The tritiated thymidine labeling of the established human normal cell line LHN 13 suggests that the DNA synthesis stimulation by the nickel sulphate is a function of nickel sulphate concentrations and a function of time. The low and high concentrations play the same role and this role is different from that played by medium concentrations. So, a band of stimulating concentrations appears that could be the result of blocked reactions and coupled oscillators.
Lectin binding on the cell surface was measured by the method of Kornfeld [16] using three tritiated lectins: Robinia pseudo acacia, Concanavalin A and Ricinus. It has been shown that the number of binding sites for Robinia and Con A decreases after the establishment of a leukaemic cell line, whereas the affinity constant increases for Robinia. The relationship between lectin binding and cell growth was carried out on 5 cell lines, 3 of leukaemic and 2 of normal origin. In all cases the maximum number of sites was determined at the time of doubling for the cell population; the affinity constant reciprocally decreases at this moment. For Robinia, a difference was found in both cell growth and the number of sites between an Epstein-Barr virus (EBV) negative leukaemic cell line and the other EBV positive cell lines. Lectins offer a quantitative method for measuring membrane structure variations during and after establishment of cell lines.
The effect was studied of purified alpha-2 macroglobulin (a potent and polyvalent protease inhibitor) on normal human lymphocytes stimulated by phytohaemagglutinin, pokeweed mitogen, or by purified protein derivative as an antigen. At physiological concentration alpha 2 M has been found to inhibit mitogenic as well as antigenic stimulation. However at lower dilutions alpha 2 M acts in synergy either with the mitogen or with the antigen. At these dilutions thymidine incorporation is enhanced. alpha 2 M has no toxic or mitogenic effect on normal human lymphocytes. These data might indicate that two proteases may play a role in the proliferation of normal human lymphocytes.
The present results of our treatment of acute lymphoid leukaemia patients are summarized: 7 out of 20 randomized patients given active immunotherapy after chemoradiotherapy are still in complete remission after periods varying from seven to ten years (compared to none in the control group). The actuarial results on 100 patients show remission and survival curves presenting a plateau between three and five years for a certain percentage, suggesting a possible cure. Several parameters studied in 200 patients indicate that the factors affecting this percentage are age, cytological type, volume of the tumour, and the localization of leukaemic cells in certain areas. Experiments with L1210 leukaemia show that immunotherapy enhances the effect of chemotherapy when administered after chemotherapy but decreases it when administered before, which is in favour of the use of the sequence chemotherapy-immunotherapy clinically.
