What is in a pebble shape?

We propose to characterize the shapes of flat pebbles in terms of the statistical distribution of curvatures measured along the pebble contour. This is demonstrated for the erosion of clay pebbles in a controlled laboratory apparatus. Photographs at various stages of erosion are analyzed, and compared with two models. We find that the curvature distribution complements the usual measurement of aspect ratio, and connects naturally to erosion processes that are typically faster at protruding regions of high curvature.

Entropic interactions in soft nanomaterials.

We review the forces that rule interactions between phospholipid membranes and other soft nanomaterials such as polymers and colloids. Contrary to traditional nanostructures, soft materials display a high susceptibility to the fluctuations of the thermal environment, leading to new forces of an essentially entropic nature.

Solubilization of homopolymers in a solution of diblock copolymers.

We study theoretically mixed solutions of homopolymer and diblock copolymer chains. The solvent is a poor solvent for the homopolymers and a selective solvent for the copolymers. We find that the formation of copolymer micelles containing also the insoluble chains allows for an increased solubility of the homopolymers in the solution. In agreement with experiments, we find also that the solubilization power of the micelles, that is, the maximum amount of total homopolymer weight solubilized per unit weight of copolymers in solution, decreases strongly with the homopolymer index of polymerization.

Permeabilisation and solubilisation of soybean phosphatidylcholine bilayer vesicles, as membrane models, by polysorbate, Tween 80.

To understand better the wide-spread pharmaceutical use of non-ionic surfactant Tween 80 (TW), the colloidal properties of the surfactant alone and in combinations with the common phospholipid, phosphatidylcholine (PC), were studied. Static and dynamic light scattering revealed that TW solubilises PC at TW/PC approximately 2.75/1 mol/mol and that TW micelle disintegration occurs on time-scale of 2.5 min, independent of amphipath concentration. This is up to nearly 300-times faster than the TW caused dissolution of PC containing unilamellar vesicles. The apparent dissolution time of TW/PC mixed aggregates, in contrast, decelerates from >700 min to <5 min upon increasing starting total amphipath concentration, with thermal activation energy > or =24 (< or =80) kJ mol(-1). The aggregate dissolution rate in highly concentrated TW/PC suspensions reflects the dissolved polysorbate-aggregate exchange rate (approximately 6.7 x 10(-3)s(-1)) rather than TW flip-flop rate across a bilayer (>0.2 min(-1)). PC solubilisation proceeds linearly with the square-root of time, and is kinetically governed by the speed of surfactant diffusion through the bulk (D approximately 2.8 x 10(-11)m2 s(-1)). Creation of small Tween-phosphatidylcholine mixed micelles is typically preceded by pre-solubilisation structures, first in the form of deformable, strongly fluctuating, bilayer vesicles and then of elongated, presumably thread-like, mixed micelles. TW/PC mixed micelles become smaller with growing surfactant/lipid molar ratio, whereas TW/PC mixed vesicles become more and more leaky with increasing surfactant concentration. Our results highlight the molecular and kinetic aspects of polysorbate-membrane interactions and provide a rationale for the popularity of Tween surfactants in pharmaceutical products: such surfactants can solubilise fatty molecules and bilayer membranes but need quite a long time for this, which is available in pharmaceutical preparations but normally not in vivo; this makes Tweens relatively efficient and safe. Furthermore, our data could help design better ultra-deformable mixed lipid-surfactant vesicles for the non-invasive transdermal drug delivery across the skin.

Rheology of giant vesicles: a micropipette study.

We develop a micropipette rheometer to study the effect of oscillatory shear flow on the spontaneous fluctuations of phospholipid bilayers. Our results on giant vesicles show that oscillatory shear flow leads to a suppression of membrane fluctuations. They also imply that the Helfrich equation is modified in the presence of the flow. This equation, a fundamental constitutive relation between the amount of area stored in the fluctuations and the membrane tension, must be supplemented under oscillatory shear by a flow excess function that we determine.

The effect of cholate on solubilisation and permeability of simple and protein-loaded phosphatidylcholine/sodium cholate mixed aggregates designed to mediate transdermal delivery of macromolecules.

