X-ray structure of nucleoside diphosphate kinase.

The X-ray structure of a point mutant of nucleoside diphosphate kinase (NDP kinase) from Dictyostelium discoideum has been determined to 2.2 A resolution. The enzyme is a hexamer made of identical subunits with a novel mononucleotide binding fold. Each subunit contains an alpha/beta domain with a four stranded, antiparallel beta-sheet. The topology is different from adenylate kinase, but identical to the allosteric domain of Escherichia coli ATCase regulatory subunits, which bind mononucleotides at an equivalent position. Dimer contacts between NDP kinase subunits within the hexamer are similar to those in ATCase. Trimer contacts involve a large loop of polypeptide chain that bears the site of the Pro----Ser substitution in Killer of prune (K-pn) mutants of the highly homologous Drosophila enzyme. Properties of Drosophila NDP kinase, the product of the awd developmental gene, and of the human enzyme, the product of the nm23 genes in tumorigenesis, are discussed in view of the three-dimensional structure and of possible interactions of NDP kinase with other nucleotide binding proteins.

Effect of sorbin on duodenal absorption of water and electrolytes in the rat.

Sorbin is a newly isolated intestinal peptide that has been purified because of its ability to induce water absorption. The effects that sorbin and some synthetic peptides corresponding to its C-terminal sequence have on duodenal absorption of water, chloride, and sodium were studied in comparison with the effects of vasoactive intestinal peptide (VIP), [D-Ala,Met]-enkephalinamide (DAMA), and angiotensin II. The technique of an in situ ligated duodenal loop in the rat was used for all peptides. Under the experimental conditions used, a low basal secretion of water, chloride, and sodium was obtained; VIP induced an increase of the secretion, whereas DAMA induced an absorption, both in the nanomolar dose range. Angiotensin II and sorbin induced an absorption in the picomolar dose range. The most effective doses of sorbin peptides but not of angiotensin induced the lowest final concentrations of Na+ and Cl- obtainable without inducing secondary water secretion. All synthetic peptides containing the C-terminal heptapeptide of sorbin were active in the picomolar dose range. Contrary to angiotensin, they had no effect on blood pressure.

Object permanence in cats (Felis catus): an ecological approach to the study of invisible displacements.

A single invisible displacement object permanence task was administered to 19 cats (Felis catus). In this task, cats watched a target object from behind a transparent panel. However, cats had to walk around an opaque panel to reach the object. While cats were behind the opaque panel, the object was hidden behind one of two screens. As cats did not perceive the disappearance of the object behind the target screen, the object was invisibly hidden. Results showed that cats solved this task with great flexibility, which markedly contrasts with what has been observed in previous research. The discussion emphasizes the difference between the typical Piagetian task in which the information necessary to succeed must be dealt with in retrospective way, whereas in our task cats had to anticipate a new position of the object. The ecological relevance of this new task is also discussed.

Crystallization and preliminary X-ray diffraction studies of nucleoside diphosphate kinase from Dictyostelium discoideum.

Nucleoside diphosphate kinase from the slime mold Dictyostelium discoideum is highly homologous to gene products that are involved in development in Drosophila and in oncogenesis in human cells. The cloned protein expressed in Escherichia coli has been purified and crystallized in a hexagonal space group with a = b = 74.9 A, c = 211.4 A. The asymmetric unit contains either one or two 17,000 Mr subunits of the hexamer.

Use of a fast protein electrophoretic purification procedure for N-terminal sequence analysis to identify S-locus related proteins in stigmas of Brassica oleracea.

In the cruciferous plant Brassica oleracea L. (cabbage), the S-locus specific glycoproteins (SLSGs) isolated only in stigmas are considered to play an important role in the normal prevention of self-fertilization. Recent molecular data have shown that the gene encoding these glycoproteins (the SLG gene) belonged to a multigenic family consisting of about 10 homologous copies among which another member is expressed, the S-locus related gene (SLR1gene). Our aim was to determine whether the SLR1-gene proteins were expressed in the stigmatic tissues. We first identified the putative SLSGs or SLR1-proteins by Con A-peroxidase detection of glycoproteins separated after isoelectric focusing in polyacrylamide gels. We describe a fast purification procedure for the glycoproteins of interest, based on analytical isoelectric focusing, electrophoresis, and electroblotting of proteins onto polyvinylidene difluoride membranes. Blotted proteins were sequenced for N-terminal amino acid determination. By comparison of the N-terminal sequences of the purified proteins with the peptide sequence predicted from the SLR1-cDNA, we demonstrate the expression of SLR1-like proteins in stigmas of B. oleracea.

Cloning and characterization of the Chlamydomonas moewusii mitochondrial genome.

