Synthesis of nitrosomethylisoamylamine from isoamylamine and sodium nitrite by fungi.

Nitrosomethylisoamylamine (NMIA), a carcinogenic N-nitroso compound was synthesized from isoamylamine (IAA) in a glucose-ammonium nitrate medium after several days' incubation with fungi and subsequent nitrosation with sodium nitrate. The nitrosamine was not produced by control reactions which lacked either IAA or fungi. The yield of NMIA varied with the length of incubation after inoculation with the fungi, and with the concentrations of IAA and NaNO2, the duration of nitrosation, the pH of the medium and the species of the fungi. The optimum conditions for the formation of the nitrosamine are reported.

[Induction of papilloma and carcinoma in the forestomach of mice by in vivo formation of N-3-methylbutyl-N-1-methylacetonylnitrosamine (MAMBNA)].

Forestomach papilloma and carcinoma, as well as liver lesion, were induced in mice by gavaging precursors of the new nitrosamine, MAMBNA (N-3-methylbutyl-N-1-methylacetonylnitrosamine) and NaNO2. The results were-similar to those in mice and rats fed on preformed MAMBNA compound. However, the induction of such tumors by in vivo formation of MAMBNA required longer time and much larger doses. Moreover, the lesions of epithelial hyperplasia in the urinary bladder, lymphoid tumor, intestinal carcinoma and interstitial-cell tumor of the testis also developed in some of the experimental animals. This may indicate that the intragastric synthesis of MAMBNA is less effective in the production of forestomach tumors in mice.

A new N-nitroso compound, N-2-methylpropyl-N-1-methylacetonyl-nitrosamine, in moldy millet and wheat flour.

A new N-nitroso compound, N-2-methylpropyl-N-1-methylacetonyl-nitrosamine (MAMPNA), was found in millet and wheat flour inoculated with Fusarium moniliforme Sheldon, a common species of fungi occurring in foods in Linxian County, after 8-day incubation and an addition of a small amount of NaNO2. The compound has been identified by GC-MS and confirmed by synthesized MAMPNA. The present paper reports the isolation and detection of MAMPNA in moldy foods, and discusses the role of the fungus in processes of nitrosamine formation and possible biosynthetic approach of the new compound.

Malignant transformation of baby hamster lung fibroblasts induced in vitro by a new nitrosamine compound.

A new nitrosamine compound, N-1-methylacetonyl-N-3-methylbutyl-nitrosamine (MAMB-NA), isolated from corn bread inoculated with moulds commonly occurring in Linxian County, a high incidence area for esophageal cancer in North China was tested for its potential carcinogenicity in vitro. Baby hamster lung fibroblasts (BHL) were treated with MAMBNA in tissue culture medium containing liver microsome preparation of Aroclor 1254 treated rat. Cell transformation was assessed by morphological changes, loss of density-dependent inhibition and unlimited growth in vitro, chromosome changes, colony formation on soft agar, growth at low serum concentration, Con A agglutinability and tumor formation after heterotransplantation into immunosuppressed newborn rats. Like the known carcinogens N-methyl-N'-nitro-N-nitroso-guanidine and diethylnitrosamine, MAMBNA was able to cause malignant transformation of cells cultured in vitro.

Mutagenicity in Salmonella typhimurium of N-3-methylbutyl-N-1-methyl-acetonyl-nitrosamine and N-methyl-N-benzylnitrosamine, N-nitrosation products isolated from corn-bread contaminated with commonly occurring moulds in linshien county, a high incidence area for oesophageal cancer in Northern China.

Two synthetic N-nitrosamines (N-3-methylbutyl-N-1-methyl acetonylnitrosamine and N-methyl-N-benzylnitrosamine), previously isolated from corn-bread which had been inoculated with moulds occurring in Linshien county, Northern China and subsequent nitrosation by sodium nitrite, were tested in Salmonella typhimurium strains TA1535 and TA100 in the presence of a liver postmitochondrial supernatant from Aroclor-treated rats. A concentration-dependent increase in the number of mutant colonies in both bacterial strains was observed when N-3-methylbutyl-N-1-methylacetonyl-nitrosamine was assayed in liquid suspension and N-methyl-N-benzylnitrosamine in plate incorporation assays. Our finding that mutagenic N-nitrosamines are present in foodstuffs that may be consumed in Linshien county are discussed in relation to the possible etiological role of these compounds in cancer of the oesophagus in that area.

