of DYRK1A was significantly higher in hibernating state in white adipose tissue, and expression level of FOXO1A was significantly higher in muscle in aroused state. These results suggest that up-regulation of the transcript levels of PDK4 during hibernation were not regulated via DYRK1A and FOXO1A in white adipose tissue and muscle, and the possible presence of another isoenzyme of PDK which is responsible for the tissue-specific regulation of Pyruvate dehydrogenase complex (PDC) activity in the bat heart during hibernation.

PIAS3 induction of PRB sumoylation represses PRB transactivation by destabilizing its retention in the nucleus.

Progesterone receptor (PR) plays a critical role in cell proliferation and differentiation, and its transcriptional activity is known to be modulated by cofactor proteins. In the present study, we demonstrated that in the presence of progesterone, protein inhibitor of activated STAT-3 (PIAS3) significantly inhibited the PR transcriptional activity and the expression of progesterone-responsive genes. Reduction of endogenous PIAS3 by PIAS3 small-interfering RNA enhanced PR transactivation in a ligand-dependent manner. PIAS3 interacted with PR both in vitro and in vivo and the interaction was enhanced by progesterone. Furthermore, our findings suggested that PIAS3 strongly induced PRB sumoylation at three sites, Lys-7, Lys-388 and Lys-531. In addition, novel roles in PRB nuclear retention and transactivation were identified for these sites. Our data also suggested that PIAS3 was recruited in a largely hormone-dependent manner in response to a progesterone-responsive promoter. Finally, we demonstrated that PIAS3 inhibited the DNA-binding activity of PR and influenced its nuclear export as well as PR transactivation. Taken together, these data strongly suggested that PIAS3 played an important physiological role in PR function.

MDM2 interacts with and downregulates a sarcomeric protein, TCAP.

Recent reports have shown that MDM2 may attenuate hypertrophy of cardiac myocytes. However, mechanism of MDM2 involving in this process is unclear. In this study, we identified a novel specific MDM2-binding protein TCAP by the yeast two-hybrid screen. It was validated by GST pull-down and co-immunoprecipitation assays. Confocal analysis showed that MDM2 and TCAP co-localized in the nucleus, and elevated MDM2 expression could alter the subcellular localization of TCAP. Notably, MDM2 downregulated the protein level of TCAP through the proteasomal pathway, and this downregulation was inhibited by p14(ARF). In addition, our results suggested that the degradation of TCAP by MDM2 was through the ubiquitin-independent pathway. Given that TCAP is a key component involving in the cardiac hypertrophy, the degradation of TCAP by MDM2 might be connected with the roles of MDM2 in cardiac hypertrophy. Further investigation will focus on the biological significance of MDM2-TCAP interaction in cardiac hypertrophy.

Proteomics-based identification of autoantibodies in the sera of healthy Chinese individuals from Beijing.

The identification of panels of tumor antigens that elicit an antibody response may have utility in cancer screening, diagnosis and in establishing prognosis. However, autoantibodies normally exist in sera of healthy individuals and are enormously diversified. To explore the reservoir of autoantibody in healthy population, we performed a proteomics investigation of autoantibody profiles in the sera of 36 healthy Chinese individuals from Beijing, which may provide valuable reference information to the identification of disease-specific autoantibodies. The results showed that autoantibody profiles varied individually, but some autoantibodies were identified at a high frequency in the healthy population. The autoantibodies against alpha-enolase and those against heterogeneous nuclear ribonucleoprotein L were positive in more than 50% of the sera samples. The autoantibodies identified in more than 20% of samples included those against annexin II, F-actin capping protein beta subunit and calreticulin. Some of these autoantibodies have been previously reported to be involved in autoimmune conditions and cancers. Autoantibodies in the healthy population are important as a foundation from which disease-specific autoantibodies can be defined. Thus our report on autoantibodies in healthy individuals may be useful as a reference for defining new autoantibody biomarkers.

ATP-dependent chromatin-remodeling complexes in DNA double-strand break repair: remodeling, pairing and (re)pairing.

The genomic integrity of a eukaryotic cell is challenged by over 10,000 chromosomal lesions per day. Therefore the cell has evolved efficient mechanisms to recognize, signal, and repair DNA breaks. Defects in any of these steps can lead to chromosomal aberrations and cancers. As these lesions must be repaired in the context of chromatin, both chromatin-modifying and nucleosome-remodeling enzymes have been implicated in DNA damage repair. We reported recently that the RSC and Swi/Snf ATP-dependent chromatin-remodeling complexes are involved in DSB repair specifically by homologous recombination. Here we discuss how such enzymes might be recruited to DNA breaks, why so many remodelers are recruited to sites of DSBs, and a possible functional connection between RSC's roles in sister chromatid cohesion and DSB repair.

Evolution of nutrient structure and phytoplankton composition in the Jiaozhou Bay ecosystem.

