Immortalization of murine primary spleen cells by v-myc, v-ras, and v-raf.

Growth factor-independent cell lines, including four lines characterized as macrophages, were isolated by infection of BALB/c mouse primary spleen cells with combinations of three retroviruses encoding v-myc, v-ras, and v-myc/v-raf. Proliferating cell lines were isolated only rarely, and after long crisis periods, following the introduction of myc and raf by infection with J2 virus, or of myc and ras by coinfection with myc309 and raszip6 viruses. However, sequential infections with all three viruses--myc plus ras cells reinfected with J2, or J2 followed by myc plus ras coinfection--resulted in rapid outgrowth of cell lines which grew at high growth rates to high densities. When cells were treated with anti-IgG F(ab')2/IL-4/IL-5 to specifically stimulate B cells, cell lines were isolated readily by infection with myc plus ras alone, J2 alone, or all three viruses. These cell lines arose after shorter crisis times and all grew at high growth rates and to high densities. Analysis of cell surface markers and immunoglobulin gene arrangement revealed no lymphoid characteristics in any of the lines. Four cell lines express all three macrophage markers analyzed (F4/80, Mac1, FcR), and many others are Mac1+ and/or FcR+. Out of 20 immortalized cell lines tested, 13 show clonal growth in soft agar, and 3/6 of these produced tumors in BALB/c mice, indicating that fully transformed cells may be isolated by these procedures. In at least one of the cell lines, integration of all three infecting viruses has occurred.

Amnesics have a disproportionately severe memory deficit for interactive context.

Two experiments were used to compare the recognition memory of amnesic and normal subjects for intentionally encoded words (targets) and for incidentally encoded words that were meaningfully related to the targets and presented at the same time (interactive context). In both experiments the target recognition of the two groups was matched at a high level by presenting the amnesics with much shorter lists of words to remember. Experiment 1 compared 20 amnesics and their matched controls and showed that whereas the amnesics' recognition of the target words did not benefit significantly when they were presented together with their interactive context words (relative to their recognition when the target words were presented alone), that of the controls did. Experiment 2 compared 14 amnesics and their matched controls and showed that when patients and their controls were matched on their target word recognition in isolation, then the patients still showed worse recognition for the interactive context words. These effects were not found only in Korsakoff patients, and their size did not correlate with behavioural measures of frontal-lobe damage. It is concluded that amnesics may be more impaired at recognizing incidentally encoded interactive context than they are at recognizing target material, and this deficit may be an essential feature of the syndrome.

Pediatric cardiovascular diagnosis.

Significant progress is being made in the field of pediatric cardiovascular imaging. The application of color Doppler transesophageal echocardiography has proven to be extremely useful in both intra- and perioperative examinations. The introduction of the long-axis pediatric probe has expanded the abilities of transesophageal imaging in clinical pediatric practice. MR imaging is continuing to evolve as a noninvasive imaging technique. The development of cine-MR imaging now permits simultaneous evaluation of cardiac anatomy and function and is forecasting a larger role for this technology. In the field of interventional cardiovascular radiology, there have been further reports describing percutaneous balloon valvoplasty both at valve and vascular levels, as well as the application of balloon expandable intravascular stents for a variety of obstructive lesions.

Studies on the phenotype and karyotype of immortalized rabbit kidney epithelial cell lines.

Differentiated mammalian cell lines can be isolated by immortalizing primary cells by transfection with DNA from plasmids containing sequences from SV40 early region. These cell lines show cytogenetic abnormalities but the degree of aneuploidy is considerably less than that observed in other established cell lines. No correlation was observed between the degree of differentiation of a clone and the extent of chromosomal damage.

Enhanced growth characteristics of hybridoma cells infected with myc- and ras-containing retroviruses.

The introduction of retrovirally encoded myc or ras genes into the mouse hybridoma cell line PQXB 1/2, in which antibody production is unstable, resulted in altered growth characteristics: infected cells showed increased growth rates, higher peak cell densities, and slower decline of viability following maximum density than did control cultures. In addition, some oncogene-infected cultures maintained initial levels of antibody production during 16 weeks in culture, while antibody levels fell by 90% in uninfected PQXB cells. Both myc and ras oncogenes were expressed at only low levels in control and infected cells. These results suggest that oncogene expression in hybridomas can lead to enhanced growth characteristics and can stabilize antibody production.

Disproportionate intentional spatial-memory impairments in amnesia.

