Repression at a distance by the global regulator KorB of promiscuous IncP plasmids.

KorB protein (358 amino acids) binds to 12 highly conserved sequences on the RK2 genome and co-ordinates the expression of at least five operons encoding genes for stable inheritance and plasmid transfer. KorB represses the trfA, korA and klaA promoters where it binds 4 bp upstream of the -35 region (class I KorB operators, OB). We show here that KorB on its own can also repress the trbA, trbB, kfrA and kleA promoters where OB is between 80 and 189 bp away from the transcription start point (class II operator). A C-terminal deletion of 17 amino acids resulted in the loss of KorB's ability to repress through class II operator but not through class I operator. This deletion reduced multimerization of His6-tailed KorB protein in vitro and greatly reduced binding specificity for fragments containing OB sequences. At the trbBp region, where OB9 lies 189 bp upstream of the transcription start point, mutagenesis of a proposed secondary binding site overlapping the trbBp -35 region had no effect on the ability of KorB to repress trbBp. Nevertheless, gel retardation analysis showed that KorB binding is promoted by sequences upstream and downstream of OB9 and that KorB can form higher order complexes on DNA. However, DNase I footprinting suggested that RNA polymerase may interact directly with KorB bound at OB9 and implied that contacts between these proteins could be responsible for the action of KorB at a distance.

A new continuous positive airway pressure (CPAP) device.

While continuous positive airway pressure (CPAP) is being increasingly employed for many forms of acute respiratory failure and postoperative hypoxaemia, most CPAP systems are either 'home made' or incorporated in sophisticated ventilators used in intensive care units. This paper describes the development of a continuous low flow CPAP system using a large latex reservoir bag and venturi generated positive end expiratory pressure (PEEP). The system has been successfully used for over twelve months in the general wards, the intensive care unit, accident and emergency department, the recovery ward and the coronary care unit.

Preparation and usefulness of frozen blood.

Red blood cell freezing is now practiced widely. Advantages of deglycerolized red cells over whole blood or packed cells include indefinite preservation, leukocyte destruction and removal, and plasma removal. Deglycerolized red cells are a purer blood component than whole blood or conventional red cells.

Effects of neonatally administered-testosterone propionate on the development of target organs of sex hormones and the induction of mammary carcinomas by 7, 8, 12-trimethylbenz (a)anthracene in female Sprague-Dawley rats.

Female Sprague-Dawley rats were given 1.25mg testosterone propionate subcutaneously within 24 hours after birth and 30mg 7, 8, 12-trimethylbenz (a) anthracene per 1 kg of body weight intravenously at 50 days of age. In these neonatally androgenized rats, the increase of the anogenital distance, aplasia or hypoplasia of the vagina, persistent estrus on the vaginal smears and eosinophilic substance in mammary glands were observed, with no corpus luteum formation in the ovaries, and the induction of mammary carcinomas by 7, 8, 12-trimethylbenz (a) anthracene was strongly suppressed.

Biochemical sequences in the transformation of normal fibroblasts in adolescent rats.

Coarse powders of acid-insoluble matrix of diaphysis and calvarial parietal bone rapidly and consistently transformed fibroblasts into masses of cartilage and bone containing hemopoietic marrow. The transformant was encapsulated by fibroblasts within 24 hr to form a plaque. Transformation was restricted to the central thicknesses of the plaque. Under the stated conditions the alteration of the phenotype, fibroblast to chondroblast, was an unstable transformation, whereas the phenotype change, fibroblast to osteoblast, was stable. The transformation occurred on a rigid timetable of sequences. Measurements of alkaline phosphatase activity and incorporation of radioactive sulfate, phosphate, and calcium were sensitive and quantitative assays for the appearance of the transformed products, cartilage and bone.

Leukemia evoked with 7,8,12-trimethylbenz(a)anthracene in rat. I. Changes in spleen and thymus.

An early sign of erythroblastic leukemia in rat was nodule formation in the spleen. Hyperplastic foci of stem cells, indistinguishable histologically from leukemic stem cells, were found in the red pulp whereas the malpighian corpuscles were uninvolved. Anemia is a normal phenomenon in immature rats and the spleen of the prepubertal rat possesses considerable hemopoietic potential. Pulse-doses of 7, 8, 12-trimethylbenz(a)anthracene prevented the physiologic hematological development of maturing rats and was associated with subsequent development of leukemic stem cells in the red pulp of the spleen. Significant enzyme changes were observed in leukemic spleens. Compared with the spleens of normal littermates, the concentration of lactate dehydrogenase rose while that of malate dehydrogenase fell; the content of alkaline phosphatase rose whereas acid phosphatase fell. Increased alkaline phosphatase activity in leukemic spleen was attributed to nonleukemic foci of myelopoiesis.

Transformation of fibroblasts by allogeneic and xenogeneic transplants of demineralized tooth and bone.

Xenogeneic transplants of powdered, dehydrated, demineralized matrix of bone and tooth were well tolerated in three species of rodents. Differences between the species were found in competence of fibroblasts to be transformed into cartilage and bone in vivo by these preparations. Rat fibroblasts were most susceptible to transformation of this sort; they were transformed by demineralized dentin of guinea pig, mouse, and rat, and to a limited extent, by a specimen of decalcified human bone.

Selective destruction of adrenal cortex by pulse doses of derivatives of 12-methylbenz(a)anthracene.

Pulse doses of emulsions of ten polynuclear aromatic hydrocarbons were found to destroy the adrenal cortex selectively. The corticolytic hydrocarbons are derivatives of 12-methylbenz(a)anthracene which possess at position 7 an alkyl, methoxymethyl, formyl, or hydroxyalkyl group. It would appear that the active corticolytic agent is 7-hydroxyalkyl-12-methylbenz(a)anthracene, of chemical source or generated in metabolism.
