Anti-inflammatory activity and conformational behavior of a branched (1 leads to 3)-beta-D-glucan from an alkaline extract of Dictyophora indusiata Fisch.

A (1 leads to 6)-branched (1 leads to 3)-beta-D-glucan (T-5-N), isolated from a M sodium hydroxide extract of the fruit bodies of Dictyophora indusita Fisch., markedly exhibited anti-inflammatory effects on both carrageenan-induced edema and scalded edematous hyperalgesia in rat's hindpaws. The activities of T-5-N (25 mg/kg i.p. X 2) were more potent than those of phenylbutazone (25-50 mg/kg i.p. X 2). The conformational behavior of T-5-N was studied. Its molecular weight in neutral solution was about three times that in 0.25M sodium hydroxide. This finding, in addition to the results of optical rotatory measurement and complex-formation with Congo Red, indicated that T-5-N has an ordered, triple-helical structure in neutral or slightly alkaline solution (less than 0.15M NaOH), and has single chains in highly alkaline solution (greater than 0.25M NaOH). The conformational transition occurs at concentrations of sodium hydroxide in the range of 0.15-0.25M.

Management of febrile outpatient neonates.

To determine whether all febrile outpatients below the age of two months should be routinely hospitalized and given parenteral antibiotics because of the possibility of occult sepsis or meningitis, 147 such admissions were reviewed over four years. None of the neonates had occult bacterial meningitis, while one may have had occult sepsis. Because serious infections may have been missed in outpatients not hospitalized, the outcome of all such illnesses seen in the clinic during a subsequent six months was monitored; none of 17 hospitalized or 20 followed as outpatients experienced sepsis or meningitis. If our findings are confirmed in other studies, febrile outpatient neonates with a well appearance, normal cerebrospinal fluid and physical examination, and dependable follow-up may not require universal hospitalization and antibiotics.

Circadian rhythm of apomorphine-induced stereotypy in rats.

The existence of circadian variations of apomorphine-induced stereotypy was determined. Male Wistar rats, standardized to a light-dark cycle (lights on from 7:00--19:00) for three weeks, were injected with apomorphine hydrochloride (1 mg/kg or 3 mg/kg) at one of six times (9:00, 13:00, 17:00, 21:00, 1:00 and 5:00). A significant time-of-day effect was found for apomorphine-induced stereotypy, with highest stereotypic score following injection at 13:00 or 17:00. The circadian rhythm of apomorphine-induced stereotypy was significantly fitted to a single cosine curve with a 24-hr cycle using the least squares method.
