Inhibitory effects of Candida albicans extracellular polysaccharides on mouse sarcoma 180.

Extracellular glycoprotein produced by Candida albicans in chemically defined medium was used to treat solid Sarcoma 180 tumor cells implanted subcutaneously in Swiss female mice. Neither toxicity nor skin ulcerations surrounding the tumors was observed. Significant tumor inhibition was obtained using different methods to evaluate treatment effectiveness. Comparison of tumor weights after a single treatment (400 mg/kg, ip) on various days after transplantation indicated significant inhibition with a 10--80% regression rate. Multiple doses (800--2,000 mg/kg, ip) regressed 60--90% of all tumors. Subsequent dosage experiments showed an increase percentage of survivors for all treated groups and a significant decrease in tumor size. Studies of the glycoprotein material indicated that it was immunologically active.

Influence of visible light on organ weights of mice.

Hau:ICR mice separated by sex, were reared for 30 days under various fluorescent lamps: pink, blue, black UV, cool white and full spectrum. Body weights and absolute organ weights were compared. After light exposure, female body weights were not significantly different between any groups; however, a difference in male body weights was observed. Light affected the weights of the pituitary, adrenals, kidneys and prostate in male mice and the adrenals, thyroid and pineal glands in females. The weight of adrenal glands of both males and females were most sensitive to changes in lighting.

A new quantitative procedure for proteinuria.

Abnormalities of tubular cells were treated by administration of nephrotoxin and studied by specialized gel filtration methods to discern early quantitative and qualitative changes in proteinuria. Dogs given nephrotoxic doses of Diglycoaldehyde, alpha-(hydroxymethyl)-alpha1-(6-hydroxy-9H-purine-9-yl) 60 mg per kg per day intravenously, for 3 days showed a marked reduction in effective renal plasma flow from 264.91 +/- 90.05 ml per min to 125.0 +/- 19.0 ml per min as measured by 131I sodium iodohippurate, and corresponding decrease in total renal perfusion by 131I xenon wash-out from 222.2 +/- 8.6 to 124.0 +/- 28.38. However, after treating four dogs with 20 mg per kg, an increase in effective renal plasma flow was obtained. Histologic changes of glomerular and tubular damage were demonstrated. Urine from this same series of animals was examined on a daily basis by sucrose-acrylamide gradient gel centrifugation together with ultraviolet scanning. Specific patterns were also obtained using dialyzed urine which eliminated molecules greater than 3500 mol wt. The ultraviolet pattern data were digitized and analyzed on computers to augment pattern recognition. These techniques indicted that molecules (3500 mol wt or less) were excreted in response to a known nephrotoxic agent, establishing early distinctive changes in tubular cells and also relating to alteration in renal function.

Lethality of Candida strains as influenced by the host.

A comparative study of the pathogenicity of Candida albicans morphological mutant strains was made to relate chlamydospore production, germ tube formation, and proteolytic activity to candidiasis in mice. It was observed that the mycelium strains were more lethal than the yeast-like strains and that neither chlamydospore production, germ tube formation, nor nutritional resquirements was related to pathogenicity in mice. Statistical analysis indicated that the culture media of the organisms and the strain and sex of the mice into which the cells were injected were important in the development of pathogenicity.
