Identification and determination of oligomeric stilbenes in the roots of Caragana species by capillary electrophoresis.

A method based on capillary electrophoresis with electrochemical detection (CE-ED) was employed for the determination of one alkaloid (hypaphorine) and the four oligomeric stilbenes, pallidol, kobophenol A, miyabenol C and (+)-alpha-viniferin in the roots of Caragana species. The five analytes could be well separated within 12 min in a 40 cm length capillary at a separation voltage of 12 kV in a 100 mmol/L borate buffer (pH 10.0). The response was linear over about 3 orders of magnitude for all investigated analytes with detection limits (S/N = 3) ranging from 0.0385 to 0.111 mg/L. The five constituents presented in Caragana sinica can also be detected in the roots of other Caragana species, but their contents were quite different. It is demonstrated that CE-ED is a useful technique for the investigation of some electroactive constituents in plants.

Influence of water on the solubility of two steroid drugs in hydrofluoroalkane (HFA) propellants.

The objective of this research work was to investigate the influence of water level, temperature, and propellant composition on the solubility of two hydrophobic steroid drugs, triamcinolone acetonide (TAA) and beclomethasone diapropionate (BDP). pMDIs containing TAA or BDP, spiked water, and propellant blend with different ratios of HFA 134a and HFA 227 were prepared. The contents of the prepared pMDIs were filtered through a 0.22 mm Acrodisc, syringe filter into a receiving canister after the pMDIs were equilibrated at 15 degrees C, 25 degrees C, 30 degrees C, and 40 degrees C. The drug concentration in the receiving canisters was determined by HPLC and the drug solubility in the propellant blend was calculated. Also, the drug crystal collected on the filter from the donor pMDIs were characterized by x-ray diffraction. The solubility of TAA and BDP varied with propellant composition at all experimental temperatures investigated. The solubility of TAA and BDP increased as the temperature was increased at all propellant compositions and water levels studied, but decreased as the water level in the propellant system was increased at all compositions and temperatures. The x-ray diffraction results indicated that the water in the propellant system had no significant influence on the crystal characteristics of TAA in HFA propellant system, but had a significant impact on the crystal characteristics of BDP was higher than TAA at all propellant compositions, experimental temperatures and water levels investigated. The solubility of TAA and BDP was not only influenced by propellant composition and storage temperature, but also depended on the water level in the propellant system. As a consequence, the crystallinity of the drugs formulated in HFA propellant was influenced by the temperature, propellant composition and the water level in the propellant system. LRP: a new adhesion molecule for endothelial and smooth muscle cells.

We recently generated a monoclonal antibody that disrupted the association of endothelial cells with their target location during kidney development. Here, we purified the antigen of this monoclonal antibody to homogeneity using rat mesangial cell cytosol. Sequence revealed that it is a previously identified protein, termed the "laminin receptor precursor" (LRP). We found that this protein is expressed in most tissues, but immunocytochemistry revealed that it is present largely or entirely in blood vessels where it is located underneath endothelial cells and in between smooth muscle cells of the vascular wall. Vascular smooth muscle cells such as mesangial cells produce and secrete LRP into their extracellular matrix where it is present in several molecular weight forms. Endothelial cells produce very little if any of the protein, but they bind avidly to LRP-coated dishes. Anti-LRP antibodies prevent the binding of smooth muscle cells to uncoated plates, implying that cells that secrete it use it for attachment. In an assay for heterologous cell-to-cell interaction, antibodies to LRP inhibited the binding of smooth muscle cells to endothelial cells. Maturation and differentiation of blood vessels require interaction between endothelial and smooth muscle cells. LRP is a new component of the mesangial matrix, and we propose that it is an adhesion molecule that mediates an interaction between smooth muscle cells and endothelia.

Exaggerated neurogenic inflammation and substance P receptor upregulation in RSV-infected weanling rats.