Formation of carcinogenic N-nitroso compounds in corn-bread inoculated with fungi.

DMNA, DENA and MBNA are formed in corn-bread which has been inoculated with the most common species of fungi found in foodstuffs of Lin Xian County, Henan Province, such as F. moniliforme, A. flavus and others, and then, added with small amount of NaNO2 after a few days of incubation. These carcinogenic N-nitroso compounds can induce cancer of the liver or/and esophagus in experimental animals. Results of the present study show the capability of some fungi to produce chemical carcinogens as well as mycotoxins in contaminated food, and thus, open a new field of research in cancer etiology.

32P-postlabeling analysis of DNA adduction in mouse skin following topical administration of (+)-7,8-dihydroxy-7,8-dihydrobenzo[a]pyrene.

32P-Postlabeling was employed for analysis of DNA adducts produced in mouse skin following topical administration of enantiomers of 7,8-dihydroxy-7,8-dihydrobenzo[a]pyrene (BP-7,8-diol). Deoxynucleoside 3'-monophosphates were isolated by digestion of epidermal DNA with micrococcal endonuclease and spleen phosphodiesterase and phosphorylated with [gamma-32P]ATP. 32P-Labeled deoxynucleoside 3',5'-bisphosphate adducts to diastereomeric benzo[a]pyrene dihydrodiol epoxides (BPDE) were separated by four-directional thin-layer chromatography on poly(ethylenimine)-cellulose plates using a recently described solvent system [Reddy, A. P., Pruess-Schwartz, D., and Marnett, L. J. (1992) Chem. Res. Toxicol. (preceding paper in this issue)]. When (+)-BP-7,8-diol was topically administered, a major adduct spot was detected that cochromatographed with a standard produced by reaction of 7(S),8(R)-dihydroxy-9-(S),10(R)-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene [(+)-syn-BPDE] with DNA. The level of this adduct increased in a dose- and time-dependent fashion and was elevated in animals pretreated with beta-naphthoflavone. Relatively small amounts of radioactivity cochromatographed with standards of deoxynucleoside 3',5'-bisphosphate adducts derived from 7(S),8(R)-dihydroxy-9(R),10(S)-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene [(-)-anti-BPDE]. Following topical administration of (-)-BP-7,8-diol, a single adduct spot was detected that cochromatographed with a standard of the major deoxyguanosine adduct derived from 7(R),8(S)-dihydroxy-9-(S),10(R)-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene [(+)-anti-BPDE]. The stereochemistry of epoxidation of the enantiomers of BP-7,8-diol indicates that cytochrome P-450 catalyzes the terminal activation step of benzo[a]pyrene activation to an ultimate carcinogen in mouse skin, a target organ for its carcinogenic activity.

Purification and characterization of an acidic beta-1,3-glucanase from cucumber and its relationship to systemic disease resistance induced by Colletotrichum lagenarium and tobacco necrosis virus.

An acidic beta-1,3-glucanase was detected in cucumber leaves inoculated with either Colletotrichum lagenarium or tobacco necrosis virus (TNV) as well as in the leaves above those inoculated with the pathogens. The enzyme is extracellular and migrates in native polyacrylamide gel electrophoresis (PAGE) together with a Class III chitinase, a bifunctional chitinase/lysozyme. The beta-1,3-glucanase was separated by ultra-narrow pH range IEF-PAGE or by SDS_PAGE and was purified to apparent homogeneity. Only one isoform of the enzyme was detected. Its apparent molecular mass in 38 kDa as estimated by SDS-PAGE, its isoelectric point is 3.6 and the specific activity is approximately 26 micromol glucose equivalents liberated from laminarin min(-1)mg(-1) protein. Partial amino acid (five peptide fragments with a total of 65 amino acids) sequencing of the beta-1,3-glucanase revealed similarities of 49% to 72% to sequences of published beta-1,3-glucanases from tobacco, tomato, soybean, barley, and rice plants. A time course study indicated that the increase of the beta-1,3-glucanase activity was associated with induced resistance against C. lagenarium. The implications of these results to coordinate defense responses in plant-microbe interactions are discussed.

