Density distribution and density conversion of neutrophils in allergic subjects.

We investigated the density distribution of neutrophils in peripheral blood of allergic subjects. We divided neutrophils into four groups: fraction 1 (density greater than 1.085), fraction 2 (1.081 less than density less than or equal to 1.085), fraction 3 (1.077 less than density less than or equal to 1.081) and fraction 4 (density less than or equal to 1.077). The percentage of neutrophils in fraction 2 in allergic rhinitis (AR) subjects or asthmatics was lower than that in normals (p less than 0.01). The percentage of neutrophils in fraction 3 and fraction 4 from AR or asthmatics was greater than that in normals (fraction 3, p less than 0.01; fraction 4, p less than 0.05). In neutrophils from AR subjects (fraction 3), chemotaxis to N-formyl-methionyl-leucyl-phenylalanine or platelet-activating factor (PAF) was enhanced compared to fraction 2. PAF (10(-7) M) changed the density of neutrophils (p less than 0.01), which were inhibited by WEB 2086 (p less than 0.05). Furthermore, granulocyte-macrophage colony-stimulating factor changed the density of neutrophils (p less than 0.01). These findings suggest that biological agents may activate neutrophils and convert their density resulting in neutrophils with lower density in allergic subjects.

Morphofunctional study of pituitary adenomas with acromegaly by immunoperoxidase technique and electron microscopy.

In attempting to establish a functional classification of acidophilic adenomas of the pituitary gland, immunoperoxidase staining techniques specific for growth hormone (GH), prolactin (PRL), and luteinizing hormone (LH) were carried out. Surgically removed specimens from 24 acromegalic patients were studied. The pathologic observations were then compared with the basal plasma levels of each of the hormones. The existence of GH was demonstrated in all patients. Immunohistologically, GH-secreting adenomas could be subdivided into three types: Type I had the highest plasma-GH level (average 121 ng/ml), while the average plasma-GH levels of types II and III were 103 ng/ml and 73 ng/ml, respectively. This result suggested that the serum-GH level may correlate with the GH cell population in a tumor. In 13 of the acromegalic patients, immunoenzyme methods were utilized in order to identify prolactin cells. No clear correlation between serum-PRL levels and the distribution of PRL cells could be demonstrated. A few immunoreactive LH cells were found in 13 patients. These LH cells may be a mixture of normal cells, based on our clinical finding.

The murine T-cell receptor uses a limited repertoire of expressed V beta gene segments.

Only 10 different V beta gene segments were found when the sequences of 15 variable (V beta) genes of the mouse T-cell receptor were examined. From this analysis we calculate that the total number of expressed V beta gene segments may be 21 or fewer, which makes the expressed germline V beta repertoire much smaller than that of immunoglobulin heavy-chain or light-chain genes. We suggest that beta-chain somatic diversification is concentrated at the V beta-D beta-J beta junctions.