Respiratory syncytial virus (RSV) infection in adult rats causes exaggerated inflammation after sensory nerve stimulation in the extrapulmonary, but not in the intrapulmonary airways. The goal of this study was to analyze neurogenic inflammation in weanling F-344 rats infected with RSV 18 +/- 2 d after birth. Five days after RSV inoculation, the extravasation of Evans blue-labeled albumin after nerve stimulation was significantly greater in the intrapulmonary airways of RSV-infected weanling rats than in pathogen-free control rats. In contrast, no difference was found in the extrapulmonary airways. The level of messenger RNA (mRNA) encoding the substance P (SP) receptor (neurokinin 1 [NK1]) increased fourfold in RSV-infected lungs, whereas mRNA encoding the VIPR1 receptor for the antiinflammatory vasoactive intestinal peptide (VIP) increased to a much lesser degree. mRNAs encoding the other neurokinin (NK2) and VIP (VIPR2) receptors were not affected by the virus. Selective inhibition of the NK1 receptor abolished neurogenic inflammation in RSV-infected intrapulmonary airways. Also, neurogenic inflammation and NK1 receptor upregulation in infected lungs were inhibited by prophylaxis with a monoclonal antibody against RSV. These data suggest that RSV lower respiratory tract infection makes the intrapulmonary airways of young rats abnormally susceptible to the proinflammatory effects of SP by selectively upregulating the expression of NK1 receptors.

Field evaluation of resistance of transgenic rice containing a synthetic cry1Ab gene from Bacillus thuringiensis Berliner to two stem borers.

Two transgenic rice (Oryza sativa L.) lines, KMD1 and KMD2 at the R4 generation, transformed with a synthetic cry1Ab gene from Bacillus thuringiensis Berliner, were first evaluated for stem borer resistance in the field during the rice growing season of 1998 in two areas of Zhejiang Province, China. Both KMD1 and KMD2 were highly resistant to the stem borers Chilo suppressalis (Walker) and Scirpophaga incertulas (Walker), and were completely undamaged during the whole rice growing season. In contrast, damage to the plants of the untransformed parental control (Xiushui 11) was in the form of deadhearts or whiteheads. Under natural infestation by the C. suppressalis, the damage to control plants reached a peak of 88.7% of plants and 20.1% of tillers encountered with deadhearts. Under artificial and natural infestation of neonate striped stem borers at the vegetative stage and booting stage, 100% of plants and 25.6% of tillers, 78.9% of plants and 15.6% of productive tillers among artificially infested control plants were observed with the symptom of deadhearts and whiteheads, respectively. Damage to the control plants from artificial infestation by the S. incertulas reached a peak of 97.0% of plants and 22.9% of tillers damaged. The field research indicated that both KMD1 and KMD2 show great potential for protecting rice from attack by these two stem borers.

Tic convulsif caused by cerebellopontine angle schwannoma.

A case is presented of painful tic convulsif caused by schwannoma in the cerebellopontine angle (CPA), with right trigeminal neuralgia and ipsilateral hemifacial spasm. Magnetic resonance images showed a 4 cm round mass displacing the 4th ventricle and distorting the brain stem in the right CPA. The schwannoma, which compressed the fifth and seventh cranial nerves directly, was subtotally removed by a suboccipital craniectomy. Postoperatively, the patient had a complete relief from the hemifacial spasm and marked improvement from trigeminal neuralgia. The painful tic convulsif in this case was probably produced by the tumor compressing and displacing the anterior cerebellar artery directly.

Serial prostate specific antigen, free-to-total prostate specific antigen ratio and complexed prostate specific antigen for the diagnosis of prostate cancer.

PURPOSE: The free-to-total prostate specific antigen (PSA) ratio and complexed PSA have been introduced as adjuncts to total PSA for prostate cancer screening. Little data exist on the use of these tests for serial PSA screening. We compared serial total PSA, the free-to-total PSA ratio and calculated complexed PSA in men diagnosed with prostate cancer and matched controls in a population based study. MATERIALS AND METHODS: We identified 90 men diagnosed with prostate cancer between 1988 and 1996 with at least 3 serial serum samples obtained at 2-year intervals who were participants in the beta-Carotene and Retinol Efficacy Trial for the prevention of lung cancer. Samples were available up to 10 years before diagnosis. A total of 90 age matched men from the same cohort without prostate carcinoma were identified as controls. Free and total PSA was measured by the Abbott AxSYM system. RESULTS: Baseline demographics of cases and controls were similar. At baseline and diagnosis the men with prostate cancer had higher total and complexed PSA, and a lower free-to-total PSA ratio than controls. Mean followup was 5.2 years in cases and 5.5 in controls. The yearly change in PSA parameters in cases versus controls was 20.7% versus 3.5% for total, -3.4% versus 0.2% for free-to-total and 21.5% versus 3.4% for complexed PSA (p <0.0001). At diagnosis PSA alone was estimated to perform with more than 90% specificity in our model. CONCLUSIONS: In this population based study total PSA was superior to the free-to-total PSA ratio for predicting the development of prostate cancer. While serial changes in free-to-total PSA ratios with time were statistically significantly different in men diagnosed with prostate cancer and controls, the magnitude of these serial changes were slight enough to render them clinically insignificant.

