Evidence for the presence of several phosphodiesterase isoforms in brown adipose tissue of Zucker rats: modulation of PDE2 by the fa gene expression.

The present study was undertaken to characterise the phosphodiesterases (PDEs) present in brown adipose tissue (BAT) of Zucker rat pups and to determine whether the capacity for degradation of cyclic nucleotides was affected by the fatty genotype. Regardless of the genotype, PDE2-4 contributed to total PDE activity, the PDE3 activity equalling the sum of PDE2 and 4 activities. In fa/fa compared to Fa/fa rats, (a) PDE2 activity was significantly increased, (b) Western blot analysis of PDE2 revealed two signals at 71 and 105 kDa, with changes in protein being in good parallelism with changes in activity, (c) the PDE2 mRNA concentration was also significantly increased. In good agreement, the cGMP concentration was decreased in BAT from fa/fa pups.

Lipid peroxidation and antioxidant status in experimental diabetes.

Oxidative stress is currently suggested as a mechanism underlying diabetes. The present study was designed to evaluate the oxidative stress related parameters in streptozotocin-induced diabetes in rats using different complementary approaches: susceptibility to in vitro oxidation (lipid peroxidation induction in liver homogenate, red blood cells hemolysis), blood antioxidant status (total antioxidant capacity by two approaches), and plasma isoprostane measurement, a new marker of lipid peroxidation in vivo. We have shown that induced liver thiobarbituric acid reactive substances increased after 4 weeks of diabetes, in spite of increased liver vitamin E content. Red blood cells hemolysis was significantly delayed after 4 weeks of diabetes. Plasma antioxidant capacity (AOC) tended to increase after 4 weeks of diabetes and was correlated with plasma vitamin E levels. Total antioxidant activity (TAA) significantly decreased after 1 week and a significant correlation was observed with plasma albumin levels. Plasma isoprostane (8-epiprostaglandinF2alpha) concentrations were not modified significantly 1 week or 4 weeks after the induction of diabetes. Levels of vitamin E in the diet and changes in its distribution among the body seems to play an important role in the development of oxidative stress during diabetes and its consequences.

Wholewheat flour ensures higher mineral absorption and bioavailability than white wheat flour in rats.

Consumption of unrefined whole flour is thought to affect mineral bioavailability because it contains high levels of fibre and phytic acid. The present experiment was designed to study the absorption of minerals from diets based on wholewheat flour and white wheat flour in rats. Two groups of male Wistar rats were fed on the diets for 3 weeks and absorption and tissue retention of minerals were studied. The rats fed on the wholewheat flour diet had significantly greater food intake, weight gain, faecal excretion and intestinal fermentation than those fed on the white flour diet. Mineral intakes, except for Ca, were significantly greater in rats fed on the wholewheat flour diet (4-fold for Mg, 2-fold for Fe and Zn). A significant rise in the apparent absorption of Fe (%) and a significant decrease in the apparent absorption of Zn (%) were observed. The amounts of minerals absorbed (mg/d) were significantly enhanced (excepted for Ca) with the wholewheat flour diet. Moreover, plasma and tibia levels of Mg and plasma, liver and tibia levels of Fe were significantly increased in rats fed on the wholewheat flour diet compared with those fed on the white flour diet. In conclusion, wholewheat flour, rich in phytic acid and minerals, did not have a negative effect on mineral absorption, but rather improved the bioavailability of some minerals. Human studies are needed to confirm these rat results before extrapolation to human nutrition.

Influence of age and hormonal treatment on intestinal absorption of magnesium in ovariectomised rats.

