Polarized trafficking of E-cadherin is regulated by Rac1 and Cdc42 in Madin-Darby canine kidney cells.

E-cadherin is a major cell-cell adhesion protein of epithelia that is trafficked to the basolateral cell surface in a polarized fashion. The exact post-Golgi route and regulation of E-cadherin transport have not been fully described. The Rho GTPases Cdc42 and Rac1 have been implicated in many cell functions, including the exocytic trafficking of other proteins in polarized epithelial cells. These Rho family proteins are also associated with the cadherin-catenin complexes at the cell surface. We have used functional mutants of Rac1 and Cdc42 and inactivating toxins to demonstrate specific roles for both Cdc42 and Rac1 in the post-Golgi transport of E-cadherin. Dominant-negative mutants of Cdc42 and Rac1 accumulate E-cadherin at a distinct post-Golgi step. This accumulation occurs before p120(ctn) interacts with E-cadherin, because p120(ctn) localization was not affected by the Cdc42 or Rac1 mutants. Moreover, the GTPase mutants had no effect on the trafficking of a targeting mutant of E-cadherin, consistent with the selective involvement of Cdc42 and Rac1 in basolateral trafficking. These results provide a new example of Rho GTPase regulation of basolateral trafficking and demonstrate novel roles for Cdc42 and Rac1 in the post-Golgi transport of E-cadherin.

Influence of ultrasonic field on microcystins produced by bloom-forming algae.

Under the background of algae removal and growth inhibition by ultrasonic irradiation, the effects of ultrasonic irradiation on removal of Microcystis, the concentration variation of microcystins (MC) produced by Microcystis in Microcystis suspension, and sonochemical degradation of microcystins in water, were studied in the paper. The results showed that ultrasonic irradiation could efficiently inhibit the growth of Microcystis, and ultrasonic irradiation shorter than 5 min would not introduce the increase of microcystins dissolved in Microcystis suspension simultaneity. Also, microcystins dissolved in Microcystis suspension would not increase as ultrasonic power increasing. Further research showed that microcystins were effectively degraded in ultrasonic fields. After 20 min ultrasonic irradiation at 150 kHz and 30 W, the removal rate of microcystins reached 70%.

Antifungal activity of camptothecin, trifolin, and hyperoside isolated from Camptotheca acuminata.

Leaf spots and root rots are major fungal diseases in Camptotheca acuminata that limit cultivation of the plant for camptothecin (CPT), a promising anticancer and antiviral alkaloid. Bioassays showed that pure CPT and flavonoids (trifolin and hyperoside) isolated from Camptotheca effectively control fungal pathogens in vitro, including Alternaria alternata, Epicoccum nigrum, Pestalotia guepinii, Drechslera sp., and Fusarium avenaceum, although antifungal activity of these compounds in the plant is limited. CPT inhibited mycelial growth by approximately 50% (EC50) at 10-30 microg/mL and fully inhibited growth at 75-125 microg/mL. The flavonoids were less effective than CPT at 50 microg/mL, particularly within 20 days after treatment, but more effective at 100 or 150 microg/mL. CPT, trifolin, and hyperoside may serve as leads for the development of fungicides.

[Expression of leptin mRNA in luteinized granulosa cells and leptin levels in serum and follicular fluid of non-obese infertile patients with polycystic ovary syndrome].

OBJECTIVE: To investigate the expression of leptin mRNA in luteinized granulose cells and the leptin level in serum and follicular fluid of non-obese patients with polycystic ovary syndrome (PCOS). METHODS: Granulosa cells were collected from the puncture fluid of follicle of 16 obese infertile patients with PCOS to undergo external fertilization. Fasting venous blood was collected while collecting the ova. RT-PCR was used to detect the expression of leptin mRNA in the granulosa cells. ELISA was used to detect the leptin level in the serum and follicular fluid. Twenty normal women undergoing external fertilization because of male infertility of their husbands were used as controls. RESULTS: The expression of leptin mRNA (A value) was 1.5 +/- 0.4 in the obese infertile patients with PCOS and was 1.8 +/- 0.6 (P > 0.05) in the controls. Both the leptin levels of serum and follicular fluid were not different between the obese infertile patients with PCOS and controls (both P > 0.05). The leptin level in follicular fluid was significantly correlated with both the serum leptin level in the obese infertile patients with PCOS (r(s) = 0.838, P = 0.000) and the control group (r(s) = 0.712, P = 0.000). The expression level of leptin mRNA in the luteinized was not correlated with the leptin level of follicular fluid in both the obese infertile patients with PCOS and the control group (both P > 0.05). The fertility rate was not different between the obese infertile patients with PCOS (7/16) and the controls (9/24) (P = 0.693). CONCLUSION: The expression of leptin mRNA is normal in the follicular fluid of the obese infertile patients with PCOS. The leptin levels in serum and follicular fluid of the obese infertile patients with PCOS are similar to those of normal women. The leptin level in follicular fluid depends mainly on the circulating leptin level.