[Effects of volatiles from different trophic level on foraging behavior of Aphidius avenae].

The responses of female Aphidius avenae to volatiles from undamaged wheat plant, Sitobion avenae and Rhopalosiphum padi, aphids-plant complexes and aphids damaged plant were investigated in wind tunnel and Y-tube olfactometer. The volatiles from undamaged wheat plant, S. avenae and R. padi were much less attractive to A. avenae than those from aphids-plant complexes and aphids-damaged plant. Although the parasitic rate was much lower, the R. padi-plant complexes and R. padi damaged plant were also attractive to A. avenae. Identification of volatiles by GC-MS showed that the main volatiles induced by aphids feeding were 2-camphene, 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol, Z-3-hexenyl-acetate, and methyl salicylate, among which, 6-methyl-5-hepten-2-one and 6-methyl-5-hepten-2-ol were more attractive to A. avenae, while methyl salicylate was not attractive.

[Effect of temperature, relative humidity and photoperiod on embryonic development and egg hatch of Pirata piraticus].

In this paper, the effects of temperature, relative humidity and photoperiod on embryonic development and egg hatch of Pirata piraticus were investigated. Temperature affected not only the whole course of embryonic development, but also its different phases. At 20 degrees C-35 degrees C, as temperature increased, the embryonic development became faster. The developmental threshold of egg was estimated to be at 11.9 degrees C. The uniform degree of egg hatch was higher at low temperature, while the highest hatching rate was happened at 28 degrees C. In order to study the factors affecting embryonic development and to determine the optimum condition for egg hatch, experiments were arranged by quadratic regression rotation composite design with three factors: temperature (X1), relative humidity (X2) and photoperiod (X3). Three models were built up, and the factors affecting the embryonic development were analyzed. After calculating the hatching rate using the model of hatching rate by computer, the optimum condition for egg hatch of the spider was given: temperature 27 degrees C-28.5 degrees C, relative humidity 94%-97%, and photoperiod 14-17 h.

[Study on expression of P53mt protein in skin of patient with arseniasis caused by coal-burning].

OBJECTIVE: To study expression of P53mt protein in skin of patient with arseniasis caused by coal-burning. METHODS: According to pathological diagnosis, the patients were divided into carcinoma group (A group, 18 cases), pre-carcinoma group (B group 11 cases) and general pathological change group (C group, 39 cases). Immuno-histochemical technique of two steps was used to detect expression of P53 mt protein. RESULTS: Among A, B and C groups, the positive cell densities of P53mt protein were 38.07 +/- 29.00, 17.16 +/- 15.00 and 4.05 +/- 8.24 respectively, but no positive cell in normal group; there was statistical significance among A, B and C groups (A and C: P < 0.01, A and B: P < 0.01, B and C: P < 0.05); positive percentages of A, B and C groups were 88.89%, 72.73% and 25.64% (A and C: P < 0.01; B and C: P < 0.05), respectively, for difference of grade constitution, there was statistical significance (P < 0.01) among three groups; grade II and III made up 38.89% in A group, but 9.09% in B group and 2.56% in C group. CONCLUSIONS: P53mt gene play a certain role in the development of skin cancer from arseniasis caused by coal-burning; the detection of P53mt protein can show whether pathological changes of skin due to the disease is malignant or benign.

[Effects of baicalin on hepatocyte apoptosis induced by TNF-alpha and act D in rats].

