Isolation and characterization of rat kidney alanine aminopeptidase.

Rat alanine aminopeptidase was purified from kidney by isolation of the brush border membrane with CaCl2 followed by differential centrifugation and tryptic proteolysis. It is a glycoprotein with a molecular weight of approximately 210,000 daltons comprising two 110,000-dalton subunits and has an amino acid composition similar to that of the human enzyme. Two zinc atoms are covalently bound to each protein subunit.

Comparative study of alanine-amino-peptidase and gamma-glutamyl-transferase activity in normal Wistar rat urine.

24 h urinary alanine-amino-peptidase (AAP) and gamma-glutamyl transferase (GGT) activities were studied from the 3rd-7th month of life in male Wistar rats. A close relationship was found between AAP and GGT activity, except at the beginning and at the end of this period. At the end there appear 2 subgroups, the larger (70%) showing a strong correlation between AAP and GGT activity and the other (30%), demonstrating no correlation at all. A good correlation between AAP and GGT activities, creatinine, 24 h urine volume and 24 h creatinine output was found.

The thymol turbidity test and determination of serum cholesterol : two tests to abandon in the routine evaluation of liver function.

The thymol turbidity test (Macglan's test) and the determination of serum cholesterol are still performed routinely as liver function tests in many laboratories. In this report, we have employed quantitative methods in order to evaluate the real usefulness of these two parameters. In a first step, the value of the thymol test and of cholesterol determination for the discrimination of the 13 most frequent hepato-biliary diseases was studied by analysis of variance, and compared to that of the following tests : serum bilirubin, serum glutamic pyruvic transaminase, serum alkaline phosphatase serum protein electrophoresis and prothrombin time. It was found that of all these parameters, the thymol test and cholesterol measurement had the lowest discriminatory powers. In a second step, the consequences of the suppression of the two tests were examined by linear discrimination analysis, which was done for all the possible pairs of diseases. It appeared that in each case, the loss of information due to the elimination of the thymol test or of cholesterol determination was nil or negligible. We conclude therefore that the thymol turbidity test and determination of serum cholesterol should be abandoned as routine tests of liver function.

[Protein assay by immunoprecipitation with Centrifichem centrifugal analyser].

The authors present an automatic technic of estimation of ten serum proteins (albumin, alpha-antitrypsin, orosomucoid, alpha-1-macroglobulin, haptoglobin, C'3 complement, transferrin, IgM, IgG, IgA) by immunoopacimetry on a Centrifichem centrifugal analyser. This technic is derived from an immunonephelometric method, with the following differences: --the use of two concentrations of polyethylene glycol permitting one to obtain for all proteins, a constant levelling-out of the reaction within 3 minutes; --by a first reading at 3 seconds avoiding the prior determination of blank reagents and sera; --by the use of two dilutions of serum permitting calculation of all concentrations. The results are reproducible and are satisfactory: 95% of the coefficients of variation are less than +/- 5% in the case of all the proteins. The correlation with IDR is good. The coefficients vary from 0.91 to 0.99.

Effect of subtotal nephrectomy on the excretion of urinary alanine aminopeptidase and gamma glutamyltransferase in the male Wistar rat. A long-term study.

The study of alanine aminopeptidase (AAP) and gamma glutamyltransferase (GGT), two integral components of the brush border membrane of the proximal kidney tubule, has generated considerable interest for research on the nephrotoxicity of drugs. AAP and GGT activity in urine correlate strongly. The three phases after a subtotal nephrectomy can be described as follows: the first phase, lasting 1 month, is characterized by an abrupt decrease of 70% in AAP and GGT activity, corresponding to the excised kidney mass. During the second phase, a partial recovery occurs representing nearly 50% of the AAP and GGT activity in the control group (30th to the 75th day after nephrectomy). These activities begin to decrease slightly in the third phase until the end of the experiment. In terms of remaining kidney mass, the results demonstrate a twofold increase of the AAP and GGT activity. The same phenomenon can be seen in the excretion of creatinine, except that the compensatory activity is multiplied by four in the remaining kidney. Good correlations between AAP activity, GGT activity, urinary creatinine, 24-hour urine volume, and 24-hour creatinine output are found in both experimental and control groups.
