Densitometric analysis of the nerve cell population of the inferior olive in aging.

Possible age-related changes in the nerve cell population of the inferior olive were studied through densitometric analysis. The mean area per nerve cell, the reciprocal of the number of cells per unit area, was derived from the distance to closest neighbor in an unselected autopsy series of specimens from 26 subjects without signs of neurological damage, aged 24 to 83 years. Kendall's rank method showed correlation between the mean area per nerve cell and the subjects' ages, with mean areal values decreasing with age. The increase in neuronal density with age was related to changes occurring in the neuropil of the adult. These changes consist of regression of olivocerebellar contacts and loss of supernumerary axonal collaterals, with a resulting decrease in the volume of the whole formation. In addition, the data supported the hypothesis that the nerve cell population of the inferior olive should not decrease with aging.

The sites of hormone storage in normal and diseased parathyroid glands: a silver impregnation and immunohistochemical study.

The results of Grimelius' silver impregnation method and of two immunohistochemical methods for the demonstration of the sites of storage of parathyroid hormone (PTH) were compared in a series of parathyroid glands from 39 patients affected by either hyperplasia or adenoma and from 10 normal subjects. An indirect immunoperoxidase method using an anti-bovine PTH serum gave unsatisfactory results. In contrast a biotin-streptavidin immunoperoxidase method using an antibody against the 44-68 amino acid sequence of human PTH yielded similar results to those with Grimelius' silver impregnation method. This supports the view that argyrophil granules correspond to the secretory granules of PTH. The degree of silver impregnation, however, varied considerably in duplicate sections, probably because of difficulties in standardization of the Grimelius' method. The biotin-streptavidin method appeared more sensitive and was reproducible. The immunohistochemical demonstration of PTH may prove useful in defining the sites of hormone storage in normal and pathological conditions.

Allometry of the perivascular spaces of the putamen in aging.

The equation of simple allometry was used to study the relative growths of the diameters of the perivascular spaces (PSs), the diameters of the arterial vessels and their wall thicknesses in specimens of the putamen from 24 subjects aged 62 to 90 years. Our findings supported the view that the diameters of the PSs were mainly related to the dimensions of their arteries, especially in the elderly, and that they grew isometrically with arterial diameters. In addition, the diameters of the PSs were basically independent of age.

The ultrastructure of the nervous tissue in a benign teratoma.

The ultrastructure of the nervous tissue in a benign ovarian teratoma is described. This tissue was organized into areas having both "meningeal" and "ependymal" surfaces, between which were found astrocytes, ependymal cells, neurones with synapses and microglia. These cells all had ultrastructural similarities to their normal counterparts in the nervous system. In addition, some signs of degenerative change--due possibly to the abnormal location of the nervous tissue--were observed. Oligodendrocytes and myelin were absent, possibly because of vascular insufficiency.

Correlation of morphological variables in germ-cell hypoplasia.

The correlations between the progression of germ-cell hypoplasia, measured with the Johnsen's score, reduction in tubular diameter, and thickening of the lamina propria of the seminiferous tubules were studied in a series of 42 biopsy specimens of the testis from 26 infertile men. There was a significant negative correlation between the mean tubular diameter and the mean thickness of the lamina propria (p less than 0.01), as well as a significant positive correlation between the Johnsen's score and the mean tubular diameter (p less than 0.01). The Johnsen's score did not correlate with the mean thickness of the lamina propria. Reduction of the tubular diameter and thickening of the lamina propria could be found across the spectrum of scores, but they prevailed in severe germ-cell hypoplasia.

The relationship between obesity and degree of arteriosclerosis. New autopsy findings.

The degree of arteriosclerotic changes in the aorta and major vessels was inversely related to obesity (P less than 0.05) in an unselected series of 300 autopsies on subjects older than 50 years who had no hypertensive diseases. In addition, out of 11 subjects with very severe obesity, 9 showed no arteriosclerosis, and only 2 showed mild arteriosclerosis.