This study was designed to assess the effect of age, ovariectomy and estrogen treatment on the absorption and the balance of Mg in the rat. Three groups of fifteen 6- (mature), 12- (old), and 30-month-old female rats (senescent), fed a diet containing 1.5 g of Mg/kg were used in the present study. Within each group, 10 rats were surgically ovariectomized (OVX). From day 2 until day 60 after OVX, they were s.c. injected with either solvant or 17 beta-estradiol (E: 10 mg/kg bw/48 h, n = 5). Five other rats were sham operated (SH, n = 5) and received solvent alone. Animals were pair fed 6 g/100 g bw/day and distilled water was available ad libitum. Food intake, urine and feces from each individual rat were measured 1 day per week, during the last 5 weeks. The results clearly showed that apparent Mg absorption (per cent) was not significantly altered with aging. Moreover, whatever the age, neither OVX nor E treatment had any significant effect on Mg apparent absorption.

Effect of oral aluminum and aluminum citrate on blood level and short-term tissue distribution of aluminum in the rat.

Aluminum (Al) absorption seems to be very low, but many factors can enhance it in animals and humans. In the present study, we investigated the acute effect of Na citrate on Al absorption by monitoring Al levels in blood and several tissues. For this purpose, 18 Wistar male rats were divided into 3 groups: control, Al, and Al + Na citrate. After a 14-h fasting period, animals were dosed orally with deionized water, or 2 mmol Al chloride, or 2 mmol Al chloride plus 2 mmol Na citrate. Blood samples were taken before and 1, 2, 4, and 6 h after the gavage. Al concentrations in blood, liver, tibia, kidney, and intestinal wall were determined by ICP-OES. In the Al and Al + citrate groups, Al blood concentrations peaked at 1 h and 2 h with higher levels in the Al + citrate group. Al gavage resulted in an increase in Al level in intestinal wall, but not in the other investigated tissues. Simultaneous gavage of citrate with Al significantly increased its tissue levels in tibia, kidney, and in intestinal wall. Our data show clearly that Al as chloride can be absorbed, but not well retained by the organism tissues. Furthermore, the model used in the present study is appropriate for acute studies to investigate the effect of various compounds on Al absorption in the rat.

Short-term ingestion of chlorogenic or caffeic acids decreases zinc but not copper absorption in rats, utilization of stable isotopes and inductively-coupled plasma mass spectrometry technique.

The amount of dietary trace elements absorbed from a meal depends, among other factors, on the quantities of certain minor plant constituents present in the meal. These substances can act as ligands and bind trace elements in the digestive tract in available or unavailable forms for absorption. The present study was designed to investigate the extent to which different polyphenols (PP) may influence Zn and Cu absorption in rats. Different PP of nutritional interest (chlorogenic acid, caffeic acid, catechin and rutin) were studied using meals extrinsically-labelled with stable isotopes 67Zn and 65Cu. Male Wistar rats were fed on a non-labelled semi-synthetic diet containing (mg/kg) 38 Fe, 35 Zn and 7.5 Cu for 8 d. PP were dissolved in dimethyl sulfoxide as the solvent and added to the meal at 1 g/kg during 3 d before isotope administration and until the end of the experiment (a further 3 d). The control group received the dimethyl sulfoxide only. After overnight food deprivation, rats were fed on the labelled test meals (4 g diet +0.1 mg 67Zn and 0.1 mg 65Cu) with 0.5 mg Dy as a faecal marker. Faeces and urine pools were collected for 3 d and analysed for 67Zn and 65Cu isotopic enrichment using the inductively-coupled plasma mass spectrometry technique. Zn absorption was significantly less in rats fed on chlorogenic acid or caffeic acid than in the control group. Catechin ingestion non-significantly inhibited 67Zn absorption. However, the PP studied were without effect on Cu absorption. The study illustrates the effect of metal-binding phenolic compounds on mineral nutrition in the rat, and the possible importance of the effects of different foods rich in these compounds on mineral absorption in man.

Intestinal fermentation lessens the inhibitory effects of phytic acid on mineral utilization in rats.