Twenty-four amnesics, including patients with Korsakoff's disease, post-encephalitic amnesia and amnesia caused by rupture of an anterior communicating artery aneurysm (ACoAA), were compared with 24 matched control subjects on a task in which words were presented in any one of four positions on a computer screen and subjects were instructed to remember both the words and their locations. The patients were tested after more learning opportunity, exposure to shorter lists, and after shorter delays than were their controls in order to match the word recognition performance of the two groups. Under these conditions, the amnesics' ability to locate recognized words was significantly worse than that of their controls. Although there was a tendency for the ACoAA patients to show more severe spatial memory deficits than Korsakoff patients, there was no clear evidence that aetiology of amnesia was a critical determinant of whether spatial memory was more impaired than word recognition. It was concluded that amnesics show a disproportionately severe memory deficit for spatial information that is intentionally encoded as well as for that which is incidentally encoded.

Korsakoff amnesics are poor at judging the sequence of two tones.

Alcoholic amnesics were given a test of temporal sequencing ability devised by Efron which has practically no memory component. These amnesics were very impaired on the task. However, the extent of this impairment did not relate to the magnitude of their "target memory" deficit nor did it relate to the ability to make temporal judgements from memory. Two other groups of amnesics (3 post-encephalitis and 4 with ruptured aneurysms of the anterior communicating artery) did not show impairments on the sequencing task. Data from the amnesic patient N.A. (who was unimpaired on the task), three other amnesics (who showed a tendency to be poor at the task) and two frontally damaged patients (one of whom performed especially badly on the sequencing task but had no obvious memory difficulties) were also presented. It is argued that deficits in temporal discrimination may not be found in all amnesics but, when they are observed, are an incidental feature related to additional (possibly cortical damage. Implications of the results for the view that amnesia results from a deficit in the use of contextual information (including temporal information) are considered.

Life course of depressive illness and characteristics of current episode in patients with double depression.

It has been suggested recently that major depression and concurrent chronic minor depression, or "double depression" (DD), may have a different course and presentation from major depression alone (MDA). The present study compares 41 patients with DD and 60 patients with MDA for differences in life course of depressive illness and characteristics of the current depressive episode. Patients with DD, as compared with patients with MDA, had an earlier age of onset of mood disturbance, more episodes of major depression, and more frequent concurrent anxiety disorders. However, patients with DD were not significantly different from patients with MDA who had greater than a 6-month history of mood disturbance, with regard to life course of illness variable. The characteristics of current depression in patients with DD and MDA were not significantly different. The clinical and theoretical implications of these findings are discussed.

Performance on tests sensitive to frontal lobe lesions by patients with organic amnesia: Leng & Parkin revisited.

Groups of amnesics with aetiologies that included chronic alcoholism, encephalitis and ruptured anterior communicating artery aneurysm (ACoAA) were examined on the Cognitive Estimation Test (CET), FAS Word Fluency Test (FAS) and the full and Nelson (1976) versions of the Wisconsin Card Sorting Test (WCST). The alcoholic amnesics were impaired on all four tests, whereas the post-encephalitic amnesics were impaired on the FAS and CET but performed normally on both versions of the WCST. The ACoAA amnesics were impaired on both the FAS and the CET, and scored at a level in between the other amnesic subgroups on the WCST.

Vascular reactivity of the coronary artery in rainbow trout (Oncorhynchus mykiss).

Vascular reactivity of the large coronary artery of a lower vertebrate was investigated for the first time in this study. The responses of vascular rings to various pharmacological agents were measured using isometric force transducers. Coronary arteries from rainbow trout, Oncorhynchus mykiss, contracted with acetylcholine and predominantly relaxed with isoproterenol, epinephrine, norepinephrine, and serotonin. Atenolol blocked the adrenergic relaxation, suggesting a predominance of beta 1-adrenoceptors. Nitroglycerin and sodium nitroprusside relaxed vascular rings. Responses to histamine and bradykinin were absent in this tissue. Adenosine, ADP, and ATP caused contractions that could be blocked by theophylline. The difference in vascular reactivity between that typically found in mammalian coronary arteries and that reported here for rainbow trout are discussed.

Thromboxane A2 and prostaglandin endoperoxide analogue effects on porcine renal blood flow.

Thromboxane A2 (TxA2) has been implicated as a mediator of renal and cardiovascular diseases. However, the direct effects of TxA2 on the renal system have been difficult to study because of the instability of the agonist. In the present study injection of synthetic TxA2 directly into the renal artery of anesthetized pigs produced dose-related decreases in renal blood flow (RBF) from 101 +/- 11 to 12 +/- 3 ml/min at the highest dose (20 ng/kg). The reductions in RBF were similar to those produced by the stable prostaglandin endoperoxide analogue, U-44069, although TxA2 was three times more potent. Under these conditions there were no effects on either mean (MAP) or pulmonary arterial pressures (PAP), or the heart rate (HR). The maximal effects produced by TxA2 and U-44069 on RBF were reproducible and the response remained unchanged after pretreatment with either heparin or indomethacin. Systemically administered TxA2 (200 ng/kg iv) increased PAP from 20 +/- 1 to 34 +/- 3 mmHg and this effect was associated with modest increases in MAP and HR. Intravenous administration of the thromboxane receptor antagonist, L-655,240, inhibited the reductions in RBF produced by intrarenal TxA2 and U-44069 and attenuated the cardiopulmonary effects of TxA2 administered intravenously. The results demonstrate directly that TxA2 is a potent agent for decreasing RBF through interaction with a thromboxane receptor.

