Activation of a nonselective cation channel by swelling in atrial cells.

Cell swelling has been shown to increase the permeability of the plasma membrane to ions such as K+, Na+, Ca2+ or Cl- in many types of cells. In cardiac cells, swelling has been reported to increase Cl- conductance, but whether cation-selective currents are activated by swelling is not known. Low Cl- or Cl(-)-free solutions were used to study the presence of such currents. Lowering the osmolarity of the extracellular medium from 299 to 219 mOsm resulted in cell swelling and concurrent activation of a cation-selective whole-cell current. When cell-attached patches were formed on swollen cells, opening of bursting single channel currents were observed in 18% of the patches studied. Ion substitution experiments indicated that the channel discriminated poorly among monovalent cations, and was impermeable to Cl-. The channel was permeable to Ca2+. In symmetrical 140 mM K+, the current-voltage relation was linear with a single channel conductance of 36 +/- 3 pS. Depolarization increased channel open probability. Interestingly, depending on the membrane patch studied, application of negative pressure to the pipette caused either an increase or a decrease in the open probability of the channel already activated by swelling. Thus, the sensitivity to tension of the swelling-activated channel was different from those of previously reported stretch-activated channels. These findings suggest that nonselective cation channels exist in rat atrial cells and may be involved in swelling-induced changes in cell function.

Immunocytochemical study with anti-muscle actin antibody (HHF35) on myocardial ischaemia and reperfusion injury in rats.

Myocardial ischaemia and reperfusion injury in 16 anaesthetized Sprague-Dawley rats, eight of which were pretreated with morphine (5 mg/kg, intraperitoneally) to prevent arrhythmias, were studied immunocytochemically with anti-muscle actin specific monoclonal antibody (HHF35). Eight cases of simple ischaemia and eight cases of sham-operated rats were used as controls. With HHF35 ABC immunocytochemical method, the left ventricular myocardium in the reperfusion group (without morphine) showed large areas of staining loss, but no loss of staining was seen in controls. No significant changes were seen with H&E stain in all hearts. Compared with HBFP, acridine orange (AO), eosin-fluorescence, HHF35 ABC staining best demonstrated myocardial reperfusion injury. The results indicate that the degree of myocardial damage may be related to the arrhythmias.

A genetic region downstream of the hydrogenase structural genes of Bradyrhizobium japonicum that is required for hydrogenase processing.

Deletion of a 2.9-kb chromosomal EcoRI fragment of DNA located 2.2 kb downstream from the end of the hydrogenase structural genes resulted in the complete loss of hydrogenase activity. The normal 65- and 35-kDa hydrogenase subunits were absent in the deletion mutants. Instead, two peptides of 66.5 and 41 kDa were identified in the mutants by use of anti-hydrogenase subunit-specific antibody. A hydrogenase structural gene mutant did not synthesize either the normal hydrogenase subunits or the larger peptides. Hydrogenase activity in the deletion mutants was complemented to near wild-type levels by plasmid pCF1, containing a 6.5-kb BglII fragment, and the 65- and 35-kDa hydrogenase subunits were also recovered in the mutants containing pCF1.

Misreading of the argI message in Escherichia coli.

It has previously been shown that either phenylalanine codon, UUU or UUC, could be misread as leucine during phenylalanine starvation, if the codons encoded residue 8 of the Escherichia coli argI gene product, ornithine transcarbamylase (OTC). However, no leucine misincorporation was detected when either of these same codons encoded residue 3. Here we report that leucine misincorporation can be directed by a UUU codon for residue 3 of OTC during phenylalanine starvation, if the argI gene has been mutated so that the codon preceding the UUU has been changed from the rarely used glycine codon GGG to the more commonly used GGC.

Effect of helium-neon laser irradiation on serum lipid peroxide concentrations in burnt mice.

The effect of helium-neon irradiation on serum lipid peroxide concentrations in mice following 6-7% body surface area burns was investigated in a controlled study. Immediately following injury by an 8-sec 100 degrees C scalding, 25 mice were irradiated by a helium-neon laser at 0.05 J/cm2. A control group of the same size underwent identical treatment but received only sham irradiation. Serum lipid peroxide concentrations increased markedly in the control group at 0.5-4 h (P less than 0.0001, two sample t-test). In the laser treated group, the lipid peroxide concentrations remained relatively constant and were significantly depressed relative to the control group 4 h following burning (P less than 0.0001, two-sample t-test).

Nickel-dependent reconstitution of hydrogenase apoprotein in Bradyrhizobium japonicum Hupc mutants and direct evidence for a nickel metabolism locus involved in nickel incorporation into the enzyme.

A double mutant (JH103K10) was created from hydrogenase constitutive mutant (JH103) by replacement of a chromosomal 0.60 kb nickel metabolism related locus with a kanamycin resistance gene. The double mutant required 10 to 20 times more nickel (Ni) to achieve near parental strain levels of hydrogenase activity. In the absence of nickel, both JH103K10 and JH103 synthesized high levels of (inactive) hydrogenase apoprotein (large subunit, 65 kDa). With nickel, the double mutant JH103K10 synthesized the same level of hydrogenase apoenzyme (65-kDa subunit) as the JH103 parent strain; however, whole cell hydrogenase activity in JH103K10 was less than half of that in JH103, and the CPM (due to 63Ni in hydrogenase) of membranes and the calculated ratio of nickel per unit of hydrogenase enzyme of the double mutant were 40% of that in JH103. Therefore, the difference in hydrogenase activities between the double mutant and the Hupc strain can be accounted for by different abilities of the strains to incorporate nickel into the hydrogenase apoenzyme. The addition of nickel ions to previously Ni-starved and then chloramphenicol-treated Bradyrhizobium japonicum whole cells (JH103 and JH103K10) resulted in (an in vivo) restoration of hydrogenase activity, suggesting that the apoprotein synthesized in the Ni-free cultures could be activated by addition of nickel even in the absence of protein synthesis. The extent of reconstitution of active hydrogenase by nickel was greater in the absence of chloramphenicol. Hydrogenase apoprotein could not be activated by nickel in vitro even with the addition of ATP. The successful in vivo but not in vitro results suggest that enzymatic but cell-disruption labile factors are required for Ni incorporation into hydrogenase.