Carriers for non-invasive administration of biologically important antioxidant enzymes Cu,Zn-superoxide dismutase (SOD) and catalase (CAT) were developed. Solubilisation and permeabilities of various soybean phosphatidylcholine/sodium cholate (SPC/NaChol) mixtures, mainly in the form of lipid bilayers, focussing on system properties relevant for non-invasive enzyme delivery were investigated in this work. Static and dynamic light scattering measurements gave information on the behaviour of the systems containing up to 40 mM NaChol and 30.6-1.2 mM SPC in the final suspension. The average size of such mixed aggregates was in the 100-200 nm range. Suspension turbidity decreased by 50% upon increasing nominal molar detergent/lipid ratio to NaChol/SPC = 7 and 1.25, in case of SPC = 1.2 and 19.6 mM, respectively. The effective NaChol/SPC molar ratio in bilayers saturated with the detergent was found to be: R(e)(sat) = 0.70 +/- 0.01; bilayer solubilisation point corresponded to R(e)(sol) = 0.97 +/- 0.02, independently of enzyme loading. Vesicles became very permeable to SOD when membrane bound NaChol concentration exceeded 13.7 mM, in case of total starting lipid concentration of 138 mM diluted to SPC = 19.6 mM. Specifically, we measured a 50% loss of SOD from the vesicles with an aggregate-associated molar detergent ratio NaChol/SPC approximately 0.7, which is near the saturation but well below the solubilisation limit. Calcein efflux from such vesicles was compared with SPC/NaChol/SOD mixed aggregates. Our results should contribute to the future design of vesicle mediated transdermal delivery of antioxidant enzymes.

Anisotropy of lamellar block copolymer grains.

The Wulff construction and the theory of Milner and Morse [Phys. Rev. E 54, 3793 (1996)] were used to calculate the shape of grains formed when a lamellar block copolymer phase is formed by a shallow quench into the ordered state. The calculations show that the grains are ellipsoids of revolution, with an aspect ratio rho=2.37. This value is in reasonable agreement with results of transmission electron microscopy and depolarized light scattering experiments, which indicate that the average value of rho of grains formed during the early stages of the disorder-to-order transition is about 2.0.

Scale-dependent rigidity of polymer-ornamented membranes.

We study the fluctuation spectrum of fluid membranes carrying grafted polymers. Contrary to usual descriptions, we find that the modifications induced by the polymers cannot be reduced to the renormalization of the membrane bending rigidity. Instead we show that the ornamented membrane exhibits a scale-dependent elastic modulus that we evaluate. In ornamented lamellar stacks, we further compute the polymer contribution to the Caillé parameter characterizing the power law singularities of the Bragg peaks.

Impact of polymer tether length on multiple ligand-receptor bond formation.

The promoters of cell adhesion are ligands, which are often attached to flexible tethers that bind to surface receptors on adjacent cells. Using a combination of Monte Carlo simulations, diffusion reaction theory, and direct experiments (surface force measurements) of the biotin-streptavidin system, we have quantified polymer chain dynamics and the kinetics and spatial range of tethered ligand-receptor binding. The results show that the efficiency of strong binding does not depend solely on the molecular architecture or binding energy of the receptor-ligand pair, nor on the equilibrium configuration of the polymer tether, but rather on its "rare" extended conformations.

[Computerized ambulatory adolescent care--the experience of PAIA/ABEB].

PAIA (Program for Adolescent Integral Assistance from ABEB) is one of the preventive medicine programs of the Employee Beneficent Association at Belgo Mineira Metallurgy Company. Since its implementation, in February 1992, it has been offering specific medical assistance and educational instruction to adolescents from 11 to 18 years old through informative group dynamics. Age and maturity level are the guidelines for group formation. The groups are supervised by a multidisciplinary team and the methodology is playful and participatory. The themes approached in the meetings are chosen according to the adolescents' interest and maturity. After two years, the program was completely computerized aiming at improving the quality of assistance and accelerating services. PAIA/ABEB Program has surpassed all expectations concerning adolescents' and parents' interest and participation and the adolescents have become extra sources of information about the program and health education in their community. Behavioral changes like more concern about health care and more search for dialogue with meaningful adults have been observed. The observations and research carried out with adolescents have become a guideline for the preventive medicine campaigns at the firms belonging to Belgo Mineira Group. Indeed, differentiated medical assistance and global approach towards adolescence have been created in the clinic centers.
