Flunixin pharmacokinetics and serum thromboxane inhibition in the dog.

Flunixin meglumine administered orally to beagle dogs at doses of 0.55, 1.10 or 1.65 mg/kg bodyweight was rapidly absorbed to produce maximum mean plasma concentrations of 2.40 +/- 0.70, 4.57 +/- 1.12 and 7.42 +/- 2.07 micrograms/ml, respectively. Thereafter, the plasma concentrations of flunixin fell rapidly to values less than 0.10 micrograms/ml from 24 hours after drug administration at all dosage levels. The maximum mean inhibition of serum thromboxane B2 was 91.5 per cent after the lowest dose of flunixin and 98.8 per cent for both the intermediate and high dose rates. At plasma concentrations of flunixin above 2 micrograms/ml there was more than 90 per cent inhibition of thromboxane.

The structure of the Escherichia coli hemB gene.

The Escherichia coli hemB gene, which encodes 5-aminolevulinic acid dehydratase, and was cloned into pTZ18U, a multicopy plasmid, was sequenced. The hemB insert was double-digested with restriction enzymes and recloned back into pTZ18U and pTZ19U to allow for sequencing in two directions. In a second procedure, used to fill in gaps and to confirm the sequence derived from the first procedure, the whole insert was cloned into M13 phages. A nested set of deletions was constructed and recloned into M13. Both the double-digested fragments cloned into plasmids pTZ18U and pTZ19U and the overlapping fragments contained in M13 phages were sequenced using the dideoxy procedure with [35S]dATP. Computer software was used to identify coding regions and the correct reading frame. Two promoter regions, two Shine-Dalgarno sequences and two possible start sites were identified. Extensive homologies with yeast (36%), human liver (40%) and rat liver (40%) amino-acid (aa) sequences were observed, especially in the 16-aa Zn-binding region (75%) and the 4 aa surrounding the essential lysine at the active site (100% for rat and human proteins). Computer analysis of promoter strength and two independent analyses of codon usage indicated that the hemB gene is moderately expressed.

5-Aminolevulinic acid synthesis in Escherichia coli.

A hemA mutant of Escherichia coli containing a multicopy plasmid which complemented the mutation excreted 5-aminolevulinic acid (ALA) into the medium. [1-14C]glutamate was substantially incorporated into ALA by this strain, whereas [2-14C]glycine was not. Periodate degradation of labeled ALA showed that C-5 of ALA was derived from C-1 of glutamate. The synthesis of ALA by two sonicate fractions which had been processed by gel filtration and dialysis, respectively, was dependent on glutamate, ATP, NADPH, tRNA(Glu), and pyridoxal phosphate. tRNA(Glu) stimulated ALA synthesis in a concentration-dependent manner. Pretreatment with RNase reduced this stimulation. The amino acid sequence of the cloned insert, derived from the nucleotide sequence (J.-M. Li, C. S. Russell, and S. D. Cosloy, J. Cell Biol. 107:617a, 1988), showed no homology with any ALA synthase sequenced to date. These results suggest that E. coli synthesizes ALA by the C5 pathway from the intact five-carbon chain of glutamate.

Availability of porphobilinogen controls appearance of porphobilinogen deaminase activity in Escherichia coli K-12.

A hemin-permeable hemB mutant had no 5-aminolevulinate dehydratase (ALA D) and extremely low porphobilinogen deaminase (PBG D) activity. When the structural gene for hemB was introduced into this strain on a single-copy plasmid, both activities were observed. When the mutant was grown on PBG, normal PBG D activity was observed. Moreover, a hemA mutant had little or no PBG D activity unless it was grown on ALA or PBG. Neither hemin nor PBG affected the level of PBG D protein produced from in vitro transcription and translation of a plasmid harboring the hemC gene as an insert. We conclude that, in Escherichia coli, PBG availability controls the activity of PBG D at some posttranscriptional level.

Cloning of the Escherichia coli K-12 hemB gene.

An Escherichia coli heme-requiring, heme-permeable mutant had no detectable 5-aminolevulinate dehydratase or porphobilinogen deaminase activities. The gene which complemented this mutation was cloned to a high-copy-number plasmid, and porphobilinogen deaminase activity was restored to normal levels, but the synthesis of 5-aminolevulinate dehydratase increased 20- to 30-fold. A maxicell procedure confirmed that the gene cloned was hemB.

Evidence for structural changes in dermatan sulfate and hyaluronic acid with aging.

