[Clinical study on effect of Chinese herbal medicine on liver damage caused by hepatic artery chemoembolization].

OBJECTIVE: To observe the effect of Chinese herbal medicine on hepatic artery chemoembolization caused liver damage. METHODS: One hundred and ten patients, who received hepatic artery chemoembolization, were divided into two groups, 60 patients in the treated group were treated with Qinggan Jiedu Sanjie Decoction and the other 50 Patients in the control group treated with routine western medicine. The changes of liver function, cirrhosis incidence and exacerbation rate of both groups were observed. RESULTS: The total liver function deterioration rate of the treated group, according to Child's grading standard, was 46.67%, while that of the control group was 68.00%, the former was much lower than the latter (P < 0.05). The cirrhosis incidence and exacerbation rate in the treated group were 35.00% (7/20) and 50.00% (20/40) respectively, while in the control group, they were 65.00% (13/20) and 76.67% (23/30) respectively. The difference between the two groups was also significant (P < 0.05). CONCLUSION: Qinggan Jiedu Sanjie Decoction has some effect on alleviating and preventing hepatic damage caused by hepatic artery chemoembolization.

[The significance of detection of hepatitis C virus RNA in peripheral blood mononuclear cells of patients with chronic hepatitis C].

OBJECTIVE: To determine the role of peripheral blood mononuclear cells (PBMCs) in hepatitis C virus (HCV) infection. METHODS: HCV RNA, HCV antigen and Fas antigen were detected in PBMCs of 22 patients with chronic hepatitis C, 21 renal dialysis patients with anti-HCV positive and 12 health blood donors as normal controls. All specimens were studied by electronic microscope (EM). RESULTS: (1) The positive rate of HCV RNA in PBMCs of 22 patients with hepatitis C was 77.3% (17/22). (2) EM showed that HCV particles replicated in PBMCs of the patients with HCV infection, but not in the serum of patients with negative HCV RNA and healthy controls. (3) HCV RNA were positive both in the serum and PBMCs of 8 patients with HCV particles positive and had a close relationship with HCV antigen and Fas antigen. The expression of HCV and apoptosis were found in the cytoplasm of PBMCs in these 8 patients. CONCLUSION: (1) Hepatitis C virus could replicate in PBMCs and produce infectious HCV particles. (2) HCV in PBMCs may act as the source of reinfection for HCV and lead to persistent infection and recurrence.

[The treatment of 432 patients with pituitary adenomas via transsphenoidal approach].

OBJECTIVE: To study some clinical features of 432 cases of pituitary adenomas treated by transsphenoidal microsurgery between July 1982 and October 1996. METHODS: Pathological examination, immunohistochemical examination and electronmicroscopic examination were done in 424, 29 and 103 cases respectively. RESULTS: Hyperprolactinemia was found in many so-called non-functioning adenomas besides in prolactinomas. The dura maters were invaded by tumors in 78 cases (the incidence of dural invasion was 65.5%). CONCLUSIONS: Hyperprolactinemia is not peculiar to prolactinomas, and differential diagnosis is necessary when hyperprolactinemia exists. The involvement of the dura mater of sellar floor is an important mark that the adjacent tissues were invaded by pituitary adenomas. The outcome of surgical treatment is significantly different between the patients with dural invasion and without invasion.

[Gene expression of TGF-beta1 and its type II receptor in hepatocellular carcinoma].