We report that the mitochondrial genome of Chlamydomonas moewusii has a 22 kb circular map and thus contrasts with the mitochondrial genome of Chlamydomonas reinhardtii, which is linear and about 6 kb shorter. Overlapping restriction fragments spanning over 90% of the C. moewusii mitochondrial DNA (mtDNA) were identified in a clone bank constructed using a Sau3AI partial digest of a C. moewusii DNA fraction enriched for mtDNA by preparative CsCl density gradient centrifugation. Overlapping Sau3AI clones were identified by a chromosome walk initiated with a clone of C. moewusii mtDNA. The mtDNA map was completed by Southern blot analysis of the C. moewusii mtDNA fraction using isolated mtDNA clones. Regions that hybridized to C. reinhardtii or wheat mitochondrial gene probes for subunit I of cytochrome oxidase (cox1), apocytochrome b (cob), three subunits of NADH dehydrogenase (nad1, nad2 and nad5) and the small and the large ribosomal RNAs (rrnS and rrnL, respectively) were localized on the C. moewusii mtDNA map by Southern blot analysis. The results show that the order of genes in the mitochondrial genome of C. moewusii is completely rearranged relative to that of C. reinhardtii.

Association of poly(ADP-ribose) polymerase with the nuclear matrix: the role of intermolecular disulfide bond formation, RNA retention, and cell type.

The recovery of the enzyme poly(ADP-ribose) polymerase (pADPRp) in the nuclease- and 1.6 M NaCl-resistant nuclear subfraction prepared from a number of different sources was assessed by Western blotting. When rat liver nuclei were treated with DNase I and RNase A followed by 1.6 M NaCl, approximately 10% of the nuclear pADPRp was recovered in the sedimentable fraction. The proportion of pADPRp recovered with the residual fraction decreased to less than 5% of the total nuclear polymerase when nuclei were prepared in the presence of the sulfhydryl blocking reagent iodoacetamide and increased to approximately 50% of the total nuclear pADPRp when nuclei were treated with the sulfhydryl cross-linking reagent sodium tetrathionate (NaTT) prior to fractionation. To determine whether this effect of disulfide bond formation was unique to rat liver nuclei, nuclear matrix/cytoskeleton structures were prepared in situ by sequentially treating monolayers of tissue culture cells with Nonidet-P40, DNase I and RNase A, and 1.6 M NaCl (S.H. Kaufmann and J.H. Shaper (1991) Exp. Cell Res. 192, 511-523). When nuclear monolayers were prepared from HTC rat hepatoma cells, CaLu-1 human lung carcinoma cells, and CHO hamster ovary cells in the absence of NaTT, pADPRp was undetectable in the nuclease- and 1.6 M NaCl-resistant fraction. In contrast, when nuclear monolayers were isolated in the presence of NaTT, from 5% (CaLu-1) to 26% (HTC cells) of the total nuclear pADPRp was recovered with the nuclease- and salt-resistant fraction. Examination of these residual structures by SDS-polyacrylamide gel electrophoresis under nonreducing conditions suggested that pADPRp was present as a component of disulfide cross-linked complexes. Further analysis by immunofluorescence revealed that the pADPRp was diffusely distributed throughout the CaLu-1 or CHO nuclear matrix. In addition, when matrices were prepared in the absence of RNase A, pADPRp was also observed in the residual nucleoli. These observations reveal that the recovery of pADPRp with a nuclease- and salt-resistant nuclear subfraction is dependent on the source of the nuclei and on the conditions used to fractionate those nuclei. In addition, these observations raise the possibility that there might be different functional classes of pADPRp molecules within the nucleus.

Cognitive development in kittens (Felis catus): an observational study of object permanence and sensorimotor intelligence.

Spontaneous behavior of kittens (Felis catus) was filmed from birth until the end of Month 5 and coded according to Piagetian criteria of sensorimotor intelligence (SI) and object permanence (OP). Data revealed that Stages 2, 3, and 4 of SI were reached at Days 10, 26, and 45, respectively, whereas Stages 2, 3, and 4 of OP were reached at Days 31, 37, and 41, respectively. Spontaneous search behavior was exhibited both in searching for an object that disappeared and in hiding while moving toward a target object. From Day 45 on, search behavior was integrated into a playful social interaction in the form of hide-and-seek. Hence, kittens' spontaneous activity provided them with contexts in which OP was necessary for activity. Lastly, it is proposed that the mobility of both social and physical objects triggered circular activity in this species.

Effect of D ala2 metenkephalinamide on feline jejunal and ileal water and electrolyte transport.