Expression of basic fibroblast growth factor in rat liver fibrosis and hepatic stellate cells.

The expression of basic fibroblast growth factor (bFGF) in rat liver fibrosis and hepatic stellate cells (HSCs) and the relationship between the expression of bFGF and rat liver fibrogenesis were studied. Sixty male SD rats (230-260 g) were divided into 4 groups randomly (the 0 week group, 1 week group, 4 week group and 8 week group). Liver fibrosis was induced by subcutaneous injection of carbon tetrachloride. The sections of rats' liver in each group were tested by Van-Gieson (V-G) staining and immunohistochemistry. The expression of bFGF mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR). HSCs were isolated by the combined methods of collagenase IV perfusion and density gradient centrifugation. The expression of bFGF protein in cultured HSCs was detected by Western blot. Images of immunohistochemistry detection, agarose gel electrophoresis of RT-PCR and SDS-polyacrylamide gel electrophoresis of Western blot were analyzed semiquantitatively by image-analyzing system. The results were analyzed by statistics. The results showed that the fibers were gradually increased in the sections of rat liver with the prolongation of the model induction. At the end of the 8th weeks, liver fibrosis was formed. The expression of bFGF detected by immunohistochemistry showed a similar tendency of gradual increase. At the end of the 8th weeks, the bFGF expression could be observed in many regions in sections and the strongest expression was in interstitial cells including HSCs and some hepatocytes in regions around the portal area and central veins. Also there was moderate expression widely in extracellular matrix (ECM). In RT-PCR detection and Western blot detection of HSCs cultured in vitro, the similar tendency of gradual increase was evident either. It is suggested that bFGF is related with liver fibrosis of rats closely and may be a fibrogenesis factor of liver. bFGF possibly regulates liver fibrogenesis through regulating metabolism of extracellular matrix (ECM) by autocrine and paracrine stimulation.

Radiotherapy of human xenograft NSCLC tumors in nude mice with a 90Y-labeled anti-tissue factor antibody.

Tissue factor (TF) is a type I transmembrane protein and the initiator of the extrinsic blood coagulation pathway. TF plays a critical role in tumor development and its overexpression is observed in many tumors. To understand the prevalence and relative level of TF expression in non-small-cell lung cancer (NSCLC), we analyzed 50 NSCLC tumors by immunohistochemical staining and found that 88% of human NSCLC tumors overexpressed TF. We then generated a high affinity anti-TF antibody, TF278, which specifically binds TF on the surface of cells and is internalized upon binding. An 111In-labeled TF278 demonstrated favorable tumor accumulation in an SW-900 xenograft tumor model with a maximum mean percent of injected dose per gram of tissue (%ID/g) of 73.1% at 96 hours postinjection. In addition, we labeled the antibody with 90Y and tested its ability to inhibit the growth of tumors in an SW-900 xenograft tumor model in immunocompromised mice. The 90Y-TF278 slowed the growth of SW-900 tumors at a 50 microCi dose and completely regressed SW-900 tumors at a 150 microCi dose with little toxicity.

Vaginal douching, condom use, and sexually transmitted infections among Chinese female sex workers.