The specific effects of phytic acid (PA) and resistant starch (RS) on mineral bioavailability, namely, Ca, Mg, Fe, Zn and Cu, were investigated in rats adapted to semipurified diets. The diets provided either 73 g/100 g digestible wheat starch (DS) alone, or 53 g/100 g DS plus 20 g/100 g crude potato starch (RS) and either 0 or 1.1 g/100 g PA. A period of 3 wk was first planned to adapt the rats to their respective diets. Compared with rats fed the DS diets, those fed the RS diets had significant cecal hypertrophy and an accumulation of short-chain fatty acids, together with greater cecal blood flow. RS enhanced the cecal absorption of Ca and Mg (from 0.15 to 0.55 micromol/min for Ca, and from 0.10 to 0.35 micromol/min for Mg). Mineral balance was enhanced significantly by RS (Ca, +46%; Mg +50%; Fe +20%; Zn, + 33% and Cu, +61%). PA had no significant effect on Ca or Mg solubility and absorption in the cecum, and it failed to alter significantly Ca or Mg balance. The apparent absorption of Fe, Zn and Cu was significantly lower in rats fed the DS + PA diet than in rats fed the DS diet (Fe, -35%; Zn, -28%; and Cu, -31%). In rats adapted to the RS diet, the inhibitory effects of PA were practically abolished and the mineral balance was restored to the control values. We conclude that the negative effects of PA on mineral balance are relatively minor compared with the stimulatory effect of RS.

[Traissac voice prosthesis. Apropos of 9 years experiences].

BACKGROUND: Long term results are discussed. PATIENTS: A retrospective study of 9 years (1/01/89, 1/01/98) concern 107 laryngectomized patients with 9 patients a part from this period, and 30% without prosthesis. Among 80 patients with prosthesis, 57% had received primary TEPs (tracheoesophageal puncture), 21% delayed primary TEPs and 22% secondary TEPs. Oesophageal voice is always learned with tracheoesophageal voice. RESULTS: We find a success rate (1 month) in 75% of cases, with a maximum follow up time of 8 years (the average is 3 years 1 month). We find a success rate with a long-term result in 66% of cases. Prosthesis removal exists in 30% of cases and oesophageal voice is successfull once out of 3 in this cases. Mean lifetime Traissac's prosthesis is 18 months. After removal, non closed fistulas can exist in 40% of cases, and surgery is always successful. DISCUSSION: Material Traissac's prosthesis explains extended lifetime. CONCLUSION: Traissac's prosthesis gave us satisfaction during a use of 8 years.

Demographic and cardiovascular risk factors in relation to antioxidant status: the EVA Study.

The aim of the study was to examine the determinants of blood antioxidant indicators on a large sample. Levels of plasma selenium and carotenoids, vitamin E in red blood cells, and thiobarbituric acid reactive substances (TBARS) were determined. The cross-sectional relationships between these markers and demographic and cardiovascular risk factors were examined in participants of the EVA study, a cohort of 1389 men and women, aged 59-71 years. Multivariable regression models including demographic (age, sex, socio-economic level), lifestyle (alcohol, tobacco), clinical and metabolic (lipids, glycemia) factors were used. Women had higher levels of plasma carotenoids, TBARS and red blood cell vitamin E. Cholesterol levels were positively associated to lipid-soluble vitamins, selenium and TBARS. Use of lipid-lowering drugs was positively associated with selenium and vitamin E and negatively with carotenoids. Body mass index was the strongest determinant of plasma carotenoids. Education and income levels were positively associated with selenium and total carotenoids. Tobacco consumption was negatively associated with red blood cell vitamin E, whereas alcohol consumption was positively associated with TBARS. This study emphasizes the respective place of the various determinants of antioxidant status. When considering tissue antioxidant indicators, analyses should take into account not only the metabolic parameters but also socio-economic factors and the subject's life style.

Selenium and antioxidant vitamin and lipidoperoxidation levels in preaging French population. EVA Study Group. Edude de vieillissement artériel.