Development of new cell lines for animal cell biotechnology.

Mammalian cell culture has been an important technique in laboratory-scale experimentation for many decades. Developments in large-scale culture have been due to the need to grow large numbers of cells to support the growth of viruses for vaccine production, and more recently, for growing hybridoma cells as a source of monoclonal antibody. Increasingly, however, pharmaceutical products such as hormones, enzymes, growth factors, and clotting factors are being produced from cell lines which have been manipulated by recombinant DNA techniques. It is clear, therefore, that the high cost of growing mammalian cells on a large scale does not necessarily prohibit their use for biotechnology, and indeed there is considerable evidence to suggest that animal cell biotechnology will continue to be a major growth area in the future.

The prevalence of cyclothymia in borderline personality disorder.

Sixty patients with personality disorders were evaluated by several different diagnostic instruments to determine the prevalence of cyclothymia in borderline personality disorder (BPD) and in other personality disorders (OPD). Cyclothymia occurred more frequently in BPD than in OPD, regardless of which diagnostic system was used. In contrast, the prevalence of major, minor, and intermittent depression, hypomania, and bipolar disorder was not significantly different in BPD as compared with OPD. Cyclothymic borderlines and noncyclothymic borderlines could not be distinguished on behavioral or functional measures. These results have implications for the diagnostic validity of both BPD and cyclothymia.

Genetic analysis of metabolic cooperation in mammalian cell lines.

Metabolites can be exchanged between cells in culture by direct transfer from the cytoplasm of one cell to that of another in a process known as metabolic cooperation. Most mammalian cell lines are able to transfer small molecules directly between adjacent cells in this way and are consequently mec+; however, a small number are defective in this ability (mec-). Results obtained from somatic cell hybrids formed between combinations of these cells have shown that the four different cell lines examined in this study can be divided into at least two different complementation groups on the basis of their ability to transfer 3H-labeled nucleotides to adjacent cells. Two of the cell lines clearly fall into a single complementation group.

Life events and mania: a case-controlled study.

Life events were evaluated in 14 patients with bipolar affective disorder in the year preceding a mania as well as in 14 patients with bipolar affective disorder without a mania matched for age, sex, and time of recall. There were a significantly greater number of uncontrolled and unanticipated life events in the patients with as compared to those without a mania. However, neither the total number of life events nor measures of distress and stress distinguished bipolar patients with as compared to those without mania.

Immunogenicity of liposome-bound hyaluronate in mice. At least two different antigenic sites on hyaluronate are identified by mouse monoclonal antibodies.

Hyaluronate (HA) was previously demonstrated to be immunogenic in rabbits. The immunogenicity of HA in mice was studied. Hyaluronidase-digested streptococcal HA (IA1) covalently linked to liposomes (IA1-liposomes) were produced for immunization. Mice immunized with IA1-liposomes developed measurable serum antibodies to IA1, while mice immunized with IA1 in Freund's adjuvant did not. mAbs produced by two stable hybridomas (10G6 and 5F11) from mice immunized with IA1-liposomes produced IgG antibody reactive with HA in ELISA. 10G6 had a much higher avidity for liposome-bound IA1 than free IA1, while 5F11 did not, suggesting that the mode of presentation of IA1 is important in HA immunogenicity and antigenicity. Both mAbs recognized terminal HA immunodeterminants exposed by hyaluronidase treatment. Sonication had no effect on HA reactivity for either mAb. However, ascorbic acid treatment significantly reduced the antigenicity of HA for mAb 5F11, but not 10G6. Only 10G6 was inhibited by glucuronic acid. Electrostatic forces appear to play a role in the binding site of 5F11, but not 10G6. 5F11 crossreacts with heparan sulfate and phosphorylcholine, while 10G6 did not crossreact with any glycosaminoglycans or phosphorylated compounds tested. These results confirm that HA is immunogenic. They suggest that the mode of presentation of HA is important for the induction of the immune response, and in HA antigenicity. At least two different antigenic sites on HA were demonstrated. 10G6 recognizes a terminal HA antigenic site expressed on IA1-liposomes that contains glucuronic acid in its immunodominant site. 5F11 recognizes an HA antigenic site in which electrostatic forces appear to play a role, is sensitive to ascorbic acid treatment, and is crossreactive with heparan sulfate. The use of mAbs should facilitate immunologic studies of HA.
