[A study on IL-5 and IL-10 in the modulation of asthmatic airway inflammation in murine models].

OBJECTIVE: To observe the time-course profile of airway inflammation in murine asthma model, and to analyze the dynamic profile of pro-inflammatory factor IL-5, and inflammtion-suppressing factor IL-10 in the airway. METHODS: An animal model of asthma was established by OVA sensitizing-challenging BALB/C mice. At seven points (0 h, 8 h, 24 h, 48 h, 96 h, days 7 and days 14) in the time course after challenge, bronchoalveolar lavage (BAL) was performed. Total cell counts, cell differential, eosinophil (EOS) counts and percentages were examined and levels of two Th2 cytokines, IL-5 and IL-10, were detected by ELISA. RESULTS: After OVA challenge, total cell counts, EOS counts and EOS percentages in BALF were increased significantly. EOS count increased at 8 h, formed a plateau from 24 h to 48 h, declined at 96 h, reduced approximately to control from days 7 to days 14. IL-5 in BALF elevated at 8 h, maitained at high level from 28 h to 96 h, decreased at days 7. The dynamic changes of IL-5 was in synchronism with that of EOS counts, a positive correlation being identified. No significant difference was found between IL-10 in BALF before and after challenge. A close correlation between EOS counts and IL-5/IL-10 ratio was demonstrated (r = 0.9, P < 0.001). CONCLUSION: IL-5 and IL-10 up- and down-regulate asthmatic airway inflammation respectively, and their interaction may play an important role in inflammatory progress and resolution.

[Preliminary investigation of effect of serum leptin on nutritional state of COPD patients].

OBJECTIVE: To explore the significance of serum leptin and TNF-alpha in malnutrition of COPD. METHODS: Serum leptin and TNF-alpha concentrations were measured by radioimmunoassay. Body mass index (BMI), percent normal body weight (NW%), triceps skin-fold thickness (TSF), subscapular skin-fold thickness (SSF), mid-upper arm circumference (MAC), serum albumin (ALB) and total lymphocytes count (LYM) were determined in 31 patients with COPD and in 11 healthy controls. The correlation between leptin and nutritional parameters was analysed. RESULTS: (1) Serum leptin concentrations (3.0 +/- 2.5) micrograms/L in 42 subjects were significantly correlated with BMI, NW%, TSF, SSF and MAC (P < 0.001). (2) Serum leptin concentrations were significantly correlated with BMI in both malnourished patients with COPD and non-malnourished patients (P < 0.05-0.001). The leptin level was significantly lower in malnourished group (1.3 +/- 1.0) micrograms/L than in non-malnourished group (4.5 +/- 2.6) micrograms/L, (P < 0.05). The differences in serum leptin level and BMI between non-malnourished group and healthy group were not significant. (3) Serum TNF-alpha level was significantly higher in COPD group (1.8 +/- 0.3) micrograms/L than in healthy group (1.0 +/- 0.5) microgram/L (P < 0.05). Serum leptin levels didn't significantly correlate with TNF-alpha levels in COPD patients. CONCLUSION: Serum leptin levels correlate with nutritional parameters in COPD patients and leptin may contribute to the malnutrition in COPD patients.

[Clinical evaluation of glutaldehyde resinifying therapy on pulp diseases].

OBJECTIVE: To evaluate clinically the glutadehyde resinifying therapy(GR) on pulp diseases. METHODS: A total of 270 teeth with pulp diseases were divided into GR and formaldehyde resinifying therapy(FR) groups. The latter served as a control. Immediate post-operation reactions, short-term (0.5-1 year) and long-term (2-4 years) results were compared between the two groups. RESULTS: Ratio of post-operation pain for GR group was 10.2%, while 19.8% for FR group. The difference was statistically significant (P < 0.05, by chi 2 test). The short-term successful rates were 85.4% and 84.9% for GR and FR groups, respectively, whilst the long-term rates were 92.0% and 90.8%, respectively, there were no significant differences between the two groups statistically. CONCLUSIONS: GR could obtain similar clinical results as FR, and has an advantage of lower rate of post-operation pain compared with FR.