Leucovorin rescue of enteric lesions induced by high-dose methotrexate: a time-related study in mice.

In this study, different groups of mice were treated with a single high-dose of methotrexate or with a leucovorin-methotrexate association, and the changes in structure and cell types of the small intestine were studied histologically. The results suggested that leucovorin rescue diminished the degree of mitotic abatement induced by methotrexate in the first days after treatment, with better preservation of enteric structures and the cell type ratio.

The sinusoids in cirrhosis. A morphometric study.

The volume fraction of the sinusoids (Vv), the sinusoidal surface:total volume ratio (Sv), and the sinusoidal length:total volume ratio (Lv) were evaluated morphometrically in twelve biopsy specimens with cirrhosis and in eleven control specimens. The Sv, which is directly related to the radius of the sinusoids, was significantly (0.01 greater than P greater than 0.001) greater in the cirrhotic group than in the controls. In contrast, the Lv, which is directly related to the length of the sinusoids, and the Vv did not vary significantly in the same groups. These data support the concept that changes of the sinusoids do not contribute to portal hypertension in cirrhosis.

Morphometry of the breast fibroadenoma.

This work is concerned with a morphometric analysis of 23 breast fibroadenomas subdivided into four histological subgroups from purely pericanalicular to purely endocanalicular forms. The volume fractions (Vv) of the lumina and the total volume: epithelial surface ratios (Sv) showed a significant (P less than 0.001) correlation (r = + 0.64). In addition, the Vv and Sv series did not correlate with any of the histological subgroups. The data reported in this work suggest that the ductal epithelium proliferates in fibroadenomas, and that the subtypes are not related to the epithelial growth.

Duct carcinoma of the minor salivary glands: a case report.

A tumour was removed from the oral vestibule of a 62-year-old woman. It showed histological features of both salivary duct carcinoma and papillary adenocarcinoma. The possibility that these entities might be but varieties of duct adenocarcinoma is suggested.

Morphometric analysis of capillary and cavernous hemangiomas.

This work is concerned with the morphometric analysis of 19 subcutaneous hemangiomas of capillary and cavernous types, to investigate their evolution. Our results suggest that hemangiomas in adults are of malformative nature, and that there is no clear-cut separation between the above types, which only designate extreme patterns of non-intermittent series. Cavernous changes are probably due to blood flow and intravascular pressure in originally capillary hemangiomas.

Epithelia of endodermal derivation in teratomas.

This work is concerned with the histological features of endoderma-derived epithelia in a series of 48 benign teratomas. The intestine-type epithelium had the histochemical characteristics of that of the large intestine. It occasionally showed argentaffin cells, Paneth cells and accessory structures, such as a basement membrane, bundles of smooth muscle fibers and lymphoid aggregates including IgA-secreting plasma cells. In a teratoma, there was transitional-type epithelium. The respiratory-type epithelium included the basic cell types of the normotopic. It often rested on a basement membrane, and was occasionally surrounded by seromucous glands, cartilage plaques, muscle fibers and lymphoid aggregates.

Microglia in teratomas.

Using an ammoniacal silver nitrate stain, we have demonstrated typical microglial cells in three teratomas. Microglia appeared identical to their counterparts in the mature human brain, both resting and slightly hypertrophic types being present in differentiated nervous tissue, which also comprised astrocytes, neurones and ependyma. The implications of the presence of microglia with ectodermal derivatives in teratomas are discussed.

Pulmonary toxicity in mice after high-dose methotrexate administration with and without leucovorin rescue.

Interstitial lung lesions were induced in mice by high-dose methotrexate with high frequency. They appeared early after treatment; their onset, evolution and recovery parallelled those of lesions to the hemopoietic tissues and the intestine. The pathogenesis of methotrexate lung toxicity in mice is discussed. Leucovorin rescue was ineffective in preventing the lung lesions induced by high-dose methotrexate.