BACKGROUND AND OBJECTIVE: Vaginal douching has been hypothesized to increase a woman's risk for human immunodeficiency virus (HIV) infection. However, data on the prevalence of this practice and its association with condom use and sexually transmitted infections (STIs) are limited. STUDY: A cross-sectional survey among 454 female sex workers (FSWs) in a Chinese county. RESULTS: Vaginal douching was reported by 64.7% of the women. The prevalence of self-reported history of STI and that of current STI was 19.4% and 41.5%, respectively. Fifteen percent of the women reported consistent use of condoms with their clients and 8.4% with their regular partners. Vaginal douching was significantly associated with decreased use of condoms (with clients: OR = 0.31; with regular partner(s): OR = 0.22) and increased rate of self-reported STI history (OR = 1.95). However, there was no direct relation between douching and current STI. Over one third of the women believed that douching can prevent STI/HIV. CONCLUSION: Vaginal douching exposes FSWs to a high risk of STI/HIV. Medical professional and public health workers should correct women's misconception about the effectiveness of douching and discourage women from douching through educational activities.

The potential of comprehensive sex education in China: findings from suburban Shanghai.

CONTEXT: More and more Chinese adolescents are engaging in premarital sexual activity. As a result, the numbers of unplanned pregnancies and sexually transmitted infections (STIs) among Chinese young adults have increased markedly. METHODS: A comprehensive sex education program, including information on abstinence, contraception and healthy sexual behaviors, was carried out in a suburb of Shanghai. The program used six methods for providing information and services to unmarried 15-24-year-olds over a period of 20 months. Sexual behavior surveys were conducted among intervention participants and among controls in a comparable town, who did not receive a similar intervention; chi-square tests and logistic regression were used to compare the results. RESULTS: Participation in the intervention was not associated with delayed sexual initiation, but was associated with reduced odds that youth coerced a partner into having sex (odds ratio, 0.3) and with increased odds of contraceptive use (6.2) and condom use (13.3) during the intervention period. The greater the level of participation, the larger the protective effects. Furthermore, the proportion of youth reporting pregnancy involvement during the intervention period was significantly lower in the intervention group than among controls (19% vs. 26%). CONCLUSION: Comprehensive, community-based interventions may be effective in reaching large numbers of Chinese youth and in promoting sexual negotiation, contraceptive use, and pregnancy and STI/HIV prevention.

[Effect of high-oxygen water on enhancing anoxia endurance and anti-fatigue function and the possible mechanism].

OBJECTIVE: To investigate the effect of high-oxygen water on anoxia endurance and anti-fatigue function in mice and explore the possible mechanism. METHODS: KM mice were used. Their body weight, serum urea nitrogen, hepatic glycogen, swimming-sustaining time of mice with a load, plasma lactic acid content, survival time under anoxia or sodium nitrite poisoning were measured. RESULTS: It was found that high-oxygen water can prolong swimming time and can protract the anoxia survival time of mice under normal pressure and being afflicted with acute cerebral ischemia (P<0.05). It can also decrease the level of serum urea nitrogen after exercise (P<0.05) and increase hepatic glycogen storage (P<0.05). CONCLUSION: The results demonstrate that high-oxygen water can enhance the emergent survival ability of mice, the ability of fatigue recovery and anoxia endurance function. It also can increase the energy storage.

[Repair of perineal and adjacent defects with thoracic biblical island flaps].

OBJECTIVE: To report the experience of repairing perineal and adjacent defects with thoracic biblical island flaps. METHODS: From January 1988 to October 2003, 7 cases of perineal and adjacent soft tissue defects with thoracic biblical island flaps, aged 17-52 years. Of 7 cases, there were 2 cases of severe scar contracture due to burn on perineal, 1 case of malignancy on perineal, 4 cases of vast soft tissue defects of trauma on the parts of groin and higher two-third thigh. The area of flaps was 9 cm x 27 cm-12 cm x 30 cm, the longest pedicle of blood vessel was 16 cm. The donor sites of flaps less than 10 cm could be sutured directly, the ones more than 10 cm could be repaired with skin grafting. RESULTS: All the flaps primarily survived. There was no ischemia and necrosis at the distal part of flaps. Four patients were followed up 6 months to 6 years. The color, texture and appearance of the flaps were good. The functions of walk and squat were satisfactory. CONCLUSION: The thoracic biblical island flap can repair perineal and adjacent soft tissue defects, moreover the donor is hidden and the effect is ideal.

[The use of superficial cervical artery island skin flaps in the repair of the maxillofacial and cervical scar].

