Alteration in the nucleosome and chromatin structures upon interaction with platinum coordination complexes.

The interaction of various platinum coordination complexes with nucleosomes and chromatin has been investigated by ultraviolet absorption spectrophotometry, circular and electric linear dichroism, and thermal denaturation, at low binding ratios (r less than 0.1-0.2). The general trend of the changes in these physicochemical properties is similar to that observed for the DNA-platinum complexes, which indicates that the same binding sites are involved in the platinum interaction with DNA and with its nucleoprotein complex. The cis-bidentate ligands, cis-dichlorodiammine, diaminocyclohexane and ethylenediamine platinum(II), showed a distinct behavior, with a more important destabilization of the DNA structure in the nucleoprotein than the trans-bidentate ligand, trans-dichlorodiammine-Pt(II), and monodentate ligand, diethylenetriamine-Pt(II). The drastic decrease of the negative electric dichroism in the 260 nm absorption band of the bases, observed with the five ligands, indicates a profound alteration of the DNA arrangement in chromatin and nucleosomes, attributed to a condensation of its superhelical structure. Some differences with previous observations on DNA complexes with the same platinum compounds indicate the possible formation of protein-DNA crosslinks in chromatin and nucleosomes. These could have some importance for the biological effects.

Do planar alkaloids from Strychnos usambarensis intercalate into the DNA helix?

The mode of binding of melinonine F and normelinonine F to DNA has been investigated through physico-chemical methods which proved most valuable in the study of dye-DNA interactions, namely absorption spectrophotometry, circular and linear dichroism, fluorescence polarization, viscosity increase of linear DNA and circular DNA unwinding. The spectroscopic properties resembled those observed with planar heterocyclic compounds intercalating between the DNA base pairs: bathochromic shift and hypochromism in the ligand absorption bands; presence of a strong binding process with an association constant of the order of 10(6)M-1 and a number of binding sites of 0.1 to 0.15; appearance of extrinsic optical activity in the ligand absorption bands with a dependence on the binding ratio typical for an excitonic origin; negative reduced electric dichrosim of the ligand chromophore of the same order of magnitude as that of the DNA heterocyclic bases. However the increase of the length of sonicated DNA determined from viscosity changes was much smaller than for most intercalating compounds and no unwinding of PM2 circular DNA was detected.

[Effects of cis-dichlorodiaminoplatinum II (cis-Pt II) on chromatin ultrastructure in animal or plant cells and repercussions on the cell cycle].

Cultivated animal cells (mouse peritoneal macrophages, chick fibroblasts or mouse Ehrlich tumor cells) or Zea mays root tips are treated with cis-Pt (II). Various effects (chromatin dispersion or condensation, pycnosis) are observed under some experimental conditions in all cell types. Cytoplasmic ribosomes in helical aggregates appear but only in vegetal cells. Mitosis and cell cycle disturbances due to cis-Pt (II) are probably related to chromatin alterations. We suggest that the latter and helical polyribosomes are produced by cis-Pt (II) reaction with nucleic acids in these structures.

The structural organization of dinucleosomes and oligonucleosomes. Electric dichroism and birefringence study.

The spatial organization of nucleosomes and linker DNA in dinucleosomes and oligonucleosomes of various chain lengths has been investigated through electric dichroism, birefringence and relaxation times measurements at low ionic strengths (0.5 to 2.2 mM). From the negative dichroism observed for all the samples, it is concluded that the nucleosome subunits in the oligonucleosome chain must lie with their disc planes closely parallel to the fibre axis. The large increase of the negative dichroism of dinucleosomes upon Hl removal is interpreted by the unwinding of the DNA tails and the internucleosomal segment. All the samples displayed, under bipolar pulses, a predominantly induced orientation mechanism.

Dissociation of double-stranded poly(I) . poly(C) by cis-diammine-dichloro-Pt(II).