Treatment of essential hypertension with auriculoacupressure.

Auriculoacupressure was used in the treatment of 291 cases of essential hypertension, in comparison with 51 similar cases treated with Fufang Jiangya (composite hypotensive) Tablets. Analysis of results showed that the therapeutic effects in the 2 groups were similar, while the rate of marked efficacy in the acupressure group was higher than that of the medication group, and the level of blood lipids also significantly dropped (P less than 0.01); there was only minor improvement in blood rheology. In addition, acupressure manifested biphasic regulatory effects on sinus arrythmia. The curative effect of acupressure on essential vascular hypertension was good.

The effect of praziquantel on the circulating antigen SJ 70 in murine schistosomiasis japonica.

A circulating schistosome 70 kDa antigen (SJ 70) has been detected in the sera of mice infected with Schistosoma japonicum. For the detection of this antigen, a double antibody sandwich enzyme-linked immunosorbent assay (ELISA) was employed. SJ 70 is first detected in the serum of S. japonicum-infected mice 4 weeks after infection. Antigen levels rise rapidly, plateau by 7 weeks after infection, and remain relatively unchanged for at least another 9 weeks. In mice infected with S. japonicum for 7 weeks and then treated with praziquantel (100 mg/kg body weight), there is a significant decrease in serum antigen levels within 2 weeks after treatment, and an almost complete disappearance of antigen from the serum 3-4 weeks after treatment.

Detection of a circulating antigen in human schistosomiasis japonica using a monoclonal antibody.

A double antibody enzyme-linked immunosorbent assay (sandwich ELISA) was used for the detection of a circulating antigen from human schistosomiasis japonica infections. This assay involves the use of polyclonal rabbit Schistosoma japonicum soluble egg antigen (SEA) antiserum to bind circulating antigen and a monoclonal antibody (MabH4) to identify and quantify this antigen. Sera from 108 S. japonicum-infected patients (acute and chronic) were tested. Sera from 93 of 95 patients with chronic infection were positive for this antigen; sera from 12 of 13 patients with acute infections were also positive. Antigen was not detectable in control human sera. Sera from 35 chronic schistosomiasis patients were collected 6-12 months after praziquantel treatment. Circulating antigen was not detectable in the sera of 33 of these patients and was dramatically reduced in 2. This ELISA system may prove valuable in differentiating past and current infections.

[Effect of praziquantel treatment on the levels of circulating SJ70 antigen in murine schistosomiasis japonica].

SJ 70 antigen was first detected in the sera of S. japonicum-infected mice at the 4th week after infection and increased rapidly, reaching their peak by the 7th week after infection, and remaining relatively unchanged for additional 9 weeks. In contrast, in mice infected with S. japonicum for 7 weeks and then treated with praziquantel (100 mg/kg body weight) there was a significant decrease in serum antigen levels within two weeks after treatment. An almost complete disappearance of the antigen from the sera occurred by the 5th week after treatment. The results indicated that the detection of SJ 70 antigen might be used as early diagnosis of schistosomiasis and evaluation of drug efficacy.

[Use of double McAb sandwich ELISA to detect circulating antigen of Toxoplasma in infected rabbits].

This paper reports on the cumulative positive frequencies of circulating antigen (CAg) detected in the sera of rabbits infected with Toxoplasma by using double-McAb sandwich ELISA. The positive frequencies of rabbits with heavy and medium infection in the incubation period are 30.8% and 11.1%. Those with medium infection in acute, subacute and early chronic period are 86.1% and 76.7%, 43.3% and 32.0% with light infection. The positive rates of CAg in rabbits of medium and light infection rose progressively in acute period, but declined in subacute and early chronic period. Cross reaction with schistosomiasis and coccidiosis was all negative. This method of high specificity, sensitivity and duplication possesses certain value in the diagnosis of acute or active Toxoplasma infection and may be useful for the diagnosis in the early period.

Rifampicin polylactic acid microspheres for lung targeting.

Rifampicin polylactic acid microspheres for lung targeting were prepared by a modified emulsion-solvent diffusion method. The microspheres were free flowing, spherical with regular surface. Drug content, particle size distribution and in vitro release properties of the prepared microspheres were evaluated. In vivo experiments on rabbits showed remarkable accumulation of microspheres in the lung.

Health effects of occupational exposures to vehicle emissions in Shanghai.

The authors investigated the health effects of occupational exposures to vehicle emissions in 745 bus drivers, conductors, and taxi drivers, compared with 532 unexposed controls, in Shanghai. Logistic regression and general linear models were used to examine the relationship between exposure and respiratory illness. Results showed that the prevalences of some respiratory symptoms and chronic respiratory diseases were significantly higher (p < 0.05) in the exposed group than in the controls. The adjusted odds ratios for throat pain, phlegm, chronic rhinitis, and chronic pharyngitis were 1.95 (95% CI 1.55-2.46), 3.90 (95% CI 2.61-5.81), 1.96 (95% CI 1.11-3.46), and 4.19 (95% CI 2.49-7.06), respectively. Also, there were exposure time response relationships for the prevalences of phlegm and chronic respiratory disease. Pulmonary function and blood lead levels were not significantly correlated with exposure status. The results suggest that occupational exposure to vehicle emissions may induce detectable adverse health effects.