OBJECTIVE: To investigate the gene expression of transforming growth factor-beta1 (TGF-beta(1)) and its type II receptor (TGF-beta R II) in hepatocellular carcinoma. METHODS: The expression of TGF-beta(1) and TGF-beta R II at mRNA level and protein level in 30 cases of HCC was separately detected using reverse transcription-PCR and immunohistochemistry technique. RESULTS: There was no difference in the expression of TGF-beta(1) at mRNA level between HCC tissue (24/30) and surrounding liver tissue (26/30). At the protein level, however, the expression of TGF-beta(1) decreased in HCC tissue compared with that in the surrounding liver tissue (P < 0.01). The expression of TGF-beta R II mRNA decreased significantly in HCC tissue (11/30) compared with that in the surrounding liver tissue (23/30) (P < 0.01). The less HCC expressed TGF-beta R II mRNA, the poorer the tumoral hepatocytes differentiated (P < 0.01) and the more likely the portal vein metastasis and cancer embolus appeared (P < 0.05). CONCLUSIONS: In the negative growth regulation of tumoral hepatocytes by TGF-beta(1), two defects take place: the expression of TGF-beta(1) at the protein level and the expression of TGF-beta R II at the mRNA level.

[Determination of cortisol in plasma and 24-hour urine of patients with central serous chorioretinopathy].

OBJECTIVE: To study the cortisol levels in patients with central serous chorioretinopathy (CSCR). METHODS: Endogenous cortisol levels in plasma and urine were determined in 44 patients with CSCR by radioimmunoassay and chromatography, and their results were compared with that of 41 controls. RESULTS: In acute CSCR, the mean values of the plasma cortisol (296.53 +/- 77.03) ng/ml and 24-hour urine 17-hydroxysteroids (the major metabolite of cortisol metabolism) (12.08 +/- 4.82) mg/24 h revealed significantly higher values in the patient group (P < 0.05). CONCLUSIONS: Increased levels of endogenous cortisol play a role in the development of CSCR.

[Wavelength selection in management of central serous chorioretinopathy].

OBJECTIVE: To compare the effects of three kinds of laser wavelength in management of central serous chorioretinopathy (CSCR). METHOD: 89 patients with CSCR were randomly divided into three groups according to different wavelengths used. Visual acuity, fundus, fundus fluorescein angiography (FFA), light sensitivity, central visual field were performed before and after laser therapy. RESULT: Compared with red and green wavelength groups, in yellow group the visual acuity, light sensitivity were improved more significantly (P < 0.05), and the disease course was shortened, the effects in recurrence rate were similar in the three wavelength groups. CONCLUSION: In the three kinds of waves, yellow and red have similar and positive effects in the treatment of CSCR.

[Effects of humic acid on lipid peroxidation in arsenosis prevalent areas].

OBJECTIVE: To explore the causes of blackfoot disease in Taiwan. METHODS: Experiments to induce lipid peroxidation by arsenic and humic acid were made in vitro. Arsenic was determined for the water sampled from the arsenosis prevalent areas in Inner Mongolia and blackfoot disease prevalent areas in Taiwan and humic acid determined for the extracts of the coal rich in arsenic sampled from Guizhou Province in China by MDA-TBA colorimetry. RESULTS: Experiments found that humic acid could induce lipid peroxidation caused by sodium salt of unsaturated fatty acids in vitro in a weak and unstable manner, which could be promoted by 0.05 mmol/L of Fe(+ +) and not by 1 mmol/L As(2)O(3). The extent of lipid peroxidation caused depended on the sources, constituents and structure of humic acid samples, those from Inner Mongolia ranked the highest, those from Guizhou and Taiwan the medium and the commercial humic acid (Aldrich Co. in the US) the lowest. The experiments also found that lipid peroxidation caused by commercial humic acid presented a dynamic process in the existence of trace amount of iron, and humic acid could decompose the products of lipid peroxidation. CONCLUSION: The experiments suggested the relations of humic acid and arsenic to iron and other transition elements in the blackfoot disease prevalent areas and areas with an environment rich in arsenic and humic acid.

[Research on the oncogenesis of cervical epithelial cells co-induced by human papillomavirus and human cytomegalovirus].