The covalent binding of cis-Pt(NH3)2Cl2 on the double stranded poly(I) . poly(C) induced an irreversible dissociation of the two strands. This dissociation was evidenced mainly by poly(I)-Agarose affinity chromatography which allowed to recover free strands of cis-Pt(NH3)2Cl2-poly(I) from a cis-Pt(NH3)2Cl2-poly(I) . poly(C) complex, by density equilibrium centrifugation where free poly(C) could be isolated, and by acid titrations of the metal-poly(I) . poly(C) complexes. The separation of the two strands of the polyribonucleotide upon cis-Pt(NH3)2Cl2 fixation was shown not to exceed 90--95%. A dissociation curve of the polynucleotide double helix as a function of the amount of bound cis-Pt(NH3)2Cl2 was determined and was shown to be of a characteristic cooperative effect. The fixation of the paltinum compound to poly(I) . poly(C) seemed also to be cooperative.

Interaction of DNA and purine nucleosides with cis-dichlorodiammineplatinum(II) and antimitotic activity of the complexes on meristematic root cells.

The appropriate experimental conditions for the preparation of complexes of cis-dichlorodiammineplatinum(II) with DNA and with purine nucleosides have been determined which leave negligible amounts of free drug in the solution. Important conformational changes of DNA upon binding to cis-Pt(NH3)2-Cl2 have been evidenced through viscosity, electric birefringence and thermal denaturation experiments. The antimitotic and antitumor activity of the drug was found to be totally inhibited by its binding to DNA and to the purine nucleosides. Enzymic degradation observations on the DNA-cis-Pt(NH3)2Cl2 complexes indicated an important inhibition of the degradation and the absence of release of free drug. The implications of the results in relation with the mode of binding of this compound to DNA, with the choice of carriers for drugs and the mechanism of action of lysosomotropic agents are discussed.

Electric birefringence of DNA and chromatin. Influence of divalent cations.

The effects of divalent cations on the DNA and chromatin conformation have been investigated by electric birefringence and birefringence relaxation measurements at low and constant ionic strength (0.001). An important decrease of the intrinsic optical anisotropy of DNA has been found in the presence of Mn2+ and Cu2+, but not with Mg2+. A complex variation of the mean relaxation time with the ratio I/P of ion to DNA-phosphate molar concentration has been evidenced in the presence of Mn2+ and Cu2+, while the mean relaxation time monotonously decreased in the presence of Mg2+. These observations are interpreted in terms of a specific organization of DNA in a compact, rigid structure, in the presence of Mn2+ and Cu2+, and a non-specific coiling in the presence of Mg2+. Drastic conformational changes encountered by chromatin in the presence of Mg2+ and Mn2+ cations have also been evidenced through electric birefringence measurements. They are interpreted by the formation of a superhelical compact arrangement of nucleosome strings which yielded a reversal of the birefringence sign with respect to the negative anisotropy observed in the presence of Na+ ions. The removal of the histone H1 prevented the appearance of this quaternary structure. More extended fragments of the chromatin chain obtained by ECTHAM chromatography of sonicated chromatin could not afford such compact arrangements.

Electro-optical studies on synthetic polyelectrolytes. IV. Electric polarizability and conformation of poly-N-methyl-2-vinylpyridinium bromide in aqueous solution.

Measurements of the relaxation time on aqueous solutions of the title polyelectrolyte as a function of the concentration and of the molecular weight show that its conformation at very high dilution can be better accounted for by a weakly bending rod or wormlike chain model, with persistence length ranging from 200 to 400 A. The analysis of the field strength dependence of the electric birefringence yields electric polarizability values which increase sharply with the dilution and are not significantly dependent upon the molecular weight. This has been tentatively interpreted on the basis of the theories derived by Oosawa and by Mandel and Van der Touw. The partially flexible model proposed by the latter authors allow to identify the electric polarizability calculated from electro-optical data to the specific dielectric increment measured in the high frequency range; both parameters appear to be molecular weight independent. The electric polarizability obtained from the Kerr effect would be originated in the induced dipoles caused by the delocalization of the bound counterions along rigid subunits whose length seems however to differ from the persistence length.

The dimerization of protochlorophyll pigments in non-polar solvents.

