[Preliminary studies on subtypes of respiratory syncytial virus in China].

An analysis of subtypes of 9 respiratory syncytial (RS) viruses isolated from Guangzhou and Nanjing areas of china was carried out with eight Sweden RS-subtype specific monoclonal antibodies (MAbs) and 7 internal anti-RS MAbs. All these MAbs directed against respectively the large Glycoprotein (G), fusion protein (F), nucleoprotein (NP), and phosphoprotein (P) components of the prototype Long strain of RS virus. The patterns of the reactions of these MAbs to the nine isolated strains of RS virus were compared with indirect immunofluorescence assay (IFA), alkaline phosphoesterase-anti alkaline phosphoesterase (APAAP) enzyme-linked assay and Western blotting. The antigenic variations were founded among the strains of RS virus, and two subtypes allocated to the subtype A and B of RS virus by using the eight RS-subtype specific MAbs. Seven out of the 9 isolated strains of RS virus belonged to the subtype A, and two were being to the subtype B. The antigenic diversities were also founded within the same subtype, and the main pronounced difference were observed on the G glycoprotein by using the internal anti-RS MAbs. These findings are potentially important both for vaccine development and for the understanding of clinical and epidemiological characteristics of RS virus.

[The enhancing effect of calcium ionophore A23187 on the accessory function of mouse dendritic cells].

In this paper, the immunoregulatory effects of calcium ionophore A23187 on the accessory function of mouse spleen dendritic cells (DC) in primary mixed lymphocyte reaction (1.MLR) and oxidative mitogenesis are reported. A23187 had a direct enhancing effect on DC. When pretreated with 0.5-1 mumol/L A23187 for 6-8 h, the accessory activity of DC was twice as high as that of the control. This is circumstantial evidence indicating an up-regulation of activity mediated by an increase in intracellular Ca2+ levels and PKC activation.

Social acceptability of methylphenidate and behavior modification for treating attention deficit hyperactivity disorder.

The social acceptability of methylphenidate, behavior modification, and methylphenidate plus behavior modification was evaluated. Fifty mothers of children with attention deficit hyperactivity disorder (ADHD) and 50 control mothers, along with 21 children with ADHD and 20 control children, read a case vignette of an 8-year-old boy with ADHD and descriptions of the three treatment conditions. Subjects then rated the acceptability of each treatment. The mothers of children with ADHD were reassessed 3.5 months later, after experience with interventions for their children. Both ADHD and control families rated behavior modification as the most acceptable, methylphenidate as least acceptable, and the combined condition intermediate between the other two. At follow-up, there was a significant improvement in the acceptability of methylphenidate and the combined condition. The increased acceptability of methylphenidate at follow-up was related to increases in parents' knowledge about ADHD but not to the significant improvements that occurred in the children's hyperactive behavior.

[Effects of semen Ziziphis Spinosae oil and Ziziphis Spinosae extract on the decrease of serum lipoprotein and inhibiton of platelet aggregation].

Our experiments have shown that oral administration of Semen Ziziphis Spinosae oil(SZSO) or Ziziphis Spinosae extract (ZSE) given to quails for 53 days can significantly reduce their TC, LDL and TG and markedly subdue the fatty degeneration in their livers. Also, SZSO pressed into the stomach of rats for five days can conspicuously inhibit their platelet aggregation, while ZSE cannot.

Evaluation of long-term weight changes after a multidisciplinary weight control program.

The American Medical Association Council on Scientific Affairs recently emphasized the importance of nutritionally sound weight reduction diets and changes in life-style to promote long-term weight control. The purpose of this study was to evaluate long-term effectiveness and to identify predictors of success of an intensive multidisciplinary program utilizing the time-calorie displacement diet, a low-fat, high-complex-carbohydrate diet, to promote and maintain reduced body weight. In 213 obese adults consecutively entering the program, including dropouts, weight loss averaged 6.3 kg (0.4 kg/week) and 16% of excess weight over 7 months of treatment. The 78 (37%) completing the recommended 15 visits lost a mean of 10.8 kg and 28% of their excess weight while under treatment. Weight loss was largely predicted by number of visits, which was more important than treatment duration. An increased number of visits was predicted by being married, being female, and reporting a usual diet low in snack foods. Of 147 patients contacted at posttreatment follow-up, 53% maintained their weight loss or continued to lose weight an average of 25 months later and 24% experienced weight rebound but were still below their pretreatment weight; only 23% regained their lost weight. Mean net weight loss from pretreatment to follow-up was 8.0 +/- 1.0 kg (mean +/- standard error of the mean). No factor significantly predicted posttreatment weight rebound. Results suggest that a nutritionally sound diet, prescribed in a multidisciplinary program and emphasizing life-style changes, can result in long-term weight control in a majority of patients treated.

Expression of transforming growth factor-alpha in primary human colon and lung carcinomas.

