Uptake of oestradiol by the rabbit hypothalamus. Specificity of binding by nuclei in vitro.

The binding of [6,7-(3)H]oestradiol-17beta to subcellular fractions of the hypothalamus and the cerebellum of the rabbit was studied in vitro. Uptake of steroid was higher in hypothalamic nuclei than in cerebellar nuclei. Lower binding was observed in other fractions of both tissues. After dialysis of the fractions, hypothalamic nuclei retained a high percentage of oestradiol whereas cerebellar nuclei lost most of the bound steroid. Supernatant fractions of both tissues retained a significant proportion of label after dialysis and after gel filtration on Sephadex G-200. No specific binding was observed in these fractions when subjected to sucrose-density-gradient centrifugation. Purification of nuclei followed by incubation with labelled oestradiol in the absence of the supernatant fraction resulted in loss of binding of steroid by hypothalamic nuclei. Incubation of the purified hypothalamic nuclei with supernatant fraction maintained the binding specificity of hormone retention.

Actinomycin D: uptake by sea urchin eggs and embryos.

Actinomycin D is excluded from unfertilized eggs and developing embryos of the sea urchin Arbacia punctulata until the blastula hatches. The rate of uptake of actinomycin D by embryos doubles as development progresses after hatching to the gastrula stage.

Partial characterization of the RNA stimulating growth of the seminal vesicle.

RNA prepared from the seminal vesicles of intact adult rats, or of testosterone-treated castrate adult rats, produced a 30 per cent increase in the weight of the seminal vesicle and a 25 per cent increase in the incorporation of labeled amino acids into protein. RNA prepared from the liver or ventral prostate of the same animals produced only a 10 per cent increase in the weight of the seminal vesicle. RNA prepared from the liver of control or testosterone-treated adult female rats was ineffective in stimulating the growth of the seminal vesicle. Polyuridylic acid instilled into the lumen had no effect on the weight of the seminal vesicle, but increased the incorporation of labeled phenylalanine into protein. RNA sedimenting in the 18S area on sucrose density gradients was most effective in stimulating growth of the seminal vesicle of three-week-old rats. Treating homogenized tissue with deoxycholate greatly increased the effectiveness of the RNA extracted. RNA extracted from seminal vesicles by treatment with deoxycholate and dextran sulfate was biologically active, whereas the usual 4S RNA in the 100,000 x g supernatant is ineffective.
