Effect of unilateral nephrectomy on renal function of diabetic rats.

Glomerular alterations of experimental diabetes mellitus are observed in animals submitted to a reduction in renal mass, suggesting that some mechanisms responsible for the progression of renal disease are common. The aim of this study was to investigate the effect of nephrectomy on the renal function and morphology of diabetic rats. Male Wistar rats were divided into 4 groups: control (C), n=8; diabetic (DM), n=8; non-diabetic nephrectomized (Nx), n=8; (DMNx), n=9. DM was induced by streptozotocin (65 mg/Kg), and animals were treated with insulin. After 12 weeks, the glomerular filtration rate (GFR), renal plasma flow (RPF) and mean arterial pressure (MAP) were evaluated in unanaesthetized animals. Glomerular volume (GV), glomerular sclerosis index (GSI), mesangial volume density (Vvmes) and glomerular capillary surface density (Svcap) were also evaluated. Results show that kidney weight increased in Nx groups, being higher in DMNx. GFR was higher in Nx groups as was RPF, being higher in DMNx. RVR was lower in Nx groups, especially in DMNx. MAP was not different among the groups. RPF and GFR showed a high correlation for the DMNx group (r=0.95, p=0.02). The DMNx group showed a correlation between RVR and GFR (r=-0.96, p=0.005). The GV increased in Nx groups, and the GSI was higher in DMNx. Vvmes and Svcap increased in DMNx group. In summary, Nx groups developed similar degrees of glomerular hypertrophy, but only DMNx showed an increased value for GSI. The present data suggest that the acceleration of glomerular lesions in DMNx animals was more closely associated to hemodynamic adaptations than to glomerular hypertrophy.

The effect of enalapril and verapamil on the left ventricular hypertrophy and the left ventricular cardiomyocyte numerical density in rats submitted to nitric oxide inhibition.

Forty male Wistar rats were separated into four groups of ten rats each (control and other three groups that have received nitric oxide (NO) synthesis inhibitor L-NAME) but the last two groups have concomitantly received antihypertensive drugs (Enalapril and Verapamil). After 40 days of experimentation, the heart and the ventricles were measured. The optical disector was used for the calculation of numerical density of the cardiomyocytes (Nv[c]). The left ventricular myocytes number (N[c]) was calculated as the product of Nv[c] and the left ventricular myocardium volume (LVMV) that was determined by using the Scherle's method. In the L-NAME group the blood pressure (BP) had a significant weekly increment. In the enalapril and the verapamil groups, BP increased in the first two weeks, but decreased in the following weeks. The LVMV increased in the L-NAME rats and decreased in the enalapril and verapamil animals. The Nv[c] and N[c] decreased in the L-NAME rats but the verapamil and enalapril treatments maintained the Nv[c] close to the control group. In conclusion, the left ventricular hypertrophy and the significant decrease of the left ventricular cardiomyocyte number caused by the NO synthesis inhibition are efficiently prevented with the use of enalapril and verapamil.

Cranial and mandibular morphometry in Leontopithecus Lesson, 1840 (Callitrichidae, primates).

In this paper, we report on a craniometric analysis comparing the species of lion tamarins, Leontopithecus Lesson, 1840. Seventeen cranial and mandibular measures were taken on skulls of 59 adult crania: 20 L. rosalia (14 females and 6 males); 13 L. chrysomelas (6 females and 7 males); 23 L. chrysopygus (8 females and 15 males), and 3 L. caissara (1 female and 2 males). All specimens were from the Rio de Janeiro Primate Center (CPRJ-FEEMA, Brazil), except the specimens of L. caissara. Statistical treatment involved a one-way analysis of variance (the Bonferroni test) and discriminant analysis, comparing cranium and mandibles separately to determine variables which best distinguished groups and to group the specimens, using size corrected methods. The Mahalanobis distance was computed from the centroids of each group. Seven measures distinguished females of L. chrysopygus with L. rosalia, six to L. rosalia with L. chrysomelas, and L. chrysopygus with L. chrysomelas. In males, the numbers of measures statistically different were 5, 4, and 3 of the pairwise comparisons above mentioned. Cranial base length and orbital breadth were the only measures that were significantly different in all three dyads, considering both sexes. For the cranium, function 1 of the Discriminant Analysis accounted for 52.4% of the variance and function 2 accounted for 40.3%. Both functions exhibited a significant value for Wilks' lambda (P<0.0001) and 96.6% of specimens were correctly classified. For the mandible, the first two functions provided a significant discrimination 51.1% and 44.9%, respectively, and 69.5% of the correct classification. Orbital breadth and cranial base length contributed most in the cranial analysis, while mandibular length and mandibular body height to mandibular ones. The analyses performed in this study (univariate and multivariate) demonstrated that cranial and mandibular morphology is significantly different among species of Leontopithecus. Despite of sample size, L. caissara shows morphological distances to L. chrysopygus in cranial analysis. However, other investigations are necessary to confirm this.