The aim of this investigation was to compare the effect of an opioid, D ala2 metenkephalinamide (DAMA), on net jejunal and ileal water and electrolyte fluxes using the gut perfusion technique in the anesthetized cat. Intestinal transport was measured during intravenous infusion of serial doses of 2, 6, and 18 micrograms.kg-1.h-1 of DAMA in 6 cats. Each cat was its own control during an intravenous infusion of 150 mmol/l NaCl preceding the first dose of peptide and following the last dose of DAMA. Both jejunal and ileal segments were isolated by inflated balloons and were studied at the same time. Fifteen ml of an iso-osmolar test solution with hypo-osmolar ion contents and complementary mannitol were administered in the upstream tube and collected 1 h later in the downstream tube. In the jejunum, water secretion was dose-dependently reversed to an absorption from a control value of +0.5 +/- 0.4 to -0.83 +/- 0.5 ml.h-1.10 cm-1; in the ileum, water absorption was increased from -0.5 +/- 0.3 to -1.5 +/- 0.2 ml.h-1.10 cm-1. The net absorption of all electrolytes, ie sodium, chloride, bicarbonate, potassium and calcium also increased during peptide administration. However, a qualitative difference in the ion transport was observed between the jejunum and the ileum.

Cloning of rodent cDNA coding the poly(ADP-ribose) polymerase catalytic domain and analysis of mRNA levels during the cell cycle.

We have isolated a partial 2.0 kb cDNA (pRATC) encoding the entire 489 amino acids of the NAD binding domain located at the C terminus of the rat poly(ADP-ribose) polymerase. pRATC sequences were analysed and compared with the human mRNA. Our analysis reveals a remarkable homology between the rat and human nucleotide and amino acid sequences. Although a few minor amino acid changes were detected, we have found that the total number of possible phosphorylation sites remained constant in the NAD binding domain of both enzymes. We have also found that a 102 amino acid sequence, containing the putative nucleotide binding site Gly-Lys-Gly (position 378), is perfectly conserved between the rat and human sequences. Strong homology was also detected between pRATC and genomic DNA isolated from various vertebrates. In addition, we have analysed the levels of poly(ADP-ribose) polymerase mRNA throughout the cell cycle. Our results show that the levels of mRNA culminate in the G1 phase. We have also found that the increase in enzymatic activity observed in rats following treatment with phenobarbital did not correspond to an increase in the mRNA levels.

Organization of stigma surface components in Brassica: a cytochemical study.

The nature of secretion products forming the pellicle of the dry-type stigma surface has been investigated in Brassica by an electron-microscopic cytochemical study. In order to determine whether the pellicle is organized like a membrane, we used a series of cytochemical methods to visualize cell membranes. Three groups of techniques presenting different degrees of specificity in terms of cell surface markers have been used: non-specific labelling of cell-surface components, characterization of glycoconjugates and localization of enzymic activities. We demonstrated that the pellicle of Brassica stigmas, although not possessing a trilamellar structure, presents numerous characteristics encountered in biological membranes. In particular, it possesses cytochemically demonstrable enzyme activities, including ATPase and adenylate cyclase, whose role remains to be elucidated in relation to the pollen-stigma interactions.

A study of 666 strains of Candida albicans: correlation between serotype and susceptibility to 5-fluorocytosine.

The minimal inhibitory concentrations (MICs) of 5-fluorocytosine (5-FC) and the serotypes were determined for 666 strains of Candida albicans isolated from white patients in Canada. Of these strains, 495 (74.3%) were serotype A and 71 (25.7%) were serotype B. Resistance to 5-FC (MIC, greater than or equal to 25 microgram/ml) was observed in 11.3% of the serotype A strains and in 49.7% of the strains of serotype B. Most specimens were from the urogenital and respiratory tracts. The results showed the difference between the distribution of the two serotypes of C. albicans in North America and that in Europe. Serotyping of C. albicans strains by indirect immunofluorescence is a rapid means of predicting the de novo sensitivity of this year to 5-FC. The specificity of this technique for the isolated strains was approximately 79%.

1 -- Respiratory depression has been studied in the no anaesthetized rabbit by analysis of the blood gases after an injection of Piritramide (R 3365) and dehydrobenzperidol. 2 -- The respiratory depression induced by Piritramide alone is less important after an injection of dehydrobenzperidol; the most interessant cases are: -- mixture dehydrobenzperidol 400 mug/kg. Pirittramide 1000 mug/kg; --dehydrobenzperidol (400 mug/kg) injected 15 minutes before Piritramide (1000 mug/kg). 3 -- The addition of Dehydrobenzperidol does not take the analgesic activity of Piritramide decrease by it increases it.

[A micromethod for determination of nicotine and cotinine in blood and urine by gas chromatography. Results obtained from various smokers].

The determination of nicotine and cotinine in plasma and urines of smokers is carried out by gas-chromatography with an Ucon-Polar 50 HB 2000 alkalinized column and a specific N detector, using diphenylamine as internal standard. The technique for extraction and chromatographic analysis is developed. The limit values for a correct determination are : 0,2 ng for nicotine and 1 ng for cotinine. The results show that the transit of nicotine and cotinine into the blood is very fast. They confirm the rapidity of the urinary excretion of nicotine, the slowness of that of cotinine, and the relation between the eliminated amounts and the urinary flow.
