Chronobiology of Oestrus ovis (Diptera: Oestridae) in Sardinia, Italy: guidelines to chemoprophylaxis.

Oestrus ovis (Linné 1761) larvae are obligatory parasites of the nasal and sinus cavities of sheep and goats. Infestation is prevalent in hot and dry regions, such as Mediterranean countries. The current work was developed to establish the chronobiology of O. ovis in Sardinia, to determine the most suitable time for chemoprophylaxis. A survey was carried out during 1998, and sheep heads were collected monthly from local flocks. A total of 443 heads was examined, and the prevalence of oestrosis was 73.8%. We collected 2,691 larvae (mean = 6.07 +/- 9.52), and the intensity was greatest in November. The humoral immune response against the nasal bot fly was analyzed by means of an indirect-ELISA using second-instar O. ovis excretory and secretory antigens. A seasonal variation in the antibody levels was observed, increasing from April and peaked in June and in September. A significant correlation was observed between first instar intensity and the mean relative humidity (r2 = 0.120; P < 0.05), and between second-instar intensity and the mean temperature (r2 = 0.241; P < 0.05). Three periods in the chronobiology of O. ovis were defined: diapause (October-February), the active phase ofthe endogenous cycle (March-September) and the exit phase (May-September). Our results showed that treatment in October-November was suitable, because first instars were in diapause, preventing the development of first into second instars, and second into third instars.

Separation and characterization of mares' milk alpha(s1)-, beta-, kappa-caseins, gamma-casein-like, and proteose peptone component 5-like peptides.

The equine alpha(s1)- and beta-caseins (CN) were purified by chromatography on DEAE-cellulose and by reversed-phase HPLC. The alpha(s1)-, beta-, and kappa-CN were characterized either by monodimensional urea-PAGE or sodium dodecylsulfate (SDS)-PAGE or by bidimensional electrophoresis. Kappa-casein was characterized after electrophoresis by glycoprotein-specific staining. To identify alpha(s1)-CN without ambiguity, internal sequences were determined after trypsin or chymosin digestion of purified alpha(s1)-CN. These sequences, that could be estimated to correspond to 62% of the full protein, presented strong identities with regions of alpha(s1)-CN primary structures of other species. In particular, 51, 48, 43, and 40% identities were obtained with corresponding regions of sow, dromedary, cow, and human alpha(s1)-CN, respectively. On the other hand, trace amounts of equine gamma-CN-like and proteose peptone component 5-like peptides were found in the whole CN. They were identified by microsequencing and corresponded to beta-CN peptides generated by plasmin action on the whole CN. The equine alpha(s1), beta-, and kappa-CN were separated by bidimensional electrophoresis in numerous isoelectric variants with apparent isoelectric points distributed between pH 4.4 to 6.3, 4.4 to 5.9, and 3.5 to 5.5, respectively. The beta- and kappa-CN displayed a more acidic character in the mare than in the cow.

Refractive acoustic devices for airborne sound.

We show that a sonic crystal made of periodic distributions of rigid cylinders in air acts as a new material which allows the construction of refractive acoustic devices for airborne sound. It is demonstrated that, in the long-wave regime, the crystal has low impedance and the sound is transmitted at subsonic velocities. Here, the fabrication and characterization of a convergent lens are presented. Also, an example of a Fabry-Perot interferometer based on this crystal is analyzed. It is concluded that refractive devices based on sonic crystals behave in a manner similar to that of optical systems.

Purification and characterization of Lip2 and Lip3 isoenzymes from a Candida rugosa pilot-plant scale fed-batch fermentation.

Previous purification of a crude extracellular enzyme preparation from Candida rugosa ATCC 14830 pilot-plant fed-batch fermentations showed the presence of two lipase isoenzymes, Lip2 and Lip3, differing in their molecular masses (58 and 62 kDa, respectively). These enzymes were purified but the lipases were forming active aggregates with a molecular mass higher than 200 kDa. In this work we developed a purification method following three steps: ammonium sulfate precipitation, sodium cholate treatment and ethanol/ether precipitation, and anion exchange chromatography which allowed the sequential disaggregation of the isoenzymes. Pure and monomeric Lip2 and Lip3 were characterized according to pI, glycosylation and activity for p-nitrophenol esters and triacylglycerols of varying acyl chain. Lip3 was the best catalyst for the hydrolysis of the simple esters and triacylglycerols with short and medium acyl chains.

Disseminated neoplasia and large foci indicating heavy haemocytic infiltration in cockles Cerastoderma edule from Galicia (NW Spain).

A histopathological survey was performed to search for the cause of high mortality in the cockle Cerastoderma edule in northern rias of Galicia (northwestern Spain). From limited sampling, 2 severe pathological conditions with high prevalence were found: disseminated neoplasia and the occurrence of numerous, large foci of heavy haemocytic infiltration. Both conditions were detected in areas affected by high mortality and were absent (foci of haemocytic infiltration) or with lower prevalence (disseminated neoplasia) in an area with unnoticeable mortality that was chosen as a reference. Advanced cases of both pathological conditions were associated with extensive tissue destruction. Because of the severity of the lesions and their high prevalence, both pathological conditions could have contributed to the high mortality.