Selenium (Se) and antioxidant vitamins might play an important role in the etiology of free radical-related diseases and aging. In the Edude de vieillissement artériel (EVA) study, we have determined the plasma thiobarbituric acid reacting substances (TBARS) as an indicator of free radical-induced lipid peroxidation, plasma selenium and carotenoids, and erythrocyte vitamin E levels in 1389 subjects aged 59-71 years. We also looked for an association between these parameters and cardiovascular risk factors in early elderly. The results show that plasma TBARS were significantly increased in elderly in comparison with values reported in younger adults. However, plasma Se and carotenoids as well as erythrocyte vitamin E in elderly people are close to those reported in adult people. If plasma Se showed no difference between men and women, the three other parameters were significantly higher in women than in men. With regard with cardiovascular risk factors, plasma TBARS were highly positively correlated with total and low-density lipoprotein (LDL) cholesterol in men and women. Plasma carotenoids were also positively correlated with plasma total cholesterol and LDL cholesterol in both sexes. Finally, plasma TBARS were highly correlated with smoking and alcohol consumption. In conclusion, this part of the EVA study shows that some cardiovascular risk factors, like smoking and cholesterol level, are associated with high free radical-induced TBARS levels in the preaging population, although plasma Se and carotenoids as well as erythrocyte vitamin E levels in elderly people were close to those reported in adult younger people.

Effect of soluble or partly soluble dietary fibres supplementation on absorption and balance of calcium, magnesium, iron and zinc in healthy young men.

OBJECTIVES: This study is aimed at investigating the effect of feeding a soluble or partly soluble fibre rich-diet on the apparent absorption and balance of calcium, magnesium, iron and zinc in healthy young men, by using a chemical balance technique. STUDY DESIGN: Nine healthy young men were given a control diet or the same diet complemented with either inulin (soluble) or sugar beet fibre (partly soluble) during 28 d periods according to a 3 x 3 latin square design with three repetitions. During the 20 d adaptation period to fibre ingestion, experimental fibres were incorporated into bread (60%) and liquid foods (40%) up to a maximum of 40 g/d. Ca, Mg, Fe and Zn were measured in diets and in a 8 d urine and faecal composites to assess mineral absorption and balance. RESULTS: The dietary mineral intake provided (mg/d) 859 +/- 196 of Ca; 311 +/- 43 of Mg; 11.6 +/- 1.7 of Fe; and 11.1 +/- 1.6 of Zn from the control diet. The apparent absorption of minerals from the control diet was (%) Ca: 21.3 +/- 12.5; Mg: 46.3 +/- 10.9; Fe: 21.8 +/- 12.3 and Zn: 14.0 +/- 14.5 (mean +/- s.d.). Ingestion of inulin significantly increased the apparent absorption and the balance of Ca. Sugar beet fibre ingestion resulted in a significant increase in Ca intake and balance, without modification its apparent absorption. Apparent absorption and balance of Mg, Fe and Zn were not significantly altered by the ingestion of either experimental fibre. CONCLUSIONS: Addition of the two experimental fibres (inulin or sugar beet fibre) to normal mixed diets can improve Ca balance without adverse effects on other mineral retention. Study of magnesium bioavailability using stable isotopes and the inductively-coupled plasma mass spectrometry technique in the rat: single and double labelling approaches.

