Autotrophic CO(2) fixation by Chloroflexus aurantiacus: study of glyoxylate formation and assimilation via the 3-hydroxypropionate cycle.

In the facultative autotrophic organism Chloroflexus aurantiacus, a phototrophic green nonsulfur bacterium, the Calvin cycle does not appear to be operative in autotrophic carbon assimilation. An alternative cyclic pathway, the 3-hydroxypropionate cycle, has been proposed. In this pathway, acetyl coenzyme A (acetyl-CoA) is assumed to be converted to malate, and two CO(2) molecules are thereby fixed. Malyl-CoA is supposed to be cleaved to acetyl-CoA, the starting molecule, and glyoxylate, the carbon fixation product. Malyl-CoA cleavage is shown here to be catalyzed by malyl-CoA lyase; this enzyme activity is induced severalfold in autotrophically grown cells. Malate is converted to malyl-CoA via an inducible CoA transferase with succinyl-CoA as a CoA donor. Some enzyme activities involved in the conversion of malonyl-CoA via 3-hydroxypropionate to propionyl-CoA are also induced under autotrophic growth conditions. So far, no clue as to the first step in glyoxylate assimilation has been obtained. One possibility for the assimilation of glyoxylate involves the conversion of glyoxylate to glycine and the subsequent assimilation of glycine. However, such a pathway does not occur, as shown by labeling of whole cells with [1,2-(13)C(2)]glycine. Glycine carbon was incorporated only into glycine, serine, and compounds that contained C(1) units derived therefrom and not into other cell compounds.

Biosynthesis of a neo-epi-verrucosane diterpene in the liverwort Fossombronia alaskana. A retrobiosynthetic NMR study.

The biosynthesis of the diterpene 8alpha-acetoxy-13alpha-hydroxy-5-oxo-13-epi- neoverrucosane in the arctic liverwort Fossombronia alaskana was studied by incorporation experiments using [1-(13)C]- and [U-(13)C(6)]glucose as precursors. The (13)C-labeling patterns of acetyl-CoA, pyruvate, and phosphoenolpyruvate in intermediary metabolism were reconstructed from the (13)C NMR data of biosynthetic amino acids (leucine, alanine, phenylalanine) and were used to predict hypothetical labeling patterns for isopentenyl pyrophosphate formed via the mevalonate pathway and the deoxyxylulose pathway. The labeling patterns observed for the neoverrucosane diterpene were consistent with the intermediate formation of geranyllinaloyl pyrophosphate assembled from dimethylallyl pyrophosphate and three molecules of isopentenyl pyrophosphate generated predominantly or entirely via 1-deoxyxylulose 5-phosphate. The experimental data can be integrated into a detailed biosynthetic scheme involving a 1,5-hydride shift. The postulated involvement of the 1,5-hydride shift was confirmed by an incorporation experiment with [6,6-(2)H(2)]glucose.

Biosynthesis of the diterpene verrucosan-2beta-ol in the phototrophic eubacterium Chloroflexus aurantiacus. A retrobiosynthetic NMR study.

The biosynthesis of verrucosan-2beta-ol in the green phototrophic eubacterium Chloroflexus aurantiacus was investigated by in vivo incorporation of singly or doubly 13C-labeled acetate. The 13C labeling of the isolated diterpene was analyzed by one- and two-dimensional NMR spectroscopy. The 13C-labeling patterns of verrucosan-2beta-ol were compared with the labeling patterns of intermediary metabolites (acetyl-CoA, pyruvate, and glyceraldehyde 3-phosphate) which were deduced from amino acids and nucleosides by retrobiosynthetic analysis. The results show that verrucosan-2beta-ol is synthesized via mevalonate and not via the deoxyxylulose pathway, which was discovered recently in some eubacteria, algae, and plants. A scheme for the formation of the unusual tetracyclic ring system is offered. The cyclization process is initiated by the solvolysis of pyrophosphate from geranyllinaloyl pyrophosphate and the mechanism involves a Wagner-Meerwein rearrangement, a 1,5-hydride shift, and a cyclopropylcarbinyl to cyclopropylcarbinyl rearrangement.

Rearrangement reactions in the biosynthesis of molybdopterin--an NMR study with multiply 13C/15N labelled precursors.

The genes moaABC of Escherichia coli were ligated into the expression vector pNCO113. The resulting plasmid was transformed into a moeA mutant of E. coli. From cultures of the recombinant strain, a pteridine designated compound Z could be isolated at 5 mg/liter. Compound Z is a product of precursor Z, a biosynthetic precursor of molybdopterin. Cultures of the recombinant E. coli strain were supplied with [U-(13)C6]glucose, [U-(13)C5]ribulose 5-phosphate, or [7-(15)N,8-(13)C]guanine. The culture medium also contained a large excess of unlabeled glucose. Compound Z as well as nucleosides obtained by hydrolysis of RNA were isolated from the bacterial cultures, and their heavy isotope distribution was investigated by one-dimensional and two-dimensional NMR spectroscopy. The labelling patterns of compound Z show that the carbon atoms of a pentose or pentulose are diverted to the ring atoms C6 and C7 and to the side chain atoms C2', C3' and C4' of compound Z. Carbon atom C1' of compound Z is derived from carbon atom C8 of a guanine derivative. The remodeling of the carbon skeleton of the pentose and purine moieties proceed via intramolecular rearrangement reactions.