Myocardial microcirculation stereological changes in rats subjected to nitric oxide synthesis inhibition.

This work aims to study stereological changes in intramyocardial blood vessels in rats submitted to nitric oxide (NO) synthesis inhibition within different periods. NO synthesis inhibition was achieved by administration of L-NAME (50 mg/kg/day); control and L-NAME rats were sacrificed 25 and 40 days after experimentation. Light microscopy and stereology [according to references 7, 13 and 14] were used for analyzing the myocardium. Arterial blood pressure and cardiac weight increased by 74.5% and 57.8% after 25 days and by 90.2% and 34.6% after 40 days, respectively. Comparing the L-NAME rats with corresponding controls revealed that the volume density of the vessels decreased by 31.3% after 40 days, and the length density by 53.5% after 25 days and by 25.7% after 40 days. The mean cross-sectional area of the vessels increased by 154.6% after 25 days. In this study on intramyocardial vessels, we observed an important decrease of the length density in L-NAME animals. Likewise, the volume density also decreased significantly in L-NAME animals. The mean cross sectional area of the vessels, which normally increases during cardiac growth between 25 and 40 days, was precociously increased in L-NAME animals at 25 days, suggesting that these animals suffer from a precocious increase of the heart (including blood vessels) due to pressure overload. Stereology of cardiac microvessels revealed remodeling of these vessels in rats under NO synthesis inhibition. Although these changes may be caused by NO inhibition and not by arterial hypertension, further comparative studies on different models of arterial hypertension are needed to confirm this hypothesis.

Quantitative study of myocardial microcirculation in arterial hypertension due to progressive inhibition of NO synthesis.

OBJECTIVE: To study the quantitative changes in intramyocardial blood vessels in rats in whom nitric oxide synthesis was inhibited. METHODS: Four groups of 10 rats were studied: control (C25 and C40) and L-NAME (L25 and L40). The animals L25 and L40 received L-NAME in the dosage of 50mg/kg/day for 25 and 40 days, respectively. On days 26 and 41 the animals in groups 25 and 40 were sacrificed. Analysis of the myocardium was performed using light microscopy and stereology. RESULTS: Arterial blood pressure and heart weight increased 74.5 and 57.8% after 25 days and 90.2 and 34.6% after 40 days, respectively. Comparing the L-NAME rats with the respective controls revealed that vessel volume density decreased 31.3% after 40 days, and the vessel length-density decreased 53.5% after 25 days and 25.7% after 40 days. The mean cross-sectional area of the vessels showed an important reduction of 154.6% after 25 days. The intramyocardial vessels decreased significantly in length-density in the L-NAME animals. The mean cross-sectional area of the vessels, which normally increases during heart growth between 25 and 40 days, showed a precocious increase by the 25th day in the L-NAME rats. This suggests an increase of the size of the heart, including blood vessels. CONCLUSION: The inhibition of the NO synthesis provokes rarefaction in the intramyocardial vessels that progresses with the time of administration of L-NAME.

[Stereology of the human epididymis in different groups of age: fetus, children, adolescents and elderly].