Immunohistochemical expression of p53 protein in invasive breast carcinoma: clinicopathologic correlations.

The results of immunohistochemical expression of p53 protein in 106 invasive breast cancers were correlated with conventional pathologic prognostic parameters. Archival formalin-fixed, paraffin-embedded tissue sections of these cases were stained with a monoclonal antibody (Ab-2), raised against p53 protein using a peroxidase-labelled streptavidin biotin kit. Fifty-six (53%) showed positive nuclear staining; 31 were considered weakly, 21 moderately and 4 strongly positive. Forty-three (77%) of these positive cases stained less than 50% of the tumor cells, with a significant association between intensity and proportion of nuclei stained (p<0. 05). p53 staining also correlated with histologic grade (p<0.005) but not with tumor size nor clinical stage (p>0.05). The follow-up data did not reveal any statistically significant survival advantage for patients with p53 negative tumors.

[Identification of a novel human MAST4 gene, a new member of the microtubule associated serine-threonine kinase family].

Human protein kinases make up a large superfamily of homologous proteins, which are related by virtue of their kinase domains (also known as catalytic domains). Here we report the cloning and characterization of a novel human MAST4 (microtubule associated serine/threonine kinase family member 4) gene, which locates on human chromosome 5q13. The MAST4 cDNA is 7587 base pairs in length and encodes a putative protein of 2435 amino acids which contains a serine/threonine kinase domain and a PDZ domain. MAST4 protein has 64%, 63%, 59% and 39% identical aminoacid residues with MAST1, MAST2, MAST3 and MASTL respectively. RT-PCR analysis revealed relatively high expression level of MAST4 in most normal human tissues, with an exception of in testis, small intestine, colon and peripheral blood leukocyte.

Endovascular treatment of fusiform aneurysms with stents and coils: technical feasibility in a swine model.

PURPOSE: To assess the biomechanical feasibility of treating experimental fusiform aneurysms endovascularly with a combination of stents and coils. METHODS: An experimental model was surgically constructed in the necks of nine swine to simulate intracranial fusiform aneurysms possessing important "perforators" or side branches. Balloon-expandable metal stents were positioned across the aneurysms in eight swine. In five of these, additional treatment was intraaneurysmal placement of detachable microcoils. Attempts were made to deposit these coils strategically away from the origin of the side branch. RESULTS: Stent placement was successful in seven swine but failed in one swine because of stent-aneurysm size mismatch. Two swine treated with only stents showed no significant alterations in blood filling of the aneurysm or side branch. Satisfactory coil placement (outside the stent, within the aneurysm sac, and away from the orifice of the side branch) was achieved in four of the five swine treated with stents and coils. Careful fluoroscopic monitoring and controlled coil delivery were necessary to avoid covering the sidebranch origin. These aneurysms could not be packed densely after detachment of the first coil because of the resultant radiographic overlap of multiple coil loops on the stent and its lumen in all projections. In one swine there was inadvertent untoward reentry of the coil tip into the expanded stent lumen during its delivery. CONCLUSION: Endovascular treatment of experimental fusiforms aneurysms using stents and coils is technically feasible. The stent maintains patency of the parent artery while allowing strategic coil placement in the aneurysm sac away from the origin of side branches. This technique may prove useful in the future treatment of intracranial fusiform aneurysms. However, potential sources of technical difficulties have been identified, and further longterm studies using an appropriate intracranial stent will be necessary before human application.
