Membrane properties of neuroglia in the optic nerve of Necturus.

The optic nerve of Necturus has proved a useful preparation for the study of glial cell membranes in vivo and in vitro with anatomical relations to axons intact and isolated following axon degeneration. The glial membrane potential behaves as a selective potassium diffusion potential; there is no evidence of a significant permeability to other naturally occurring ions. The specific membrane resistance of the glial cells is high compared to that of neurones; there are low-resistance intercellular connexions among the cells which permit the passage of both ions and the dye Lucifer Yellow. The cells are readily and reversibly uncoupled by procedures which decrease the intracellular pH. There is no evidence for voltage-sensitive sodium channels in the membrane. Following sodium gain and potassium loss the membrane displays a potassium-dependent strophanthidin-sensitive electrogenic sodium pump. The glial membrane is depolarized by potassium released from active axons as well as by glutamate. The glial depolarization contributes to potentials recorded with surface electrodes. Depolarization by K+ plays a role in the redistribution of K+ which locally accumulates around active neurones and also affects glial metabolism and glucose uptake.

Sodium channels in axons and glial cells of the optic nerve of Necturus maculosa.

Experiments investigating both the binding of radioactively labelled saxitoxin (STX) and the electrophysiological response to drugs that increase the sodium permeability of excitable membranes were conducted in an effort to detect sodium channels in glial cells of the optic nerve of Necturus maculosa, the mudpuppy. Glial cells in nerves from chronically enucleated animals, which lack optic nerve axons, show no saturable uptake of STX whereas a saturable uptake is clearly present in normal optic nerves. The normal nerve is depolarized by aconitine, batrachotoxin, and veratridine (10(-6)-10(-5) M), whereas the all-glial preparation is only depolarized by veratridine and at concentrations greater than 10(-3) M. Unlike the depolarization caused by veratridine in normal nerves, the response in the all-glial tissue is not blocked by tetrodotoxin nor enhanced by scorpion venom (Leiurus quinquestriatus). In glial cells of the normal nerve, where axons are also present, the addition of 10(-5) M veratridine does lead to a transient depolarization; however, it is much briefer than the axonal response to veratridine in this same tissue. This glial response to veratridine could be caused by the efflux of K+ from the drug-depolarized axons, and is similar to the glial response to extracellular K+ accumulation resulting from action potentials in the axon.

Detection of gene-disruption events in Aspergillus transformants by polymerase chain reaction direct from conidiospores.

We describe a rapid method for the identification of gene disruption events after DNA-mediated transformation of Aspergillus fumigatus. This involves a polymerase chain reaction in which the target DNA is added in the form of intact conidiospores. Using one primer specific to the plasmid DNA and a second primer specific to the target gene on the chromosome, it is possible to identify gene disruption events among the more common ectopic integrations approximately 4 h after sporulating transformants appear on selective medium.

Alcoholic carbowax prefixation and formal alcohol fixation. A new technique for urine cytology.

OBJECTIVE: To compare the cellular yield and sensitivity of cancer detection by urine cytology using alcoholic carbowax as a prefixative and formal alcohol as a final fixative with the conventional centrifugation-and-smearing method. STUDY DESIGN: Four hundred seventeen voided urine specimens were examined. Each specimen was divided into two equal portions. One portion was centrifuged and wet fixed (CS). The other was treated with alcoholic carbowax as a prefixative and then fixed in formal alcohol (CW). The smears were studied for cellular yield, morphology, cancer detection rate and typing. RESULTS: The CW method yielded significantly more cells with excellent morphology. It detected 25 more abnormal cases out of a total of 86, including 5 more transitional cell carcinomas, as compared to the CS method. This finding was statistically significant (P < .0000003). CONCLUSION: The CW method is simple and enhances cellular yield and the cancer detection rate as compared to the conventional centrifuge-and-smear technique.

L-myc genotypes in Hong Kong Chinese colorectal carcinoma patients.

L-myc genotypes have been correlated with prognosis in different human tumors. Its role in colorectal carcinoma (CRC) is still unclear. This study aimed to assess the L-myc genotypes in 99 Hong Kong Chinese CRC patients by PCR-RFLP techniques. The results obtained were correlated with clinical, histological and pathological parameters and genetic alterations. The observed frequency of L-myc genotypes (LL:LS:SS) was 27:46:26. The ratio of S to L alleles was 0.51:0.49. Distribution of L-myc genotypes and alleles in Hong Kong Chinese CRC was similar to that of healthy Chinese and CRC patients of other ethnic origins. The homozygous SS genotype was significantly associated with Dukes' stages C versus B. Other parameters including sex, differentiation status and survival, and genetic alterations such as p53 and Ki-ras mutations and Dcc LOH had no significant association with L-myc SS genotype.

Sensitivity of sputum cytology after homogenization with dithiothreitol in lung cancer detection. Two years of experience.

OBJECTIVE: To compare the sensitivity of sputum cytology using the dithiothreitol homogenization method (DTT) with that of the standard pick-and-smear (PS) method in lung cancer detection. STUDY DESIGN: In a two-year period, 2,428 sputum specimens were examined. Standard PS smears were prepared as controls; the remainder of the specimens were homogenized with DTT before smears were prepared. The sensitivity of the two methods in detecting positive cases was compared. RESULTS: Among 260 positive cases, the DTT method detected 256, while the PS method detected only 212, representing a difference of 17%. The DTT method also detected 28 more suspicious cases and permitted definitive typing of the tumor in a further 11 cases. CONCLUSION: Homogenization of sputum with DTT is not only a simple and versatile method but also a more sensitive method as compared to the PS method in lung cancer detection.