Critical state theory for nonparallel flux line lattices in type-II superconductors.

Field penetration profiles in type-II superconductors with nonparallel vortex configurations are obtained by a proposed least action principle. We introduce a functional C[H-->] which is minimized for the current density constrained by J-->inDelta(H-->,x-->), where Delta is a bounded set. Within the isotropic case /J-->/< or =J(c)(H), the profiles H-->(x-->,t) are derived for an applied changing excitation. The model can reproduce the physical phenomena of flux transport and consumption, and the magnetization collapse in crossed field measurements.

Bronchopulmonary helminths of roe deer (Capreolus capreolus) in the northwest of Spain.

From 1993 to 1995, 148 hunter killed roe deer (Capreolus capreolus L.) from game reserves in Lugo (northwestern Spain) were examined for lungworm infection. The overall prevalence of infection was high (66.2%). Infections were comprised to two species, Varestrongylus capreoli and Dictyocaulus noerneri with a prevalence of 62.0 and 18.2%, respectively. Significant differences in the prevalence and larval burdens in lungs and faecal samples were observed between different locations. The highest overall prevalence of lungworm infection was observed in the large reserve at Ancares (78%) in comparison to that in smaller reserves (61.7%). Considering that the species recorded in this study are highly host-specific, it is unlikely that roe deer represent a significant reservoir of lungworm transmissible to domestic ruminants in northwestern Spain.

Influence of the conformational flexibility on the kinetics and dimerisation process of two Candida rugosa lipase isoenzymes.

We have investigated the interfacial activation process of two isoenzymes from Candida rugosa (Lip1 and Lip3) using triacetin as substrate. Kinetics were coupled to inhibition experiments in order to analyse the transition between the open and closed conformers. This process was slow, particularly for Lip1, in the absence of an interface provided by the substrate or a detergent. Dimers of Lip3 were also purified and their catalytic action was closer to that of a typical esterase. In spite of the high sequence homology between Lip1 and Lip3, small changes enhance hydrophobicity in the binding pocket of Lip3 and increase the flexibility of its flap. We postulated that these factors account for the higher tendency of Lip3 to dimerise fixing its open conformation.

Distribution of aflatoxin M1 in cheese obtained from milk artificially contaminated.

Small-scale manufacture of cheese using artificially AFM1 contaminated milk as raw material to study the distribution of such toxin both in whey and in cheese, was carried out. Whole milk with undetectable levels of AFM1 was used. The toxin was added in concentration that varied from 1.7 to 2.0 microg/l of milk. After the home-made production of cheese, the concentration of AFM1 was determined both in whey and in cheese, using the enzymatic immunoassay technique. The greatest proportion, 60%, was detected in whey while 40% AFM1 remained in cheese.

Relationship between atmospheric pressure and mortality in the Madrid Autonomous Region: a time-series study.

This study set out to determine the possible influence of variations in atmospheric pressure on mortality in the Madrid Autonomous Region (MAR), taking into account the possible confounding effect of other atmospheric variables. The study was based on daily mortality data from the MAR Revenue and Excise Authority, meteorological data from Getafe Observatory and air pollution data from the Madrid Municipal Automatic Air Pollution Monitoring Grid. A time-series analysis was performed, using Box-Jenkins modelling and controlling for the respective confounding variables. Furthermore, the different variables studied (pressure, temperature, pollutants, etc.) were used to produce a multivariate model of the different causes of mortality. A significant association was found between anticyclonic trend and mortality with circulatory causes in the medium-long term and anticyclonic trend and mortality with respiratory causes in the long term.

Polymorphism of the 5' terminal region of Citrus tristeza virus (CTV) RNA: incidence of three sequence types in isolates of different origin and pathogenicity.

Sequences of the 5' terminal region of the genomic RNA from eight isolates of Citrus tristeza virus (CTV) were previously classified into three types (I, II and III), with intragroup sequence identity higher than 88% and intergroup sequence identity as low as 44%. Sequencing of an additional 58 cDNA clones from 15 virus isolates showed that all sequences could be unequivocally assigned to one of the three types previously established. The relative frequency of each sequence type was assessed in 57 CTV isolates of different geographic origin and pathogenic characteristics by RT-PCR with sets of type-specific primers using CTV dsRNA as template. None of the isolates yielded amplification of the type I or II sequences alone, but in 19 of them type III sequences were the only amplification product detected. Within isolates containing more than one sequence type, eight had type II and III sequences, 11 had type I and III sequences, and 19 had sequences of the three types. Isolates containing only type III sequences caused only mild to moderate symptoms in Mexican lime, an indicator species for most CTV isolates, whereas isolates causing stem pitting in sweet orange an/or grapefruit, generally contained sequences type II. None of the sequence types could be traced to a precise geographic area, as all types were detected in isolates from at least nine of the 12 countries from which samples were taken.