[Treatment of 121 heroin addicts with Han's acupoint nerve stimulator].

OBJECTIVE: To observe the Han's Acupoint Nerve Stimulator (HANS) for the treatment of heroin addicts. METHODS: One hundred and eighty-one cases of heroin addiction with obvious withdrawal syndrome were selected. Among them, 121 cases were randomly assigned to be treated with HANS, and the other 60 cases was taken as the control group. In the HANS group, patients were treated with HANS from the second day of their admission in the treatment center (when the withdrawal symptoms were obvious), they were given 4 treatments (30 min each) a day at the first 3 days, twice a day for 3 more days, and once a day 7 days later. The total treatment lasted 15 days. The acupoints selected were Hegu (LI4) and Laogong (PC 8) on one hand, Neiguan (PC 6) and Waiguan (SJ 5) on the other hand, and Zusanli (ST 36), Sanyinjiao (SP 6) on both legs. The frequency was 2/100 Hz, the intensities of the stimulations were 12-16 mA on arms and 16-26 mA on legs. All the manipulations in the control group were the same as in the HANS group, except that electrodes were placed at the acupoints without any electrical stimulation. RESULTS: There was no significant difference (P < 0.05) on the 8 indices (heart rate, body weight, sleeping time, chilling, pain, anxiety, catarrh and craving) observed between the 2 groups before treatment. After the HANS treatment, all the 8 indices improved significantly as compared with those observed before treatment (P < 0.01), while in the control group only a moderate improvement was observed. CONCLUSION: HANS is obviously effective in relieving the withdrawal syndrome in heroin addicts.

[Effect of ligustrazine injection on vascular endothelial cell of the patients with arteriosclerosis obliterans].

OBJECTIVE: To study the effect of ligustrazine injection (LI) on vascular endothelial cell of the patients with arteriosclerosis obliterans (ASO) and explore the new pathway of investigating effective vascular protective agents in Chinese medicinal herbs. METHODS: Forty-six patients with ASO in the LI group treated by LI were observed, their circulating endothelial cells (CEC) were detected quantitatively before and after treatment. The results were compared with the CEC of 53 cases of healthy persons (control group) in the same period. RESULTS: In the LI group, the immediate cure rate was 45.7% (21 cases), markedly effective rate 36.9% (17 cases) and the effective rate 17.4% (8 cases). The CEC of patients before treatment was 4.39 +/- 1.76/0.9 microliter, which was significantly higher than that of the healthy persons (1.53 +/- 0.42/0.9 microliter). It could be reduced after treatment, along with the improvement of symptoms and signs, to 2.43 +/- 0.87/0.9 microliter, P < 0.01. CONCLUSION: LI in treating ASO not only displays extraordinary effect, but also has good effect in curing the damage of endothelial cells.

[Changes of leukocyte rheologic characteristics and cell adhesion molecules in patients with multiple organ failure after severe trauma].

OBJECTIVE: To explore the changes of leukocyte rheologic characteristics and of cell adhesion molecule in patients with multiple organ failure (MOF) after severe trauma. METHODS: By using the erythrocyte deformability apparatus, platelet and thrombus adhesion dual-purpose apparatus and enzyme-linked immunosorbent assay (ELISA), We measured the leukocyte deformability (LD), leukocyte adhesion function (LAF), leukocyte CD18 expression, soluble intercellular adhesion molecule-1 (sICAM-1) and soluble vascular cell adhesion molecule (sVCAM-1) concentration in 36 MOF patients, 31 trauma patients, and 35 to be controls. RESULTS: The leukocyte filtration index (LFI), leukocyte adhesion rate (LAR), leukocyte CD18 expression, and sICAM-1 and sVCAM-1 concentration were significantly higher in MOF patients than in controls and trauma patients (F = 68.45 - 116.20, q = 12.161 - 21.374, P < 0.00), and the changes of these indicators in MOF deaths were more obvious than those in MOF survivors (t = 6.920 - 11. 665, P < 0.00). The LFI and LAR in MOF patients were positively related to leukocyte CD18 expression, sICAM-1, and sVCAM-1 concentration (r = 0.691 - 0.844, P < 0.001); LFI was positively related to LAR (r = 0.711, P < 0.001). CONCLUSION: The abnormalities of leukocyte rheologic characteristics and CAMs might be closely related to the occurrence of MOF and the severity of pathologic changes.