OBJECTIVE: The carcinogenesis of the human cervical carcinoma is known closely associated with human papillomavirus (HPV). The purpose of this article is to identify whether another sexually transmitted factor, human cytomegalovirus (HCMV) plays as a co-factor role in the carcinogenesis. METHODS: Human cervical epithelial cells immortalized by HPV16 were transfected with IE gene of HCMV. The condition and efficiency of IE gene were examined. RESULTS: It was showed that the cervical epithelial cells could be completely transformed into malignant by HPV 16 and IE gene of HCMV and tumor was formed by injecting the transformed cells in nude mice. CONCLUSION: The co-operation effect of HPV and HCMV auailed the oncogenesis of human cervical carcinoma and moreover, the cellular and molecular biological mechanism was discussed.

[The effect of defibrase in prevention of experimental atherosclerosis in rabbits].

OBJECTIVE: To study the anti-atherosclerosis effect of difibrase in rabbits. METHODS: Use of computer, light microscope, electron microscope, biochemistry and histochemical methods to identify the total cholesterol (TC) content, mean plaque area in aorta and its cholesterol content, degree of stenosis of coronary arteries of the experimental animals and control animals. RESULTS: Defibrase could decrease the mean plaque area of aorta and the TC content in the experimental animal group was less than that in the control group (P < 0.01). The findings in aortic arches and thoracic aorta were similar to those in the aorta (P < 0.05, P < 0.01). The number of plaques at the small artery openings in the thoracic aorta and abdominal aorta of the experimental group were less than that of the control group (P < 0.01). The cross sections made at the upper third of all hearts showed that the degree of stenosis of coronary artery branches in the heart wall of the experimental group was much less than that of the control group. CONCLUSION: Defibrase can significantly inhibit atherosclerosis.

[Purification and identification of pili of Vibrio cholera O139].

To provide the base of manufacturing V. cholera O139 subunit vaccine, purification and identification methods of V. cholera O139 Toxin-coregulated pili (TCP) were studied. The results showed the optimal expressing condition of TCP was in AKI or CFA medium, at 30 C for 24-36 h with stationary cultivation. The molecular weight of the TCP subunit protein measured by sodium dodecyl sulfate-poly acrylamide gel electrophoresis was about 20.5 kd. By using mouse anti-TCP monoclone antibody and sheep anti-mouse IgG-HRP antibody, the TCP was confirmed by dot blot immunoassay. Rabbit immune serum of TCP purified from O139 strain agglutinated both O139 biotype and El Tor biotype Vibrio cholera strains. These indicate that TCP may be an effective and protective antigen to conduct the preparation of V. cholera vaccine.

[Preparation of oral microspheres carrying V. cholera vaccine and its target's distribution].

To prepare oral biodegradable microspheres carrying V. cholera vaccine, the major outer membrane protein (OMP, MW = 41 kd) as a common antigen of cholera Vibriae was obtained from the classical strain Inaba 569 B, and the OMP was encapsulated in the biodegradable delivery system comprising Poly (DL-Lactide)-Co-Poly(ethylene glycol)microspheres. The average size of the microspheres was less than 5 microns, the amount of OMP encapsulated in microspheres was 15.3%. It was found that microspheres were taken up in Peyer's patches and then distributed in spleen, liver and mesenteric lymph nodes after oral administration.

[Chemical studies on the polysaccharide in fructus Corni].

OBJECTIVE: To study the tonic constituents in Fructus Corni, the pulp of Cornus of ficinalis. METHOD: Hot-water extraction, ethanol precipitation and column chromatography on DEAE-cellulose and Sephadex G-200 were used to isolate and purify the polysaccharides in Fructus Corni. HPGPC was used to determine the purity and measure the mean molecular weight of the polysaccharides. TLC, PC, GC, GC-MS, 13CNMR, partial acid hydrolysis and permethylation to elucidate the structures. RESULT: A water-soluble polysaccharide with mean molecular weight of 2.47 x 10(14), named Co-4, was obtained. It's main chain was (1-->4) linked beta-D-glucan, with branching points at position 6. CONCLUSION: A thorough study on the polysaccharides of Fructus Corni has been made for the first time and the structure of Co-4 has been elucidated.

