[Lysine and arginine requirement of "Ustilago cynodontis" 4001 yeast-like cells. I. --Growth in the presence and in the absence of lysine (author's transl)].

For optimal growth, the yeast-like cells of Ustilago cynodontis 4001 (originating from the mycelium 4001 prototroph forms) require the presence of both arginine and lysine. However, in the absence of lysine, growth does occur, but two exponential growth phases can then be observed: a pseudo-lag phase during which the growth rate is slow, and a second, true exponential phase. The initial OD of the culture and the arginine concentration of the medium do not appear to affect the duration of the pseudo-lag phase. The arginine concentration does, however, affect the growth yield. Upon addition of lysine to the culture medium, the period of the pseudo-lag phase is reduced and the growth rate increased. The pseudo-lag phase can be completely suppressed by the addition of adequate amounts of lysine. In the presence of lysine and in the absence of arginine, cells are not capable of multiplication. Addition of arginine initiates growth; whether or not a lag phase occurs depends on the quantity of arginine added. The successive inoculation of cells into lysine-free medium leads to suppression of the pseudo-lag phase but only after three or four passages. Therefore it does not seem that there is selection of cells which do not require lysine during the pseudo-lag phase. It is shown that it is not the modification of the culture medium which suppresses the lysine requirement: the cells which are put to grow in the supernatant of a culture in which a pseudo-lag phase had taken place behave in the same way that when they are inoculated in a new medium. The modification of the cell metabolism itself is discussed.

Melatonin therapy of advanced human malignant melanoma.

We undertook a study to investigate the therapeutic potential of orally administered melatonin in patients with advanced melanoma. Forty-two patients received melatonin in doses ranging from 5 mg/m2/day to 700 mg/m2/day in four divided doses. Two were excluded from analysis. After a median follow-up of 5 weeks, six patients had partial responses, six additional patients had stable disease. Sites of response included the central nervous system, subcutaneous tissue and lung. The median response duration was 33 weeks for the partial responders. There was a suggestion of a dose-response relationship. The toxicity encountered was minimal and consisted primarily of fatigue in 17 of 40 patients. Melatonin also appeared to reduce basal levels of follicle-stimulating hormone (FSH). No significant changes were encountered in serum levels of luteinizing hormone (LH) or thyroid stimulating hormone (TSH). We conclude that further study of melatonin as a potentially useful agent in metastatic melanoma is warranted.

Proinflammatory effects in experimental mesangial proliferative glomerulonephritis of the immunosuppressive agent SDZ RAD, a rapamycin derivative.

BACKGROUND/AIM: The new immunosuppressant SDZ RAD, a rapamycin derivative, inhibits growth factor driven cell proliferation. SDZ RAD designed for transplantation may also be a candidate agent to treat inflammatory kidney diseases. Therefore, we investigated the effects of SDZ RAD in two different animal models of glomerulonephritis, in anti- Thy1.1 nephritis and in acute puromycin aminonucleoside (PAN) nephrosis. METHODS: Eighty-seven male Wistar rats were investigated. Anti-Thy1.1 nephritis: healthy rats (n = 9), SDZ RAD-treated healthy rats (n = 6), nephritic rats (n = 9), SDZ RAD placebo treated nephritic rats (n = 6), SDZ RAD-pretreated nephritic rats (n = 9), and early (n = 6) as well as delayed (n = 6) SDZ RAD-posttreated nephritic rats. PAN nephrosis: healthy rats (n = 6), SDZ RAD-treated healthy rats (n = 6), nephritic rats (n = 12), and SDZ RAD-pretreated nephritic rats (n = 12). In a separate study, 12 male Sprague-Dawley rats were analyzed in anti-Thy1.1 nephritis: healthy rats (n = 3), nephritic rats (n = 3) and pretreated nephritic rats (n = 6). SDZ RAD and SDZ RAD placebo were given at single doses of 2.5 mg/kg body weight per day by gavage. The experiments lasted until days +2 and +9 after induction of anti-Thy1. 1 nephritis and until day +13 in the case of PAN nephrosis. RESULTS: In anti-Thy1.1 nephritis, SDZ RAD demonstrated marked proinflammatory effects in a time-dependent manner, as reflected by severe focal damage to glomerular histology including inhibition of mesangial cell proliferation, reduction of creatinine clearance, and increase in plasma creatinine levels as well as proteinuria. Almost identical results were obtained in both rat strains. In contrary, SDZ RAD ameliorated significantly the development of PAN nephrosis. Animals pretreated by this agent showed a significant reduction of proteinuria and of glomerular invasion of monocytes/macrophages. CONCLUSION: Some caution is warranted for the use of SDZ RAD in inflammatory glomerular diseases, since it accentuated glomerular damage induced by anti-Thy1.1 antibodies.

