Distribution, levels and phosphorylation of Raf-1 in Alzheimer's disease.

Extracellular signal-regulated kinase (ERK), a member of the mitogen-activated protein kinase pathway, has been increasingly implicated in the pathogenesis of Alzheimer's disease due to its critical role in brain function. While we previously demonstrated that ERK is activated in Alzheimer's disease, the upstream cascade leading to its activation had not been fully examined. In this study, we focused on Raf-1, one of the physiological activators of the ERK pathway. Raf-1 is activated by phosphorylation at Ser338 and Tyr340/341 and inhibited by phosphorylation at Ser259. Interestingly, phosphorylation at all three sites on Raf-1 was increased as evidenced by both immunocytochemistry and immunoblot analysis in Alzheimer's disease brains compared to age-matched controls. Both phospho-Raf-1 (Ser259) and phospho-Raf-1 (Ser338) were localized to intracytoplasmic granular structures, whereas phospho-Raf-1 (Tyr340/341) was localized to neurofibrillary tangles and granules in pyramidal neurons in Alzheimer's disease hippocampus. There is extensive overlap between phospho-Raf-1 (Ser338) and phospho-Mek1/2, the downstream effector of Raf-1, suggestive of a mechanistic link. Additionally, increased levels of Raf-1 are associated with Ras and MEK1 in Alzheimer's disease as evidenced by its coimmunoprecipitation with Ras and Mek1, respectively. Based on these findings, we speculate that Raf-1 is activated to effectively mediate Ras-dependent signals in Alzheimer's disease.

[The value of detection of gene rearrangement of immunoglobulin heavy chain and immunohistochemistry in pulmonary mucosa-associated lymphoid tissue type lymphoma].

OBJECTIVE: To investigate the features of gene rearrangement of immunoglobulin heavy chain (IgH) and immunophenotypes of pulmonary mucosa-associated lymphoid tissue type lymphoma (MALTLoma). METHODS: The clinical and pathological data of 12 cases with pulmonary MALTLoma and follow-up information were retrospectively reviewed, and the paraffin-embedded samples were examined with immunohistochemistry staining (12 cases) and semi-nested polymerase chain reaction (PCR) for IgH and T-cell receptor gamma (TCRgamma) gene rearrangement (7 cases). RESULTS: The patients included 9 cases confirmed by open or video-assisted thoracoscopic lung biopsy and 3 cases by needle lung biopsy. Histopathologically, the tumors were composed of a spectrum of cell types that included mainly centrocyte-like cells and small lymphocytes. Lymphoepithelial lesions were identified in 12 cases, reactive colliculus lymphaticus in 11 cases, follicular colonization in 10 cases, vascular infiltration in 9 cases, and pleura involvement in 4 cases. All cases showed immunoreactivity for B-cell correlative markers. Positivity for FR2 and FR3A primers in MALTLoma were found in 6 case and 5 case respectively. The detection of TCRgamma1 and TCRgamma2 was negative in 7 cases. The combined positive rate was 100%. Chemotherapy alone was administered in 3 patients, surgery alone was performed in 8 patients, and chemotherapy after operation was carried out in 6 patients. Follow-up data were available in 11 patients. Eight of them were alive and stable, one experienced relapse and two died of the disease within 11 years and 12 years after diagnosis respectively. CONCLUSIONS: Most of the cases of pulmonary MALTLoma can be diagnosed with morphology and immunohistochemistry staining if the lesions are typical. PCR detection of IgH gene rearrangement would be helpful in differential diagnosis from benign lymphoplasia of the lung.

HIV-1 subtype B' dictates the AIDS epidemic among paid blood donors in the Henan and Hubei provinces of China.

OBJECTIVE: To investigate the genetic background of HIV-1 strains among infected paid blood donors (PBD) in the Henan and Hubei provinces of China. METHODS: The polymerase chain reaction was used to amplify HIV-1 gag p17 fragments and some 5'-half or full-length viral genomes from peripheral blood mononuclear cells. The HIV-1 sequences obtained were analysed using phylogenetic and recombinant approaches. RESULTS: Among the total of 62 samples studied, 59 (95.2%) came from PBD. Of those 59 PBD, 45 were collected from 14 different geographical locations in Henan, whereas the remaining 14 were from five locations in Hubei. A total of 62 HIV-1 gag p17 fragments, three 5'-half and one full HIV-1 genome sequences were obtained. Phylogenetic analysis of these sequences suggests that PBD from Henan and Hubei, despite being geographically distant, harbored a group of genetically closely related HIV-1 B' strains. No recombinant forms have been detected in this cohort. CONCLUSION: Our data indicate that the AIDS epidemic among PBD in Henan and Hubei is caused largely by HIV-1 subtype B', which contrasts greatly with the CB' recombinant strains CRF-07 and CRF-08 currently dominating among the drug abusers in other regions of China. Our report has revealed the first complete genome background of the most dominant circulating HIV-1 strain in Henan and Hubei, which is essential for the design and development of an effective AIDS vaccine for the region.