Role of intrapartum hypoxia in carnitine nutritional status during the early neonatal period.

We analyze markers of carnitine insufficiency and deficiency, lysine (LYS) and methionine (MET), in 39 neonates with intrapartum hypoxia (selection criteria: umbilical artery pH <7.20, lactate >1.8 mmol/l and PaO2 <25 mm Hg), and in 35 healthy newborn infants (control group) in the early neonatal period (1-7 days of life). Free (FC), total (TC) carnitine and acylcarnitines (AC=short-chain+long-chain acylcarnitines) were measured using a radioisotopic micromethod; LYS and MET were determined by high-pressure liquid chromatography. AC and TC plasma concentrations and AC/FC ratio were higher while FC/TC ratio was lower in the hypoxic neonates than in the control group. Hypoxic newborn infants (59%) presented "carnitine deficiency" (FC/TC <0.7) and 60% of them "carnitine insufficiency" (AC/FC ratio >0.4) vs. 31% and 28%, respectively, for the neonates of the control group (p<0.05). In the healthy neonates group, MET correlated with FC/TC and the AC/FC ratio. FC, TC, AC, AC/FC and umbilical artery pH (pHua) were inversely correlated. FC/TC and MET correlated with pHua. We conclude that: (1) an important percentage of newborn infants with intrapartum hypoxia suffer carnitine deficiency and carnitine insufficiency in the early neonatal period, related to MET plasma levels; (2) the carnitine deficiency or insufficiency in the neonate is determined by the degree of intrapartum acidosis.

Cidofovir treatment of human polyomavirus-associated acute haemorrhagic cystitis.

We report the case of an 18-year-old patient who received an allogeneic bone marrow transplant from an HLA-identical unrelated donor for a Ph+ acute lymphoblastic leukemia, in his third complete remission. Cyclophosphamide and busulfan were used as conditioning treatment. Acute graft-versus-host disease developed on day +9, and the response to adequate treatment (steroids) was favourable. On day +45 the patient developed an acute severe haemorhragic cystitis, and BK polyomavirus was demonstrated in urine samples using electron microscopy and polymerase chain reaction. Urinary symptoms did not improve in spite of palliative treatment, but a response was evident after 2 weeks of cidofovir treatment.

Case report. Cutaneous cryptococcosis in a patient with systemic erythematous lupus.

The first case of a cutaneous cryptococcosis associated with systemic erythematous lupus (SLE) diagnosed in our Mycology Reference Centre is presented: a 24-year-old female patient diagnosed with SLE, nephrotic syndrome, arterial hypertension, renal insufficiency due to glomerulonephritis type IV and cellulitis in the right thigh and gluteus. Cryptococcus neoformans was isolated by cutaneous biopsy and haemoculture. Cryptococcal antigen was detected in serum by the latex agglutination test. As the patient did not respond to fluconazol intravenous treatment, amphotericin B administration was performed. She died of acute renal insufficiency.

Novel carbocyclic nucleosides containing a cyclobutyl ring: adenosine analogues.

(1S,1'R)-cis-1-(3'-aminomethyl-2',2'-dimethylcyclobutyl)ethanol (1) and (1S,1'R)-cis-1-[3'-(2-aminoethyl)-2',2'-dimethylcyclobutyl]ethanol (2) were used as precursors in the synthesis of cyclobutyl nucleoside analogues containing adenine and 8-azaadenine moieties, which were tested as antiviral and antitumoral agents in a variety of assay systems. Compounds 8 and 9 displayed significant activity against respiratory syncytial virus, and compounds 14 and 15 were moderately active against vaccinia virus.

Dynamics of the antibody response in cattle infested by Hypoderma (Diptera: Oestridae) after a treatment with injectable and pour-on ivermectin.

The dynamics of the anti-Hypoderma antibody response after a treatment with injectable and pour-on ivermectin against first instars of Hypoderma sp. were determined in naturally infested cows. Cattle were randomly assigned to three groups: group 1, which served as an untreated control; group 2, treated with pour-on ivermectin at a dose of 500 microg/kg of body weight, and group 3, which received 200 microg/kg of body weight of ivermectin by subcutaneous injection. Both ivermectins showed total efficacy against L-1 of Hypoderma sp. In untreated control animals, grubs became detectable in the 2 mo after treatment. Indirect enzyme-linked immunosorbent assay (ELISA) showed a significant increase in group 3 antibody levels 1 mo after treatment, whereas in group 2 the increase was not significant. Following the peak of anitibody activity in group 3, antibody levels declined at a faster rate than in those treated topically. In both treated groups, cattle remained serologically positive for 4 mo after ivermectin treatment. Thus, serological surveillance programs for cattle grubs conducted during this posttreatment period will not be influenced by treatment with ivermectin. The persistence of antibody levels after larval destruction makes the indirect ELISA unsuitable for the prediction of the efficacy of treatment.