[Diagnosis and treatment of thoracic myelopathy due to degenerative thoracic spine disease].

OBJECTIVE: To summarize the experience in the early diagnosis and treatment of chronic thoracic myelopathy due to the compression of degenerative thoracic spine disease. METHODS: 21 patients with thoracic spinal stenosis were operated on from January 1995 to April 2000. Etiologically 16 patients were found to have ossification of the ligamentum flovum, 3 Scheuermann's disease, and 2 DISH. They were confirmed by CT, CTM, MRI and Magnetic MEP. Decompression was performed by en block unroof laminectomy for the posterolateral wall of the thoracic canal. The epstein criteria were used to judge the operative results. RESULTS: In the 21 patients 5 belonged to localized type, 10 continuous type, and 3 skip type in 21 cases. Three patients showed a compression from the anterior side, and 3 were complicated by OPLL of the cervical spine. Results were excellent in 4 patients, good in 8, improved in 2, and poor in 1. One patient was operated on two times because the extent of laminectomy was not enough for the first operation. CONCLUSIONS: Early diagnosis can be made if orthopedists are familiar with the clinical characteristics of thoracic myelopathy, and are alert to thoracic spine stenosis. En block unroof laminectomy is effective and safe to treat thoracic spine stenosis.

[Study on the epidemiological features of Kawasaki disease in Jiangsu].

OBJECTIVE: To investigate the incidence of Kawasaki disease and epidemiological characteristics in Jiangsu province. METHODS: Questionnaire and diagnostic guidelines for Kawasaki disease with color - printed photographs of typical clinical symptoms were sent to all county hospitals with pediatric beds. All the in - patients with Kawasaki disease hospitalized from January, 1993 to December, 1997 were studied regarding age, sex, season of illness, cardiac sequelae, outcome etc. RESULTS: There were 509 cases (329 males and 180 females) diagnosed with 489 cases (96%) clinically typical. The incidence of the 5 - year period was averaged 2.53 per 100,000 children under 5. The number of male patients with Kawasaki disease was higher than that in female. The age peak of incidence was between 6 month and 2 - year olds. 13.6% of all cases developed cardiac sequelae which mostly occurred in males and in those younger than 2 years of age. There was one patient recurred among total patients reported (recurrence rate 0.2%) and 2 patients died (fatality rate 0.4%). CONCLUSION: The epidemiological characteristics of Kawasaki disease in Jiangsu province were similar to those in Japan in sex, season distribution and cardiac sequelae. The number of patients in male was higher than in female. Patients were more commonly seen in spring than in winter. However, the incidence rate in Jiangsu was significantly lower than that in Japan and the age peak was between 6 month and 2 years old.

[Epidemiological and clinical characteristics of Kawasaki disease in Shanxi province].

