Cyclo-oxygenase- and capsaicin-sensitive afferent fibres affect Beta-adrenoceptor-evoked response in the rat urinary bladder.

As reported previously, the beta-adrenoceptor-evoked response in the in vitro rabbit detrusor is inhibited by non-steroidal anti-inflammatory drugs (NSAIDs). Prostaglandins can restore this response. This study was designed to better define physiological influences on beta-adrenoceptors in the urinary bladder and to ascertain whether the observations made in the rabbit urinary bladder in vitro were valid in vivo, as well as in a different animal species, i.e. in the rat. The NSAID indometacin partially inhibited the isoproterenol-evoked response in the rat detrusor muscle. When the neurokinin A (NKA) antagonist MEN-10376 was administered after indometacin, the beta-adrenoceptor-evoked response was completely abolished. The isoproterenol-evoked beta response was dose dependently reduced following treatment with the capsaicin antagonist ruthenium red. Taken together, these data indicate an involvement of prostanoids, NKA- and capsaicin-sensitive fibres in the beta-adrenoceptor-evoked response in the rat urinary bladder.

Family backgrounds and eating disorders.

The aim of this study was to assess the possible relationship between the presence of a pathological family background and various eating disorders subgroups. A semi-structured interview was used to assess the socio-demographic and clinical characteristics and the presence of psychological complaints among family members of 79 subjects with anorexia nervosa (AN) and 34 subjects with bulimia nervosa (BN). The subjects were also administered the following self-assessment questionnaires: BITE, EDI, and HSCL-90. There were nonsignificant differences between AN and BN in terms of parental mental disorders. A further subdivision of the patients (as indicated in DSM-IV) revealed significant differences in the distribution of psychiatric family history. In particular, it seems that the presence of purgative behavior is associated with a higher incidence of a pathological family background. These results suggest that pathological family histories are not responsible for the development of ED, but they are an aggravating factor both in AN and BN.

[Prognostic factors in acute nonlymphoid leukemias].

Our retrospective study was aimed at assessing parameters affecting the prognosis of acute non lymphoid leukemia (ANLL). Since 1988 to 1994 we observed 84 patients: 52 males, 32 females. For each patient we considered at diagnosis: age, fever, performance status, platelets, hemoglobin and white blood cell count, extramidollary disease, bone marrow blastosis, phenotype and cytogenetic abnormalities of blasts cells. All the parameters listed above were correlated with the time to achieve the complete remission (CR), CR duration and the overall survival. Statistical tests as t-student and chi square test were used. Statistical analysis of the parameters considered revealed that the only value affecting the achievement of a CR was the age. The prognostic significance of immunophenotyping in ANLL has been a controversial issue, with a number of conflicting reports. In our study only the terminal deoxynucleotidyl transferase was significantly associated with prognosis. Our study, as data reported in literature, confirms that the prognostic impact of the various parameters in ANLL is controversial. The study of prognostic factors and of the immunophenotype is important to identify the clinical and the biologic profile of the disease and to evaluate the optimal post-remission treatment.

Photoexcitation of the methionine-iron bond in iron(III) cytochrome c: bimolecular reaction with NADH.

When iron(III) cytochrome c aqueous solutions containing NADH are irradiated with polychromatic light (wavelength greater than 280 nm), iron(II) cytochrome c and NAD+ in the stoichiometric ratio 2/1 are observed to be the principal reaction products, independently of the presence of oxygen; in addition, a minor process due to direct photodegradation of the nucleotide is observed. The selection of monochromatic 290 nm irradiation light (at which NADH has an absorbance minimum) and an adequate reactant concentration allowed parallel reactions to be minimized and new information to be obtained on the mechanism of the photoredox process. The experimental results are consistent with a reaction mechanism whereby NADH donates one electron to a "reactive intermediate" of the hemoprotein formed from the light-induced methionine-to-iron charge transfer excited state. In this process an NAD. radical is formed which, in deaerated solution, immediately reduces another molecule of the hemoprotein, and is itself oxidized to NAD+. In aerated solution, the NAD. radical rapidly reacts with oxygen to give NAD+ and superoxide O2- anion radical which, in turn, reduces the second iron(III) cytochrome c molecule.