The present work aimed to investigate the feasibility of using stable isotopes and inductively-coupled plasma mass spectrometry (ICP/MS) to study Mg absorption in rats. Male Wistar rats, aged 7 weeks and weighing 180 g, were used. They were fed on a semi-purified diet containing 1070 mg Mg/kg, and had free access to feed and distilled water. In the first experiment, after a 16 d adaptation period, two doses of enriched 25Mg (6 and 12 mg) were administered by oral intubation, faeces and urine were collected daily and blood was sampled. Isotope ratios were determined by ICP/MS. 'True' absorption values, using the faecal isotope data, were 0.63 and 0.56 in rats receiving 6 and 12 mg 25Mg respectively, while apparent absorption was 0.50 for two successive periods of metabolic balance studies. Moreover, the oral isotope administration resulted in a measurable isotopic enrichment in plasma within hours which was still detectable on the third day following the isotope administration. In the second experiment, investigating the double labelling technique, similar rats were dosed simultaneously with 5 mg 26Mg orally (premixed with diet) and 0.29 mg 25Mg intravenously. The calculated Mg true absorption values were very similar when calculated from blood or urine data (0.38) but were lower than that obtained from faecal data (0.50). The possible causes of such an unexpected difference and limits of the double labelling technique for Mg absorption are discussed here. Together these results indicate that although 25Mg and 26Mg isotopes have high natural abundance, the described methodology permits meaningful investigations of Mg bioavailability and metabolism.

Combined effects of lipid peroxidation and antioxidant status on carotid atherosclerosis in a population aged 59-71 y: The EVA Study. Etude sur le Vieillisement Artériel.

There are few epidemiologic studies of the effects of lipid peroxidation and antioxidant status on atherosclerosis. The relation of lipid peroxidation evaluated by thiobarbituric acid-reactive substances (TBARS) and biological markers of antioxidant status to ultrasonographically assessed carotid atherosclerosis was examined from baseline data of a longitudinal study on cognitive and vascular aging (Etude sur le Vieillisement Artériel, the EVA Study). The study sample was composed of 1187 mean and women aged 59-71 y without any history of coronary artery disease or stroke. Ultrasound examination included measurements of intima-media thickness (IMT) on the common carotid arteries (CCAs) and at the site of plaques. After adjustment for conventional cardiovascular risk factors, erythrocyte vitamin E was significantly and negatively associated with CCA-IMT in both men and women whereas plasma selenium and carotenoids were not. No association was found between TBARS and CCA-IMT in either sex. However, TBARS were significantly higher in men with carotid plaques than in those without. This association was strengthened in men with concentrations of erythrocyte vitamin E, plasma selenium, and carotenoids below the lowest quartile. Our findings give some epidemiologic support to the hypothesis that lipid peroxidation and low antioxidant status are involved in the early phases of atherosclerosis.

Lipid peroxidation level and antioxidant micronutrient status in a pre-aging population; correlation with chronic disease prevalence in a French epidemiological study (Nantes, France).

OBJECTIVE: The general objective of the Etude du Viellissement Arterial (EVA) program is to follow vascular aging and the decline in cognitive functions at the cerebrovascular level longitudinally over a 4-year period. One of the specific objectives of this EVA study is to examine epidemiologically the relationship between the markers of oxidative stress (lipid peroxidation), the antioxidant micronutrient status (particularly of selenium, vitamin E, and the carotenoids) and the prevalence of chronic disorders occurring during the pre-aging period. METHODS: 1389 subjects aged from 59 to 71 years were studied. RESULTS: The concentration of plasma lipid peroxides was higher than in young adults (2.91 +/- 0.38, men; 2.97 +/- 0.40, women (mumol/l). On the other hand, plasma Se (1.09 +/- 0.21, men; 1.10 +/- 0.19, women (mumol/l)), erythrocyte vitamin E (5.32 +/- 1.29, men; 5.52 +/- 1.28, women (mumol/l)), and total plasma carotenoids (2.19 +/- 0.98, men; 3.07 +/- 1.33, women (mumol/l)) were comparable to values in young adults. In our cohort, 40% of subjects had unremarkable medical histories. The disorders most often encountered were lipemia (29.8% of men, 36.1% of women), and hypertension (28.9% of men, 30.4% of women). CONCLUSION: Se and vitamin E levels were raised in cases of lipemia, especially in those treated with fibrates. The mechanism of the increase is unknown. In hypertensives and diabetics, there was a decrease in total carotenoids associated with increased peroxidative risk.

5-Hydroxymethyluracil excretion, plasma TBARS and plasma antioxidant vitamins in adriamycin-treated patients.