The expression of TGF-alpha in human colon and lung carcinoma cell lines has been reported previously, but its expression in primary tumours has not been described in detail. We have used the radio-immunoassay method to measure the specific content of immunoreactive TGF-alpha in the acid ethanol extracts of normal and cancerous tissues of human colon and lung. The average TGF-alpha content of colon carcinomas is 4 times that of the normal mucosa, and for non-small cell lung carcinomas it is twice that of the normal parenchyma. Because of variability in the TGF-alpha expression among individuals and in different segments of colon and lobes of lung, the ratio of TGF-alpha content of paired tumour and normal tissue was also calculated. On average, the tumour/normal ratio for colon carcinoma is higher than that for lung carcinoma. Although 55% of colon tumours show a ratio 4 times, or greater, only 33% of lung carcinomas demonstrate this ratio. The level of TGF-alpha in both colon and lung carcinomas does not correlate with histological type stage, grade nor degree of desmoplasia of these tumours. Northern blot analysis of total cellular RNA confirms the expression of an approximately 4.8 kb TGF-alpha mRNA in normal colonic mucosa and lung parenchyma. However, in contrast to the results of radio-immunoassay, significant over-expression of TGF-alpha mRNA is uncommon in primary human colon carcinomas.

Magnetic resonance imaging of flow dynamics in the circle of Willis.

Magnetic resonance angiography was applied to the study of blood flow dynamics in the circle of Willis in nine patients with cerebrovascular disease and two normal volunteers. In conjunction with two-dimensional or three-dimensional gradient-echo acquisitions, selective presaturation of individual vessels was used to determine the direction of blood flow and the origin of the vascular supply. Presaturation causes signal loss within the territory supplied by the presaturated artery, without affecting vessels not crossing the presaturation slab. The results were correlated with those from transcranial Doppler sonography and conventional angiography. Magnetic resonance angiography was able to demonstrate the direction of blood flow, the presence or absence of collateral blood flow, and the blood supply to the pericallosal arteries, as well as the presence of a fetal posterior circulation. Magnetic resonance angiography is a noninvasive means for imaging the blood supply of the major intracranial arteries.

Nonenterococcal group D streptococcal septicemia: association with unrecognized endocarditis.

68 patients presented to the Veterans General Hospital, Taipei with nonenterococcal group D streptococcal septicemia in the years 1985-1987. 36 patients (53%) had nonenterococci as part of a polymicrobial bacteremia. The large intestine was not examined in most patients. Five patients (7%) had associated colonic carcinoma, and 17 patients (25%) had colorectal diseases. Only 7/68 patients (10%) were clinically diagnosed as having infective endocarditis by the doctors in charge. The others were regarded as having septicemia. The charts of these patients were reviewed retrospectively to diagnose infective endocarditis based on strict definitions. One (1%) had definite endocarditis proved at autopsy. 16 patients (24%) had probable endocarditis due to the presence of either a new regurgitant murmur or both a predisposing heart disease and embolic phenomena; 39 (57%) had possible endocarditis based on evidence of having either a predisposing heart disease or embolic phenomena; and only 12 (18%) had no evidence of endocarditis. 27 patients (40%) had at least one predisposing heart disease associated with endocarditis. 51 patients (75%) had at least one lesion suggesting embolic phenomena. 30 patients (44%) had electrocardiographic abnormalities. This high incidence of arrhythmia in nonenterococcal septicemia is of particular interest and could be related to cardiac involvement in some patients. The overall mortality, 62% (42/68), was extremely high in our series, but in those who were clinically diagnosed and treated as infective endocarditis, the mortality was low, 14% (1/7). We suggest all patients with nonenterococcal septicemia associated with either heart disease or lesions of CNS, lung, heart, kidney or limbs suggesting embolic phenomena should be regarded as having possible or probable endocarditis. Treating such patients as having infective endocarditis may reduce the mortality in nonenterococcal septicemia.

Short-course treatment of bacteremia with ceftriaxone monotherapy.

The efficacy of short-course ceftriaxone monotherapy in treatment of bacteremia was evaluated in an open protocol. Patients with laboratory-proven bacteremia were randomly treated with one of three dosing schedules for a duration of 5 to 7 days. Fifty-seven (62%) out of the 92 evaluable infections had successful results. Successful responses were seen in 20 (59%) out of 34 infections given 4 g every 24 hours, 15 (54%) out of 28 given 2 g every 12 hours, and 22 out (73%) of 30 given 2 g every 24 hours. The results showed no significant differences. The cases evaluated as failures were largely due to infections with resistant organisms or inadequate drainage of the primary infectious foci. Forty-nine (94%) of the 52 infections had successful results with one of the short-course treatment regimens, provided that they had no factors indicative of a poor prognosis. We stress the importance of anti-microbial susceptibility and adequate removal of the primary foci in the treatment of bacteremia. Our experience indicates that once-daily administrations of 2 g ceftriaxone as monotherapy is preferred for short-course treatment of bacteremia since it is equally effective, but more economical than higher dose regimens.

[Effects of radix Salviae Miltiorrhizae on visceral pain discharges in the posterior nucleus of the thalamus in cats].

Extracellular microelectrode method and stereotaxic technique of brain were used to investigate the effects of Radix Salviae Miltiorrhizae on the discharges of visceral pain in the posterior nucleus of thalamus(po)in cats. Experiments showed that Radix Salvia Miltiorrhizae inhibited the discharges of visceral pain in po, Radix Salviae Miltiorrhizae acts directly on the peripheral nerve trunk and cannot block its exciting conduction. This suggests that the analgesic effect of Radix Salviae Miltiorrhizae is exerted via central nervous system.
