Pyrazinoylguanidine: antihypertensive, hypocholesterolemic, and renin effects.

In a single-blind, placebo-controlled study of 12 subjects diagnosed as having mild to moderate hypertension and hypercholesterolemia, pyrazinoylguanidine (PZG) in a dose of 600 mg twice daily for 4 weeks reduced systolic blood pressure and heart rate. Pyrazinoylguanidine also reduced diastolic pressures, but to a lesser extent. Pyrazinoylguanidine reduced total serum cholesterol and low-density lipoprotein (LDL). Regression analysis indicated a dose-dependent reduction of both total cholesterol and LDL by PZG, i.e., the higher the presenting serum concentration, the greater the reduction by PZG. The extent of the reductions produced by PZG in elevated cholesterols and LDLs was highly correlated (r = .949). Normal high-density lipoprotein levels were unchanged by PZG. Pyrazinoylguanidine increased 24-hour urine volume and urinary excretion of sodium. Serum Na+, K+, or Cl- concentrations were unaltered. Means for plasma aldosterone and renin activities tended to decrease, but these trends did not attain statistical significance. Pyrazinoylguanidine was well tolerated. An activity profile that includes antihypertensive effects as well as reduction in hypercholesterolemia without major impact on serum renin or electrolyte balance makes PZG an attractive candidate for the management of hypertension.

[The problem of deeply situated parotid tumors].

Of 804 surgically treated parotid gland tumours, every fifth (19.9%) was situated medially from the 8th cranial nerve. Among these deep lobe tumours (DLT) there are more malignant tumours (36.8%) than in superficial lobe tumours (SLT; 11.4%). This fits especially the carcinoma arising in pleomorphic adenomas (14.3% in DLT; 3.1% in SLT). Postoperative complications of the 8th cranial nerve, defective tumour capsules, and necessary second or third look operations are more frequently in DLT than with SLT. Early diagnosis of tumours that are still small, is therefore very important.

External ocular inflammatory effects of lipoxygenase enzyme products.

We systematically observed the effects of lipoxygenase enzyme products (5-, 8-, 9-, 12-, and 15-HETE and leukotrienes (LT) C4, D4, and B4) on the external ocular inflammatory process in rabbits. Our results, using 1 and 10 micrograms enzymatic preparations topically applied to the conjunctiva, were consistent with the potent chemotactic activity of 12-HETE and LTB4. Modulation of the inflammatory process can be accomplished better, as a result of our findings, by inhibition of both the lipoxygenase and cyclooxygenase pathways.

Top concentrations of dynorphin-like immunoreactivity in fractions of rat anterior pituitary cells enriched in gonadotrophs.

A possible relationship between anterior pituitary cells containing luteinizing hormone (LH) and those containing the endogenous opioid dynorphin and other proenkephalin B-derived peptides was examined. Gonadotroph-enriched and gonadotroph-depleted cell fractions were prepared from cell suspensions of adult female rat anterior pituitary glands by centrifugal elutriation. Fractions with high or low concentrations of LH contained also high or low concentrations of dynorphin-like immunoreactivity. A positive correlation was found between the content in the eluted cell fractions of LH and dynorphin-like immunoreactivity with a correlation coefficient and a slope of the regression line close to one. Therefore, LH-containing and dynorphin-containing cells of the rat adenohypophysis exhibit almost the same characteristics under the conditions of centrifugal elutriation. This is consistent with the suggestion that dynorphin and other proenkephalin B-derived peptides may be colocalized with LH and/or follicle-stimulating hormone in at least some of the gonadotrophs of the rat anterior pituitary gland.

[First experiences with a new electroaerosolgenarator produced by VEB TuR Dresden (author's transl)].

First clinical experiences with a new developed electroaerosolgenator connecting with an ultrasonicaerosolgenerator are described. Propositions of corrections concern the material of the inonizating tube, which be produced of non fragile plastics and the position of exhalationway. By ionometer measuring we found a directly relation between chargetransport and inhaled volume and the particleconcentration. The transportcapacity was measured by 110.10(-9) Coulomb (C) per ml. One aerosolparticle of 1 micrometer carried more than 100 elementary charges.