[A survey of children with HIV/AIDS in highly epidemic villages of AIDS].

OBJECTIVE: To estimate prevalence of HIV/AIDS among children and the transmission routes in a highly endemic villages of AIDS. METHODS: Totally 208 high-risk women of child bearing age and 159 of their children aged 0 - 14 years were investigated. Their medical histories of blood donation or transfusion were collected, blood samples were taken and sera were separated for HIV test. Enzyme-linked immunosorbent assay (ELISA) and Western blot assay were performed for HIV antibody. The Nested-polymerase chain reaction (PCR) assay amplifying gag gene p17 was performed on samples of children aged less than 18 months. RESULTS: Thirty-seven HIV infected cases were found among 159 children aged 0 - 14 years of whom 33 were infected by mother-to-child transmission (89.2%, 33/37), 3 by blood transfusion (8.1%, 3/37) and one by iatrogenic route (2.7%, 1/37). Sixty seven mothers who were seropositive for HIV and their 86 children who were born after 1992 were investigated, 33 cases of them were infected with HIV. The rate of vertical transmission was 38.4% (33/86). The HIV vertical transmission rate among mothers with AIDS (68.8%, 22/32) was significantly greater than that among mothers with asymptomatic HIV infection (20.4%, 11/54, P < 0.05). The number of children infected with HIV through vertical transmission increased from 1993 to 2001. Among 37 children infected with HIV, 12 cases developed AIDS and 4 of them died, of whom 2 cases died from tuberculosis. The morbidity of AIDS was 27.3% (9/33). Ninety three point nine percent (31/33) of infected mothers didn't know their HIV seropositive status before pregnancy and delivery. Of 8 pregnant women infected with HIV, one had aggravation of AIDS, 2 miscarried, 2 terminated their pregnancy and 3 continued their pregnancy. CONCLUSION: Mother-to-child transmission of HIV was the major route of HIV/AIDS transmission to the children. The main reason leading to HIV infection in children was the lack of prenatal HIV counseling and testing for the high-risk women of childbearing age and lake of interventions. The countermeasures must be taken to control the further transmission of AIDS in order to protect the health of women and children in the highly endemic areas of AIDS.

[The role of nitric oxide in the cytotoxic effect of macrophages].

AIM: To explore the role of nitric oxide (NO) in cytotoxicity of macrophages. METHODS: 20 mice of Kunming species were divided into two groups. One group were injected subcutaneously with rhabdosarcoma cells S180 and the other were normal controls. The macrophages in peritoneal lavage fluid of two groups of mice were co-cultured with tumor cells K562. L-arginine(L-Arg) or NG nitro L-arginine methyleste(L-NAME) was added to the culture medium, and the cytotoxicity of macrophages was measured. RESULTS: Macrophages from tumor bearing mice had stronger cytotoxicity than those from normal mice. L-Arg increased and L-NAME decreased the cytotoxicity of macrophages from tumor bearing mice, but both had no effect on the cytotoxicity of macrophages from normal mice. CONCLUSION: NO is an effector molecule in cytotoxicity of activated macrophages.

Rapid detection of epidermal growth factor receptor mutations in non-small cell lung cancer using real-time polymerase chain reaction with TaqMan-MGB probes.

UNLABELLED: We investigated somatic mutations of the epidermal growth factor receptor gene in non-small cell lung cancer tumor tissue and their detection using real-time polymerase chain reaction with TaqMan-MGB probes. METHODS: The DNA was extracted from surgically resected non-small cell lung cancer tumor specimens. Genes encoding for epidermal growth factor receptor tyrosine (exons 18, 19, and 21) were amplified by nested polymerase chain reaction, sequenced, and analyzed by chromatograms with manual review. TaqMan-MGB probes were designed to detect the epidermal growth factor receptor gene mutations in the tumor tissues using real-time polymerase chain reaction. RESULTS: Somatic point mutations and deletions were identified in the tyrosine kinase domain of the epidermal growth factor receptor gene in 21 of 80 non-small cell lung cancer patients, including 13 patients with deletion mutations occurring in exon 19 and 8 patients with point mutations occurring in codon 858 (exon 21). The results from real-time polymerase chain reaction with TaqMan-MGB probes were completely consistent with sequencing outcomes. Both the sensitivity and specificity for detecting the epidermal growth factor receptor gene mutations using real-time polymerase chain reaction with TaqMan-MGB probes were 100%. The mutation incidence was significantly higher in female patients, nonsmokers, and patients with adenocarcinoma than in male patients, smokers, and those with nonadenocarcinomas (P < 0.05). The mutations were not related to patient's age or tumor nodal metastasis staging. CONCLUSIONS: Somatic mutations of the epidermal growth factor receptor gene that develop in non-small cell lung cancer patients are more common in female patients, nonsmokers, and patients with adenocarcinoma. Real-time polymerase chain reaction using TaqMan-MGB probes is effective, simple, and fast in the detection of epidermal growth factor receptor gene mutations.
