Erythromycin, miocamycin and clindamycin towards adhesivity and phagocytosis of gram-positive bacteria.

To estimate the antibacterial activity of sub-MIC concentrations of Erythromycin and Miocamycin in the mucosal surfaces, we studied the adhesivity of Gram-positive pathogenic strains (S. pyogenes, S. aureus) towards oral and urinary epithelial cells. Erythromycin strongly inhibits the adhesivity of Staphylococci to oral cells (50% of inhibition) and to a lesser extent the adhesivity of Streptococci (21%), while Miocamycin reduced the adhesivity of Staphylococci by about 16%. In some cases, an increase in the adhesivity on urinary cells was found mainly for Miocamycin. Therefore Erythromycin, at sub-MIC concentrations, is able to induce a marked reduction in the adhesivity of Staphylococci and Streptococci; this may interfere with the invasivity and subsequent pathogenicity of these bacteria. As far as phagocytosis is concerned, one strain of enterotoxic coagulase + S. aureus bearing A-protein on its surface but lacking the capsule was taken into consideration. The presence of A-protein induces resistance to phagocytosis and opsonization by normal serum. The addition of antibiotics which inhibit protein biosynthesis, such as Clindamycin, Miocamycin and Erythromycin, increases either uptake of this strain or intracellular killing. In particular Clindamycin and Erythromycin, at sub-MIC concentrations, are able to affect the interaction of Staphylococcus with phagocytic cells.

Incidence of hepatitis B virus DNA and DNA-polymerase in sera of Italian asymptomatic carriers with the serological markers of HBV.

The presence of Hepatitis B virus (HBV) DNA in sera of patients with HBV related diseases is considered a reliable marker of virus active replication. In this paper HBV DNA was assayed by the molecular hybridization method with a 32P labeled nick translated HBV probe. The assay was positive in sera of 21 out of 22 HBeAg-positive and 4 out of 8 HBeAg-negative asymptomatic HBsAg carriers. 15 HBsAg-negative sera obtained from healthy donors showed no HBV DNA. Almost 80 per cent of HBeAg and HBV DNA positive sera revealed a DNA-polymerase activity. In order to determine the infectivity of HBsAg carriers, it appears that, whenever possible, the HBV DNA spot hybridization should be performed in conjunction with the DNA-polymerase, HBsAg and HBeAg tests.

Seroepidemiology of infection with hepatitis A virus and hepatitis B virus in the Seychelles.

A batch of 417 serum samples obtained from native-born subjects were tested for the presence of hepatitis B surface antigen (HBsAg) and corresponding antibody (anti-HBs), by enzyme-linked immunosorbent assay (ELISA); and antibodies to hepatitis B core antigen (anti-HBc), e-antigen (anti-HBe), and hepatitis A virus (anti-HAV), by radioimmunoassay (RIA). HBsAg was found in only two of the 417 subjects studied. Anti-HBs was detected in 112 samples (26.8%), anti-HBc in 114 (27.3%) and anti-HBe in 31 samples (7.4%). Serologic evidence of a previous or present infection by hepatitis B virus (HBV) was found in 34.5% of the samples studied. Males showed a greater prevalence of anti-HBs and anti-HBc, while anti-HBe was more common in females; however, these differences were not significant. With regard to age, a significantly higher prevalence of anti-HBs (p less than 0.05), anti-HBc (p less than 0.025) and anti-HBe (p less than 0.025) was found in the older age groups. Anti-HAV antibodies were present in 90% of the subjects studied, with no variation between the sexes. The anti-HAV rate in the group under 20 years was similar to that found in the older age groups. The total infection rate of hepatitis B virus in the Seychelles is lower than in other tropical areas, HBs antigen/antibody ratio approaching that in temperate areas. Elucidation of the reasons for the low prevalence of hepatitis B virus carriers among the Sevchelles population requires further investigation.

[Multicenter study on the distribution of Streptococcus pneumoniae strains in Italy].

Pneumococcal serotyping by quellung reaction has been performed in five Italian areas on 267 strains, isolated from throat swab, sputum, blood, liquor, etc. 37 serotypes have been identified. The most frequent types resulted 19, 6, 3, 20, 9, 4 (in 50% of total samples), with no difference from those detected in other European countries. The proportion of type 19 was the highest among all age group, in four out of five different areas and in bacteremic pneumonia and meningitis. 36 strains were isolated from infection sites among which type 19 resulted more frequent than in isolates from throat swab and sputum (27.7% versus 16.1%). Pneumococcal serotyping should be routinely done in the light of forthcoming use of polysaccharide vaccine.

[Histochemical technics for detecting intrahepatic HBsAg by a rapid diagnosis of HBV infection].

On 44 liver biopsy specimens a trichromic staining for histochemical detection of HBsAg in cytoplasm of hepatocytes has been performed. In 34 subjects their sensitivity and availability has been compared with aldehyde-fuchsin Gomori's staining and IF. Trichromic staining is easy to perform, makes it possible simultaneous morphological diagnosis and is sufficiently sensitive for HBsAg detection especially in chronic hepatitis. It can be considered as elective method for a rapid diagnosis of HBV infection.

[HBAg-RIA as a parameter of functional evaluation of viral hepatitis].

The investigation of HBAg by RIA appears to be very helpful in the diagnosis and follow-up of viral hepatitis. In most cases, a prolonged antigenemia is more strongly indicative of chronic evolution of the illness than other parameters. The persistence of HBAg, therefore, is significant for the persistence of viral infection.