The thymine oxidative lesion-5-hydroxymethyluracil (HMUra)-was measured in urine collected from cancer patients. These patients all received chemotherapy using Adriamycin. Adriamycin (ADR) intercalates DNA coils and interferes with normal cell metabolism through diverse biochemical mechanisms that may explain its different actions. The anticancer action of ADR could derive from its interaction with topoisomerase II, resulting in DNA nicking followed by DNA fragmentation and apoptosis. Side effects of ADR-mainly its cardiotoxicity-may derive from the fact that ADR generates superoxide and hydroxyl radicals in two ways: redox-cycling and a Haber-Weiss type reaction due to Fe-ADR complexes. The oxygen free radicals, particularly .OH, are thought to be produced by ADR directly in genomic material and attack all its components. 5-Hydroxymethyluracil is a thymine lesion provoked by these attacks, and it has been proposed as a marker of DNA alterations. In this article, we report the results of a study involving 14 cancer patients treated with ADR. We found that urine HMUra is significantly increased by the anticancer therapy (HMUra (nmol/24 h): 74.4 9.46 vs. 96.3 8.74; p < .01), this increase reveals a higher risk of mutagenesis. Our study is the first to show an in vivo alteration of DNA by ADR. Results also show that thiobarbituric acid reactants increase significantly, and that the vitamin levels for retinol and alpha-tocopherol, which are antioxidant vitamins, are lower at the end of chemotherapy. We suggest to supplement these patients with vitamins A and E, and selenium to reduce the side effects of ADR.

Effect of selenium supplementation on biological constants and antioxidant status in rats.

Oxygen-derived free radicals are currently suspected to be widely involved in the aetiology of several clinical disorders. In animals as well as in man, antioxidant trials are often undertaken to prevent oxidative stress. Among antioxidant molecules selenium has been largely studied. This study shows that plasma Se level is not a good index of Se status in the organism, at least at high levels of selenium. Red blood cell Se seems to be a more reliable index of Se status and could replace plasma Se level in the supplementation trials both in animals and humans. Se supplementation did not result in a significant decrease in oxidative stress markers as evaluated by blood and tissue malondialdehyde contents in healthy animals. Furthermore, heart function was altered and plasma Alanine aminotransferase activity was significantly increased in the selenium-supplemented group, which could reflect a slight subtoxic effect of selenium supplementation at the level used here. In view of the results presented, the maximum selenium content in animal diet in selenium supplementation experiments should not be higher than 2 mg/kg.

Rapid high-performance liquid chromatographic assay for salicylic acid in plasma without solvent extraction.

The in vivo measurement of highly reactive free radicals, such as the hydroxyl radical (oOH), in humans is very difficult, if not impossible. Specific markers, based on the ability of oOH to attack aromatic molecules and produce hydroxylated compounds, are under investigation. In vivo radical metabolism of salicylic acid produces two main hydroxylated derivatives: 2,3- and 2,5-dihydroxybenzoic acid (DHBA). The measurement of 2,3-DHBA, following oral administration of salicylic acid or its acetylated form (aspirin), is proposed for the assessment of in vivo oxidative stress. The intensity of oxidative stress is a function of the ratio of dihydroxylated derivatives to salicylic acid rather than the absolute dihydroxylated derivatives levels. Consequently, a simple, accurate, and sensitive assay of the salicylic acid level in plasma is needed to investigate the in vivo free radical production. In this work, a rapid and sensitive method is presented that is useful for the quantitation of salicylic acid in biological fluids. The methodology uses high-performance liquid chromatography with spectrophotometric detection for the identification and quantitation of salicylic acid without organic extraction. A detection limit of less than 5 mumol is achieved with spectrophotometric detector responses that are linear over at least 6 orders of magnitude. Plasma concentrations of salicylic acid determined by the present technique are reported following the administration of 1000 mg aspirin in 20 healthy subjects.