OBJECTIVE: To investigate the method of harvesting of superficial cervical artery island skin flap, and its clinical application in the repair of the maxillofacial and cervical scars. METHODS: The origin, course, branches and distribution of the superficial cervical artery were studied in 10 adult cadavers. The superficial cervical artery could be divided into three segments: i.e. the segment before entering the trapezius muscle, the segment in the trapezius muscle, and the segment emerging from the trapezius muscle. Fourteen patients with maxillofacial and cervical scar contracture were enrolled in the study. The postburn scars were removed, and the wounds were covered by superficial cervical artery island skin flaps. The survival of the flap after the operation and the recovery of the neck movements were observed. RESULTS: The length of superficial cervical artery before entering the trapezius muscle was 5.1 +/- 0.4 cm, while that inside the trapezius muscle was 2.1 +/- 0.5 cm, and that after emerging from the trapezius muscle was 4.7 +/- 0.7 cm, respectively. The internal perforating branch was located 7.3 +/- 0.6 cm beside the seventh cervical vertebrae, and 3.9 +/- 0.7 cm above the spine of scapula. The flap was from 16 cm x 7 cm to 35 cm x 12 cm in size. All the flaps survived, except one with partial necrosis in the distal part (3.0 cm x 1.5 cm) of the flap. Follow-up observation for 4 months to 3 years showed satisfactory results. CONCLUSION: Superficial cervical artery island skin flap is simple in the flap preparation, and there is no necessity to cut the pedicle, thus it is particularly suitable for the reconstruction of severe postburn maxillofacial and cervical contracture.

[Screening for lymphatic metastasis-associated genes in mouse hepatocarcinoma cell lines Hca-F and Hca-P using gene chip].

BACKGROUND & OBJECTIVE: Metastasis is a complex process involving multiple genetic changes. The high mortality and poor prognosis caused by metastasis in malignant tumor patients and the uncertain mechanisms are always the prominent problems in the field of oncology. In order to screen for lymphatic metastasis-associated genes, the gene expression profiles of mouse hepatocarcinoma cell lines Hca-F (highly metastatic) and Hca-P (low metastatic) were compared by gene chip. METHODS: Total RNA was isolated from Hca-F and Hca-P cells, and synthesized into double-stranded cDNA, then synthesized into biotin-labeled cRNA probes by in vitro transcription. The cRNA probes were separately hybridized with Affymetrix GeneChip MOE430A (containing 22,690 transcripts, including 14,500 known mouse genes and 4,371 ESTs), and the signals were scanned by the GeneArray Scanner. The results were analyzed by bioinformatics. RESULTS: Compared with the gene expression profile of Hca-P cells, 901 (6.2%) genes and 129 (3%) ESTs were up-regulated by at least 2 folds in Hca-F cells; 33 genes, including endoglin (EDG; CD105), Mcam (Muc18; Mel-CAM; CD146), Cdc42ep5 (CEP5; Borg3), Ptprr (protein tyrosine phosphatase, receptor type, R), F2r [coagulation factor II (thrombin) receptor; Par1; ThrR], D7Ertd458e (necl-5), NR1D1, Serpin h1 (HSP47), AXL, Mak, and Areg (AR), were up-regulated 13.93-29.86 folds. According to Gene Ontology and Treeview analysis, these 33 genes were involved in angiogenesis, cell adhesion, signal transduction, cell motility, transcription, chaperone activity, protein kinase activity, receptor binding, and so on. CONCLUSION: Many lymphatic metastasis-associated genes were screened by high-throughput gene chip method; validating their cellular functions will help to identify the key or candidate gene/pathway responsible for lymphatic metastasis, which might be used as diagnostic markers and therapeutic targets for lymphatic metastasis.

[Expression of glycoprotein 90K in non-Hodgkin's lymphoma and its clinical significance].

BACKGROUND & OBJECTIVE: 90K, a kind of secretory glycoprotein, is related to prognosis of some tumors. So far, little is reported on its relation to prognosis of non-Hodgkin's lymphoma (NHL). This study was to explore the 90K expression in naive NHL tissues and its clinical significance. METHODS: 90K expression in 110 specimens of NHL was detected by immunohistochemistryû its correlations to short-term treatment efficacy of CHOP regimen and long-term survival of the patients were analyzed retrospectively. RESULTS: Of the 110 NHL specimens, 36 (32.7%) showed negative expression of 90K, 27 (24.5%) showed weak expression, 17 (15.5%) showed fairly strong expression, and 30 (27.3%) showed strong expression; the positive rate of 90K in NHL tissue was 67.3%. 90K expression had no correlation to gender, age, IPI and other clinicopathologic characteristics. All patients received treatment of standard CHOP regimen; the response (complete remission and partial remission) rate was significantly higher in low 90K expression (negative and weak expression) group than in high 90K expression (fairly strong and strong expression) group [82.5% (52/63) vs. 65.2% (30/46), P=0.039], but 90K expression had no correlation to long-term survival (P > 0.05). CONCLUSIONS: 90K expression may be applied to predict short-term efficacy of CHOP regimen on NHL. However, whether 90K could become a prognostic tumor marker of NHL needs further research.