OBJECTIVE: The objective of this paper is to describe the epidemiological and clinical characteristics of Kawasaki disease (KD) in Shanxi province, China during a 5 year - period from January 1993 to December 1997. METHODS: A province - wide epidemiological survey on Kawasaki disease (KD) was carried ont by the China - Japan Kawasaki disease study group. The questionnaire form and the diagnostic criteria of KD which was prepared by the Japan Kawasaki disease research committee and translated into Chinese were sent of the departments of pediatrics of all the hospitals with 100 or more beds in Shanxi province. All the KD patients who were diagnosed during the observation period from 1993 to 1997 were asked to report to this survey. The database of reported KD in this survey was analyzed at the Department of pediatrics of the Shanxi provincial people's hospital, Xian, China. All the patients that satisfied the diagnostic criteria were included. RESULTS: A total of 105 (70%) hospitals responded and 376 cases of KD were confirmed. More cases were reported in 1993 and 1994. Of the total patients reported, 69% were children under 3 years old with male to female ratio of 1.6:1. The proportion of patients with cardiac sequelae was 19% with male to female ratio: 3:1. There were 4 fatal cases with a fatality rate of 1%. CONCLUSIONS: Since KD is common in China continuous surveillance is necessary to maintain high awareness of KD so as to identify possible risk factors and its association with other disease.

[Analysis on the characteristic and trend of AIDS epidemics in Shandong].

OBJECTIVE: To analyse the data thru labs and field epidemiological investigation from 1987 to 1999, in order to find out the characteristics, sources of infections and its trend which would lead to the development of strategy, prevention and control of AIDS in Shandong. METHODS: Anti-HIV antibody was detected by ELISA and WB in the serum of high-risk groups. When Anti-HIV antibody was positive, personal and epidemiological history was investigated. The anticoagulated blood was repeatedly collected to verify the infected people. PBMCs were separated from the anticoagulated blood to collect previrus DNA. Gene sequence was then detected and subtype analysed by PCR using ABI company kit. RESULTS: Eight hundred five thousand eight hundred and one specimens were detected in the whole province from 1987 to 1999. 52 cases showed Anti-HIV antibody positive, with a rate of 0.06 per thousand. Of the 52 cases, 2 died. Sequence detection and subtype analysis of PBMC in 24 infected people showed that there were five HIV-1 subtypes M: A, B, B', C, E which were from 7 cities of the province. Blood and sexual transmission took the majority with proportions, 59.62% and 30.76% respectively. The subtypes from 13 blood donors were all B'. Among 10 labourers refurned from abroad and 1 spouse six cases were C, two were B, and another two were A and E subtypes respectively. Epidemiologic data showed that most of the infected people were mobile, making sexual transmission easy to occur. CONCLUSION: The moods of HIV/AIDS transmission in Shandong were mainly through blood and sexual contacts.

[Experimental study on suppressive effect of ginkgo extract on adhesion of vascular endothelial cell to monocyte induced by minimally modified low density lipoprotein].

OBJECTIVE: To observe the effect of Ginkgo extract on the interaction between minimally modified low density lipoprotein (MM-LDL) induced human umbilical vein endothelial cell (HUVEC) and U937 monocyte-like cell line. METHODS: The adhesive percentage between HUVEC and U937 by counting and the expression of VCAM-1, ICAM-1, P-selectin by ELISA. RESULTS: Treatment of HUVEC with MM-LDL (75 micrograms/ml) for 4 hrs could significantly increase the adhesion of U937 to HUVEC (P < 0.01) and Ginkgo extract could suppress the adhesion in dose-dependent manner. The surface expression of VCAM-1, ICAM-1, P-selectin was not induced by MM-LDL, while recombination tumor necrosis factor-alpha 5.0 ng/ml, as a positive control, could do. CONCLUSION: Ginkgo extract afforded protection against HUVEC damage induced by MM-LDL and the adhesion of monocytes to endothelial cells induced by MM-LDL is not mediated by VCAM-1, ICAM-1 and P-selectin.

[Repair of collateral ligament rupture in the interphalangeal joint using superficial flexor tendon of finger].

