Clinical significance of glucocorticoid receptors in acute leukaemia. Preliminary observations in Hungary and review of the literature.

Glucocorticoid receptor (GR) levels were quantitated in leukaemic blast cells separated from peripheral blood of 15 children with acute lymphocytic leukaemia (ALL) and of 6 children with acute non-lymphocytic leukaemia (ANLL). Using a whole-cell assay, it was found that specific (3H)-dexamethasone binding exhibited a wide range in both types of acute leukaemia: GR levels scattered between 0-22,346/cell and 0-8772/cell in ALL and ANLL patients, respectively. In this paper we discuss our observations together with current knowledge on GR levels in leukaemic blast cells and their relationship to glucocorticoid sensitivity and disease outcome.

Focal nodular hyperplasia of the liver after clomiphene treatment in a young boy.

The case of a 3.5-year-old boy treated unsuccessfully with clomiphene for cryptorchidism is reported. Nine months later focal nodular liver hyperplasia developed. Clomiphene promotes testicular descent by stimulating endogenous production of gonadotropins and their releasing factors. A causal relationship between the hepatic lesion and clomiphene treatment has been supposed. In our case subsequent fertility can only be expected from orchidopexy performed as soon as possible. Hepatic function must be checked during and after clomiphene treatment also in children.

Detection of main core proteins of simian C-type viruses and human retrovirus HTLV and antibodies to them in patients with lymphoid malignancies.

Peripheral leukocytes or lymph node cells and blood plasma samples from patients with lymphoid malignancies were investigated for immunological markers of BaEV, GaLV and HTLV. Antigens and antibodies were shown with radioimmunoassay. Antigen related to the p30 core protein of BaEV could be detected in each cell type of leukaemias and lymphomas. Antigen related to the GaLV p30 was found mainly in B- and O-cell forms, while that related to the p24 protein of HTLV could be detected only in two T-cell malignancies. Antibodies reactive with these antigens showed a similar distribution.

Differential translation of virogenic and oncogenic sequences in malignant lymphoproliferative diseases and transfection of coding DNAs into NIH 3T3 cells.

The expression of oncoviral p30 polypeptides and onc gene-specific proteins has been examined in different human lymphoid malignancies. The distribution of antigen(s) related to the p30 of BaEV lacked any specificity. Antigen(s) related to the main core polypeptide of GaLV could be detected mainly in B- and O-cell malignancies. The myc-encoded protein was translated at higher levels in malignant than in normal lymphoid cells. An active src gene was identified in three acute lymphoid leukaemias and in one non-Hodgkin lymphoma of T-cell origin. Human DNAs coding oncoviral antigens or onc gene-specific proteins could be transfected into NIH 3T3 cells. These data suggest that the synergistic effect of the myc and src genes would operate in malignant transformation of some progenitors of T-cell lineage.

Natural antibodies to simian type-C viruses and human retrovirus HTLV in patients with lymphoid malignancies.

Blood plasma samples from adult patients and children with malignant lymphoproliferative diseases were studied for presence of antibodies to primate type-C viruses by membrane immunofluorescence. Antibodies to baboon endogenous virus could be detected in various types of lymphoid leukaemias and lymphomas. Gibbon ape leukaemia virus-specific antibodies were mainly found in B- and O-cell leukaemias and lymphomas. Anti-HTLV antibodies could be detected only in a few cases of T-cell malignancy. There was no evidence of a horizontal transmission of such viruses.

New theory of the development of myocardial infarction.

Solutions of KCl (1%, 10% and 31.2%) administered into the pericardial fluid or applied onto the surface of the heart evoked a dome-like change of the ECG in dogs, rats and guinea-pigs and led to myocardial infarction in 3-5 days. Both the acute changes in ECG and the infarction itself could be prevented by the application of pericardial fluid samples and by administration of three synthetic compounds onto the heart surface. The same substances also inhibited the development of ECG changes elicited by general hypoxia due to stopping artificial respiration. The existence of a peculiar myocardial space beginning with outer pores and reaching the myocardial cells through connective tissue pathways is postulated. Earlier studies showed that 125I-labelled albumin applied to the epicardial surface through a filter paper reached the endomyocardium through some intramyocardial pathways beginning with epicardial pores. In the present experiments intrapericardial application of India ink led to obstruction of these pores and thus prevented the infarction elicited by intrapericardial administration of KCl solutions. This space, being distinct from that accessible from the coronary arteries, serves for transfer of various substances into the myocardium, while other compounds (e.g. noradrenaline) are not effective through this pathway. Oxygen reaching this space from the epicardial surface protects the myocardium from the damaging effects of hypoxia and KCl.

Compensatory hypertrophy in radiothyroidectomized dwarf rats.

The aim of the study was to clarify whether growth hormone and thyroid hormones play a role in compensatory organ hypertrophy. Radiothyroidectomized dwarf rats with serious disorder of growth hormone production due to the loss of thyroid function were used in the experiments. As regards growth hormone production these animals may be considered as having been "hypophysectomized". The loss of thyroid function and growth hormone deficiency did not affect the degree of compensatory hypertrophy of the kidney, the adrenal and the gonad. The observation indicates that the hormones under study are not of decisive importance in the mechanism of compensatory organ enlargement. Further investigations are needed to clarify whether organ enlargement in the hypometabolic rat occurring at the level observed in the controls is associated with enhanced function.

Gas chromatographic determination of unconjugated dehydroepiandrosterone, androstenedione, testosterone, pregnenolone and progesterone in human ovarian tissue.

A gas chromatographic method has been presented for the determination of unconjugated dehydroepiandrosterone, androstenedione, testosterone, pregnenolone and progesterone in human ovarian tissue. The procedure utilized radioactive tracers added to homogenate for correcting methodological loss, preliminary separation of steroids by thinlayer chromatography, acetylation, rechromatography on chromatoplate and gas chromatography on 3% SE-30 or 1% XE-60 columns with flame ionisation detection of steroids by using internal standards. Results of control experiments and representative clinical findings on normal and polycystic ovaries are reported.
