Pathology of experimental chlamydiosis in chicks.

Twelve one-day-old chicks were experimentally inoculated with Chlamydia psittaci derived from turkeys. Acute chlamydial septicemic lesions were induced by the inoculation into the air sac and trachea. No lesions were produced by the esophageal injection. Clinically, the affected chicks showed emaciation and mouth breathing, and were inactive while some birds died. Grossly, they had hepatomegaly, splenomegaly and airsacculitis. Histopathologically, fibrinopurulent airsacculitis, pneumonia and bronchitis, multiple fibrinous serositis in the hepatic and splenic capsules, peri- and epicardium, and mesenterium, focal endoarteritis in the aortae, activation of reticuloendothelial cells in the spleen, and hepatic necrosis were noted. Immunohistochemically, chlamydial antigen granules were present in the cytoplasm of epithelial cells of the respiratory system, hepatocytes, macrophages in the air sac, lung, serous membrane, liver, spleen, aortae, reticuloendothelial cells in the spleen, and mesothelial cells in various organs or tissues. Chlamydial multiplication in the cells of the organs or tissues involved was preceded to form the lesions.

Evaluation of pathogenicity and protective efficacy of serotype 2 Marek's disease virus from birds belonging to genus Gallus in Japan.

The new cloned serotype 2 Marek's disease viruses (MDV) of ML-6, ML-9, and ML-22 strains were inoculated in specific-pathogen-free (SPF) chicks to evaluate the pathogenicity and protective efficacy. Chicks inoculated or contact-infected with ML strains showed no gross and histological lesions in lymphoid organs, sciatic plexuses and other visceral organs during 10 weeks of observation periods, indicating that the viruses were non-pathogenic. Moreover, the viruses were found to be spread horizontally among chicks by demonstrating the presence of viremia in contacted chicks at 2 weeks-old. Chicks vaccinated with ML-6 at one day-old were protected against subsequent challenge by inoculation with virulent MDV strain of Md/5 at 4 or 7 days old or by contact infection at 7 days old with chickens previously inoculated with the same strain.

Human focal nodular hyperplasia-like lesion in the liver of a cat.

A young cat clinically displaying marked ascites had multiple nodule hyperplasia in the liver. One of the nodules sampled at exploratory laparotomy was studied histopathologically. The nodule consisted mainly of hyperplastic hepatocytes with a central stellate scarlike connective tissue. The connective tissue had thick-walled blood vessels and radiating bands of fibrous connective tissue extending peripherally, dividing the nodule into pseudolobules. The lesions resembled focal nodular hyperplasia (FNH) in man. Serial section revealed a single large artery with the splitting of the internal elastic lamina running into the central connective tissue and branching to connect with the main vessels. The arterial branchings formed a spider-like structure. From these findings, it was suggested that the nodule had developed from a hepatic response to pre-existing vascular anomalies as had been reported in human FNH.

Bone lesions in pigs with locomotor dysfunction of hind legs.

A study was made of the femur, sternum and rib of 10 male Yorkshire x Landrace pigs weighing 87 to 179 kg at 9 months. Locomotor dysfunction of the hind legs occurred in all animals from 5 to 9 months of age. Noteworthy gross changes were a markedly open angle between the long axis of the femoral head and that of the great trochanter, and an abnormally high position of the top of the great trochanter in relation to the level of the femoral head. Histological changes in the femoral growth plate were characterized by oblique arrangement of chondrocytic columns, necrosis of the cartilage in the area adjacent to the metaphysis or epiphysis, eosinophilic streaks, and loosening of the cartilage matrix. Cracks with fractures of the cortex, and eosinophilic streaks and loosening of the cartilage matrix were observed in the growth plate of the sternum. Oblique arrangement of chondrocytic columns, eosinophilic streaks and loosening of the cartilage matrix were found in the growth of the rib.

Histogenesis of neoformation in the endocrine pancreas of aging horses.

Pancreatic tissue from 20 horses was examined using immunocytochemical techniques. In aged horses, neogenesis of endocrine cells, neoformation, and hyperplasia of islets occurred closely associated with the pancreatic duct; these changes were regarded as nesidioblastosis. In addition, pancreatic fibrosis accompanied by ductal proliferation and endocrine neogenesis was considered a regenerative change. Thus, the origin of neoformation in the endocrine pancreas was in the ductal system, and it is suggested that the pancreatic endocrine cells were of endodermal origin.

Immunocytochemical component of endocrine cells in pancreatic islets of horses.