Immunity assessment in the early stages of amyotrophic lateral sclerosis: a study of virus antibodies and lymphocyte subsets.

Serum and CSF from 32 patients with idiopathic ALS, 30 age-matched controls and 30 MS patients were investigated regarding immunoglobulin concentration and virus-specific antibodies, the lymphocytes in the peripheral blood and lymphocyte subsets were also investigated. ALS patients' results were compared with findings in MS and controls. The ALS patients had significantly higher IgG concentration in serum than the controls, marked lymphopenia, reduction of CD2, CD8 and Leu 7 positive cells and increase of the CD4/CD8 ratio and of SIg-positive lymphocytes. Compared with the MS patients, the ALS patients showed similarity in T-subset distribution with a lower standard deviation. No HTLV-I and HIV antibodies were found in any group and no significant differences in antibody distribution to Toxoplasma G, herpes simplex, cytomegalovirus, measles and mumps viruses were evident. All ALS patients were investigated at an early disease stage, therefore, our findings seem to support the conclusion that the immune alterations are related to the mechanisms of the disease and not to complications of its evolution.

[Reduction of cytochrome c by NADH induced by light].

We have studied the behaviour of Fe(III) cytochrome c upon irradiation in the 290-360 nm wavelength range either in the presence or in the absence of NADH; in both cases the photoexcitation caused the reduction of the heme iron. When the irradiation was performed in the absence of NADH, the iron reduction was coupled to a non reversible modification in the protein structure; the photoreduction quantum yield was decreasing with the increase of the irradiation wavelength. Irradiation in the presence of NADH gave heme iron reduction coupled to NADH oxidation and the protein resulted finally unmodified; the quantum yield depended on the irradiation wavelength in a way similar to the observed in the absence of NADH, but it was tenfold higher. We propose that in both cases the active species is an electronic excited state of the heme iron.

[Acupuncture and micro-massage in the treatment of idiopathic nocturnal enuresis].

1) Goals of the Research. We have treated 15 cases of night enuresis (4-13 year) 9 of which had already been treated with other therapies without success, in order to compare the two methods. 2) Methods employed. A) Acupuncture in 10 cases at the following loci, alternatively every other day, with an average number of treatments of 10 per patient: treatment a) C7-E36-RP6-VC3; treatment b) V23-V28-V32. B) Micromassage on the points "Nicturia" in the creases between the first and the second and the third phalanx of the 5th finger (P. Chang) in 5 cases; the first treatment at the presence of parents who have then repeated treatments at home every night for ten minutes, for an average of 20 days. 3) Results and summary. With acupuncture: 7:10 complete recovery, 2 partial recoveries, 1 negative result. With micromassage: 2:5, complete recovery, 1 partial recovery, 2 negative results. Both groups were followed-up up to 4 months after the last treatment. Acupuncture produces better results in percentage, however the second method is, for its simplicity, a valid alternative mostly in little children who reject acupuncture and it should be further investigated.

Interactions between A-9THC and capsaicin on isolated lamb bladder detrusor.

A number of studies have demonstrated that capsaicin, a capsicum alkaloid, can affect isolated bladder tissue with either a relaxation or a contraction, depending on the species, by acting on VR1 receptors. In a previous work on isolated lamb detrusor, we demonstrated that capsaicin generally produces a relaxation of the tissue; this relaxation seems to be mediated by CGRP. Endogenous cannabinoids, such as anandamide, produce some of their actions by stimulating VR1 receptors and this seems to cause the release of peptides, e.g. CGRP. The aim of this work was to ascertain whether a cannabinoid, delta-9-tetrahydrocannabinol (delta-9THC), was able to interfere with the response of the isolated lamb detrusor to capsaicin. A-9THC, at concentrations between 1.6 x 10(-7) and 1.3 x 10(-6) M, displayed no activity on tissues. Instead, following delta-9THC, most of the tissues responded to capsaicin with a contraction that was abolished by atropine (9.0 x 10(-7) M). It has been reported that cannabinoids can inhibit the release of CGRP by stimulation of CB1 and CB2 cannabinoid receptors. Delta-9THC could act stimulating these receptors and thus inhibiting CGRP release and vesical relaxation. The muscle relaxing component removal could favour the contracting component, usually not active.