[Hypermethylation of fragile histidine triad gene and 3p14 allelic deletion in ovarian carcinomas].

OBJECTIVE: The FHIT (fragile histidine triad) is a candidate tumor suppressor gene (TSG) located on chromosome 3p14.2. Hypermethylation and loss of heterozygosity (LOH) are major mechanisms in the inactivation of tumor suppressor genes. In this study, the methylation status of FHIT and LOH of 3p14 in 61 cases of human sporadic ovary carcinomas were investigated. METHODS: Sixty-one primary ovary carcinomas and 10 borderline ovarian tumors were analyzed with methylation specific PCR (MSP) to detect the CpG island methylation status in the FHIT promoter region. In addition, 45 cases of ovary carcinomas and their corresponding non-tumor ovary tissues were investigated with D3S1287 microsatellite polymorphic marker for LOH. RESULTS: Hypermethylation of FHIT gene was observed in 39.3% (24/61) of ovarian carcinomas. The frequencies of hypermethylation in serous ovarian carcinoma, mucinous ovarian carcinoma, endometrioid ovarian carcinoma and ovary borderline tumor were 45.2% (19/42), 14.3% (1/7), 33.3% (4/12) and 60.0% (6/10), respectively. Ten of twenty-three (43.5%) informative tumors showed LOH and 6 of 18 (33.3%) informative cases showed homozygous deletions. The status of FHIT methylation was not associated with clinical stage and differentiation grade, there was no significant difference between the malignant and borderline tumors. CONCLUSION: Hypermethylation and allelic deletion of FHIT are frequent events in ovarian carcinomas and are important mechanisms for the loss of expression of this gene.

[Protective effect of protease inhibitor on renal function of rat challenged by lipopolysaccharide].

OBJECTIVE: To investigate the protective effects of protease inhibitor on lipopolysaccharide (LPS)-induced kidney injury in rats and its possible mechanism. METHODS: Thirty-two male Wistar rats were divided into four groups randomly: normal control (n=6); LPS group (n=10), receiving intravenous endotoxin (LPS, O55:B5, 5 mg/kg); low-dose ulinastatin (UT) intervention group (n=8), receiving intraperitoneal UT 50 000 U/kg and LPS 5 mg/kg as above; high-dose UT intervention group (n=8), UT 100,000 U/kg and LPS 5 mg/kg. The following examinations were carried out: blood gas analysis, kidney pathological changes, plasma endothelin-1, plasma lactic acid, N-acetylglucosaminidase (NAG) in urine, and plasma creatinine (Cr) level. RESULTS: Blood gas analysis showed that pH, partial pressure of oxygen in artery (PaO(2)) and base excess (BE) lowered significantly (all P<0.01) in LPS group compared with normal control group, and elevated in low-dose and large dose UT intervention groups compared with LPS group, the differences were statistically significant (P<0.01). The plasma endothelin-1 increased significantly (P<0.01) in LPS group compared with normal control group and intervention groups. Plasma lactate increased significantly in LPS group compared with normal control group (P<0.001), decreased significantly in intervention groups compared with LPS group (P<0.01). No significant difference was found between two intervention groups (P>0.05). Plasma Cr and urine NAG level increased in LPS groups, and the difference was significant compared with normal control groups and intervention groups (P<0.01). Pathohistologic examination revealed normal glomeruli but vacuolar degeneration of tubular epithelial cells, and part of them disrupted and desquamated, and also tubular dilatation. Only mild pathological changes were seen in the intervention groups. There was no obvious difference in morphology between two intervention groups. CONCLUSION: The protease inhibitor, UT, may alleviate LPS-induced inflammatory reaction and damage to kidney in rats.