The endocrine cell components in the pancreatic islets of the following 4 pancreatic regions of the horse were investigated by immunohistochemical methods: lobus pancreatis sinister (left lobe); lobus pancreatis dexter (right lobe); and 2 regions of Corpus pancreatis (body), the duodenal lobe which lies along the cranial duodenal flexure and descending duodenum, and the intermediate lobe which is situated around the portal vein. The islets in the left and intermediate lobes contained a central mass of glucagon cells surrounded by insulin cells, a few somatostatin cells and sporadic pancreatic polypeptide (PP) cells. On the other hand, the islets in the duodenal lobe were small in size compared with the other 3 regions, and were predominant in insulin and pancreatic polypeptide (PP) cells, but almost lacked in glucagon cells. These findings suggested that the duodenal lobe was derived from the ventral pancreatic primordium, and the left and intermediate lobes were originated from the dorsal pancreatic primordium. In the right lobe, the composition and distribution of the islet cells were almost the same as those in the left and intermediate lobes, but there were several lobules containing numerous PP cells as seen in the duodenal lobe.

Bone lesions in clinically normal and in lame pigs.

A pathological study was made of the femur, rib and sternum of 76 clinically normal Yorkshire X Landrace pigs weighing 2.8 to 100.0 kg (10 days to 6 months of age) and on 26 lame Yorkshire X Landrace weighing 134.9 to 149.1 kg (9 months). Locomotor dysfunction of the hind legs occurred in all animals 6 months old or more and progressed with increase in age. In clinically normal pigs, unevenness of the width of the femoral growth plate was seen in pigs weighing 20 kg or more. Furthermore, the top of the great trochanter was lower than that of the femoral head, and the angle between the long axis of the femoral head and that of the great trochanter became gradually larger with increasing body weight. Histological changes in the femoral growth plate were characterized by necrosis of the cartilage in the area adjacent to the metaphysis or epiphysis, eosinophilic streaks, loosening of the cartilage matrix, and oblique arrangement of chondrocytic columns. These changes were seen in the pigs weighing 2.8 kg or more. In the rib, changes in the growth plate were essentially similar to those in the femur. No appreciable changes were seen in the sternum. In the lame pigs, noteworthy gross changes were an abnormal high position of the top of the great trochanter in relation to the level of the femoral head and a markedly open angle between the long axis of the femoral head and that of the great trochanter. Although the nature of the histological changes in the femur is essentially the same as those of clinically normal pigs, the changes such as oblique arrangement of chondrocytic columns and loosening of the cartilage matrix were more marked than those of the clinically normal pigs. Loosening of the cartilage matrix and oblique arrangement of chondrocytic columns were found in the growth plate of the rib. Cracks, eosinophilic streaks and loosening of the cartilage matrix were also observed in the growth plate of the sternum.

Aseptic necrosis of bone in ICR mice.

Aseptic bone necrosis was observed in the tibia of 23 ICR mice. Histological changes were characterized by a loss of marrow tissue with proliferation of connective tissue and bone necrosis with empty osteocytic lacunae. Focal necrosis was confined beneath the articular cartilage. Extensive necrosis was present in half or all of the epiphysis. Massive necrosis was noted in the diaphysis of one animal. It was considered that focal necrosis might be related to degenerative osteoarthritis, and that extensive and massive necroses might have been caused by a disturbance of the blood supply.

Osteosclerosis in F344/DuCrj rats.

Osteosclerosis was observed in the tibia and sternum in F344/DuCrj rats of both sexes at 6, 18 and 30 months of age. The lesion first seen was a proliferation of osteogenic tissues on the marrow surface of the cortical bone and bone trabeculae, resulting in replacement of the marrow cavity by lamellar bone. Most of the affected rats had associated degenerative osteoarthrosis and regressive changes of the growth plate. Osteosclerosis was considered to be an aging change, lesions were observed at 6 months and increased in frequency with age.

Experimental infection of chickens with Cryptosporidium.

To study the clinical and pathological manifestations of Cryptosporidium infections, 24 h-old chickens were inoculated via four routes: orally, nasally, cloacally and by contamination, using 10(5) oocysts of Cryptosporidium in each case. The chickens began to shed oocysts of Cryptosporidium on days 7 to 8 after inoculation. There was no difference in the clinical manifestations or histological damage with respect to the inoculation routes. Various developmental stages of Cryptosporidium were also demonstrated microscopically in the smears or sections of bursa of Fabricii, larynx, trachea and cecal tonsils of inoculated chickens. Although the intestinal mucosa and respiratory tract mucosa were scarcely infected with the parasites, the bursa Fabricii was heavily infected in association with hypertrophy and hyperplasia of the lining epithelial cells.