OBJECTIVE: To introduce the surgical procedure and clinical result of microsurgical repair on collateral ligament rupture in the interphalangeal joint using superficial flexor tendon of finger. METHODS: From 1987, 23 cases with collateral ligament rupture in the interphalangeal joint were repaired by part of superficial flexor tendon of finger. The initial point of collateral ligament was drilled two holes, 2 to 3 mm in distance, and crossed by steel wire. Then the superficial flexor tendon of finger was crossed under the steel wire and sutured to the terminal point of superficial flexor tendon of finger by 5/0 to 7/0 nontraumatic suture thread to maintain suitable tension, and sutured to collateral ligament by 8/0 nontraumatic suture thread. Extension splint fixation was performed 4 to 6 weeks postoperatively. RESULTS: Followed up 3 to 6 months, 15 cases were excellent, 6 cases were better, 2 cases were moderate, and the excellent rate was 91.34% according to Saetta standard. CONCLUSION: Microsurgical repair on collateral ligament rupture in the interphalangeal joint using superficial flexor tendon of finger is a convenient and effective surgical method.

[Studies on antitussive, antiasthmatic, expectorant and anti-inflammatory effects of Jin'an kechuan granules].

OBJECTIVE: To study the pharmacologic effect of Jin'an Kechuan granules. METHOD: Routine antitussive, antiasthmatic and expectorant methods were used. RESULT: Jin'an Kechuan granules in the doses of 16, 8, 4 g.kg-1 ig could significantly decrease the frequency of cough caused by ammonia water in mice, and increase the secretion of phenol red from mouse tracheas in a dose-dependent manner. The granules also help increase the threshold voltage to induce cats to cough, prolong the latent period of asthma induced by histamine and acetylcholine mixture, and antagonize the contraction of isolated trachea strips induced by histamine and acetylcholine in guinea pigs. Jin'an Kechuan granules had inhibitory effects on common bacteria in the respiratory tract as well as on the swelling of acute inflammation in mice and rats. CONCLUSION: Jin'an Kechuan granules have antitussive, antiasthmatic, expectorant and anti-inflammatory, antibacterial effects.

[Study of the distribution feature of the endocrine cells of human islet and its relation to microvasculature].

Double immunohistochemical stain was used for the study of distribution feature of the endocrine cells of human islets and its relation to the microvasculature. The results were as follows (1) Sheath islet was found in human adult islets and the B cells situated the central part of the islets and arrounding it were the A and D cells which formed the sheath of the islets. (2) The identical distribution of the A and D cells was found. (3) Some islets were divided into several "subunite" by connective tissue in which the B cells and A(or D) cells situated the central and peripheral part respectively. (4) The A and D cells were near the microvasculature. These results will provide morphologic basis for the research of the functional relation among the endocrine cells.

[Effects of sulpiride on morphine-induced reward and its relation to cyclic AMP levels in mice].

This study examined the effects of sulpiride, a dopamine D2 receptor antagonist, on morphine rewarding properties and its relation to cyclic AMP levels in brain. The morphine-induced reward in mice was observed in a conditioned place preference test. Cyclic AMP was estimated by radioimmunoassay. The time of staying in morphine pairing compartment and the cAMP levels in brain of mice in the morphine group increased as compared with those in other groups (P < 0.05), but there was no significant difference in the time of staying in morphine pairing compartment and the cAMP levels in brain between the saline group and the sulpiride plus morphine groups (P > 0.05). These suggest that the inhibition of morphine-induced reward by sulpiride be due to the prevention of an increase in cyclic AMP levels in brain.

[The descriptions identification of "yimu cao"].

Remained but entire bracts beneath the inflorescence and the persistent calyxes were selected as primary characteristics to distinguish "Yimu Cao", which derived from 8 species and 1 variety in the same genus Leonurus. The results were reliable.

[Influences of counterfeits on RAPD results of Semen Cuscutae].

Compared the differences of the RAPD results between pure Semen Cuscutae and 12 commercial samples, it comes into the conclusion that the exiting other sample influences the RAPD results when its content is over 60% and the results is same as its content being under 60%.