OBJECTIVE: To study the effects of baicalin on rat hepatocyte apoptosis induced by tumor necrosis factora (TNF-alpha) and actinomycin D (Act D) in vitro. METHODS: Hepatocytes were cultured with different concentrations of baicalin and the cell apoptosis was induced by TNF-alpha and Act D in vitro. After culturing for 24 h, the activity of hepatocysts was examined by MTT assay and the function of hepatocytes for secreting albumin was detected by bromocresolum Viride method. Agarose gelelectrophoresis and flow cytometric analysis were used to determine the apoptotic cells. RESULTS: The actively of the hepatocyte (A) and content of albumin (ALB) in the supernatant of cultured cells in the groups of 0.2 microgram.ml-1 and 2.0 micrograms.ml-1 of baicaline were significantly higher than those in apoptotic model group (A: P < 0.01; ALB: P < 0.01); respectively Even the ALB value in the group of 0.2 microgram.ml-1 baicalin was higher that of black control group (P < 0.01); The clear "ladder-like" streak-band of DNA fragments was found only in the apoptosis model group by the agarose gel electrophoresis. The flow cytometry showed that all the apoptosis rates in the three baicalin groups were lower than that of the apoptotic model group (P < 0.01). CONCLUSION: Baicaline can restrain rat hepatocyte apoptosis induced by TNF-alpha and ActD in vitro.

[Molecular cloning and expression in E. coli of the surface-exposed lipoprotein LipL41 gene of Leptospira lai].

OBJECTIVE: To construct a recombinant expressive plasmid using LipL41 gene of leptospira lai for further research of subunit vaccine of leptospira. METHODS: A pair of oligonucleotide primers were designed based on the sequence of LipL41 of leptospira kirschneri RM 52 in Genbank. With genomic DNA of leptospira lai 017 as template, a fragment was amplified by PCR and DNA sequencing analysis showed this fragment to be the gene that encodes LipL41. A recombinant vector was constructed using plasmid pGEX1 lambda T and the expression of LipL41 gene was tested. RESULTS: The production of PCR was LipL41 gene. The recombinant plasmid was constructed and testified by nuclease digestion and PCR, and LipL41 gene could express in E. coli. CONCLUSION: The recombinant plasmid has been constructed successfully and LipL41 gene can express stably at high level.

[DNA sequence analysis and expression of the recombinant plasmid pBX1 from Borrelia burgdorferi B31 strain].

OBJECTIVE: This study was to provide the target antigen for the development of a Lyme disease vaccine and serodiagnosis reagent. METHODS: We used the automatic DNA sequencing machine (Model 377) to detect the nucleotide sequence of the inserted part of the recombinant plasmid pBX1 from Borrelia burgdorferi B31 strain. The restriction enzyme map of the inserted part of pBX1 was analysed by using computer software. The expressed product of pBX1 in E. coli XLI-Blue MRF was analysed by using SDS-PAGE and western-blotting. RESULTS: 1. DNA sequencing showed that pBX1 contained a 477bp inserted gene fragment, and when it was compared with the published sequence of the specific region of the gene of the 83 kd antigen protein from Borrelia burgdorferi B31 strain, only one amino acid codon was different. 2. The restriction enzyme map of the inserted part of pBX1 was successfully constructed. 3. The recombinant plasmid pBX1 expressed a 29 kd fusion protein in E. coli XL1-Blue MRF' after induced with IPTG. The recombinant fusion protein could be recongnized by rabbit polyclonal antiserum against Borrelia burgdorferi B31 strain. CONCLUSION: A recombinant plasmid which contains the gene fragment encoding the specific region of the 83 kd antigen protein from Borrelia burgdorferi B31 strain has been successfully constructed. The recombinant plasmid can stably express 29 kd fusion protein in E. coli XL1-Blue MRF'. These results could serve as a base of further studies on the usefulness of the fusion protein in serodiagnosis and vaccine for Lyme disease.

Proniosomes: a novel drug carrier preparation.

A procedure is described for producing a dry product which may be hydrated immediately before use to yield aqueous niosome dispersions similar to those produced by more cumbersome conventional methods. These 'proniosomes' minimize problems of niosome physical stability such as aggregation, fusion and leaking, and provide additional convenience in transportation, distribution, storage, and dosing. This report describes the preparation of dispersions of proniosome-derived niosomes, comparison of these niosomes to conventional niosomes, and optimization of proniosome formulations. In addition, conventional and proniosome-derived niosomes are compared in terms of their morphology, particle size, particle size distribution, and drug release performance in synthetic gastric or intestinal fluid. In all comparisons, proniosome-derived niosomes are as good or better than conventional niosomes.