The infrared, visible and nuclear magnetic resonance spectra of protochlorophyll a and vinylprotochlorophyll a in dry non-polar solvents (carbon tetrachloride, chloroform, cyclohexane) are presented and interpreted in terms of dimer interaction. The infrared spectra in the 1600-1800 cm-1 region clearly show the existence of a coordination interaction between the C-9 ketone oxygen function of one molecule and the central magnesium atom of another molecule. Infrared spectra in the OH stretching region (3200-3800 cm-1) provide a valuable test of the water content in the samples. The analysis of the absorption and circular dichroism spectra of protochlorophyll a and vinylprotochlorophyll a in carbon tetrachloride demonstrates the existence of a monomer-dimer equilibrium in the concentration range from 10(-6) to 5.10(-4) M. The dimerization constants are (6 +/-2).10(5)1.M-1 for protochlorophyll a and (4.5 +/-21.10(5) 1.M-1 for vinylprotochlorophyll a at 20 degrees C. The deconvolution of visible spectra in the red region has been performed in order to obtain quantitative information on the dimer structure. Two models involving a parallel or a perpendicular arrangement of the associated molecules are considered. From 1H NMR spectra, it appears that the region of overlap occurs near ring V, in agreement with the interpretation of the infrared spectra.

Interaction of ethidium bromide with ribosomes. Absorption, fluorescence, circular dichroism and sedimentation studies.

It is shown in this work that the binding of ethidium bromide to yeast ribosomes occurs through intercalation in the double-stranded rRNA regions and produces changes in the ribosomes structure, yielding unfolded subparticles, and even partial separation of proteins from rRNA at very high binding ratios. The addition of Mg2+ prevents these structural changes, probably by partial inhibition of the dye binding.

[Effects of the cis-dichloro-diammino-platinum(II)-deoxyribonucleic acid on normal and cancerous cells].

The effects of cis-dichloro-diammino-platinum(II) (cis-Pt) bound to DNA have been compared to those of free cis-Pt in mouse Ehrlich tumour cells, in peritoneal macrophages and in chick embryo fibroblasts cultivated in vitro. Cis-Pt has no antimitotic activity anymore when linked to DNA. This would be due to the fact that free cis-Pt is not released from cis-Pt-DNA complex inside lysosomes.

Binding of ethidium bromide to ribosomal RNA. Absorption, fluorescence, circular and electric dichroism study.

The interaction between ethidium bromide and ribosomal RNA has been studied by means of absorption, fluorescence, circular and electric dichroism measurements in the near ultraviolet and visible regions at low ionic strength (1 . 10(-3) and 6 . 10(-3). The results have been interpreted on the basis of a model of interaction involving the intercalation of the phenanthridinium ring of the dye in the double-stranded regions of the RNA molecule, resulting in an increase of the dye-dye interactions as compared to DNA, and a stiffening of the intercalation regions.

Electro-optical studies on synthetic polyelectrolytes. I. Ultraviolet electric dichroism of poly-N-alkylvinylpyridinium bromides in aqueous solution.

The electric dichroism of aqueous solutions of poly-2- and 4-vinylpyridinium bromides has been measured in the 220 to 300 nm region. From the knowledge of the assignments of the absorption bands, the orientation of the pyridinium ring with respect to the macromolecular axis was determined for the poly-N-butyl-4-vinylpyridium bromide. An estimation of the birefringence contribution in the visible region originating from these ultraviolet dichroic bands revealed that the negative birefringence observed mainly arose from electronic transitions at shorter wavelengths.

Physico-chemical study of the complexes of "33258 Hoechst" with DNA and nucleohistone.

The degree of binding of "33258 Hoechst" to DNA and nucleohistone has been determined by equilibrium dialysis and the properties of the complexes have been followed by different optical and electro-optical methods, after determining the orientation of the main transition moments within the dye molecule. The binding isotherm was found composed of a Langmuir-type and of a strongly cooperative component. The existence of two bound species yielded a continuous variation of most of the properties of the complexes studied as the amount of binding increased, while the hydrodynamic properties of the macromolecules were not affected. At low binding, the strongly bound dye molecules appeared to bind to highly fluorescent sites with their long axis oriented at 45 degree to the helix axis. As the binding proceeds, less fluorescent sites are cooperatively occupied and the inclination of these ligand molecules becomes closer to that of the base planes. These results are compatible with the formation of two external complexes with the double helical structure.
