Pharmacokinetics of cadmium in ewes: a preliminary study.

A pharmacokinetic analysis of cadmium was carried out by administering cadmium chloride i.v. to 3 ewes. Each received 0.033, 0.1 and 0.33 mg cadmium/kg body weight, with each administration separated by 21 d. Pharmacokinetic parameters were determined using a 3-compartment open model (multiple dosage analysis). The half life of elimination reached 40 to 50 d, the body clearance was 0.3 to 0.4 L/kg/d, and the steady-state volume of distribution was 6 to 28 L/kg. The invariability of the values of the elimination parameters and the good fitting of the mathematical model to the experimental values are in agreement with linear kinetics for cadmium in the ewe.

Effects of fluoride on secretory and postsecretory phases of enamel formation in sheep molars.

Effect of fluoride was assessed on molars during and after mineralization. Two groups of 7 sheep each were dosed orally with 3.5 mg of fluoride/kg of body weight daily for 4 months (from 5 to 9 months after birth). Sheep of the first group were slaughtered immediately after fluoride administration; those of the second group were slaughtered 4 months later at the age of 13 months. Three control groups of 7 sheep each were slaughtered at 5 months (to determine the state of the teeth at the beginning of fluoride administration), and at 9 and 13 months. During fluoride administration, plasma fluoride concentration rapidly increased to about 0.50 microgram/ml; after fluoride administration, it stabilized at 0.20 microgram/ml in treated sheep, whereas controls had concentration of 0.10 micrograms/ml (P less than 0.01). Parts of the molars that were in the process of mineralization during fluoride administration (mainly second molars) had thinning enamel, with pits, mainly close to the apex, marked decrease in hardness throughout the layer (less than 100 Vickers U, compared with 240 Vickers U), and fluoride accumulation twice as high as that in controls (1,000 to 2,500 mg/kg [dry weight]). Fluoride accumulation was higher in dentine (2,700 to 4,200 mg/kg), but hardness was less affected. On parts of the molars that were already mineralized (mostly, the first molar), changes in the appearance of enamel and cementum, decreased hardness (less important than in teeth during mineralization) affecting outer enamel more than inner enamel, high fluoride concentration (4,000 to 5,500 mg/kg [dry weight]) in outer enamel extending over 200 microns were observed.(ABSTRACT TRUNCATED AT 250 WORDS)

Proximal vergence and perceived distance.

Proximal vergence was measured haploscopically in response to a change in physical distance of a large screen of blurred horizontal lines. These responses were compared to proximal vergence measurements made using a clinical method. Perceived distance of the horizontal stripes was estimated according to the perceived size of a projected afterimage. For 18 normal observers the change in vergence measured haploscopically (in prism diopters per meter angle change in test distance) averaged 2.59 (SD = 1.44) binocularly and 2.11 (SD = 1.22) monocularly. Clinical measures averaged 2.88 (SD = 1.57) binocularly and 2.10 (SD = 1.27) monocularly. Haploscopic and clinical measures of proximal vergence correlated only when the clinical technique was modified by viewing through pinholes. Taking estimated perceived distance into consideration only partially reduced the differences between proximal vergence measured haploscopically and clinically as well as binocularly and monocularly. Both haploscopic and clinical measures of proximal vergence were sizeable, indicating that this vergence component can contribute significantly to the normal vergence response.

Lens-induced fixation disparity curves.

Change in fixation disparity was measured as lens sphere power was changed at distance and near on a young adult population. The results were graphed to yield a lens-induced fixation disparity curve. The lens curves found were grouped into four basic types (A through D). Two types (A and B) had increasing eso fixation disparity with increasing minus lens power. Type C had little change in fixation disparity with any change in lens power, whereas type D had an initial increase and then no further change with increasing minus power. In addition, approximately 74% of the 38-subject sample had different curve types at distance and near. Possible clinical uses of the lens-induced fixation disparity curve include the prescribing of near additions for pre-presbyopes based on measures of the binocular vergence response to various lens sphere combinations.

[Long-term (6 months) treatment of chronic bronchitis with respiratory insufficiency using almitrine dimesylate. Double-blind study versus placebo].

The purpose of this controlled, double-blind therapeutic trial in chronic bronchitis patients with hypoxia and normocapnia was to evaluate the beneficial effects of a 6-month treatment with almitrine bismesylate on dyspnoea and blood gases, as well as the clinical, spirometric and pulmonary haemodynamic acceptability of the drug. Patients were allotted at random to either almitrine bismesylate 100 mg/day in two divided doses (n = 10) or a placebo similarly administered (n = 9). On entering the trial, both groups had comparable clinical symptoms, lung function, blood gas values and pulmonary haemodynamic values. A significant (p = 0.014) reduction of dyspnoea was observed in the treatment group but not in the placebo group. A significant (p less than 0.05) improvement in PaO2 was noted after 3 and 6 months of treatment while PaCO2, initially normal, was not significantly modified. There were no significant changes in respiratory rate and external ventilation in either group, and the mean pulmonary arterial pressure remained unchanged in all patients. No significant variations were observed in the patients' weight, systemic arterial pressure, heart rate or signs of right ventricular failure. In conclusion, this trial in chronic bronchitis patients with hypoxia and normocapnia demonstrated sustained improvement in hypoxaemia and clinical symptoms during a 6-month treatment with almitrine bismesylate, without changes in pulmonary haemodynamic values.

Age-dependent effects of a chronic ultramild stress procedure on open-field behaviour in B6D2F1 female mice.

Few studies have been devoted to the interaction between age and stress. However, in view of the age-related changes in various components of the stress responses, the effects of stress may not be constant with age. In this study, we used a dimensional approach to compare open-field behaviour of B6D2F1 female mice, aged 5-6, 11-12, 17-18 and 23-24 months, exposed to a chronic ultramild stress (CUMS) procedure, solely based on nonnociceptive socioenvironmental stressors. Three behavioural dimensions emerged from the principal-component analysis; these were labelled as motor reactivity, exploratory activity, and emotional reactivity. Despite a major effect of age on the three dimensions, we could not conclude that CUMS had any influence as a function the age of the subjects. At all ages, CUMS increased motor activity and had no clear-cut effect on emotional reactivity and exploratory activity. The results are discussed in terms of the influence of the nature of the stressors on behavioural responses to novelty.