Mycobacterium tuberculosis infection in complement receptor 3-deficient mice.

Complement receptor type 3 (CR3) present on macrophages is used by Mycobacterium tuberculosis as one of its major phagocytic receptors. In this study, we examined the in vivo significance of CR3-mediated phagocytosis on the pathogenesis of disease caused by M. tuberculosis. The outcome of tuberculous infection in mice deficient in the CD11b subunit of CR3 (CR3-/-) on a mixed 129SV and C57BL background and control wild-type counterparts was comparable with respect to survival, bacterial burden, granulomatous lesion development, and cytokine expression in the spleen and lungs. M. tuberculosis infection was also examined in CR3-/- mice on C57BL/6 and BALB/c backgrounds and was found to be similar. In conclusion, our results suggest that in the absence of CR3, M. tuberculosis is able to gain entry into host cells via alternative phagocytic receptors and establish infection. The data also indicate that absence of CR3 does not alter disease course in either the relatively resistant C57BL/6 or the relatively susceptible BALB/c strains of mice.

Epidermal growth factor (EGF) suppresses staurosporine-induced apoptosis by inducing mcl-1 via the mitogen-activated protein kinase pathway.

Overexpression of epidermal growth factor receptor (EGFR) and establishment of transforming growth factor alpha (TGF alpha)/EGF autocrine system are frequently detected in tumor cells. In addition to mitogenic ability, we demonstrate in this report that EGF protects a human esophageal carcinoma (CE) cell line, CE81T/VGH, from staurosporine-induced apoptosis. The anti-apoptotic signal of EGF is alleviated by a MEK inhibitor PD98059 or an ERK2 dominant negative mutant but not by a phosphatidylinositol-3'-kinase (PI-3K) inhibitor wortmannin. Furthermore, v-raf blocks apoptosis induced by staurosporine. This evidence implies that the survival signal of EGF is mediated via the Raf-MEK-ERK pathway but not the PI3-K pathway. The survival effect of EGF is coincident with the induction of mcl-1, an antiapoptotic gene in the bcl-2 family. PD98059 also suppresses the induction of Mcl-1 by EGF, implying that EGF may up-regulate Mcl-1 via the MAP kinase pathway. Overexpression of mcl-1 is sufficient to protect against apoptosis, while transfection of a mcl-1 antisense plasmid causes cell death. The expression of mcl-1 antisense plasmid also suppresses the anti-apoptotic effect of EGF. Taken together, these results indicate that EGF may up-regulate Mcl-1 through the MAP kinase pathway to suppress apoptosis.

Radiolabeling brachytherapy sources with Re-188 through chelating microfilms: stents.

Rhenium-188 (Re-188, T(1/2) = 17 h) emits beta particles (E(max) = 2. 12 MeV) having an ideal range for intravascular brachytherapy and certain cancer brachytherapies. Re-188 was attached to metal wafers and stents via a chelating microfilm, and these brachytherapy sources characterized in vitro and in vivo. To prepare the sources, a siloxane film containing reactive amines was plasma deposited on the metal, a chelating microfilm conjugated to the amines, and the chelating microfilm used to attach Re-188. Re-188 was selectively bound to materials coated with the chelating microfilm. Binding correlated with the amount of radionuclide used. Wafers (1 cm(2)) bound up to 62.9 MBq (1.7 mCi) of Re-188 with yields generally near 30%. Stents bound up to 26.6 MBq (720 microCi). Typically, stents were labeled to bind 4-12 MBq and deposit 10-30 Gy at 2 mm in the arterial wall. In phantom studies, the longer nitinol stents deposited doses of 2.3 Gy/MBq (0.085 Gy/microCi), while shorter stainless steel stents deposited 4.62 Gy/MBq (0.171 Gy/microCi). After placement in arteries of pigs, only the Re-188-stents were detected by scintigraphy at times up to 24 h. Scintigraphy did not detect activity in other organs. Blood sampling (0.1-24 h) detected maximum radioactivity (up to 388 cpm/mL/100micro Ci) at 6 h. We conclude that on-demand radiolabeling of stents and other brachytherapy sources with Re-188 can be performed routinely.