The importance of abnormalities of function and epididymis structure in the etiology of male infertility is still not well understood. We studied 52 individuals distributed in five age groups: fetuses, children, adolescents, adults and the elderly. The region of the body of epididymis was obtained by autopsy and immediately fixed by immersion in a solution of 10% buffered formaldehyde, embedded in paraffin and histologically prepared. The samples were observed under an optic microscope. Test-points were counted in 12 random microscopic fields with the M42 test-system. The following stereological parameters were determined: ductal area, volumetric densities (Vv) of the duct, smooth muscle, connective tissue, epithelial duct and blood vessels. The main results distinguished by those whose averages were statistically significant (p < 0.05), showed that the ductal area is 9.7 times greater in the adolescent/adult/elderly group than the children's group. The Vv of the lumen of the epididymis duct occupies 11.7% of the epididymis body in the fetal period, 5.3% in the child and in individuals after puberty this figures reaches more than 15%. The Vv of smooth muscle occupies 28.3% of the body of the epididymis in the fetus and 35.9% in children, but after puberty this figures stays around 22%. The Vv of the connective tissue occupies 26% in prenatal life, 37% in children, and after puberty these figures range from 21 to 27.5%. Comparing the results of the adult group with that of the elderly group there is an increase in the volumetric density of the connective tissue by 18.1%. In conclusion, the epididymal duct area and the Vv of the ductal lumen, smooth muscle and connective tissue were significant comparing the different groups. However, the quantitative relative differences of the duct's epithelium and the blood vessels were not significant comparing these groups. The study of quantitative aspects of the normal human epididymis can increase our knowledge about male fertility.

[The collector system of the kidney. Applied anatomy based on the analysis of 3-dimensional casts].

The kidney collector system were studied in 120 three-dimensional casts obtained by the injection-corrosion method. To the procurement of the first hundred casts, we made use of a polyester resin; however on the preparation of the last twenty we made use of a mixture containing latex and radiographic contrast as we intended to obtain, besides the casts, the roentgenograms. Two anatomic details, which have a great importance for endourology, were observed: a. the calices' cross draining the meso-renal region and the consequent inter-pelvis-calice (space (IPC) formation which is seen in the roentgenograms, b. a smaller calice which penetrates perpendicularly the surface of the renal pelvis or into the surface of a great calice. The urologists must appreciate these two anatomic structures, before the surgical procedures on the collector system.

Mast cells in tissue response to dentistry materials: an adhesive resin, a calcium hydroxide and a glass ionomer cement.

Synthetic materials used in dentistry may trigger various inflammatory responses. In order to evaluate biocompatibility, standardized implants of Calcium Hydroxide (CH), Glass Ionomer Cement (GIC) and Light-activated Dental Adhesive (LDA) were surgically introduced into Wistar rats' back bone. Six (experimental) animal groups, five each, and two Sham (S) groups were studied after 15 and 30 days from surgery. In each animal, the density of mast cells and interstitial fibrosis volume was evaluated by quantitative light microscopy. In addition, the interaction between the disk material and its fibrous capsule was evaluated by scanning electron microscopy. The density of mast cells per area (N(A)[mast cells]) was lower in CH group than in LDA group. GIC group displayed N(A)[mast cells] results intermediate between CH and LDA groups (p<0.05). The smallest interstitial fibrosis volume density (Vv[f]) was observed in CH group, then in GIC group, while the greatest in LDA group. After 30 days, the fibrosis in LDA group was 30% higher than in CH group (p<0.05). In S group, discreet fibrosis restricted to surgical area was present, with few mast cells near the vessels. Significant interaction between fibrous capsule and the surrounding disk material was most evident in CH group. The implanted materials induced mast cell migration, distinct fibrosis development, suggesting that CH is the most biocompatible material among those tested.

Numerical density of cardiomyocytes in chronic nitric oxide synthesis inhibition.

