Extracellular calcium is a mediator of astroglial injury during combined glucose-oxygen deprivation.

We tested the hypothesis that extracellular calcium is a mediator of astroglial injury during combined glucose-oxygen deprivation. Both differentiated and undifferentiated astroglial cultures were exposed to combined glucose-oxygen deprivation in the presence and absence of extracellular calcium. Lactate dehydrogenase efflux was used as an index of cellular injury. Both types of cultures exhibited significantly less cellular injury when exposed to combined glucose-oxygen deprivation in the absence of extracellular calcium (e.g. lactate dehydrogenase efflux in undifferentiated cultures after 12 h of exposure: presence of calcium, 65.2 +/- 2.5% vs. absence of calcium, 21.4 +/- 1.3%). To further elucidate the mechanism by which extracellular calcium produces injury, we studied the effect of nimodipine, an L-type calcium channel blocker, on astroglial injury resulting from combined glucose-oxygen deprivation. Nimodipine decreased cellular injury in both types of cultures (e.g. lactate dehydrogenase efflux in undifferentiated cultures after 12 h of exposure: untreated, 65.4 +/- 2.2% vs. 10 nM nimodipine, 44.6 +/- 4.2%). Extracellular calcium appears to be a mediator of astroglial injury during combined glucose-oxygen deprivation. These results suggest that influx of extracellular calcium via L-type voltage-gated calcium channels may contribute to astroglial injury during cerebral ischemia.

Response of pharyngeal gonorrhoea to single dose penicillin treatment.

The prevalence of gonococcal infection of the pharynx in 205 women, 331 heterosexual men, and 11 homosexual men with gonorrhoea was 6.8%, 4.2%, and 27.3% respectively. In only one patient, a heterosexual man, was the pharynx the sole site of infection. Throat symptoms were found in 7% of women, 21% of heterosexual men, and none of the homosexual men. Orogenital contact was reported by 29% of women, 36% of heterosexual men, and all the homosexual men. A single intramuscular dose of 1.8 g Bicillin (procaine penicillin 1.5 g plus benzyl penicillin 300 mg) cured 90% of patients.

Kidney development: regulatory molecules crucial to both mice and men.

Although the study of embryonic kidney development began in the 1950s, three decades passed until scientists began identifying the molecular controls of renal organogenesis. Most of these advances have come from mouse gene targeting and rodent kidney explant manipulation. Translation of the rodent data to human congenital kidney disease has only just begun. The activities of those regulatory molecules proven to be used in common appear remarkably similar in mouse and human renal development. Examples of these genes include glial cell line-derived neurotrophic factor (GDNF), RET, PAX2, Wilms tumor suppressor (WT1), and components in the renin-angiotensin pathway. Other factors that participate in mouse renal organogenesis, such as N-Myc, may later be proven important in human kidney development.
