Advance shift of feeding circadian rhythm induced by obesity progression in Zucker rats.

To determine the relation between the circadian rhythm of ingestive behavior and the progression of obesity in Zucker rats, ingestion and ambulation were analyzed at four different developmental stages. The obese rats were disrupted gradually in nocturnal patterns of feeding, drinking, and ambulation with the progression of obesity, although the lean littermates maintained the patterns during whole test periods. Analysis of autocorrelogram revealed that circadian rhythms of those behaviors remained throughout the whole test periods. Least-squares spectrum ascertained the following. 1) The obese made advance shift of acrophases in feeding and drinking circadian cycles, but not in ambulation. 2) Amplitudes in those behavioral measures decreased with the progression of obesity. 3) Mesor in the obese feeding was not affected, although that in the lean feeding decreased. The findings indicate that disruption of the light-dark cycle in ingestion of the obese was not due to disappearance of circadian rhythm but to transformation by both decreased amplitude and advance shift of the circadian cycle.

Impaired hypophagia induced by fecal anorexigenic substance in Zucker obese rats.

Influence of fecal anorexigenic substance (FS-T) on feeding by Zucker obese rats was compared to that by their lean littermates and Wistar King A rats. FS-T, which has been found to suppress food intake mainly by activation of glucoreceptor neurons in the ventromedial hypothalamus, was injected intraperitoneally in a dose of 7 U/kg at 1930 hr, immediately before the dark period. Potency of FS-T in feeding suppression was much less in the obese rats than in their lean littermates or the Wistar King A rats. Meal size of the obese rats was decreased after the injection, but meal duration was unaffected. The suppressive effect on the lean rats and the Wistar King A rats included decrease of both size and duration of the meal. These results suggest that chemosensitivity in the ventromedial hypothalamus of Zucker obese rats may be impaired, which may be one explanation of the obesity in Zucker obese rats.

Statistical evaluation of factors influencing prognosis of gastric cancer patients: Predication of prognosis on patient clusters.

We found ten clusters of gastric cancer patients in Imanaga's group under a cancer research project organized by the Ministry of Health and Welfare, and evaluated the prediction of prognosis of those patients in each cluster by using the censored regression of postsurgical survival time on a "prognosic" factor which has been extracted from nine explanatory variables observed mainly at the time of surgery. Consequently, the ten clusters were interpreted and confirmed to be useful for prediction of the patient prognosis by comparison of the failure rates among those clusters and between treated (administration of chemotherapy) group and control group.

Statistical principle and methodology in the NISAN system.

The NISAN system is a new interactive statistical analysis program package constructed by an organization of Japanese statisticans. The package is widely available for both statistical situations, confirmatory analysis and exploratory analysis, and is planned to obtain statistical wisdom and to choose optimal process of statistical analysis for senior statisticians.