OBJECTIVE: The myocardial effect on the normalization of arterial blood pressure in animals with hypertension previously induced by nitric oxide synthesis (NOS) inhibition is still unknown. The purpose of this study was to estimate the numerical density of cardiomyocytes that is able to show cardiomyocyte loss as a consequence of NOS inhibition. METHODS: Sixty rats were divided into the following groups: control for 25 days (C25), control for 40 days (C40) control for 80 days (C80) and three other groups in which the rats received the NOS inhibitor N(G)-nitro-L-arginine-methyl-ester hydrochloride L-NAME; 50 mg/kg/day for 25 days (L25), 40 days (L40) and 40 days, respectively, the latter group having another 40 days of only water and food ad libitum (without L-NAME; L80 group). The detection of apoptotic cells was performed using a monoclonal antibody. RESULTS: In the L25, L40 and L80 groups, blood pressure was 74.5, 90.2 and 56.3% higher than the respective age-matched control rats, the myocardium had hypertrophy of the cardiomyocytes and scattered areas of fibrosis, and apoptosis occurred in isolated cells. Compared to the controls, the heart mass/body mass ratio was significantly greater in the L-NAME groups L25, L40 and L80, i.e. 31, 26 and 21%, respectively, the numerical density of cardiomyocytes in L-NAME rats was 32.7, 48.8 and 41.7% lower and the mean volume of cardiomyocytes was 33, 53 and 48% greater. CONCLUSION: The cardiomyocyte loss in NOS inhibition seems to be mainly due to necrosis, although apoptosis is also present.

[Role of nitric oxide synthase in the etiopathogenesis of hypertrophic pyloric stenosis in infants]

OBJECTIVE: To experimentally reproduce, in rats, the findings corresponding to the histopathology of infantile hypertrophic pyloric stenosis (IHPS), using nitric oxide synthase (NOS) inhibitor (L-NAME). METHODS: L-NAME was administered to pregnant rats (L-NAME group), from the 14th gestational day on in order to reproduce the model of NOS inhibition in the production of IHPS. This group was then compared to control animals. After birth, all the animals in the L-NAME group were maintained under NOS inhibition until the 42nd day of life, when they were sacrificed. The control animals, which did not receive any kind of drug, were also sacrificed on the 42nd day of life. The animals and their internal organs were analyzed and weighed. The pyloric region was technically prepared and observed through light microscopy. RESULTS: The L-NAME group presented lower body and intestinal weight and higher gastric weight than the control group. Light microscopy revealed hypertrophy of the circular smooth muscle layer of the pyloric muscle in L-NAME animals. CONCLUSIONS: This work reproduced an experimental model of an IHPS study, confirming the effect of NOS blockade on the pyloric musculature.

Stereology of the pterygopalatine ganglion of the rat.

The right pterygopalatine ganglia (PG) of 9 male Wistar-strain rats were dissected, embedded in Epon (3 specimens) or paraffin (6 specimens), and prepared for stereological examination under light microscopy. The perikarya were quantitatively characterized, and the ganglionic volume was determined. Stereology is an efficient method for the quantitative evaluation of the perikarya of the PG. The results(expressed as mean +/- standard deviation) were: a) areal fraction occupied by the perikarya = 53.8 +/- 7.4%; b) the perikaryal surface area per volume = 0.101 +/- 0.013 microns-1; c) the number of perikarya per volume x 10(-5) = 5.26 +/- 0.99 microns-3; d) the mean profile area of the perikarya (apk) = 505.93 +/- 78.29 microns 2; e) the mean perikaryal volume (vpk) = 9,179.33 +/- 1,533.52 microns 3; and f) the ganglionic volume = 0.210 +/- 0.127 mm3. The low coefficient of variation the apk and vpk values suggests the presence of only one population of neurons in the PG of the rat. The number of perikarya in the PG is about 11,046 per ganglion. As compared to analogous data in the otic ganglion of the rat, the PG did not show statistically significant stereological differences, but the relatively higher number of neurons found in the PG is probably associated with the higher functional activity of this ganglion.

Allometry of hepatic weight growth in human staged fetuses.

The growth of the hepatic weight was correlated to fetal parameters of maturity (gestational age, crown-rump length and weight) in 70 human fetuses ranging from 14 to 39 weeks post-conception using the allometric method. The growth of the hepatic weight presents moderate positive allometry relative to age and C-R length, and isometry relative to fetal weight. The coefficiens of correlation are very high (p less than 0.001, Table 1). This study presents growth curves of the hepatic weight useful in medical branches such as anatomy, forensic medicine, fetopathology, medical imagery, onstetrics and pediatrics.