Sonic and ultrasonic scalers in a clinical comparison. A study in non-instructed patients with gingivitis or slight adult periodontitis.

In the present study, the Cavitron 2002 ultrasonic scaler was compared with the Titan-S air scaler in 20 subjects with gingivitis or slight periodontitis, whereby the majority of the patients suffered from gingivitis. A split mouth experimental design was used. However, patients did not receive any oral hygiene instructions during the study in order to allow the observation of the true effect of instrumentation. The outcome of a one-time treatment was assessed after 4, 14, 28, and 56 days. Gingival crevicular fluid (GCF), papilla bleeding index (PBI), plaque index (Pl-I), probing depth (PD), and relative attachment level (AL) were measured. Both treatments resulted in a statistically significant decrease of clinical signs of inflammation (PBI: p < 0.001). Probing depths decreased (p < 0.001) and a small gain of attachment of 0.11 mm +/- 0.05 mm (p < 0.001) was observed. Following treatment, a statistically significant (p < 0.001) decrease in GCF and Pl-I was observed between baseline and day 4. No statistically significant difference between the instruments' influences on the evaluated clinical parameters could be found. Thus it can be concluded indirectly that the Cavitron 2002 and the Titan-S are both useful instruments for scaling of tooth and root surfaces.

[Clinical evaluation of co-tetroxazine in acute purulent sinusitis and acute tonsillitis in a double-blind comparison].

127 outpatients, 78 with acute purulent sinusitis and 49 with acute tonsillitis, were treated for 7 days with a benzylpyrimidine -sulphonamide combination. In this double-blind and randomized study 59 patients received co- tetroxazine (100 mg tetroxoprim and 250 mg sulphadiazine) b.i.d., whilst the reference substance, co-trimoxazole (160 mg trimethoprim and 800 mg sulphamethoxazole) was given to the remaining 68 patients b.i.d. The test criteria were the therapeutic efficacy and both subjective and objective tolerance. An improvement in clinical symptoms and signs occurred in both conditions under each therapeutic regimen. Clinical therapeutic success was rated very good or good in 96.6% treated with co- tetroxazine and in 97.1% of patients treated with co-trimoxazole. In the former group therapy failed in 1 patient with sinusitis and in 1 with acute tonsillitis . In 98.3% of patients treated with co- tetroxazine the tolerance was very good or good, whilst the respective figure for co-trimoxazole was only 91.2%. 6 patients suffered from side effects ( gastric spasm, gastralgia , nausea, vomiting, diarrhoea) which were so severe in 2 cases that treatment had to be prematurely terminated. The generally good tolerance to both preparations was confirmed by the results of the laboratory investigations.

The structure of fantasied movement in suicidal children and adolescents.

Guided by the hypothesis that imagining actions is a way of rehearsing actions in the service of effective adaptations to reality, various characteristics of imagined motion, assessed by the Rorschach Test, were compared in hospitalized suicidal and nonsuicidal preadolescents and adolescents, and in public school children. A number of differences were found for example, suicidal children imagined less vigorous motion than distinguished suicidal and nonsuicidal children and preadolescent and adolescent suicidal children. Further, the scale devised to assess imagined motion successfully predicted about 75 percent of the suicidal children. Implications for diagnosis and treatment of suicidal children are discussed.

[A factor analysis of cochleovestibular findings. (author's transl)].

Using the variables auditory threshold, speech audiometry, SISI, ABLB, stapedius reflex, Carhart-test, Békésy audiometry and calorization three factors could be extracted: 1. The quality of peripheral speech-coding in combination with central processing; 2. the degree of damage of the inner ear (threshold, distance between hearing-threshold and reflex-threshold, SISI and calorization); 3. a retrocochlear factor. SISI and ABLB are both tests concerning the inner ear, but must be regarded separately as it is to be supposed that these two findings are caused be two different pathologic processes.

Heat-induced reversible hexagonal packing of spindle microtubules.

Epithelial cells cultured from the lung of the Northwest rough-skinned newt (Taricha granulosa granulosa) were subjected to brief (10-15 min) elevated temperature shocks of 33 degrees-36 degrees C during metaphase. Electron microscope studies on these cells reveal that the spindle microtubules (Mts) are differentially stable to heat treatment. The great majority of nonkinetochore Mts are destroyed within the first few minutes of the shock while kinetochore and adjacent Mts rearrange to form hexagonal closely packed structures before disassembling, the latter occurring only after prolonged heat treatment. The significance and theoretical implications of the formation of hexagonal closely packed Mt structures and of the differential stability of spindle Mts to heating are discussed. The data suggest the existence of one or more heat-sensitive structural component(s) which maintain the individual minimum spacing seen between spindle Mts. To our knowledge, this is the first reported instance of the experimental rearrangement of kinetochore Mts into reversible, hexagonal closely packed bundles.