Renal matrix metalloproteinase activity is unaffected by experimental ischemia-reperfusion injury and matrix metalloproteinase inhibition does not alter outcome of renal function.

BACKGROUND: Ischemia-reperfusion injury can lead to organ damage, such as delayed graft function in kidney transplantation. Reactive oxygen species that play a key role in this disorder may directly activate latent matrix metalloproteinases (MMP). In the kidney, little is known about the role of MMP in ischemia-reperfusion. Therefore, the aim of our study was to analyze activity/expression of MMP and to assess their functional role by the use of the MMP inhibitor BB-94 (Batimastat). METHODS: Renal ischemia was induced by left renal pedicle occlusion for 60 min, preceded by right nephrectomy. Thirty-two female Sprague-Dawley rats were analyzed: sham-operated rats (n = 8), treated sham-operated rats (n = 4), ischemic rats (n = 12), and treated ischemic rats (n = 8). Batimastat therapy (30 mg/kg body weight/day) was initiated 2 days prior to induction of ischemia. Animals were sacrificed 12 h (n = 8) and 24 h (n = 24) after ischemia for analyses of MMP activity/expression and of plasma creatinine levels. RESULTS: We found no evidence for an alteration in the activity or expression of MMP as a result of renal ischemia-reperfusion. Importantly, plasma creatinine levels significantly increased to a mean of 374 +/- 61 micromol/l in ischemic rats after 24 h, almost identical to the BB-94-treated ischemic rats (384 +/- 36 micromol/l). The creatinine levels in sham-operated rats remained within normal limits. CONCLUSION: MMP play no role during the early phase of experimental renal ischemia-reperfusion injury.

Mental health: counselling sexual abuse survivors.

Some nurses are now becoming involved in helping adult survivors of child sexual abuse to overcome problems caused by an early abusive relationship. Developing a therapeutic partnership between victim and counsellor is a fundamental part of resolving such problems. Using a psychodynamic perspective, the author examines the effects of child sexual abuse on female survivors, and discusses the benefits of group therapy and individual counselling in helping the survivor to come to terms with the abuse.

[Do the eosinophilic granulomas observed in Brittany represent a form of anisakiasis? The larvae of Thynnascaris aduncum do not produce these granulomas experimentally].

To explain the enigmatical origin of gastric eosinophilic granulomas prevailing among patients in Brittany (France), we have tried to experiment with the larvae of Thynnascaris (Thynnascaris aduncum (Rud.)), a Nematode found in the sardine (Sardina pilchardus (Walbaum)), a fish sometimes eaten raw in our country. In a preliminary experiment, larvae of Thynnascaris were observed to penetrate the mucosa of the rabbit stomach in vitro at room temperature. But in vivo, whatever the means, the ways and the experimented mammals of our study, we have failed to experimentally reproduce such tumours. We think that this result belongs to the fact that the larvae of Thynnascaris in the conditions of our experimentation could not perform an evolution in homeothermal animals.

[A critical study of the penile Doppler test. Considerations apropos of a personal series of over 850 patients tested for the evaluation of impotence since 1983].

The penial Doppler is, in 1990, the first intention examination the most asked for when there is a suspicion of a vascular origin in an erection problem. In fact, due to the technical difficulties to carry it out, this examination presents many uncertainties in its interpretation, and can even be the source of diagnostic errors. The aim of this study is to argue in favour of a systematic intracavernous injection of 8 mg of papaverine when the penial Doppler is carried out, enabling an increase in its diagnostic value and often a therapeutic orientation straightaway.