[Immunohistochemical detection of hepatitis C virus antigen from hepatocellular carcinoma].

To investigate the infection of HCV and the expression of HCV antigen (HCAg) in HCC, immunohistochemical protocol was performed on tumorous liver tissues from 40 patients with HCC to detect HCAg and hepatitis B virus surface antigen (HBsAg) simultaneously. The results showed that the positive rates for HCAg and HBsAg were 17.5% (7/40) and 70% (28/40) respectively. HBsAg and HCAg were simultaneously detected in six of 40 cases and only one case had HCAg positive alone in the liver tumorous tissue. The positive signals were localized in diffuse cytoplasm of hepatocytes and the positively stained cells were mainly distributed in local pattern. The results suggested that HCV infection did exist in some cases of HCC, so HCV might be a viral risk factor in addition to HBV in the genesis of HCC in China.

[Effects of Dexamethasone, Fluorometholone and Florex on intraocular pressure after photorefractive keratectomy].

To evaluate the results of elevated intraocular pressure (IO) secondary to dropping of 0.1% Dexamethasone, Fluorometholone and Florex for 1-3 months after photorefractive keratectomy, we summed up and analysed the 3-month follow-up data on 546 eyes in three groups. The rates of IO elevated in Dexamethasone, Fluorometholone and Florex were 36%, 12.4% and 31.9% respectively. After topical use of corticosteroid, some of the elevations of IO took place as early as two weeks, but most of them occurred around 1 month. Eleven patients with IO elevation of both eyes had visual acuity decreased associated with eye-ache and halos and the other patients had no symptoms. After discontinuation of corticosteroid and institution of antiglaucoma therapy, the IO became normal in 3-7 days. In practice, this means that patients receiving corticosteroid eyedrops for more than two weeks should be checked for the possibility of ocular hypertension to prevent the corticosteroid glaucoma.

[Determination of gallic acid in garden burnet by HPLC].

OBJECTIVE: To establish a HPLC method for the determination of gallic acid in garden burnet. METHOD: The sample was extracted with methanol under ultrasonic management for 25 min. Chromatographic conditions: Shimadzu LC-6A with SPD-6AV detector at 270 nm and auto injector SIL-6A, methanol-0.025 mol/L phosphoric acid(15:85) as the mobile phase. Shin-pack CLC-ODS column(6.0 mm x 15 mm). RESULT: Gallic acid in garden burnet was baseline separated and determined. The average content was 0.22%, recovery rate 97.4% and relative standard deviation 1.3%. CONCLUSION: Simple, rapid and accurate, the method provides new indexes for quality control of Garden Burnet.

[Polymorphism analysis of D19S253 locus in Guangzhou area population].

Frequency data for STR system D19S253 were obtained from 105 unrelated individuals of Guangzhong population. PCR products were detected by horizontal native polyacrylamide gel electrophoresis and a total of 9 alleles were identified by side-by-side comparison with a sequenced allelic ladder prepared by ourselves. The observed genotype distribution conformed with Hardy-Weinberg equilibrium. The high information content found in this system(heterozygosity rate was 0.8353, the mean exclusion chance was 0.7499, the discrimination power was 0.9211, the polymorphism information content was 0.8203) indicated it is a useful means in forensic routine casework both in criminal and paternity cases.

[Polymorphism and haplotype study of Y-chromosomal multi-STRs loci in Han population in Guangzhou].

Using polymerase chain reaction and silver stain, polymorphism and haplotype study of Y-chromosomal multi-STRs loci: DYS19, DYS389I/II, DYS390 were studied. 111 samples of male were collected from Guangzhou area. 5 alleles were determined in DYS19 locus, 4 alleles in DYS389I locus, 5 alleles in DYS389II locus and 5 alleles in DYS390 locus. Compared with other racial populations, differences of distribution of allele frequencies existed significantly. 72 haplotype were present in this study.