The inventories of nutrients in the surface water and large phytoplankton( > 69 microm) were analyzed from the data set of JERS ecological database about a typical coastal waters, the Jiaozhou Bay, China, from 1960s for N, P and from 1980s for Si. By examining long-term changes of nutrient concentration, calculating stoichiometric balance, and comparing diatom composition, Si limitation of diatom production was found to be more possible. The possibility of Si limitation was from 37% in 1980s to 50% in 1990s. Jiaozhou Bay ecosystem is becoming serious eutrophication, with notable increase of NO2-N, NO3-N and NH4-N from 0.1417 micromol/L, 0.5414 micromol/L, 1.7222 micromol/L in 1960s to 0.9551 micromol/L, 3.001 micromol/L, 8.0359 micromol/L in late 1990s respectively and prominent decrease of Si from 4.2614 micromol/L in 1980s to 1.5861 micromol/L in late 1990s; the nutrient structure is controlled by nitrogen; the main limiting nutrient is probably silicon; because of the Si limitation the phytoplankton community structure has changed drastically.

[Investigation on comparison method of chromatographic fingerprints of lacquer coat of cars and its application].

The comparison method of fingerprints of the lacquer coat of cars (LCC) was established by using thermal desorption instrument with gas chromatography. The actual LCC samples were also analyzed. The samples were cut out to proper size and placed in the desorption furnace of the thermal desorption instrument. The volatile organic compounds in LCC were desorbed from the lacquer coat samples in the furnace under the chosen temperature, then separated in the capillary column and detected on a flame ionization detector of gas chromatography. The incipient judgment whether the two fingerprints of LCC were the same can be made from the contour and figure of the chromatograms. To make farther study of the two similar fingerprints, the overlap ratio of the peaks and relative retention values were given in the article. The two LCC samples can be regarded as the same if the overlap ratio of peaks was more than 90%, and the similarity of the ratio of relative retention times r(t2) and r(t1) and relative peak areas r(A2) and r(A1) in the two fingerprints were more than 99% and 70%, respectively. The method is good in repeatability and is easily carried out. The peaks in the fingerprint can be readily recognized. The fingerprint was characterized quantitatively. The method can be used in the department of traffic police and the comparison result can be used as material evidence in the court.

Sequence and expression of cytochrome P450 aromatase and FTZ-F1 genes in the protandrous black porgy (Acanthopagrus schlegeli).

In this study, a cDNA encoding cytochrome P450 aromatase (P450arom) was cloned from black porgy Acanthopagrus schlegeli ovary. The deduced amino acid sequence had high homology with ovarian P450arom of other teleost fish. Moreover, we partially cloned two FTZ-F1 homologues (asff1a and asff1b) from black porgy. Comparative sequence analysis grouped asff1a and asff1b in NR5A2 and NR5A4 clades, respectively. Among the various tissues tested, P450arom mRNA was highly expressed in the ovary and weakly in the brain and testis, asff1a was expressed in brain, liver, intestine, kidney, testis, and ovary, asff1b was expressed in brain, kidney, testis, and ovary. The transcript levels of P450arom, asff1a, and asff1b were measured in the ovary and testis of 1+ -year-old, 2+ -year-old, and 5+ -year-old black porgy. The transcript level of P450arom in the ovary of 2+ -year-old fish was significantly higher than those of 1+ -year-old and 5+ -year-old fish. The results suggest that P450arom gene may be involved in the mechanism of natural sex change of protandrous black porgy. No change in ovarian expression of asff1a or asff1b was observed among different ages. These results suggest that up-regulation of the transcript levels of P450arom during the course of natural sex change of black porgy was not regulated via FTZ-F1.

[Technology of biodesulfurization from flue gases].

The desufurization in dilute sulfuric acid solution, acidic ferric solution, and microbial solution (Thiobacillus ferrooxidans) was studied in this paper. The concentration of ferric, pH value and time all affect the rate of desulfurization. The experimental results showed that in dilute sulfuric acid solution the absorption of SO2 was a physical absorption. In acidic ferric solution, the ion of ferric can not only catalyze but also oxidize SO2. The effects of desulfurization became obvious along with high concentration of ferric and pH value. Desulfurization rate was up to 90% when pH value was 2.0 and the concentration of ferric was 1.5 g/L. In microbial solution, the effects were also influenced by the concentration of ferric and pH value. Thiobacillus ferrooxidans had an ability of oxiding the ion of ferrous and S(IV). The results of desulfurization was the best at the concentration of ferric 1.5 g/L and pH 2.0.

[Determination of purity of different types of aprotinin by micellar electrokinetic capillary chromatography].

A sensitive and rapid method was developed for purity determination of different types of aprotinin by micellar electrokinetic capillary chromatography (MECC). MECC was performed with 80 mmol/L Na2HPO4-H3PO4(pH 7.00) buffer solution containing 4 mmol/L cetyltrimethyl ammonium bromide (CTAB) at an applied voltage of 15 kV. The electrophoresis was monitored at 277 nm, and completed in 6 minutes. The relative standard deviations of migration time and area for aprotinin were below 0.3% and below 5% respectively within 1 day (for 6 repetitive runs). The effect of high concentration of salt in the samples has been studied. The results obtained by capillary zone electrophoresis, MECC and high performance liquid chromatography were compared, and those of MECC were the best.