Three dermatan sulfates (DS18, DS28, and DS35) were isolated from women's skin of ages 19 +/- 2.5, 35 +/- 3.5, 47 +/- 1.7, 60 +/- 0.8, and 75 +/- 5 years. They sequentially precipitated with 18, 28, and 35% ethanol. Their sulfate content was: 23.5, 25.3, and 29.0% (w/w) for DS18 at ages 19-35, 47, and 60 years, respectively; 29.0, 24.0, and 18.8% for DS28; and 18.0, 20.0, and 20.6% for DS35 at ages 19-47, 60, and 75 years, respectively. Both DS18 and DS28 decreased, respectively, from 0.030% (of wet-skin weight) to traces at age 75, and from 0.020 to 0.010% at 60 years. At age 75, DS28 apparently increased by 30%. The DS35 values (traces-0.006%) had no age-related trend. Hyaluronic acid (HA) precipitated with 45% ethanol, was 0.030% of skin-weight at ages 19-47, and decreased to 0.015 and 0.007% at 60 and 75 years, respectively. Its electrophoretic mobility was slower at age 47. In the oldest group, i.r. spectra of HA and DS35 displayed no bands at 1650-1600, 1380, and 1320 cm-1, and a new band at 1560 cm-1. Moreover, ninhydrin-positive material of HA and DS35 increased by 75 and 95%, respectively, and the reducing GlcNAc content of HA decreased. These data showed three chemically different dermatan sulfates (two of which were preponderant) and N-deacetylation of HA and DS35 of the oldest group. After age 47, total DS and HA considerably decreased, DS18 and DS35 were oversulfated, and DS28 became undersulfated with aging.

Chemical alterations of hyaluronic acid and dermatan sulfate detected in aging human skin by infrared spectroscopy.

Age-mediated deacetylation of hyaluronic acid and dermatan sulfate, and shift of sulfate ester configuration were indicated by infrared spectroscopy. Hyaluronic acid and the three dermatan sulfates (DS18, DS28 and DS35), sequentially precipitated from adult skin with 18%, 28% and 35% ethanol, were analyzed at varying ages. At age 75 years, loss of infrared bands in the 1,650-1,600 cm-1 region, at 1,380 cm-1 and 1,320 cm-1 and appearance of a band at 1,560 cm-1 were characteristic of hyaluronic acid and DS35; moreover, in DS28 and DS35 the intensities of the bands at 840 cm-1 and 860 cm-1 were, respectively, decreased and increased. A low intensity band in the 805-785 cm-1 region was observed in the spectra of DS18 (19-35 years), DS28 (70-80 years) and DS35 (all ages). It intensified in DS28 of the 80-year-olds. In the 75 +/- 5-year-old group, ninhydrin-positive material of hyaluronic acid and DS35 increased, while reducing GlcNAc of hyaluronic acid decreased. The data demonstrated hyaluronic acid and DS35 deacetylation and suggested a decrease of equatorial sulfates with infrared band at 840 cm-1 and an increase of axial sulfates with band at 860 cm-1 in DS28 and DS35 of the 75 +/- 5-year-old set. Equatorial Equatorial sulfates with band in the 805 +/- 785 cm-1 region apparently decreased in DS18 after 35 years and increased in DS28 of the oldest group.

Hemagglutinin activity in Nereis coelomic fluid.

1. Nereis coelomic fluid agglutinates rat, mouse, chicken, guinea pig and rhesus monkey erythrocytes (RBC). 2. Lipid fractions of the particulate matter from coelomic fluid are hemagglutinins exhibiting different activity inhibition profiles with complex polysaccharides. 3. The high mol. wt hemagglutinin from coelomic fluid supernatant is not a protein and is inhibited by bovine submaxillary mucin (BSM), thyroglobulin, transferrin and their asialo derivatives. 4. Coelomic fluid supernatant has a population of low mol. wt protein hemagglutinins inhibited by BSM, fetuin, antiserum to coelomic fluid and some mannan preparations. 5. Hemagglutination by lipids characterized by RBC specificity and specificity for inhibition by carbohydrate is noteworthy and may be significant in studies of cellular interactions and immunity in invertebrates.

Circumplex model of marital and family systems: VI. Theoretical update.

This paper updates the theoretical work on the Circumplex Model and provides revised and new hypotheses. Similarities and contrasts to the Beavers Systems Model are made along with comments regarding Beavers and Voeller's critique. FACES II, a newly revised assessment tool, provides both "perceived" and "ideal" family assessment that is useful empirically and clinically.

Circumplex model of marital and family system: I. Cohesion and adaptability dimensions, family types, and clinical applications.

The conceptual clustering of numerous concepts from family therapy and other social science fields reveals two significant dimensions of family behavior, cohesion and adaptability. These two dimensions are placed into a circumplex model that is used to identify 16 types of marital and family systems. The model proposes that a balanced level of both cohesion and adaptability is the most functional to marital and family development. It postulates the need for a balance on the cohesion dimension between too much closeness (which leads to enmeshed systems) and too little closeness (which leads to disengaged systems). There also needs to be a balance on the adaptability dimension between too much change (which leads to chaotic systems) and too little change (which leads to rigid systems). The model was developed as a tool for clinical diagnosis and for specifying treatment goals with couples and families.

Circumplex model of marital and family systems: III. Empirical evaluation with families.

This study was designed to test the circumplex model of family systems that hypothesizes moderate family cohesion and moderate adaptability to be more functional than either extreme. Thirty-one Catholic family triads with daughters ranging in age from 14 to 17 years participated in a structured family interaction game (SIMFAM) and filled out questionnaires that measured the variables of cohesion and adaptability and the facilitative variables of support and creativity. All families were considered normal but were subdivided into those that had more and less difficulty with this adolescent. Analysis of the data yielded considerable support for the circumplex model. High family functioning was associated with moderate family cohesion and adaptability, and low family functioning had extreme scores on these dimensions. As predicted, high family support and creativity were also related to high family functioning. Implications of these findings for family therapy are discussed.
